Royal-jelly-based apitherapy can attenuate damages to male reproductive parameter following nicotine administration

Background:Nicotine administration can generate severe oxidative stress and lipid peroxidation.Royal jelly,with its antioxidant properties,acts as a scavenger of reactive oxygen species.This study describes the apitherapy effects of royal jelly on testicular damage following nicotine administration.Methods:Forty-eight male BALB/c mice were divided into 8 groups(n=6):saline,3 different doses of royal jelly(100,150,and 200 mg/kg body weight(BW)per day),nicotine(1.5 mg/kg),and 3 different groups of Nic+Roy(1.5 mg/kg of Nic+100,150,and 200 mg/kg BW per day of royal jelly).Nicotine was administrated intraperitoneally,and royal jelly was prescribed orally for 10 consecutive days.Serum levels of hormones(testosterone,luteinizing hormone,and follicle-stimulating hormone),total antioxidant capacity,nitric oxide(NO)status,malondialdehyde levels,sperm DNA fragmentation,sperm parameters,histopathological changes(H&E staining),immunohistochemistry against apoptotic proteins,and gene expression of Bcl-2,p53,Caspase-3,and Nrf2(real-time PCR)were assessed to evaluate the molecular and histological changes.Results:Hormone levels,sperm parameters,and status of antioxidants were decreased significantly(p<.05)following nicotine administration.Moreover,royal jelly treatment normalized hormonal and antioxidant characteristics,decreased apoptotic gene expression,increased Nfr2 gene expression,and restored histopathological alteration to the physiological status significantly(p<.05).Conclusion:Royal jelly upregulates the antioxidant status,inhibits the mitochondrialdependent apoptosis pathway,and increases the rate of proliferation.This therapeutic agent effectively protected the testis against nicotine-associated damages by antioxidant and anti-apoptotic effects.

Alteration of Apoptotic Gene Expression and Reproductive Parameters through Thymus vulgaris Administration Following Paclitaxel-Induced Testopathy in Mice

Objective:Paclitaxel(PAC)is a chemotherapy drug and has an active role in the treatment of lung,breast,and ovarian cancers;however,its use causes increased levels of free radicals,leading to severe and irreversible organ damage.Thymus vulgaris is a species of flowering plant that contains various antioxidant chemical compounds.This study aimed to investigate the effect of T.vulgaris on PAC-induced oxidative damage in mice testis and sperm parameters.Methods:In this study,40 BALB/c mice were divided into five groups:normal control(NC);positive control(20 mg/kg of PAC);and three treatment groups(4.5,9,and 18 mg/kg doses of T.vulgaris extract+20 mg/kg PAC).Treatments were administered intraperitoneally daily for 50 days.Finally,the levels of serum luteinizing hormone(LH),follicle-stimulating hormone(FSH),and testosterone were measured using immunoassay method.Testicular stereological features and sperm parameters were also calculated.Antioxidant parameters were measured using nitrite oxide(NO)and perioxidation levels and ferric-reducing ability of plasma assay.The expressions of p53,caspase-3,Bax,and Bcl-2 were measured through real-time quantitative polymerase chain reaction.Results:T.vulgaris+PAC treatments at all doses significantly increased all parameters in NC and three treatment groups compared with the positive control group(20 mg/kg of PAC)(P<0.05),except the LH,FSH,and NO levels,which were decreased.Further,significantly downregulated levels of p53,caspase-3,and Bax genes and upregulated levels of Bcl-2 gene expression were noted in NC and three treatment groups in comparison with the positive control group(P<0.05).Conclusions:T.vulgaris administration attenuates the toxic effects of PAC on male reproductive parameters.

Single intratesticular injection of blood-serum-derived exosomes can potentially alleviate testopathy following testicular torsion

Background:Testicular torsion(TT)is an acute inflammatory process leading to male infertility.Today,anti-i nflammatory effects of exosomes derived from blood serum are used in various laboratory procedures.In the present study,the anti-i nflammatory effects of blood-serum-derived exosomes in treatment of acute inflammation following TT in mice were evaluated.Materials and Methods:Eighteen male mice were grouped as healthy control,TT,and TT+exosome.TT was induced surgically,and exosomes were extracted from blood serum and administrated by a single intratesticular injection(10 IU).Malondialdehyde(MDA)and Griess assays were used to evaluate the level of oxidative stress.Sperm indices,testosterone(Tes),and apoptotic gene expression(p-53,Bcl2,and Caspase-3)were also assessed.H&E and immunohistochemistry(IHC)stainings were used for histopathological investigations.Data analysis was applied by SPSS(v.19)software.Results:Oxidative stress and apoptotic genes expression were increased significantly(p<0.05)in TT group compared with control.Sperm parameters and Tes were significantly increased,and expression of apoptotic genes was significantly reduced in TT+exosome group(p<0.05).Conclusion:Since the blood-serum-derived exosomes have anti-i nflammatory features,the intratesticular application of blood-serum-derived exosomes can be used clinically in acute phase of orchitis following TT to inhibit testicular inflammation.

