Homeostatic relevance of vitamin D in maintaining male fertility in human: Down-regulation of oxidative stress and up-regulation of anti-oxidative defense and steroidal hormones

Objective:To evaluate correlation between the levels of vitamin D and male infertility as well as to determine the efficacy of vitamin D in improving the male fertility by up-regulating the levels of testosterone and ***: In the present study, 130 male patients (aged 25-70 years) having fertility defects were screened and 145 healthy individuals were taken as control. All human subjects were screened for 4-hydroxynonenal, isoprostane-F2α, 8-hydroxy-2′-deoxyguanosine, vitamin D, luteinizing hormone, follicle stimulating hormone, testosterones, malondialdehyde, superoxide dismutase, catalase, glutathione peroxidase, and nitric ***: The screening analysis revealed that the levels of luteinizing hormone, follicle stimulating hormone, and testosterone were lower in male infertile subjects compared to healthy subjects. Similarly, the levels of vitamin D [(17.17 ± 2.30) ng/mL] and calcium[(6.29 ± 0.31) mg/dL] were significantly lower in infertile groups compared to the normal healthy groups. Moreover, the study revealed that the levels of superoxide dismutase, catalase, and glutathione peroxidase were significantly higher in healthy subjects compared to the infertile ***:Vitamin D exhibits strong relevance to male fertility by maintaining the levels of sex hormones (luteinizing hormone, follicle stimulating hormone, and testosterone), up-regulating the antioxidant defense (superoxide dismutase, catalase, and glutathione peroxidase), and down-regulating the oxidative stress (malondialdehyde, nitric oxide, and inducible nitric oxide synthase species).

Characterization of the Ubiquitin E2 Enzyme Variant Gene Family in Arabidopsis

Ubiquitin E2 enzyme variant （UEV） proteins are similar to ubiquitin-conjugating enzyme （E2） in both sequence and structure, but the lack of a catalytic cysteine residue renders them incapable of forming a thiolester linkage with ubiquitin. While the functional roles of several UEVs have been defined in yeast and animal systems, Arabidopsis COP10, a photomorphogenesis repressor, is the only UEV characterized in plants. Phylogenetic analysis revealed that the eight Arabidopsis UEV genes belong to three subfamilies. The expresslon of those genes is supported by either the presence of ESTs or RT-PCR analysis. We also characterized the other members of the COP10 subfamily, UEV2. Semi-quantitative RT-PCR analysis indicated that the UEV2 transcripts can be detected in most organs of Arabidopsis. Analysis of UEV2：：GUS transgenic lines also showed its ubiquitous expression in nearly all the developmental stages of Arabidopsis. Transient expression analysis indicated that the sGFP-UEV2 fusion protein can localize to both the cytoplasm and nucleus. A T-DNA insertion mutant, uev2-1, which abolished the transcription of UEV2, displays no visible phenotype. Further, the cop10-4 uev2-1 double mutant exhibits the same phenotype as the cop10-4 mutant in darkness. UEV2 is therefore not functionally redundant with COP10.

Prediction of C-glycosylated apigenin （vitexin） biosynthesis in Ficus deltoidea based on plant proteins identified by LC-MS/MS

Background Plant secondary metabolites act as defence molecules to protect plants from biotic and abiotic stresses. In particular, C-glycosylated flavonoids are more stable and reactive than their O-glycosylated counterparts. Therefore, vitexin (apigenin 8-C glucoside) present in Ficus deltoidea is well-known for its antioxidant, anti-inflammatory, and antidiabetic *** Phenol based extraction was used to extract proteins (0.05% yield) with less plant pigments. This can be seen from clear protein bands in gel electrophoresis. In-gel trypsin digestion was subsequently carried out and analysed for the presence of peptides by LC-MS/*** Thirteen intact proteins are identified on a 12% polyacrylamide gel. The mass spectra matching was found to have 229 proteins, and 11.4% of these were involved in secondary metabolism. Proteins closely related to vitexin biosynthesis are listed and their functions are explained mechanistically. Vitexin synthesis is predicted to involve plant polyketide chalcone synthase, isomerization by chalcone isomerase, oxidation by cytochrome P450 to convert flavanone to flavone, and transfer of sugar moiety by C-glycosyltransferase, followed by dehydration to produce *** Phenol based extraction, followed by gel electrophoresis and LC-MS/MS could identify proteome explaining vitexin biosynthesis in F. deltoidea. Many transferases including β-1,3-galactosyltransferase 2 and glycosyl hydrolase family 10 protein were detected in this study. This explains the importance of transferase family proteins in Cglycosylated apigenin biosynthesis in medicinal plant.

Circulating hTERT mRNA as a tumor marker in cholangiocarcinoma patients

AIM： To investigate human telomerase reverse transcriptase （hTERT） mRNA in the serum of cholangiocarcinoma patients. METHODS： The serum of thirty three cholangiocarcinoma patients, forty one benign biliary tract disease patients and ten healthy volunteers were collected and analyzed for the expression of hTERT mRNA by real-time reverse transcriptase-polymerase chain reaction （RT-PCR）. We then examined the correlation between values of serum hTERT mRNA and the pathological staging of cholangiocarcinoma. RESULTS： hTERT mRNA was detected in 28 of 33 （84.85%） of serum obtained from cholangiocarcinoma patients and 9 of 41 （21.9%） of serum obtained from benign biliary tract disease patients, hTERT mRNA was not detected in any serum obtained from healthy volunteers, on the other hand the common tumor marker, CA19-9 was detected in 20 of 33 （60.6%） of serum obtained from cholangiocarcinoma patients and 8 of 41 （19.5%） of serum obtained from benign biliary tract disease patients. However, no correlation was found between the present of serum hTERT mRNA and tumor staging. CONCLUSION： These results indicate that the detection of circulating hTERT mRNA was identified in almost all cholangiocarcinoma patients. It offers a novel tumor marker, which can be used as a complementary study for diagnosis of cholangiocarcinoma .
