Thioredoxins Play a Crucial Role in Dynamic Acclimation of Photosynthesis in Fluctuating Light

Sunlight represents the energy source for photosynthesis and plant growth. When growing in the field, plant photosynthesis has to manage strong fluctuations in light intensities. Regulation based on the thio- redoxin （Trx） system is believed to ensure light-responsive control of photosynthetic reactions in the chlo- roplast. However, direct evidence for a role of this system in regulating dynamic acclimation of photosyn- thesis in fluctuating conditions is largely lacking. in this report we show that the ferredoxin-dependent Trxs ml and m2 as well as the NADPH-dependent NTRC are both indispensable for photosynthetic acclimation in fluctuating light intensities. Arabidopsis mutants with combined deficiency in Trxs ml and m2 show wild- type growth and photosynthesis under constant light condition, while photosynthetic parameters are strongly modified in rapidly alternating high and low light. Two independent trxmlm2 mutants show lower photosynthetic efficiency in high light, but surprisingly significantly higher photosynthetic efficiency in low light. Our data suggest that a main target of Trx ml and m2 is the NADP-malate dehydrogenase involved in export of excess reductive power from the chloroplast. The decreased photosynthetic efficiency in the high-light peaks may thus be explained by a reduced capacity of the trxm lm2 mutants in the rapid light acti-vation of this enzyme. in the ntrc mutant, dynamic responses of non-photochemical quenching of excita- tion energy and plastoquinone reduction state both were strongly attenuated in fluctuating light intensities, leading to a massive decrease in PSii quantum efficiency and a specific decrease in plant growth under these conditions. This is likely due to the decreased ability of the ntrc mutant to control the stromal NADP（H） redox poise. Triosephosphate isomerase （TPi） catalyzes the interconversion of glyceraldehyde-3-phosphate to dihydroxyacetone phosphate. Photosynthetic organisms generally contain two isoforms of TPi located in both cytoplasm and chloroplasts. While the cytoplasmic TPi is involved in the glycolysis, the chloroplastic isoform participates in the Calvin-Benson cycle, a key photosynthetic process responsible for carbon fixation. Compared with its cytoplasmic counterpart, the functional features of chloroplastic TPi have been poorly investigated and its three-dimensional structure has not been solved. Recently, several studies proposed TPi as a potential target of different redox modifications including dithiol/disulfide interchanges, glutathionylation, and nitrosylation. However, neither the effects on protein activity nor the molecular mechanisms underlying these redox modifications have been investigated. Here, we have produced recombinantly and purified TPi from the unicellular green alga Chlamydomonas reinhardtii （Cr）. The biochemical properties of the enzyme were delineated and its crystallographic structure was determined at a resolution of 1.1 A. CrTPi is a homodimer with subunits containing the typical （β/α）8-barrel fold. Although no evidence for TRX regulation was obtained, CrTPi was found to undergo glutathionylation by oxidized glutathione and trans-nitrosylation by nitrosoglutathione, confirming its sensitivity to multiple redox modifications.

Aims Studies that investigate the space-filling heterogeneity of biological struc-tures in plant communities remain *** main objective of this study was to evaluate the relationship between newly developed photo-graphic measures of structural heterogeneity in digital images and plant species composition in the context of a long-term grassland *** We tested a close-range photographic protocol using measures of structural heterogeneity in gray-tone images,namely mean infor-mation gain(MiG)and spatial anisotropy,to assess differences in the compositional(species richness)and functional characteristics(plant height and flowering)of 78 managed grassland *** also implemented a random placement model of community assembly to explore the links between our measures of structural complexity and the geometric pattern of plant *** Findings MiG and spatial anisotropy correlated with the growth and species richness of grassland *** showed that struc-tural heterogeneity in gray-tone digital images is a function of the size distribution and orientation pattern of plant *** easy,fast and non-destructive methodological approach could eventually serve to monitor the diversity and integrity of various ecosystems at different resolutions across space and time.

Over the last decade virtual reality （VR） setups for rodents have been developed and utilized to in-vestigate the neural foundations of behavior. Such VR systems became very popular since theyallow the use of state-of-the-art techniques to measure neural activity in behaving rodents that can-not be easily used with classical behavior setups. Here, we provide an overview of rodent VR tech-nologies and review recent results from related research. We discuss commonalities and differ-ences as well as merits and issues of different approaches. A special focus is given to experimental（behavioral） paradigms in use. Finally we comment on possible use cases that may further exploitthe potential of VR in rodent research and hence inspire future studies.

Role of ethylene and proteolytic N-degron pathway in the regulation of Arabidopsis seed dormancy

Primary dormant seeds of Arabidopsis thaliana did not germinate in darkness at temperature higher than 10–15℃.Ethylene improved the germination of dormant wild-type(Col-0)seeds at 25℃ in darkness but seeds of the mutant affected in the proteolytic N-degronpathway,proteolysis6(prt6),were insensitive to ethylene suggesting that PRT6 was involved in dormancy release by *** substrates of the N-degron pathway,the Ethylene Response Factors from group Vii(HRE1,HRE2,RAP2.2,RAP2.3,and RAP2.12),were identified to be involved in this insensitivity with an increased germination in prt6 rap2.2 rap2.3 rap2.12 rather than in prt6 hre1 hre2,which also indicated that the three RAPs acted downstream of PRT6,while the two HREs acted upstream of *** reduced the expression of the three RAPs in Col-0 seeds but they were maintained or induced by ethylene in prt6 *** promoting effect of ethylene was associated with a down-regulation of dormancy-related genes in gibberellins(GAs)and abscisic acid(ABA)signaling,such as RGA,RGL2,and ABi5,and with a strong decrease in ABA/GA4 ratio in the presence of *** contrast,we show that the insensitivity of prt6 seeds to ethylene was mainly related to GA signaling disturbance.

