Aminoglycosides are a widely used class of antibacterials renowned for their effectiveness and broad antimicrobial ***,their use leads to irreversible hearing damage by causing apoptosis of hair cells as their direct *** addition,the hearing damage caused by aminoglycosides involves damage of spiral ganglion neurons upon *** investigate the mechanisms underlying spiral ganglion neuron degeneration induced by aminoglycosides,we used a C57BL/6J mouse model treated with *** found that the mice exhibited auditory deficits following the acute loss of outer hair *** ganglion neurons displayed hallmarks of pyroptosis and exhibited progressive degeneration over *** profiling of these neurons showed significant upregulation of genes associated with inflammation and immune response,particularly those related to the NLRP3 *** of the canonical pyroptotic pathway in spiral ganglion neurons was observed,accompanied by infiltration of macrophages and the release of proinflammatory *** intervention targeting NLRP3 using Mcc950 and genetic intervention using NLRP3 knockout ameliorated spiral ganglion neuron degeneration in the injury *** findings suggest that NLRP3 inflammasome-mediated pyroptosis plays a role in aminoglycoside-induced spiral ganglion neuron *** of this pathway may offer a potential therapeutic strategy for treating sensorineural hearing loss by reducing spiral ganglion neuron degeneration.

Noise-induced hearing loss is the primary non-genetic factor contributing to auditory ***,there are currently no effective pharmacological interventions for patients with noise-induced hearing ***,we present evidence suggesting that the lysine-specific demethylase 1 inhibitor–tranylcypromine is an otoprotective agent that could be used to treat noise-induced hearing loss,and elucidate its underlying regulatory *** established a mouse model of permanent threshold shift hearing loss by exposing the mice to white broadband noise at a sound pressure level of 120 d B for 4 *** found that tranylcypromine treatment led to the upregulation of Sestrin2(SeSN2)and activation of the autophagy markers light chain 3B and lysosome-associated membrane glycoprotein 1 in the cochleae of mice treated with *** noise exposure group treated with tranylcypromine showed significantly lower average auditory brainstem response hearing thresholds at click,4,8,and 16 k Hz frequencies compared with the noise exposure group treated with *** findings indicate that tranylcypromine treatment resulted in increased SeSN2,light chain 3B,and lysosome-associated membrane glycoprotein 1 expression after noise exposure,leading to a reduction in levels of 4-hydroxynonenal and cleaved caspase-3,thereby reducing noise-induced hair cell ***,immunoblot analysis demonstrated that treatment with tranylcypromine upregulated SeSN2 expression via the autophagy *** treatment also reduced the production of NOD-like receptor family pyrin domaincontaining 3(NLRP3)*** conclusion,our results showed that tranylcypromine treatment ameliorated cochlear inflammation by promoting the expression of SeSN2,which induced autophagy,thereby restricting NLRP3-related inflammasome signaling,alleviating cochlear hair cell loss,and protecting hearing *** findings suggest that inhibiting lysine-specific demethylase 1 is a

Trimetallic mesoporous PdCuRu for efficient formate electrosynthesis from polyethylene terephthalate recycling

electrochemical recycling of waste polyethylene terephthalate（PeT） plastics represents a green and sustainable route to produce valuable fuels and address serious environmental issues. Despite some encouraged achievements, their progress is still slower because of the lack of highly active and selective electrocatalysts. Here, we reported an efficient PeT recycling route with trimetallic PdCuRu mesoporous nanospheres（MSs） as a high-performance electrocatalyst that achieved selective electrosynthesis of valuable formate from PeT hydrolysate（PeTH） electrooxidation. Penetrated metal mesopores and trimetallic compositions of PdCuRu MSs synergistically optimized electronic structures of Pd sites and changed chemisorption properties of intermediates, which disclosed a high formate Faradic efficiency of 97% and impressive yield rate of 0.49 mmol h-1cm-2in an alkaline solution. Meanwhile, this electrocatalyst held good electrocatalytic stability, retaining high formate Faradic efficiency and yield rate even being performed for 10 cycles, which highlighted its potential applications for practical PeT recycling. Our study provides a powerful strategy for designing high-performance electrocatalysts toward selective electro-synthesis of valuable fuels from various waste plastics.

Interleukin-6 role in head and neck squamous cell carcinoma progression

Interleukin-6 (IL-6) is a pleiotropic cytokine which plays an important role in a num-ber of cellular processes including proliferation, survival, differentiation, migration and inva-sion. IL-6 mediates its downstream effects by activating a number of signaling cascades including JAK/STAT, PI3K/AKT and MAPK pathways. In addition to its effects on tumor cells, IL-6 also regulates tumor progression and tumor metastasis by modulating tumor angiogenesis and tumor lymphangiogenesis. A number of studies have shown that IL-6 levels are markedly upregulated in cancer patients. We and others have shown that high IL-6 expression indepen-dently predicts tumor recurrence, tumor metastasis and poor survival in head and neck cancer patients. Therefore targeting IL-6 signaling is a potential therapeutic strategy for the treat-ment of head and neck squamous cell carcinoma (HNSCC). In this review, we discuss the cur-rent understanding of the role of IL-6 in HNSCC progression and potential therapeutic strategies to target IL-6 signaling for the treatment of head and neck cancer patients. The impact of round window vs cochleostomy surgical approaches on interscalar excursions in the cochlea:Preliminary results from a flat-panel computed tomography study

