Transcriptional changes in mesenteric and subcutaneous adipose tissue from Holstein cows in response to plane of dietary energy

Background： Dairy cows can readily overconsume dietary energy during most of the prepartum period, often leading to higher prepartal concentrations of insulin and glucose and excessive body fat deposition. The end result of these physiologic changes is greater adipose tissue lipolysis post-partum coupled with excessive hepatic lipid accumulation and compromised health. Although transcriptional regulation of the adipose response to energy availability is well established in non-ruminants, such regulation in cow adipose tissue depots remains poorly characterized.Results： Effects of ad-libitum access to high [HIGH; 1.62 Mcal/kg of dry matter（DM）] or adequate（CON; 1.35 Mcal/kg of DM） dietary energy for 8 wk on mesenteric（MAT） and subcutaneous（SAT） adipose tissue transcript profiles were assessed in non-pregnant non-lactating Holstein dairy cows using a 13,000-sequence annotated bovine oligonucleotide microarray. Statistical analysis revealed 409 and 310 differentially expressed genes（DEG） due to tissue and diet. Bioinformatics analysis was conducted using the Dynamic Impact Approach（DIA） with the KEGG pathway database. Compared with SAT, MAT had more active biological processes related to adipose tissue accumulation（adiponectin secretion） and signs of pro-inflammatory processes due to adipose tissue expansion and macrophage infiltration（generation of ceramides）. Feeding the HIGH diet led to changes in m RNA expression of genes associated with cell hypertrophy（regucalcin）, activation of adipogenesis（phospholipid phosphatase 1）,insulin signaling activation（neuraminidase 1） and angiogenesis（semaphorin 4 G, plexin B1）. Further, inflammation due to HIGH was underscored by m RNA expression changes associated with oxidative stress response（coenzyme Q3, methyltransferase）, ceramide synthesis（N-acylsphingosine amidohydrolase 1）, and insulin signaling（interferon regulatory factor 1, phosphoinositide-3-kinase regulatory subunit 1, retinoic acid receptor alpha）. A number of feed additives are marketed to assist in boosting the pigs' immune system, regulate gut microbiota, and reduce negative impacts of weaning and other environmental challenges.The most commonly used feed additives include acidifiers, zinc and copper, prebiotics, direct-fed microbials, yeast products, nucleotides, and plant extracts.Inclusion of pharmacological levels of zinc and copper, certain acidifiers, and several plant extracts have been reported to result in improved pig performance or improved immune function of pigs.It is also possible that use of prebiotics, direct-fed microbials, yeast,and nucleotides may have positive impacts on pig performance, but results have been less consistent and there is a need for more research in this area.

Ruminal bacteria lipopolysaccharides:an immunological and microbial outlook

Lipopolysaccharides(LPS)are outer membrane components of Gram-negative bacteria made of three regions:the O-antigen;the core oligosaccharide;and a glucosamine disaccharide linked to hydroxy fatty acids,which is named lipid A.The number phosphate groups,and hydroxy fatty acid chains is associated with the immunopotency and the immunomodulatory activity of LPS,where six-acyl chain lipid A with two phosphate groups is found in virulent strains and five-or four-acyl chain lipid A with one phosphate group are found in non-virulent bacteria strains.Ruminal bacteria are predominantly Gram-negative and their LPS have not been thoroughly investigated.In the rumen,LPS is comprised of mixed ruminal LPS.Drawing upon a body of theoretical and applied work,this paper aims to critically review the scientific literature regarding single-species and mixed ruminal bacteria LPS,highlighting the importance of ruminal LPS to the host.Lastly,future research directions are suggested in order to further our understanding of the roles of LPS in the rumen.Possible suggestions for further understanding ruminal LPS include(1)in silico evaluation of major bacteria contributing to ruminal LPS,(2)structural characterization of LPS from prominent ruminal bacteria species,such as ruminal selenomonads and Megasphaera elsdenii,and,(3)ruminal epithelial tissue immune response evaluation from single-species and mixed ruminal LPS.In conclusion,this review identifies numerous areas for future research,including setting the basis for future modeling and simulation of host microbiome interactions in ruminants.

Blood neutrophil extracellular traps:a novel target for the assessment of mammary health in transition dairy cows

