A Dosimetric Comparison of Radiotherapy Techniques in the Treatment of Carcinoma of Breast

Objectives of present study are a) to compare the planning and delivery aspects of five different techniques, planned by a) forward, inverse planning and electronic tissue compensation methods;and b) to evaluate and verify the accuracy of the planning system using phantom to estimate the skin dose for target and contraletral breast from five techniques. in-vivo skin dosimetry is planned with TL detectors. Five different radiotherapy techniques for treatment of carcinoma breast were studied using archived computed tomography (CT) scans of 25 breast conserving surgery patients (leftsided whole breast), planned for 50 Gy in 25 fractions. Linear accelerator (Clinac 2300 CD) photon beams were used and thermoluminescent detectors (TLD) [LiF:Mg, Ti] estimated dose on humanoid phantom. Dose coverage (95%) (to PTV) and hot spot (105%) covering volumes did not show differences (p > 0.05) in all 5 plans;Electronic compensator plans are better than others. iP-iMRT plan showed the worst Homogeneity index (Hi) (p itor units (MU) (437 ± 84), than other techniques. The mean doses to ipsi-lateral lung, contra-lateral breast (CB) and heart OARs (V20 ***, CB, V30 Heart,) are the least with iP-iMRT. iP-iMRT and E-COMP plans resulted in significantly lower mean dose to the superficial skin (Dmean, V40skin, 45skin, 50skin) (p imated by TLDs were comparable or higher in 3D-CRT (D) and 3D-CRT (P) for PTV and CB;less for iP-iMRT and E-COMP compared to TPS. iP-iMRT and E-COMP techniques provide good target coverage, low doses to OARs, the least doses to the skin of PTV and contra-lateral breast and less hot spots;E-COMP showed better homogeneity, fewer MUs, and the least dose in non-target zones.

OBJECTiVE To determine the transcriptional expression of mitochondrial genome （mtDNA） in MGC803 cell lines subjected by various time-phase hypoxic dispositions, and further to discuss the influence of mtDNA transcripts on hypoxic resistance to irradiation. METHODS The MGC803 cells exposed to anoxic environment were divided into control group （0 h）, hypoxic group （2 h, 8 h, 16 h, 24 h） and irradiated group after exposing the hypoxia. RTPCR was applied to detect the transcripts of cytochrome oxidase subunit Ⅰ （COi）, NADH dehydrogenase subunit 4 （ND4）, ND5, cytochrome b （cyt-b） and ATPase6 （ATP-6） in MGC803 cell lines at various time-phases of hypoxic, and after X-ray irradiation. Flow cytometry and colony formation assay were conducted to evaluate the cell cycle phase and survival fraction. RESULTS COi and ND4 transcripts of MGC803 cell lines were influenced remarkably by hypoxia. COi transcripts were decreased remarkably with the elongation time of exposing the hypoxic, and reduced to one fourth of its original amount of prehypoxia 24 h after exposing the hypoxia. ND4 transcripts were increased initially, and elevated to two folds 8 h after exposing the hypoxia, and then reduced to one second 24 h after exposing the hypoxia. Hypoxia resulted in G1 phase blockage, especially after hypoxia for 16 h. The survival fraction of MGCS03 ceils exposing the hypoxia in irradiated group showed that as the time of exposing the hypoxic before irradiation is prolonged, the survival fraction of MGC803 cells may have an elevated tendency. CONCLUSiON The tumor cells with lower expression levels of the COi and the ND4 after exposing the hypoxic have stronger resistance to the radiation, which indicates that increasing the expression levels of the COi and the ND4 might be advantageous to enhance the sensitivity of hypoxic tumor cells to the radiotherapy.

AiM. To explore the role of the matrix metalloproteinase-9 （MMP-9） polymorphism in colorectal cancer （CRC） in a northeast Chinese population.METHODS： Genotyping of MNP-9-1562C〉T and 279R〉Q polymorphisms was carried out on blood samples from 137 colorectal cancer patients and 199 controls using polymerase chain reaction-restriction fragment length polymorphism （PCR-RFLP）. Multivariate logistic regression models were used to calculate adjusted odds ratios （OR） and 95% confidence intervals （95% Ci）.RESULTS： The distribution of iVllVlP-9 -2562C〉T and 279 R〉Q genotype was not significantly associated with the risk of CRC. However, the risk of ilymph node metastasis of CRC was increased in patients with the -1562T allele （OR = 2.601; 95% Ci = 1.160-5.835; P = 0.022）. The frequency of MMP-9 279RR ＋ RQ genotype was higher than the QQ genotype among CRC patients younger than sixty years old （OR = 0.102, 95% Ci = 0.013-0.812; P = 0.012）.CONCLUSiON： Our results indicated that the MMP-9- 1562C〉T polymorphism affects lymph node metastasis of CRC. in addition, the MMP-9 279R allele may lead to a younger age of onset of colorectal cancer.

Currently there are many unanswered questions concerning contouring a target with PET/CT in radiotherapy planning. Who should contour the PET volume-the radiation oncologist or the nuclear medicine physician？ Which factors will contribute to the dual-observer variability between them？ What should be taken as the optimal SUV threshold to demarcate a malignant tumor from the normal tissue？ When the PET volume does not coincide with the local area CT findings, which portion should be contoured as the target？ if a reginal lymph node,draining area or a remote region is shown to be PET positive but CT negative, or PET negative but CT positive, how is the target identified and selected？ Further studies concerning the relationship between PET/CT and the cancerous tissue are needed. The long-term clinical results showing an increased therapeutic ratio will finally verify the applicability of guidelines to contour the target with PET/CT in radiotherapy planning.