Objective:To evaluate incidence of interscalar excursions between round window (RW) and cochleostomy approaches for cochlear implant (CI) insertion. Methods:This was a retrospective case-comparison. Flat-panel CT (FPCT) scans for 8 CI users with Med-el standard length electrode arrays were collected. Surgical technique was identified by a combination of operative notes and FPCT imaging. Four cochleae underwent round win-dow insertion and 4 cochleae underwent cochleostomy approaches anterior and inferior to the round window. Results:In our pilot study, cochleostomy approaches were associated with a higher likeli-hood of interscalar excursion. Within the cochleostomy group, we found 29% of electrode contacts (14 of 48 electrodes) to be outside the scala tympani. On the other hand, 8.5%of the electrode contacts (4 of 47 electrodes) in the round window insertion group were extra-scalar to the scala tympani. These displacements occurred at a mean angle of occur-rence of 364? ? 133?, near the apex of the cochlea. Round window electrode displacements tend to localize at angle of occurrences of 400? or greater. Cochleostomy electrodes occurred at an angle of occurrence of 19?e490?.

Overview of smartphone applications for sleep analysis

Objective:To review and assess the current selection of sleep analysis smartphone applications (apps) available for download. Methods:The iOS and Google Play mobile app store were searched for sleep analysis apps tar-geted for consumer use. Alarm clock, sleep-aid, snoring and sleep-talking recorder, fitness tracker apps, and apps geared towards health professionals were excluded. App information and features were obtained from in-store descriptions, and the app developer website. Results:A total of 51 unique sleep apps in both iOS and Google Play stores were included. The apps were rated 3.8/5 in both stores, and had an average price of$1.12 in the iOS store and$0.58 in the Google Play store.>65%of sleep apps report on sleep structure, including dura-tion, time awake, and time in light/deep sleep, while reporting of ReM was limited. The avail-ability of extra features was variable, ranging from 4%to 73%of apps. Conclusions:There are a variety of sleep analysis apps with a range of functionality. The apps with the most reviews from the each store are featured. Many apps provide data on sleep structure;however the algorithms are not validated by scientific literature or studies. Since patients may inquire about their sleep habits from these apps, it is necessary for physicians to be aware of the most common apps and the features offered and their limitations in order to properly counsel patients. Primary chordoma of the nose

Chordoma as a rare malignant tumor arising from remnants of the notochord, accounts for 4% of malignant bone tumors. It occurs twice as frequently in men compared with women and is uncommon in individuals less than 40 years of age.1-3 Topographic distribution is spheno-occipital (35% to 44%), vertebral (15%), and sacrococcygeal (42% to 50%).4 Unusual locations include the mandible and the maxilla, and lesions in these locations have been called ectopic or dental chordomas. Some chordomas with nasal and paranasal presentations have been reported, but primary chordoma of the nose occurs rarely. To our knowlege only two cases have been reported so far according to the literature.4,5 In this paper, we present primary chordoma of the nose, its characteristics and our experiences with it.

Advancing theranostics with tumor-targeting peptides for precision otolaryngology

Worldwide, about 600,000 head and neck squamous cell carcinoma (HNSCC) are de-tected annually, many of which involve high risk human papilloma virus (HPV). surgery is the primary and desired first treatment option. Following surgery, the existence of cancer cells at the surgical margin is strongly associated with eventual recurrence of cancer and a poor outcome. Despite improved surgical methods (robotics, microsurgery, endoscopic/laparo-scopic, and external imaging), surgeons rely only on their vision and touch to locate tumors during surgery. Diagnostic imaging systems like computed tomography (CT), magnetic reso-nance imaging (MRI), single-photon emission computed tomography (SPeCT) and positron-emission tomography (PeT) are too large, slow and costly to use efficiently during most sur-geries and, ultrasound imaging, while fast and portable, is not cancer specific. This purpose of this article is to review the fundamental technologies that will radically advance Precision Otolaryngology practices to the benefit of patients with HNSCC. In particular, this article will address the potential for tumor-targeting peptides to enable more precise diagnostic imaging while simultaneously advancing new therapeutic paradigms for next generation image-guided surgery, tumor-specific chemotherapeutic delivery and tumor-selective targeted radiotherapy (i.e., theranostic).

Normal values of nasal NO and exhaled NO in young Chinese people aged 9 e 22 years

Objective:To assess the normal levels of nasal nitric oxide (NNO) and fractional exhaled nitric oxide (FeNO) in healthy Chinese young people, and to determine whether the obtained values were associated with age, sex, height, weight, BMI (body mass index) or BSA (body surface area). Methods:One hundred and twenty healthy people were selected from a total of 436 Chinese young people based on their answers to a questionnaire. An electrochemical analyzer (NIOX MINO system) was used to measure NNO and FeNO. The relationship between NNO, FeNO and age, sex, height, weight, BMI, BSA was analyzed using SPSS software. Results:The values of NNO were normal distributed (mean 273.5 ppb;SD 112.3). The values of FeNO were non-normally (Skewed) distributed (median:14.00 ppb;interquartile range:7.00 ppb). The ob-tained NNO values were independent of age, sex, height, weight, BMI and BSA, but were positively correlated to lnFeNO (FeNO log base e);lnFeNO values were also independent of age, height, weight, BMI and BSA, but correlated with NNO and sex. Conclusions:NNO values positively correlate with lnFeNO in healthy people and the levels of each may be predicted by the other. The results of this study are expected to serve as a reference for future studies in China. Copyright a 2016 Chinese Medical Association.