Genistein Improves Liver Damage in Male Mice Exposed to Morphine

Background： Morphine is commonly used to treat severe pain. This substance is significantly metabolized in the liver and causes disturbing effects. Genistein is an isoflavone and has antioxidant properties. The aim of this study was to evaluate the effects of genistein against morphine damages on mouse liver. Methods： Between May 2017 and March 2018, 48 male mice were divided into six groups （n = 8 in each group）. Various doses of genistein （25 and 50 mg/kg） and morphine plus genistein （25 and 50 mg/kg） were administered intraperitoneally to 48 male mice for 20 consequent days. Aspartate aminotransferase （AST）, alanine aminotransferase （ALT）, alkaline phosphatase （ALP）, serum nitric oxide （NO） levels, liver weight, and the diameter of hepatocytes and central hepatic vein were studied and compared using one-way analysis of variance. Results： Morphine administration significantly increased the mean diameter of the central hepatic vein （22.76 ± 1.9 μm vs. 15.04 ± 0.60μm, x^2= 21.814, P = 0.001） and hepatocytes （3.03 ± 0.10 μm vs. 1.10 ± 0.05 μm, x^2 = 9.873, P = 0.001） respectively, blood serum NO level （38.00% ± 2.09% vs. 18.72% ±4.40%, x^2 = 20.404, P 〈 0.001 ）, liver enzyme level （AST： 111.80 ± 5.10 ng/ml vs. 81.93 ±2.20 ng/ ml, x^2 = 32.201, P 〈 0.0001; ALT： 45.14 ± 4.10 ng/ml vs. 35.49 ± 2.50 ng/ml, x^2= 18.203, P 〈 0.0001; and ALP： 3.28 ± 0.20 ng/ml vs. 2.14± 0.10, x^2= 5.04, P 〈 0.0001, respectively）, and decreased liver weight （18.50 ± 0.90 g vs. 27.15 ± 0.50 g, x^2 = 22.415, P=0.001 ） compared to saline group （0.535±3.750, P 〈 0.0001）. However, administration of genistein plus morphine significantly enhanced liver weight （25 mg/kg： 21.15 ±2.13 g vs. 18.50 ±0.90 g, x^2= 19.251 P 〈 0.0001 ; 50 mg/kg： 21.20 ±1.00 g vs. 18.5± 0.9 g, x^2= 19.502, P 〈 0.0001, respectively） and reduced the mean diameter of hepatocyte （25 mg/kg： 2.17±0.30 μm vs. 3.03 ± 0.10 μm, x^2 = 22.780, P =0.001 ; 50 mg/kg： 2.01± 0.20 μm vs. 3.03 ±0.10 μm x^2 = 7.120, P = 0.001, respectively）, central hepatic vein （25 mg/kg： 19.53 ± 1.00 μm vs

The dynamics of adult neurogenesis in human hippocampus

The phenomenon of adult neurogenesis is now an accepted occurrence in mammals and also in humans.At least two discrete places house stem cells for generation of neurons in adult brain. These are olfactory system and the hippocampus. In animals, newly generated neurons have been directly or indirectly demonstrated to generate a significant amount of new neurons to have a functional role. However, the data in humans on the extent of this process is still scanty and such as difficult to comprehend its functional role in humans. This paper explores the available data on as extent of adult hippocampal neurogenesis in humans and makes comparison to animal data.

Hair follicle stem cells: In vitro and in vivo neural differentiation

Hair follicle stem cells(HFSCs) normally give rise to keratinocytes, sebocytes, and transient amplifying progenitor cells. Along with the capacity to proliferate rapidly, HFSCs provide the basis for establishing a putative source of stem cells for cell therapy. HFSCs are multipotent stem cells originating from the bulge area. The importance of these cells arises from two important characteristics, distinguishing them from all other adult stem cells. First, they are accessible and proliferate for long periods. Second, they are multipotent, possessing the ability to differentiate into mesodermal and ectodermal cell types. In addition to a developmental capacity in vitro, HFSCs display an ability to form differentiated cells in vivo. During the last two decades, numerous studies have led to the development of an appropriate culture condition for producing various cell lineages from HFSCs. Therefore, these stem cells are considered as a novel source for cell therapy of a broad spectrum of neurodegenerative disorders. This review presents the current status of human, rat, and mouse HFSCs from both the cellular and molecular biology and cell therapy perspectives. The first section of this review highlights the importance of HFSCs and in vitro differentiation, while the final section emphasizes the significance of cell differentiation in vivo.

