Predicting the Prognosis and Immunotherapeutic Response of Triple-Negative Breast Cancer by Constructing a Prognostic Model Based on CD8+T Cell-Related Immune Genes

Objective Triple-negative breast cancer(TNBC)poses a significant challenge for treatment ***8+T cells,which are pivotal immune cells,can be effectively analyzed for differential gene expression across diverse cell populations owing to rapid advancements in sequencing *** leveraging these genes,our objective was to develop a prognostic model that accurately predicts the prognosis of patients with TNBC and their responsiveness to *** Sample information and clinical data of TNBC were sourced from The Cancer Genome Atlas and METABRIC *** the initial stage,we identified 67 differentially expressed genes associated with immune response in CD8+T ***,we narrowed our focus to three key genes,namely CXCL13,GBP2,and GZMB,which were used to construct a prognostic *** accuracy of the model was assessed using the validation set data and receiver operating characteristic(ROC)***,we employed various methods,including Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway,immune infiltration,and correlation analyses with CD274(PD-L1)to explore the model's predictive efficacy in immunotherapeutic ***,we investigated the potential underlying biological pathways that contribute to divergent treatment *** We successfully developed a model capable of predicting the prognosis of patients with *** areas under the curve(AUC)values for the 1-,3-,and 5-year survival predictions were 0.618,0.652,and 0.826,*** this risk model,we stratified the samples into high-and low-risk *** KEGG enrichment analysis,we observed that the high-risk group predominantly exhibited enrichment in metabolism-related pathways such as drug and chlorophyll metabolism,whereas the low-risk group demonstrated significant enrichment in cytokine ***,immune landscape analysis revealed noteworthy variations between(PD-L1)expression and risk scores,indicating that o

Rational therapeutic strategy for T2 gallbladder carcinoma based on tumor spread

AIM:To evaluate the adequacy of surgical treatment of T2 gallbladder carcinoma(GBCa)according to tumor spread in the subserosal layer. METHODS:A series of 84 patients with GBCa were treated at Saga University Hospital,Japan between April 1989 and October *** tumor stage was graded according to the TNM staging for GBCa from the American Joint Committee on Cancer Manual 6th edition. Tumor staging revealed 30 patients with T2 tumors.T2 GBCa was divided into three groups histologically by the extent of tumor spread in the subserosal layer,using a score of ss minimum(ss min),ss medium(ss med)or ss massive(ss mas). RESULTS:For ss min GBCa,there was no positive pathological factor and patient survival was satisfactory with simple cholecystectomy,with or without extra-he- patic bile duct *** ss med GBCa,some pathological factors,h-inf(hepatic infiltration),ly(lymphatic invasion)and n(lymph node metastasis),were positive. For ss mas GBCa,there was a high incidence of positive pathological *** patient group with extra-hepatic bile duct resection with D2 lymph node dissection (BDR with D2)and those with S4a5 hepatectomy had significantly better survival ***:We suggest that radical surgery is not necessary for ss min GBCa,and partial hepatectomy and BDR are necessary for both ss med and ss mas GBCa.

Proper Accelerations of Time-Like Curves near a Null Geodesic

It is well known that when given a null geodesic γ0(λ) with a point r in (p,q) conjugate to p along γ0(λ), there will be a variation of γ0(λ) which can give a time-like curve from p to q. Here we prove that the time-like curves coming from the above-mentioned variation (with the second derivative β2≠0) have a proper acceleration A = (AaAa)1/2 which approaches infinity as the time-like curve approaches the null geodesic. Because the curve obtained from variation of the null geodesic must be everywhere time-like, we also discuss the constraint of the `acceleration' B0a of the variation vector field on the null geodesic γ0(λ). The acceleration B0a of the variation vector field Za on the null geodesic γ0(λ) cannot be zero.

