Molecular characterization and expression patterns of Nanog gene validating its involvement in the embryonic development and maintenance of spermatogonial stem cells of farmed carp,Labeo rohita

Background: The homeobox containing transcription factor Nanog plays crucial roles in embryonic development/proliferation and/or maintenance of spermatogonial stem cells(SSCs) via interacting with transcription factors such as Oct4 and Sox2 in mammals. However, knowledge of its exact mechanistic pathways remains unexploited. Very little is known about teleost Nanog. Information on the Nanog gene of farmed rohu carp(Labeo rohita) is lacking. We cloned and characterized the Nanog gene of rohu carp to understand the expression pattern in early developmental stages and also deduced the genomic organization including promoter ***: Rohu Nanog(LrNanog) cDNA comprised an open reading frame of 1,161 nucleotides bearing a structural homeodomain; whereas, the genomic structure contained four exons and three introns suggesting that it is homologous to mammalian counterparts. Phylogenetical y, it was closely related to freshwater counterparts. Protein sequence(386 AA of42.65 kDa) comparison revealed its low similarity with other vertebrate counterparts except that of the conserved homeodomain. Tissue distribution analysis revealed the existence of LrNanog transcripts only in adult gonads. The heightened abundances in the ovary and proliferating spermatogonia suggested its participations in maternal inheritance and male germ cell development. The potentiating abundances from fertilized egg onwards peaking at blastula stage vis-à-vis decreasing levels from gastrula stage onwards demonstrated its role in embryonic stem cell development. We also provided evidence of its presence in SSCs by western blotting analysis. AIM: To examine the effect of doxycycline on the activity of matrix metalloproteinases (MMPs) and oxidative stress in gastric tissues of rats following gastric ***: Gastric ulcers were generated in rats by administration of 70% ethanol,and activity of doxycycline was tested by administration 30 min prior to ***,the effect of doxycycline was tested in an indomethacin-induced gastric ulcer *** activities and expression of MMPs were examined by zymography and Western blot ***: Gastric injury in rats as judged by elevated ulcer indices following exposure to ulcerogen,either indomethacin or ethanol,was reversed significantly by ***-induced ulcerated gastric tissues exhibited about 12-fold higher proMMP-9 activity and about 5-fold higher proMMP-3 activity as compared to control ***,ethanol induced about 22-fold and about 6-fold higher proMMP-9 and proMMP-3 activities,respectively,in rat gastric *** proMMP-9 and MMP-3 activities were markedly decreased by doxycycline in ulcerogen treated rat gastric *** contrast,the reduced MMP-2 activity in ulcerated tissues was increased by doxycycline during ulcer *** the other hand,doxycycline inhibited significantly proMMP-9,-2 and -3 activities in *** addition,doxycycline reduced oxidative load in gastric tissues and scavenged H2O2 in *** results suggest a novel regulatory role of doxycycline on MMP-2 activity in addition to inhibitory action on MMP-9 and MMP-3 during prevention of gastric ***: This is the first demonstration of dual action of doxycycline,that is,regulation of MMP activity and reduction of oxidative stress in arresting gastric injury.

The ventral pallidum(VP) is a crucial component of the limbic loop of the basal ganglia and participates in the regulation of reward, motivation, and *** the VP receives afferent inputs from the central histaminergic system, little is known about the effect of histamine on the VP and the underlying receptor mechanism. Here, we showed that histamine, a hypothalamicderived neuromodulator, directly depolarized and excited the GABAergic VP neurons which comprise a major cell type in the VP and are responsible for encoding cues of incentive salience and reward hedonics. Both postsynaptic histamine H1 and H2 receptors were found to be expressed in the GABAergic VP neurons and co-mediate the excitatory effect of histamine. These results suggested that the central histaminergic system may actively participate in VP-mediated motivational and emotional behaviors via direct modulation of the GABAergic VP neurons. Our findings also have implications for the role of histamine and the central histaminergic system in psychiatric disorders.