Faulty homocysteine recycling in diabetic retinopathy

Background:Although hyperglycemia is the main instigator in the development of diabetic retinopathy,elevated circulating levels of a non-protein amino acid,homocysteine,are also associated with an increased risk of retinopathy.Homocysteine is recycled back to methionine by methylenetetrahydrofolate reductase(MTHFR)and/or transsulfurated by cystathionineβ-synthase(CBS)to form cysteine.CBS and other transsulfuration enzyme cystathionine-γ-lyase(CSE),through desulfuration,generates H_(2)S.Methionine cycle also regulates DNA methylation,an epigenetic modification associated with the gene suppression.The aim of this study was to investigate homocysteine and its metabolism in diabetic retinopathy.Methods:Homocysteine and H_(2)S levels were analyzed in the retina,and CBS,CSE and MTHFR in the retinal microvasculature from human donors with established diabetic retinopathy.Mitochondrial damage was evaluated in retinal microvessels by quantifying enzymes responsible for maintaining mitochondrial dynamics(fission-fusionmitophagy).DNA methylation status of CBS and MTHFR promoters was examined using methylated DNA immunoprecipitation technique.The direct effect of homocysteine on mitochondrial damage was confirmed in human retinal endothelial cells(HRECs)incubated with 100μM L-homocysteine.Results:Compared to age-matched nondiabetic control human donors,retina from donors with established diabetic retinopathy had~3-fold higher homocysteine levels and~50%lower H_(2)S levels.The enzymes important for both transsulfuration and remethylation of homocysteine including CBS,CSE and MTHFR,were 40–60%lower in the retinal microvasculature from diabetic retinopathy donors.While the mitochondrial fission protein,dynamin related protein 1,and mitophagy markers optineurin and microtubule-associated protein 1A/1B-light chain 3(LC3),were upregulated,the fusion protein mitofusin 2 was downregulated.In the same retinal microvessel preparations from donors with diabetic retinopathy,DNA at the promoters of CBS and MTHFR were hypermethylated.Incu

Falcaria vulgaris extract attenuates diabetes-induced kidney injury in rats

Objective: To assess the effects of Falcaria vulgaris(F. vulgaris) as an antioxidant on damage to kidney of diabetic rats. Methods: Diabetic rats were established via streptozotocin(60 mg/kg). Various doses of F. vulgaris extracts(50, 100 and 150 mg/kg) and streptozotocin + F. vulgaris extracts were administered via intraperitoneal(i.p) injection to 48 rats(n=8 per group) for 28 d. Subsequently, ferric ion reducing antioxidant power(FRAP) of renal tissue, thiobarbituric acid reactive species, blood glucose concentrations, insulin, nitrite oxide, the weight of animals, glomeruli characteristics and kidney function were evaluated. Results: Compared with the control rats, diabetic rats showed significant increase in malondialdehyde, blood urea nitrogen, creatinine, blood glucose, nitrite oxide contents in renal tissues, and glomerular diameter. Furthermore, tissue FRAP level, body weight, number of glomeruli and plasma insulin were markedly reduced in diabetic rats when compared with the control group(P < 0.05). However, in all F. vulgaris and F. vulgaris + streptozotocin groups, malondialdehyde level, blood urea nitrogen, creatinine, glomerular diameter, nitrite oxide, and glucose levels were decreased significantly; meanwhile, tissue FRAP level, body weight, glomeruli number and insulin serum level were increased, compared to the control diabetic group(P < 0.05). Conclusions: F. vulgaris extract alleviates renal damage in diabetic rats.

Peripheral nerve injury has remained a substantial clinical complication with no satisfactory treatment options.Despite the great development in the field ofmicrosurgery,some severe types of neural injuries cannot be treated without causing tension to the injured nerve.Thus current studies have focused on the new approaches for the treatment of peripheral nerve injuries.Stem cells with the ability to differentiate into a variety of cell types have brought a new perspective to this matter.In this review,we will discuss the use of three main sources of mesenchymal stem cells in the treatment of peripheral nerve injuries.

Human chorionic gonadotropin in vitro: Effects on rat sperm motility and fertilization outcome

Objective:To investigate the effect of human chorionic gonadotropin (hCG) on the motility of rat sperm and the fertilization rate following in vitro fertilization. Methods: hCG concentrations of 25, 50, 75 and 100 ng/mL with incubation time of 1 h and 2 h were used in medium to study sperm motility. Then, 25 and 100 ng/mL of hCG with incubation time of 2 h were applied for in vitrofertilization. After 6 h, the number of two pronuclei was counted. Obtained data was subjected to one way ANOVA and Bonferroni-Dun hoc-post test.Results:Total motility and progressive motility of sperms were increased significantly (P<0.05) in hCG experimental group with concentration of 25 ng/L and incubation time of 2 h compared to control group, however, total motility and progressive motility of sperms were decreased significantly (P<0.05) with increasing dose of hCG. Fertilization rate was decreased significantly (P<0.05) in hCG experimental group with dose of 100 ng/mL compared to 25 ng/mL and control group. Conclusions:The effect of hCG on the rat sperm motility and the rate of their fertility is dose-dependent so that hCG with dose of 25 ng/mL leads to an increase in rat sperm motility and to some extent fertilization rate.
