Vascular endothelial growth factor/connective tissue growth factor and proteomic analysis of aqueous humor after intravitreal conbercept for proliferative diabetes retinopathy

AIM:To investigate the role of connective tissue growth factor(CTGF)and vascular endothelial growth factor(VEGF)in the protein profile of the aqueous humor in patients with proliferative diabetic retinopathy(PDR)following intravitreal injection of ***:This study included 72 PDR patients and 8 cataract patients as *** patients were divided into 3 groups according to the intervals of 3,5,and 7d between intravitreal conbercept(IVC,0.5 mg/0.05 mL)injection and pars plana vitrectomy(PPV)*** humor samples were collected before and after IVC and PPV for VEGF and CTGF levels detected with enzyme-linked immunosorbent assay(ELISA).The differential proteomics of 10 patients who underwent PPV surgery 5d after IVC and 8 normal controls was studied,Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis were performed on the data,and the protein interaction network of 23 differential proteins was ***:Post-IVC,VEGF levels decreased and CTGF levels increased significantly in aqueous humor,with the CTGF/VEGF ratio rising significantly at all *** chromatography tandem mass spectrometry(LC-MS/MS)identified differentially expressed proteins between preand post-IVC *** and KEGG analyses revealed involvement in immune response,stress response,complement and coagulation cascades,ferroptosis,and PPAR signaling *** analysis highlighted key proteins like APOA1,C3,and transferrin(TF).ELISA assay confirmed the differential expression of proteins such as HBA1,SERPINA1,COL1A1,and ACTB,with significant changes in the IVC ***:The study demonstrates that IVC effectively reduces VEGF levels while increasing CTGF levels,thereby modifying the CTGF/VEGF ratio,and IVC significantly alters the protein profile in the aqueous humor of patients with *** analysis reveals that these changes are associated with critical biological pathways and protein interactions involved in immune respons

Hepatocellular carcinoma(HCC)is a common malignant tumor in the Chinese *** to its high degree of malignancy,rapid progression,and poor prognosis,it mainly requires multi-disciplinary treatment(MDT)in the *** December 2019,COVID-19,a novel coronavirus pneumonia,broke out in Wuhan,*** has rapidly spread across the country,with various places launching a level I response to major public health emergencies and traffic being *** patients with HCC were only able to attend primary hospitals,while the MDT model for HCC in provincial hospitals was ***,it was a huge task for clinicians in primary hospitals to ensure MDT was given to patients with HCC during the level I response to major public health *** to formulate a reasonable MDT mode for patients with HCC according to local conditions was worthy of consideration by hepatobiliary surgeons in primary hospitals.

Long noncoding RNAs(lncRNAs) have emerged as critical regulators in a variety of human cancers. However, the function and mechanisms responsible for these molecules in hepatocellular carcinoma(HCC) have not been thoroughly elucidated to date. In this study, we investigated the expression pattern and prognostic values of HOXA11-AS in different HCC cohorts. HOXA11-AS was overexpressed in HCC tissues and was associated with poor prognosis. The potential gene and miRNA targets affected by HOXA11-AS were predicted by employing in silico analysis. The impacts of HOXA11-AS on the malignant characteristics of HCC cells and downstream signalling pathways were assessed using in vitro and in vivo. HOXA11-AS promoted the proliferative activities of HCC cells by upregulating CCND2(encoding cyclin D2) and proliferating cell nuclear antigen(PCNA), ***, HOXA11-AS functioned as a molecular sponge for let-7 a,-7 b and-7 c to affect their target, CCND2. Taken together, our data suggested that HOXA11-AS plays a pivotal role in enhancing HCC oncogenic capacity by functioning as a let-7 s sponge to regulate CCND2 and regulating *** LncRNA HOXA11-AS is significantly up-regulated and is associated with poor outcome in HCC. Overexpression of HOXA11-AS accelerates proliferation of HCC of cells in vitro and in vivo. HOXA11-AS functions as a molecular sponge for let-7 a,-7 b and-7 c. one novel signalling pathway and one proliferation-related molecule regulated by the same lncRNA for HCC progression is proposed.

Diagnosis of chest pain with foregut symptoms in Chinese patients

AIM:To evaluate the diagnosis of chest pain with foregut symptoms in Chinese patients. METHODS:Esophageal manometric studies, 24-h introesophageal pH monitoring and 24-h electrocardiograms(Holter electrocardiography)were performed in 61 patients with chest pain. RESULTS:Thirty-nine patients were diagnosed with non-specific esophageal motility disorders(29 patients with abnormal gastroesophageal reflux and eight patients with myocardial ischemia).Five patients had diffuse spasm of the esophagus plus abnormal gastroesophageal reflux(two patients had concomitant myocardial ischemia),and one patient was diagnosed with nutcracker esophagus. CONCLUSION:The esophageal manometric studies, 24-h intra-esophageal pH monitoring and Holter electrocardiography are significant for the differential diagnosis of chest pain,particularly in patients with foregut *** cases of esophageal motility disorders,pathological gastroesophageal reflux may be a major cause of chest pain with non-specific esophageal motility *** of the esophageal smooth muscle might affect the heart-coronary smooth muscle,leading to myocardial ischemia.