ROS and nerves play together during the regeneration process： For many years, the role of reactive oxygen species （ROS） in neurobiology has mainly focused on its pathological implications in neurodegenerative diseases. Contrasting with this view, ROS were recently put forward as key positive signals for axon growth and repair, highlighting beneficial functions of ROS signalling in the vertebrate adult brain （Borquez et al., 2016）.

The major human bacterial pathogen Pseudomonas aeruginosa causes multidrug-resistant infections in people with underlying immunodeficiencies or structural lung diseases such as cystic fibrosis(CF).We show that a few environmental isolates,driven by horizontal gene acquisition,have become dominant epidemic clones that have sequentially emerged and spread through global transmission networks over the past 200 years.

Activation of Defense Response Pathways by OGs and Fig22 Elicitors in Arabidopsis Seedlings

We carried out transcriptional profiling analysis in 10-d-old Arabidopsis thaliana seedlings treated with oligogalacturonides （OGs）, oligosaccharides derived from the plant cell wall, or the bacterial flagellin peptide Fig22, general elicitors of the basal defense response in plants. Although detected by different receptors, both OGs and Fig22 trigger a fast and transient response that is both similar and comprehensive, and characterized by activation of early stages of multiple defense signaling pathways, particularly JA-associated processes. However, the response to Fig22 is stronger in both the number of genes differentially expressed and the amplitude of change. The magnitude of induction of individual genes is in both cases dose-dependent, but, even at very high concentrations, OGs do not induce a response that is as comprehensive as that seen with Fig22. While high doses of either microbe-associated molecular pattern （MAMP） elicit a late response that includes activation of senescence processes, SA-dependent secretory pathway genes and PR1 expres- sion are substantially induced only by Fig22. These results suggest a lower threshold for activation of early responses than for sustained or SA-mediated late defenses. Expression patterns of amino-cyclopropane-carboxylate synthase genes also implicate ethylene biosynthesis in regulation of the late innate immune response.

Glycine-extended gastrin activates two independent tyrosine-kinases in upstream of p85/p110 phosphatidylinositol 3-kinase in human colonic tumour cells

AiM： To investigate whether Src, JAK2 and phosphatidylinositol 3-kinase （Pi3K） pathways are involved in the proliferation of human colonic tumour cells induced by glycine-extended gastrin （G-gly）, the precursor of the mature amidated gastrin and to elucidate the molecular interaction between these three kinases in response to this peptide. METHODS： Using the human colonic tumour cell line HCT116 as a model, we first measured the activation of Pi3K, p60-Src and JAK2 in response to G-gly by in vitro kinase assays. Then we investigated the involvement of these kinases in G-gly-induced cell proliferation by MTT test. RESULTS： G-gly stimulation induced p60-Src, JAK2 and Pi3K activation in HCT116. The different pathways were involved in proliferation of human colon cancer cells induced by G-gly. Furthermore, we found that both Src and JAK2 were necessary to Pi3K regulation by this peptide. However, we did not find any cross-talk between the two tyrosine kinases. CONCLUSiON： Our results suggest that the p60-Src/ Pi3K and JAK2/Pi3K pathways act independently to mediate G-gly proliferative effect on human colonic tumour cells.

Prevalence of intestinal parasite infections among patients in local public hospitals of Hail,Northwestern Saudi Arabia

Objective:To evaluate the prevalence of intestinal parasites among patients in Hail,Northwestern Saudi ***:Stool samples were collected from 130 patients(69females and 61 males) in Hail General *** sample was examined by direct wet mount microscopic examination using both normal saline and Lugol's iodine preparation and concentration techniques using salt and formol-ether *** stained smears were performed for intestinal coccidian using modified Ziehl-Neelsen ***:The overall prevalence of intestinal parasitic infection was 45.38%(59 cases).Forty-four(33.84%)were found to be infected with one or more intestinal protozoa.5(3.84%) were infected with helminthes and 10(7.69%) had mixed infection with both helminthes and *** most common intestinal helminth detected was Ancyloxtoma duvdenale(n=5.3.84%),followed by Ascaris *** *** Trichuris trichiura(n=2 for each species,1.5%).For intestinal protozoa,the coccidian Cryptosporidium parvum(n=25,19.23%) was the most common followed by Entamoeba histolytica/dispur(n=21,16.15%),Giurdia lamblia(n=15,11.54%),Entamoeba coli(n=5.3.85%) and Blastocysts hominis(n=3,2.30%).The prevalence of intestinal parasitic infections in females was significantly higher than in males(Pions:This is the first study highlighting that intestinal parasites are still an important public health problem in Northwestern Saudi Arabia Therefore,health education would be the best way to prevent from intestinal parasite infections which are mainly food borne diseases.