Background:Mammary health is important for transition dairy cows and has been well recognized to exert decisive effects on animal welfare.However,the factors influencing mammary health are still unclear.Differential somatic cell count(DSCC)could reflect the mastitis risk since it is the percentage of neutrophils plus lymphocytes in total somatic cells and could be reflective of mammary health of dairy cows.This work aimed to investigate the assessment and prognosis of the health of transition cows based on blood neutrophil extracellular traps(NETs).Results:Eighty-four transition Holstein dairy cows were selected.The serum was sampled in all the animals at week 1 pre-and postpartum,and milk was sampled at week 1 postpartum.Based on the DSCC in milk at week 1,cows with lower(7.4%±4.07%,n=15)and higher(83.3%±1.21%,n=15)DSCCs were selected.High DSCC cows had higher levels of red blood cell counts(P<0.05),hemoglobin(P=0.07),and hematocrit(P=0.05),higher concentrations of serum oxidative variables[reactive oxygen species(P<0.05),malondialdehyde(P<0.05),protein carbonyl(P<0.05),and 8-hydroxy-2-deoxyguanosine(P=0.07)],higher levels of serum and milk NETs(P<0.05)and blood-milk barrier indicators,including serumβ-casein(P=0.05)and milk immunoglobulin G2(P=0.09),than those of low DSCC cows.In addition,lower concentrations of serum nutrient metabolites(cholesterol and albumin)(P<0.05)and a lower level of serum deoxyribonuclease I(P=0.09)were observed in high DSCC cows than in low DSCC cows.Among the assessments performed using levels of the three prepartum serum parameters(NETs,deoxyribonuclease I andβ-casein),the area under the curve(0.973)of NETs was the highest.In addition,the sensitivity(1.00)and specificity(0.93)were observed for the discrimination of these cows using NETs levels with a critical value of 32.2 ng/mL(P<0.05).Conclusions:The formation of NETs in blood in transition dairy cows may damage the integrity of the blood-milk barrier and thereby increase the risk for mastitis in postpartum cows.

Genome-wide identification and comparative analysis of lipocalin families in Lepidoptera with an emphasis on Bombyx mori

Lipocalins exhibit functional diversity,including roles in retinol transport,invertebrate cryptic coloration,and stress response.However,genome-wide identification and characterization of lipocalin in the insect lineage have not been thoroughly explored.Here,we found that a lineage-specific expansion of the lipocalin genes in Lepidoptera occurred in large part due to tandem duplication events and several lipocalin genes involving insect coloration were expanded more via tandem duplication in butterflies.A comparative analysis of conserved motifs showed both conservation and divergence of lepidopteran lipocalin family protein structures during evolution.We observe dynamic changes in tissue expression preference of paralogs in Bombyx mori,suggesting differential contribution of paralogs to specific organ functions during evolution.Subcellular localization experiments revealed that lipocalins localize to the cytoplasm,nuclear membrane,or nucleus in BmN cells.Moreover,several lipocalin genes exhibited divergent responses to abiotic and biotic stresses,and 1 lipocalin gene was upregulated by 30o fold in B.mori.These results suggest that lipocalins act as signaling components in defense responses by mediating crosstalk between abiotic and biotic stress responses.This study deepens our understanding of the comprehensive characteristics of lipocalins in insects.

This present study aimed to determine the efficacy of supplementing layer diets with vitamin A(0, 8,000 and16,000 IU/kg diet) and vitamin E(0, 250 and 500 mg/kg diet) either individually or in combination on egg production and quality, and blood hematology and chemistry of birds reared under summer conditions. A total of 135 Bovans Brown laying hens were distributed to 9 treatment groups with 5 replicates of 3 hens/pen in a 3 × 3 factorial design. A significant improvement in feed conversion ratio(FCR) was observed as supplementary vitamin A or E increased(P 0.01). Hens fed diets supplemented with 16,000 IU vitamin A plus 500 mg vitamin E/kg diet had the best FCR among all groups. Egg quality traits were not significantly affected by the interaction of vitamin A and vitamin E levels. There was a significant increase in monocytes(P ≤ 0.01)and a decrease in basophils counts(P ≤ 0.05) in response to vitamin E. Significant decreases were observed in packed cell volume(PCV), thyroxine(T_4), alanine transferase(ALT), albumin, total cholesterol and total lipids((P ≤ 0.05 or P ≤ 0.01) P 0.01), and increases were observed in serum concentrations of globulin(P ≤ 0.05)and calcium(P ≤ 0.01) due to vitamin A. The combination of 0 IU vitamin A and 500 mg vitamin E/kg diet had the highest values of PCV(40.09%) and hemoglobin(Hb)(10.33 mg/100 mL) among all groups. Vitamin E raised serum values of total protein, total cholesterol and total lipids(P ≤ 0.05 or P≤ 0.01). Feed intake, FCR,PCV, Hb, lymphocytes, monocytes, eosinophils, T_4, ALT and total protein were significantly affected by the interaction of vitamins A and E(P ≤ 0.05 or P ≤ 0.01). The interaction of vitamins A and E was only significant with respect to serum total protein(P ≤ 0.05). It can be concluded that layer diets supplemented with vitamins A and E had good results in alleviating the harmful impacts of high ambient temperature. Bacteroides-derived isovaleric acid enhances mucosal immunity by facilitating intestinal IgA response in broilers