Hepatocellular carcinoma:Surgical perspectives beyond the barcelona clinic liver cancer recommendations

The barcelona clinic liver cancer(BCLC)staging system has been approved as guidance for hepatocellular carcinoma(HCC)treatment guidelines by the main Western clinical liver *** to the BCLC classification,only patients with a small single HCC nodule without signs of portal hypertension or hyperbilirubinemia should undergo liver *** contrast,patients with intermediate-advanced HCC should be scheduled for palliative therapies,even if the lesion is *** studies report good short-term and long-term outcomes in patients with intermediate-advanced HCC treated by liver ***,this classification has been criticised because it excludes many patients who could benefit from curative *** aim of this review was to evaluate the role of surgery beyond the BCLC *** liver resection can be performed in patients with portal hypertension and well-compensated liver function with a 5-year survival rate of 50%.surgery also offers good long-term result in selected patients with multiple or large HCCs with a reported 5-year survival rate of over 50%and 40%,*** macrovascular invasion is associated with a poor prognosis,liver resection provides better long-term results than palliative therapies or best supportive ***,researchers have identified several genes whose altered expression influences the prognosis of patients with *** genes may be useful for classifying the biological behaviour of different tumours.A revision of the BCLC classification should be introduced to provide the best treatment strategy and to ensure the best prognosis in patients with HCC.

Liver resection versus radiofrequency ablation in the treatment of cirrhotic patients with hepatocellular carcinoma

BACKGROUND: Hepatocellular carcinoma is the most common type of primary liver tumor and its incidence is increasing worldwide. The study aimed to compare patients subjected to liver resection or radiofrequency ablation. METHODS: One hundred and forty cirrhotic patients in stage A or B of Child-Pugh with single nodular or multinodular hepatocellular carcinoma were included in this retrospective study. Among them, 87 underwent surgical resection, and 53 underwent percutaneous radiofrequency ablation. Patient charac-teristics, survival, and recurrence-free survival were analyzed. RESULTS: Recurrence-free survival was longer in the resection group in comparison to the radiofrequency group with a median recurrence-free time of 36 versus 26 months, respectively (P=0.01, HR=1.52, 95% CI: 1.05-2.25). In the resection group, median survival was 46 months, with the 1-, 3- and 5-year survival rates of 89.7%, 72.4% and 40.2%. In the radiofrequency group, median survival was 32 months, with the 1-, 3- and 5-year survival rates of 83.0%, 43.4% and 22.6% (Psurgery.

Detection of autoantibodies in the serum of primary hepatocarcinoma patients

Objective: To study the significance of detecting au-toantibodies in primary hepatocarcinoma(PHC) ***: Autoantibodies were detected by indirect im-munofluorescence assay. Antigens and antibodies ofHBV were determined by enzymeimmune assay. Antibodyto HCV IgG was detected by enzyme-linked immunoab-sorbent ***: The positive rate of autoantibody was 27.3%(38/139) in 139 PHC patients. The main type of au-toantibodies in PHC was anti-nuclear antibody (36/38, 94.7%); others included anti-smooth muscle anti-body(2/38, 5.3% ), anti-mitochondria antibody(1/38, 2.6%), anti-midbody antibody (1/38, 2.6%), andanti-liver cell membrane antibody(2/38, 5.3%).Conclusions: Detecting autoantibodies in PHC patientsis of significance in studying the mechanism of au-toimmune reaction and etiology in PHC. The diversityof autoantibodies might result from a wide variety ofetiological factors involved in PHC development, andfrom a wide variety of overexpressed or mutated pro-teins involved in repeated cycles of necrosis and regen-eration in hepatocarcinoma development.

Novel sericin-based hepatocyte serum-free medium and sericin's effect on hepatocyte transcriptome

AIM To develop a novel hepatocyte serum-free medium based on sericin,and to explore the effect of sericin on the hepatocyte *** A controlled trial comparing novel serum-free medium and other media: C3 A cells were cultured in our novel serum-free medium,Hepato ZYME,complete medium(DMEM/F12 with 100 m L/L FBS),and DMEM/F12,andthen cell attachment,proliferation,and function as well as the biocompatibility of the media were assessed.A comparative study of serum-free media with or without 2 mg/m L sericin: the effect of sericin on C3 A growth was assessed by cell viability and proliferation,the effect of sericin on C3 A cell cycle distribution was determined by flow cytometry,and the effect of sericin on the C3 A transcriptome was assessed by gene-chip array and RT-q *** More C3 A cells attached to the plate containing our serum-free medium than to those containing Hepato ZYME and DMEM/F12 at 24 h *** the viability and proliferation rate of C3 A cells in sericin-based serum-free medium were superior to those of cells in Hepato ZYME and DMEM/F12(P < 0.001).The content of albumin and urea in our serum-free medium was significantly higher than that in Hepato ZYME and DMEM/F12 throughout the whole culture period(P < 0.001) and was similar to that in complete medium at day 3,4,and *** part 2,cell viability and proliferation were greater in the presence of 2 mg/m L sericin(P < 0.001),as was the proportion of cells in S phase(16.21% ± 0.98% vs 12.61% ± 0.90%,P < 0.01).Gene-chip array analysis indicated that the expression of CCR6,EGFR,and FOS were up-regulated by 2 mg/m L sericin,and RT-q PCR revealed that the expression of CCR6,EGFR,FOS,AKT1,JNK1,NFk B1,MMP-9,MEK2,ERK1/2 and MYC was upregulated by 2 mg/m L sericin(P < 0.05).CONCLUSION We developed a novel hepatocyte serum-free *** probably enhances cell attachment through the CCR6-Akt-JNK-NF-κB pathway and promotes cell proliferation through CCR6-mediated activation of the E