Unlike the peripheral nervous system (PNS), the central nervous system (CNS) has a low intrinsic regenerative capacity and has mechanisms that actively suppress axon regrowth, for example, glial scarring and myelin inhibition (Fischer, 2012). Even in the PNS, which has the principle ability to regenerate injured axons, functional recovery remains limited, particularly in cases where the nerve target has become unreceptive to re-innervation over time due to an insufficient axonal growth rate (Diekmann and Fischer, 2015). Progress towards robust neuroregenerative therapies depends upon an understanding of the relevant signaling and cytoskeletal proteins that drive and control axon extension. Muscle LIM protein (MLP), also known as cysteine and glycine-rich protein 3, was recently discovered to be one such protein that is expressed in regenerating rat neurons and whose overexpression can promote the axon regeneration of adult central, and peripheral neurons of different species (Levin et al., 2019).

[Objectives] This study was conducted to investigate the scientific prevention and control of Solanum nigrum L. [Methods] Through experiments on S. nigrum from different sources, it was found that glyphosate stress had significant effects on antioxidant enzyme activity and oxidative damage of sensitive S. nigrum plants. [Results] Sensitive S. nigrum showed oxidative damage under glyphosate stress, while resistant S. nigrum responded to adversity damage by improving its antioxidant enzyme activity. The experimental results showed that the antioxidant enzymes and reduced glutathione of S. nigrum had certain metabolic detoxification effects under glyphosate stress. [Conclusions] This study provides a theoretical basis for scientific prevention and control of S. nigrum , and has a certain reference value for revealing the glyphosate resistance mechanism of S. nigrum .

This study was conducted to evaluate the effects of different levels of dietary phytase supplementation in the layer feed on egg production performance,egg shell quality and expression of osteopontin(OPN) and calbindin(CALB1) ***-five White Leghorn layers at 23 weeks of age were randomly divided into 5 groups consisting of a control diet with 0.33% non-phytate phosphorus(NPP) and 4 low phosphorus(P) diets: 2 diets(T1 and T2) with 0.24% NPP t 250 FTU/kg laboratory produced phytase or commercial phytase and another 2 diets(T3 and T4) with 0.16% NPP t 500 FTU/kg laboratory produced phytase or commercial phytase with complete replacement of inorganic *** results indicated that there were no significant differences(P > 0.05) in egg production performance and quality of egg during the first 2 months of ***,in next 2 months,a significant drop in egg production and feed intake was observed in birds fed diets with low P and 500 FTU/kg supplementation of laboratory produced *** gene was up-regulated whereas the CALB1 gene was down regulated in all phytase treatment groups irrespective of the source of *** current data demonstrated that250 FTU/kg supplementation of laboratory produced phytase with 50% less NPP supplementation and500 FTU/kg supplementation of commercial phytase even without NPP in diet can maintain the egg *** up-regulation of OPN and down regulation of CALB1 in egg shell gland in the entire phytase treated group birds irrespective of the source of enzymes is indicative of the changes in P bioavailability at this site.

Aim:To investigate the ultrastructural localization of atrial natriuretic peptide(ANP)-synthesizing cells and the relationship between ANP-synthesizing cellsand microvessels in rat gastric ***:Immunohistochemistry tech-niques and postembedding immunoelectron microscopy techniques were used tovalidate the findings regarding the expression of ANP-synthesizing cells and theultrastructural localization of ANP-synthesizing cells in the gastric ***-tochemistry techniques and the tannic acid-ferric chloride method(TA-Fe stain-ing method)were used to reveal microvessel density and the distribution of ANP-synthesizing cells in different regions of the ***:Cells expressingANP were localized and ANP-synthesizing cells were identified as enterochroma-ffin(EC)cells in the gastric ***-synthesizing cells existed in differentregions of the *** percentage ANP-synthesizing cells in the mucosawas greatest in the fundus(46.7%e5.3%),intermediate in the antrum(40.1%±4.5%),and least in the body(21.6%±3.6%).There was a positive relationshipbetween the percentage of ANP-synthesizing cells and the density of microvesselsin the antral mucosa,but not in the fundus or body ***:ANP issynthesized by EC cells in rat gastric mucosa,and ANP-synthesizing cells aremost dense in the gastric *** may act not only as a regional autocrineand/or paracrine regulator,but also as an endocrine regulatory peptide in thegastrointestinal tract.