OBJECTiVE To evaluate the applicability of combined therapyand the prognostic factors in patients with carcinoma of thecervical stump (CCS).METHODS The clinical records of 60 CCS patients whounderwent combined treatment in our hospital during a periodfrom January 2000 to December 2007, were collected andretrospectively analyzed. The prognostic factors were studiedusing univariate analysis. Analytical evaluation of the independentprognostic factors was performed using COX proportionalhazardsregression *** The 1-, 3- and 5-year survival rates of the 60 patientswere 95%, 78% and 68%, respectively, with a median survivaltime of 32 months. Univariate survival analysis showed thatthese independent prognostic factors included positive pelviclymph nodes (P = 0.001), lymphovascular tumor embolus (P =0.001), and adjuvant chemotherapy (P = 0.011). in the 60 cases,postoperative local recurrence in the pelvic cavity occurred in1 and distant metastasis in 3. Related complications, such asradiocystitis, recto-vaginal fistula and vesico-vaginal fistula werefound in 6 of the total cases (10%). The serum levels of squamousepithelium antigen detected before and after treatment weresignificantly different (P = 0.000). The incidence of CCS is low;however, the disease is difficult to cure due to the high incidenceof complications and to the frequency of distant ***, individualized treatment is needed. Complications fromsubtotal hysterectomy (STH) should be treated and controlledaggressively. Palliative radiotherapy(pRT) is primarily employed for palliation of bone pain in patients with castrate-resistant prostate cancer(CRPC). However, evidence that p RT influences prostate-specific antigen response in patients with CRPC on systemic therapy is lacking. We describe three cases of CRPC progressing after treatment with docetaxel(n=2) and abiraterone(n=1), who responded unusually after p RT for bone pain with the development of a significant biochemical response and restoration of response to systemic therapy. The possibility of p RT influencing metastatic disease in CRPC has not been previously reported, and raises the possibility of radiation-induced modulation of anti-tumor immune response mechanisms that may play a role in the restoration of response to systemic treatment.

Chuankezhi(CKZ),a new Chinese medicine,plays an important role in *** killer(CiK)cells have been commonly used for immunotherapy in recent *** this study,we aimed to investigate the immunoregulatory effect of CKZ on CiK *** blood monocytes were isolated from healthy donors,and CiK cells were generated by culturing monocytes with interferon-gamma(iFN-γ)and interleukin *** concentrations of CKZ were added on day *** incubation for 14days in culture,the antitumor effects of CiK cells were measured by cytotoxicity *** cytometry was used to explore the effect of CKZ on CiK cell immunophenotype,intracellular cytokine production,and *** effect of CKZ on the antitumor activity of CiK cells in nude mice was also *** increased the percentage of CD3+CD56+CiK cells but did not significantly change the percentage of CD4+,CD8+,or CD4+CD25+CiK ***-conditioned CiK cells showed a greater ability to kill tumor cells,as well as a higher frequency of iFN-γand TNF-αproduction,compared with the CiK cells in the control *** also suppressed the apoptosis of CiK cells in ***,CKZ combined with CiK cells had a stronger suppressive effect on tumor growth in vivo than the CiK,CKZ,or normal saline control *** results indicate that CKZ enhances the antitumor activity of CiK cells and is a potential medicine for tumor immunotherapy.

Let q be a prime power. By PL（Fq） the authors mean a projective line over the finite field Fq with the additional point ∞. in this article, the authors parametrize the conjugacy classes of nondegenerate homomorphisms which represent actions of △（3, 3, k） = （u, v： u^3 = v^3 = （uv）^k = 1〉on PL（Fq）, where q ≡ ±1（modk）. Also, for various values of k, they find the conditions for the existence of coset diagrams depicting the permutation actions of △（3, 3, k） on PL（Fq）. The conditions are polynomials with integer coefficients and the diagrams are such that every vertex in them is fixed by （u^-v^-）^k. in this way, they get △（3, 3, k） as permutation groups on PL（Fq）.

AiM:To investigate the silencing effects of pAdshRNA-pleiotrophin(PTN) on PTN in pancreatic cancer cells,and to observe the inhibition of pAd-shRNA-PTN on neurite outgrowth from dorsal root ganglion(DRG) neurons in ***:PAd-shRNA-PTN was used to infect pancreatic cancer BxPC-3 cells;assays were conducted for knockdown of the PTN gene on the 0th,1st,3rd,5th,7th and 9th d after infection using immunocytochemistry,real-time quantitative polymerase chain reaction(PCR),and Western blotting *** morphologic changes of cultured DRG neurons were observed by mono-culture of DRG neurons and co-culture with BXPC-3 cells in ***:The real-time quantitative PCR showed that the inhibition rates of PTN mRNA expression in the BxPC-3 cells were 20%,80%,50% and 25% on the 1st,3rd,5th and 7th d after *** and Western blotting analysis also revealed the same *** contrast to the control,the DRG neurons co-cultured with the infected BxPC-3 cells shrunk;the number and length of neurites were significantly ***:Efficient and specific knockdown of PTN in pancreatic cancer cells and the reduction in PTN expression resulted in the inhibition of neurite outgrowth from DRG neurons.
