Objective:Nutraceutical products are widely used for their claimed therapeutic ***,falsified or adulterated nutraceuticals present a major health threat to *** study investigates the pharmaceutical quality,safety and anti-inflammatory effects of six male enhancement nutraceuticals that claim to be 100%***:Three batches of six male enhancement products were tested to detect the presence and levels of adulterants via high-performance liquid chromatography(HPLC).The pharmaceutical quality of the selected nutraceuticals was tested with near infrared spectroscopy(NIR)and Se De *** cytotoxic effects of these products on HepG2 cells were determined through cell proliferation(XTT)and lactate dehydrogenase(LDH)cytotoxicity ***,the in vitro inflammatory effects of these products were investigated using murine J774 macrophages through cytokine release ***:HPLC analysis detected the presence of sildenafil citrate,a vasodilator,and the active ingredient in Viagra and Revatio,in all batches of the products we *** of sildenafil citrate ranged from 0.45 mg to 51.85 mg among different *** assessment showed inter-and intra-batch heterogeneity in product *** of the XTT and LDH assays showed significant cytotoxic effects of the analyzed *** analysis revealed that the viability of HepG2 treated with tested products varied from 27.57%to 41.43%.Interestingly,the male enhancement products also showed anti-inflammatory ***:Despite their labeling as 100%natural,all products tested in this study contained levels of sildenafil citrate,which was not reported on the *** was a lack of pharmaceutical uniformity among products of the same batch and across different ***,the products we tested had cytotoxic *** study findings highlight the adulteration,poor quality and hazard of these ***,strict regulation of these product

Amperometric Hydrogen Peroxide Biosensor Based on Horseradish Peroxidase Entrapped in Titania Sol-Gel Film on Screen-Printed Electrode

We report the fabrication of disposable and flexible Screen-Printed Electrodes (SPEs). This new type of screen-printed electrochemical platform consists of Ag nanoparticles (AgNPs) and graphite composite. For this purpose, silver nanoparticles were first synthesized by a chemical reduction method. The morphology and structure of the AgNPs were analyzed using a Scanning Electron Microscope (SEM) and UV-Visible spectroscopy. Graphite was chosen as the working electrode material for the fabrication of a thick-film. The fabrication of a screen-printed hydrogen peroxide biosensor consisting of three electrodes on a polyethylene terephthalate (PET) substrate was performed with a spraying approach (working, counter and reference: enzyme electrode, graphite, pseudo reference: Ag/AgCl). This biosensor was fabricated by immobilizing the peroxidase enzyme (HRP) in a Titania sol-gel membrane which was obtained through a vapor deposition method. The biosensor had electrocatalytic activity in the reduction of H2O2 with linear dependence on H2O2 concentration in the range of 10-5 to 10-3 M;the detection limit was 4.5 × 10-6 M.

Phenotypic and Antimicrobial Susceptibility Studies of Proteus mirabilis Isolates from Fresh Water Fishes in FCT, Abuja-Nigeria

Our study was carried out to determine the phenotypic characterization antimicrobial susceptibility of Proteus mirabilis from fish in FCT, Abuja using isolation, selective plating, preliminary observation, complete biochemical method and antimicrobial susceptibility testing. The biochemical tests conducted includes include Citrate Utilization test, Triple Sugar Iron test, Urea test, Methyl Red test, Indole test, and Voges Proskauer test. The isolates were confirmed by MicrobactTMGNB24E identification kit (Oxiod, UK). A total of 400 fish samples were bought in the market from three area council of the FCT. The result of the study showed overall prevalence rate of (13) 3.25% of Proteus mirabilis isolates. Distribution based on Area councils showed that AMAC had higher prevalence rate of 4.81%, while Bwari had 2.99% and Gwagwalada with 2.57% prevalence. All isolates were subjected to antimicrobial susceptibility testing using the modified single disc diffusion method. From the antimicrobial susceptibility testing done it was discovered that Proteus mirabilis are resistant to Amoxyclav (100%), Erythromycin (92.3%), Tetracycline (92.3%) and Ceftriaxone (23.1%). However, the isolates were susceptible to Ofloxacin (100%), Netillin (92.3%), Levofloxacin (92.3%), Ceftazidime (76.9%), Co-trimoxazole (69.2%) and Gentamicin (61.5%). Since Proteus mirabilis sources of zoonotic diseases and can potentially be dangerous to humans and other animals, our research was able to isolate it from fresh water fish sold in the Federal Capital Territory. This makes public health awareness of the risks associated with Proteus mirabilis in Nigeria necessary.

Isolation, purification and characterization of carboxymethyl cellulase (CMCase) from endophytic Fusarium oxysporum producing podophyllotoxin

Endophytic fungus Fusarium oxysporum is a rich source of cellulases. In the present study, the highest activity was reported at 28 ° C, pH 5.6 with 2% Carboxymethyl cellulose (CMC) as carbon source. CMC was purified using Sephadex G and DEAE cellulose chromatography to 15.9 folds and the molecular weight was determined to be 84 kDa by SDS-PAGE analysis and was subsequently characterized. The purified enzyme was stable over the pH range from 4.0 to 8.0 and at temperatures below 50 ° C. The enzyme was highly active on CMC and reduced or no activity on Avicel, cellobiose and it was suggested to be CMCase/endoglucanase. The activity of endoglucanase was enhanced in the presence of MgCl2, CoCl2, FeCl3, CaCl2, FeCl2 and intensive to HgCl2. The purified enzyme showed its optimum activity at pH 5.0 - 6.0 and was quite stable at 50 ° C for 30 min and retained 45% of original activity.