Background:The interaction between nutrition and immunity plays a vital role in nutrient digestion,absorption,and metabolism during poultry production.Recent studies showed that the gut microbiota contributes to the development of intestinal mucosal immunity.However,the mechanisms by which gut microbes regulate this process remain unclear.Methods:We compared the intestinal mucosal immunity and gut microbiota of Arbor Acre broilers AA(lower mucosal immunity)and Chinese native Wuliang Mountain Black-bone chickens(WLMB)(higher mucosal immunity)using 16S rDNA sequencing,transcriptomic analysis,and immunoglobulin A(IgA)antibody repertoire sequencing.We then combined 16S rDNA sequencing with transcriptomics to identify the key microbes and found that they were positively correlated with IgA production.Next,we transplanted candidate microbes into 1-day-old broiler to explore their role in intestinal mucosal immunity.Finally,we verified the function of candidate microbial metabolites in regulating the immune function of macrophages and the intestinal-epithelial cells(IECs)using in vitro experiments.Results:WLMB performs stronger mucosal immunity than AA,including higher IgA levels,more diverse IgA antibody repertoire,and higher bacterial affinity.Bacteroides was identified as the key microbes related to the intestinal IgA response.Bacteroides transplantation could increase IgA concentration in the duodenal contents by enhancing the expression of IgA,polymeric immunoglobin receptor(PIgR),B cell-activating factor of the TNF family(BAFF),and activation-induced cytidine deaminase(AID)in the duodenum.Additionally,Bacteroides-derived isovaleric acid promoted M2 macrophage polarization of macrophage via mTOR/PPAR-γ/STAT3 signaling pathways and regulated the immunologic function of IECs to produce cytokines,including interleukin(IL)-10,IL-4,BAFF,and transforming growth factor-beta(TGF-β),thus promoting IgA production in B cells by facilitating AID expression.Conclusion:Our study revealed that Bacteroides modulate the intestinal

Recent advances in fermented feeds towards improved broiler chicken performance, gastrointestinal tract microecology and immune responses: A review

Previously, fermentation has been associated with methods that improve the nutritional value of unconventional feed ingredients for broilers. In recent decades, the fermentation process has been employed to produce functional feeds that have the potential to improve broiler gastrointestinal tract microecology, health and production performance. Some of the functional ingredients found in fermented feed include lactic acid bacteria(LAB), lactic acid and other organic acids, and appear to play major roles in determining the beneficial effects of fermented feed on broiler gut health and performance. Unlike the pig, the available literature on broiler fermented feed is still rather limited. This review describes recent advances in the use of fermented feed(on the basis of conventional and unconventional feed ingredients) in broilers. Similarly, this review also shows that additional research is necessary to exploit fermented feed as a viable food source in broiler nutrition.

Husbandry practices and gut health outcomes in weaned piglets:A review

The immediate post-weaning period is one of the most stressful phases in a pig's life, and during this period, piglets are usually exposed to environmental, social and psychological stressors which have direct or indirect effects on gut health and overall growth performance. In this review, the impact of husbandry practices on gut health outcomes and performance of piglets is discussed. Husbandry practices in the swine barn generally include nutrition and management practices, maintenance of hygienic standards and disease prevention protocols, and animal welfare considerations. Poor husbandry practices could result in reduced feed intake, stress and disease conditions, and consequently affect gut health and performance in weaned piglets. Reduced feed intake is a major risk factor for impaired gut structure and function and therefore a key goal is to maximize feed intake in newly weaned piglets. In weaned piglets, crowding stress could reduce pig performance, favor the proliferation of pathogenic bacteria resulting in diarrhea, stimulate immune responses and interfere with beneficial microbial activities in the gut. Sanitation conditions in the swine barn plays an important role for optimal piglet performance, because unclean conditions reduced growth performance, shifted nutrient requirements to support the immune system and negatively affected the gut morphology in weaned piglets. Appropriate biosecurity measures need to be designed to prevent disease entry and spread within a swine operation,which in turn helps to keep all pigs and piglets healthy. Collectively, husbandry practices relating to feeding and nutrition, animal welfare, biosecurity and disease prevention are important determinants of gut health and piglet performance. Thus, it is suggested that adopting high husbandry practices is a critical piece in strategies aimed at raising pigs without the use of in-feed antibiotics.

Techniques for evaluating digestibility of energy, amino acids,phosphorus,and calcium in feed ingredients for pigs

Sound feed formulation is dependent upon precise evaluation of energy and nutrients values in feed ingredients. Hence the methodology to determine the digestibility of energy and nutrients in feedstuffs should be chosen carefully before conducting experiments. The direct and difference procedures are widely used to determine the digestibility of energy and nutrients in feedstuffs. The direct procedure is normally considered when the test feedstuff can be formulated as the sole source of the component of interest in the test diet. However, in some cases where test ingredients can only be formulated to replace a portion of the basal diet to provide the component of interest, the difference procedure can be applied to get equally robust values. Based on components of interest, ileal digesta or feces can be collected, and different sample collection processes can be used. For example, for amino acids(AA), to avoid the interference of fermentation in the hind gut, ileal digesta samples are collected to determine the ileal digestibility and simple T-cannula and index method are commonly used techniques for AA digestibility analysis. For energy, phosphorus, and calcium, normally fecal samples will be collected to determine the total tract digestibility, and therefore the total collection method is recommended to obtain more accurate estimates. Concerns with the use of apparent digestibility values include different estimated values from different inclusion level and non-additivity in mixtures of feed ingredients. These concerns can be overcome by using standardized digestibility, or true digestibility, by correcting endogenous losses of components from apparent digestibility values. In this review, methodologies used to determine energy and nutrients digestibility in pigs are discussed. It is suggested that the methodology should be carefully selected based on the component of interest, feed ingredients, and available experimental facilities.