Joint application of feature extraction based on EMD-AR strategy and multi-class classifier based on LS-SVM in EMG motion classification

This paper presents an effective and efficient combination of feature extraction and multi-class classifier for motion classification by analyzing the surface electromyografic(sEMG) signals. In contrast to the existing methods,considering the non-stationary and nonlinear characteristics of EMG signals,to get the more separable feature set,we introduce the empirical mode decomposition(EMD) to decompose the original EMG signals into several intrinsic mode functions(IMFs) and then compute the coefficients of autoregressive models of each IMF to form the feature set. Based on the least squares support vector machines(LS-SVMs) ,the multi-class classifier is designed and constructed to classify various motions. The results of contrastive experiments showed that the accuracy of motion recognition is improved with the described classification scheme. Furthermore,compared with other classifiers using different features,the excellent performance indicated the potential of the SVM techniques embedding the EMD-AR kernel in motion classification.

Bi-directional hepatic hydrothorax

A 59-year-old male with alcoholic cirrhosis presented to our hospital with an acutely painful umbilical hernia,and 4 mo of exertional *** was noted to be tachypneic and *** had a massive right sided pleural effusion with leftward mediastinal shift and gross ascites,with a tense,fluid-filled,umbilical *** paracentesis with drain placement and a large volume thoracentesis were *** improvement in dyspnea and drainage of 15 L of ascitic fluid,the massive transudative pleural effusion remained largely *** underwent a repeat large volume thoracentesis on hospital day *** patient subsequently developed a tension pneumothorax,which resulted in a dramatic reduction in the effusion.A chest tube was placed and serial radiographs demonstrated resolution of the pneumothorax but recurrence of the *** radiographs illustrate the movement of fluid between the peritoneal and pleural *** this case,the mechanism of pleural effusion was confirmed to be a hepatic hydrothorax via an unintended tension *** to elucidate a hepatic hydrothorax include Tc99m or indocyanine green injection into the ascitic fluid followed by its demonstration above the *** unintended tension pneumothorax in this case additionally demonstrates bi-directional flow across the diaphragm.

Synthesis of Cobalt Containing Block Copolymers Via Ring Opening Metathesis Polymerizastion (ROMP)

In order to realize a metal containing nano-structure, a block-copolymer containing alkynyl groups in one block was designed and synthesized by living ring opening metathesis polymerization (ROMP) technique. The newly developed bis-3-bromopyridine complex of Grubbs catalyst was employed in the series of polymerization and it yielded the desired polymers with great molecular weight control and narrow polydispersities. The characteristics of the block copolymers were investigated by gel permeation chromatography, H- and 13H- NMR spectroscopy. Complexation of the alkynyl group containing block copolymers by treatment with dicobalt octacarbonyl occurred smoothly in 5 min at room temperature. GPC analysis before and after cobalt complexation indicated a significant increase of the hydrodynamic volume. AFM images of the films before and after the complexation also showed a noticeable change in its morphology where grain sizes become smaller and more regular upon complexation.

Under a quite general assumption imposed upon f（t） and h（x）, it is shown that φn（t） is an asymptotically optimal EBE of θ.

Effects of High Erucic Acid Rapeseed Oil on Fatty Acid Oxidation in Rat Liver

The effects of high erucic acid rapeseed oil (HER) on fatty acid oxidation in rat liver compared with low erucic acid rapeseed oil (LER) were studied. Weanling male SD rats were fed diets containing 20% HER or LER for 1 week or 4 weeks, or 5% HER diet for 4 weeks. The hepatic oxidation capacity of butyric acid or palmitic acid was determined by titrating the propanone produced by their oxidation. The results showed that feeding HER to rats led to an increase in the weight of liver and a decrease in the hepatic oxidation capacity of palmitic acid. Hepatic oxidation of butyric acid was not influenced by the intake of HER. The inhibitory action of HER on the oxidation of long-chain fatty acids probably resulted from the incorporation of erucic acid into mitochondrial membranes, interfering the fatty acyl-CoA transfering system on the membranes, but not from the β-oxidation enzyme system in mitochondria being directly inhibited.