Utilization of Wood Biomass for Organic Soil Based on the Soil Fertility Index (SOFIX)

Possibility of wood biomass for preparing organic soil was examined to construct reproducible and stable organic standard soil. Seven organic soils were constructed from base soils and additive materials based on the recommended values of the soil fertility index (SOFIX) (total carbon ≥ 25,000 mg/kg, total nitrogen ≥ 1500 mg/kg, total phosphorus ≥ 1100, and total potassium of 2500 to 10,000 mg/kg). Base soils were prepared from two types of wood biomass (big- and small-sized wood chips) at 50%, 60%, and 70% (v/v) and other organic materials such as peat moss, black soil, and mountain soil. Additive materials (soybean meal, oil cake, cow manure, and bone meal) were amended into all organic soils at the same amount. Incubation experiment showed that bacterial biomass in all organic soil was greater than 6 × 108 cells/g-soil after addition of 30% of water content for 1 week. In addition, polymerase chain reaction denaturing gradient gel electrophoresis (PCR-DGGE) analysis resulted in a stable bacterial diversity of the organic soil prepared from the small size wood chip at 70%. Chemical properties of all organic soils were within the recommended values of SOFIX. The plant cultivation experiment showed that fresh Brassica rapa var. peruviridis weights in the organic soils with 50%, 60%, and 70% of small-sized wood chip were 5%, 16%, and 27% higher than that of the chemical fertilizer-amended soil. The organic soil with 70% of small wood chip was the best in the seven organic soils in this study.

NMR Study of Damage on Isolated Perfused Rat Heart Exposed to Ischemia and Hypoxia

Myocardial ischemia is the most common and primary cause of myocardium damage. Numerous conventional techniques and methods have been developed for ischemia and reperfusion studies. However, because of damage to the heart sample, most of these techniques can not be used to continuously monitor the full dynamic course of the myocardial metabolic pathway. The nuclear magnetic resonnance (NMR) surface coil technique, which overcomes the limitations of conventional instrumentation, can be used to quantitatively study every stage of the perfused heart (especially after perfusion stoppage) continuously, dynamically, and without damage under normal or designed physiological conditions at the molecular level. In this paper, 31 P NMR was used to study the effects of ischemia and hypoxia on isolated perfused hearts. The results show that complete hypoxia caused more severe functional damage to the myocardial cells than complete ischemia. 

Isolation and Identification of Potential Probiotic Bacteria from Cattle Farm Soil in Dibrugarh District

Several studies have been done to isolate probiotic bacteria from different sources. In this present study, an attempt was made to isolate, screen and identify potential probiotic bacteria from cattle farm soil in Dibrugarh district, Assam, India. At the level of screening, the result showed the isolates designated as DUA4, DUD3 and DUE2 showed a percent survival rate of 75.36, 69.14 and 52.36 respectively at a pH of 2.5. Similarly survival rate of the same isolates in 0.5% bile salt condition was found to be 117.17%, 144.59% and 118.10% for the isolates DUA4, DUD3 and DUE2 respectively. Antimicrobial activity of the isolates towards the indicator organisms tested showed that DUA4 inhibited gram positive organisms while DUD3 showed activity against both gram positive and gram negative bacteria. All the three isolates showed activity against L. monocytogenes. Autoaggregation ability of the isolates DUA4, DUD3 and DUE2 was found to be 44.15%, 54.11% and 9.42% respectively. The adhesion ability of the isolates DUD3, DUA4 and DUE2 to xylene was 61.78%, 45.37% and 14.83% respectively. Antimicrobial susceptibility test of the isolates showed that the isolates are in general sensitive to antibiotics tested. Phenotypic and genotypic analysis based on the 16S rRNA gene sequence of the isolates DUA4, DUD3 and DUE2 resulted in the identification and designation of the isolate DUA4 as Bacillus spp., DUD3 as Enterococcus faecium and DUE2 as Enterobacter sp. In conclusion, the study has indicated the possibility of isolating potential probiotic bacterial strains from cattle farm soil.

Production and Optimization of <i>Pseudomonas fluorescens</i>Biomass and Metabolites for Biocontrol of Strawberry Grey Mould

Pseudomonas species have been widely studied as biological agents (BCAs) and it is alternative to the application of chemical fungicides. Our objective was to optimize nutritional and environmental conditions of the isolated Pseudomonas fluorescens fp-5 for biomass and metabolites production and to evaluate itsagainst the grey mould disease caused by Botrytis cinerea on strawberry plants under field conditions. Pseudomonas fluorescens, showed antagonistic properties, in vitro, against thepathogen Botrytiscinerea. Effect of the separated secondary metabolites on the fungal growth by broth dilution technique and antifungal activity by agar well diffusion technique was studied. Response surface methodology was used to investigate the effects of four fermentation parameters (pH, incubation time, carbon and nitrogen concentrations) on biomass and bioactivemetabolites [antibiotic phenazin and siderophore] production. Glycerol was found to be the best carbon source for improved biomass and metabolites production. Meanwhile, peptone and yeast extract were found to be the best nitrogen source. Analysis of each formulation revealed that glycerol oil at 0.01% the best oil used for protect P. fluorescens for 3 months Under natural condition, P. fluorescens formulation was effective in reducing B. cinerea disease in strawberry leaves and fruits. Pre-harvest treatment protected fruits from Botrytis post-harvest disease in comparing of fungicide. In addition, the obtained results showed that bacterial treatment significantly increased thegrowth parameters as well as dry weights and yield.
