Antihyperglycemic and hepatoprotective properties of miracle fruit(Synsepalum dulcificum) compared to aspartame in alloxan-induced diabetic mice

Objective: This study was undertaken to investigate the antihyperglycemic potential of miracle fruit(MF)as well as its hepatic safety as compared to aspartame in alloxan-induced diabetic ***: MF extracts were prepared and screened for their phytochemical composition using highperformance liquid chromatography(HPLC). Total phenolic, flavonoid and tannin contents and antioxidant potential were also determined. Additionally, MF was evaluated for its sensory attributes. For in vivo work, MF ethanol extract at high(MFH: 500 mg/kg body weight [BW]) and low(MFL: 250 mg/kg BW) doses as well as aspartame were injected intraperitoneally into alloxan-induced diabetic *** glucose levels were determined following acute and subchronic treatment. At the end of the study,animals were sacrificed, serum was collected for biochemical analysis and liver tissues were obtained for histopathological ***: MF ethanol extract contained more flavonoids and tannins, and had higher 1,1-diphenyl-1-picrylhydrazyl radical-scavenging activity(79.61%) compared to MF aqueous extract(P < 0.05). HPLC analysis of MF ethanol extract also revealed the presence of 10 antioxidants with quercetin comprising the major polyphenol. Additionally, sensory analysis of MF showed that its intake is effective in masking undesirable sourness. Subchronic administration of MFH proved amelioration of hyperglycemia in mice as compared to aspartame. Moreover, aspartame treatment significantly elevated(P < 0.05) the level of alanine aminotransferase and had destructive effects on the liver histopathology;however, hepatic architecture was restored by low and high doses of ***: MF is an effective antihyperglycemic with hepatoprotective properties that can be used as a healthier alternative sweetening agent in place of aspartame for sour beverages.

Phytochemical and anti-neuropathic investigations of Crocus sativus via alleviating inflammation, oxidative stress and pancreatic beta-cells regeneration

Objective:The aim of this study is to investigate the phytochemical and the long-term anti-neuropathic potentials of Crocus sativus cultivated in the university botanical garden,and explore its most bioactive compounds and their underlying mechanisms of ***:Phytochemical analysis and bio-guided isolation-procedures including RP-HPLC and1 H and13 C NMR utilizing biological models of diabetes,inflammation,and diabetic-neuropathy were *** saffron(S-RCED)and Spanish-saffron stigma(S-SP)alone or in combination with Camellia sinus(CS)were ***:The RP-HPLC analyses showed the presence of picrocrocin,crocin I,crocin II,crocin I’,crocin II’,and safranal(SAF)in both S-SP and S-RCED extracts with *** had been shown that SAF was the most bioactive-compound in Crocus *** S-SP and S-RCED possessed significant(P<0.05)anti-diabetic activities in acute(6 h),subchronic(8 d)and chronic(8 weeks)models.S-RCED had been proven with more hypoglycemic potentials when compared to S-SP and SAF.S-SP,S-RCED,and SAF produced significant anti-inflammatory and anti-nociceptive activities against carrageenan-induced inflammatory,hyperalgesic and tactile diabetic-neuropathy models,respectively.S-SP,S-RCED,and SAF elevated serum catalase,reduced glutathione,and insulin serum levels,ameliorated lipid peroxidation and Hb A1 c levels,and histopathologically regenerated the pancreatic *** with CS showed more significant efficacy than the single ***:The oxidative stress reduction,insulin secretagogue,and pancreatic beta-cells regeneration potentials might be responsible for the mechanism underlying the anti-diabetic,anti-inflammatory and anti-diabetic neuropathy ***,the cultivated Crocus sativus might be clinically useful for protecting against many serious-disorders.

Antioxidant activity of water-soluble polysaccharide extracted from Eucalyptus cultivated in Lebanon

Objective: To extract and identify the chemical composition of the polysaccharide isolated from the Eucalyptus cultivated in Lebanon and to evaluate its antioxidant ***: The water-soluble polysaccharide was isolated from Eucalyptus leaves, and its structure was identified by Fourier transform infrared spectroscopy, proton nuclear magnetic resonance, and carbon-13 nuclear magnetic resonance. The antioxidant activity of the active ingredient was screened for its radical scavenging ability using 2,2-diphenyl-1-picryl-hydrazyl radical(DPPH) ***: The results of the DPPH test have shown that fucoidan, the polysaccharide isolated from Eucalyptus, exhibited almost the same antioxidant activity against DPPH$as the ascorbic acid did at 100 mg/***: This natural molecule extracted from a medicinal plant has a promising antioxidant activity and could be used in pharmaceutical and medical applications.

Simultaneous Determination of Amlodipine with H<sub>1</sub>-Receptor Antagonists by Reversed Phase High Performance Liquid Chromatography and Application to Interaction Studies

A rapid, fast and precise method has been developed and validated for the simultaneous determination of amlodipine with H1-receptor antagonists (cetirizine, fexofenadine, and buclizine) from dosage forms. The chromatography was performed on a Purospher? Star, C18 (5 mm, 250 × 4.6 mm) column using acetonitrile: buffer (0.01 mM) (40:60, v/v, pH adjusted to 3.0), as a mobile phase. The mobile phase was pumped at a flow rate of 1.0 mL·min-1 and UV detection was performed at 240 nm. The method was validated for linearity, accuracy, precision and specificity. The method was applied to study the interaction between amlodipine and H1-receptor antagonists. These interactions were carried out in simulated gastric juice (pH 1), simulated full stomach (pH 4), blood pH (pH 7.4) and simulating GI (pH 9). The interacting drugs were heated at 37℃ with intermit-tent shaking and the samples were withdrawn every thirty minutes for three hours and drug contents were analyzed by RP-HPLC techniques. In most cases the in vitro availability of amlodipine was decreased. It was observed that the change in in vitro availability was pH dependent.

Patient Perspectives of the Role of the Community Pharmacist in the Middle East: Jordan, United arab Emirates and Iraq

Objectives: To explore patient’s perspectives of the role of the community pharmacist in view of other health care professionals (specialist physician, general practitioner, nurse and others (e.g. herbalist)) in Jordan, UAE and Iraq. The study also investigated patient’s willingness to pay for specialized pharmaceutical care services. Methods: This study followed a single phase cross sectional survey methodology, conducted in three Middle Eastern countries. Data were collected over a period of 6 months on two consecutive years (March to May 2009 and 2010). A questionnaire was designed and validated, then completed by patients walking into community pharmacies. The source of advice (specialist physician, general practitioner, pharmacist, nurse, or herbalist) regarding patient’s medication use and medical management were investigated. Key findings: Patients visiting community pharmacies in Jordan (n = 1000), UAE (n = 1000) and Iraq (n = 968) were recruited into the study (mean age 35.9 ± 13.1, 50.6% males). Significant difference between the three countries was shown, as more patients chose the pharmacist as their primary source of advice on medication use vs. the specialist physician in Jordan (50.8% vs. 37.3%) and Iraq (41.9% vs. 36.7%) compared to UAE (38.0% vs. 40.1%), P pharmaceutical care services (Jordan 19.5%;UAE 24.7%;Iraq 2.3%). Conclusion: Current situation resulting in the lower socioeconomic status in Jordan and Iraq seems to lead to patients’ higher reliance on the pharmacist, as compared to the UAE. Majority of patients, regardless of the country of origin, are not willing to pay for specialized pharmaceutical care services. These findings are important for future social pharmacy research in the area.
