Exhaled ammonia(NH_(3))is an essential noninvasive biomarker for disease diagnosis.In this study,an acetone-modifier positive photoionization ion mobility spectrometry(AM-PIMS)method was developed for accurate qualitative and quantitative analysis of exhaled NH_(3)with high selectivity and sensitivity.Acetone was introduced into the drift tube along with the drift gas as a modifier,and the characteristic NH_(3)product ion peak of(C_(3)H_(6)O)_(4)NH_(4)^(+)(K_(0)=1.45 cm^(2)/V·s)was obtained through the ion-molecule reaction with acetone reactant ions(C_(3)H_(6)O)_(2)H^(+)(K_(0)=1.87 cm^(2)/V·s),which significantly increased the peak-to-peak resolution and improved the accuracy of exhaled NH_(3)qualitative identification.Moreover,the interference of high humidity and the memory effect of NH_(3)molecules were significantly reduced via online dilution and purging sampling,thus realizing breath-by-breath measurement.As a result,a wide quantitative range of 5.87-140.92μmol/L with a response time of 40 ms was achieved,and the exhaled NH_(3)profile could be synchronized with the concentration curve of exhaled CO_(2).Finally,the analytical capacity of AM-PIMS was demonstrated by measuring the exhaled NH_(3)of healthy subjects,demonstrating its great potential for clinical disease diagnosis.

Source apportionment and ecological risk assessment of PAHs in surface sediments from the Liaodong Bay,northern China

The sources and ecological risk from sixteen polycyclic aromatic hydrocarbons in surface sediment in the Liaodong Bay were investigated from 2014 to 2015.The total concentrations of PAHs ranged from 88.5 to 347.1 ng/g,and the high value occurred in the central region of the Liaodong Bay.Cluster analysis identified two site clusters representing the coastal region affected by land-based pollution and the central region of the Liaodong Bay.Principal component analysis-multiple linear regression and diagnostic ratios suggested that PAHs contaminants originated from a mixture of combustion and petroleum sources,and the major was combustion sources.Based on sediment quality guideline,naphthalene,acenaphthylene,acenaphthene,phenanthrene and dibenz[a,h]anthracene may occasionally cause adverse biological effects in some stations.The toxic equivalent concentrations of carcinogenic PAHs indicated low carcinogenic risk for the Liaodong Bay.The ecological risk and toxic pollution levels of PAHs were higher in the central region than in the coastal region along the Liaodong Bay.

Rapid Isolation and Multiplexed Detection of Exosome Tumor Markers Via Queued Beads Combined with Quantum Dots in a Microarray

Tumor-derived exosomes are actively involved in cancer progression and metastasis and have emerged as a promising marker for cancer diagnosis in liquid biopsy.Because of their nanoscale size,complex biogenesis,and methodological limitations related to exosome isolation and detection,advancements in their analysis remain slow.Microfluidic technology offers a better analytic approach compared with conventional methods.Here,we developed a bead-based microarray for exosome isolation and multiplexed tumor marker detection.Using this method,exosomes are isolated by binding to antibodies on the bead surface,and tumor markers on the exosomes are detected through quantum dot(QD)probes.The beads are then uniformly trapped and queued among micropillars in the chip.This design benefits fluorescence observation by dispersing the signals into every single bead,thereby avoiding optical interference and enabling more accurate test results.We analyzed exosomes in the cell culture supernatant of lung cancer and endothelial cell lines,and different lung cancer markers labeled with three QD probes were used to conduct multiplexed detection of exosome surface protein markers.Lung cancer-derived samples showed much higher(~sixfold-tenfold)fluorescence intensity than endothelial cell samples,and different types of lung cancer samples showed distinctive marker expression levels.Additionally,using the chip to detect clinical plasma samples from cancer patients showed good diagnostic power and revealed a well consistency with conventional tests for serological markers.These results provide insight into a promising method for exosome tumor marker detection and early-stage cancer diagnosis.

Objectives:The difficulties in the early detection consequent to the lack of sensitive biomarkers render patients with cholangiocarcinoma(CCA)to have poor outcomes.Recently,sensitive and specific volatile organic compounds(VOCs)were identified in several cancers.However,the VOC profiles in CCA are not well-studied.Thus,we investigated the VOC profiles in exhaled breath of CCA patients and controls.Methods:We prospectively collected exhaled breath samples from 30 consecutive patients newly diagnosed with CCA and 30 controls who did not have CCA(seven had benign biliary strictures and 23 had other medical conditions).Exhaled VOCs were identified using gas chromatography mass spectrometry Triple Quadrupoles system.Analysis of the significant differences in VOCs between cases and controls was conducted using supervised multivariate regression analysis.Further validation was performed for these VOCs in another cohort of 18 CCA patients and 22 controls.Results:Levels of six compounds were significantly different between CCA patients and controls,namely,acetone,isopropyl alcohol,dimethyl sulfide,1,4-pentadiene,allyl methyl sulfide,and N,N-dimethylacetamide.Acetone and dimethyl sulfide were independently associated with CCA as demonstrated in the multivariate analysis.Using the cut-off value of 8.59107 arbitrary unit(AU),acetone had a sensitivity and specificity of 82.1%and 75.8%,respectively,with an area under the receiving operator curve(AUROC)of 0.85 for the CCA diagnosis.Acetone level was also significantly different between cases and controls in the validation cohort.Using the same cut-off value,the sensitivity,specificity,and AUROC was 59.1%,66.7%,and 0.85,respectively.Conclusion:breath analysis may potentially be useful for CCA diagnosis.A cohort of patients with earlystage CCA in further studies is needed to confirm the ability of exhaled VOCs for the early detection of CCA.

Magnetic probe diagnostics of nonlinear standing waves and bulk ohmic electron power absorption in capacitive discharges

It is recognized that standing wave effects appearing in large-area,very-high-frequency capacitively coupled plasma(CCP)reactors cause center-high plasma non-uniformity.Using a high-frequency magnetic probe,we present a direct experimental diagnostic of the nonlinear standing waves and bulk ohmic electron power absorption dynamics in low pressure CCP discharges for different driving frequencies of 13.56,30,and 60 MHz.The design,principle,calibration,and validation of the probe are described in detail.Spatial structures of the harmonics of the magnetic field,determined by the magnetic probe,were used to calculate the distributions of the harmonic current and the corresponding ohmic electron power deposition,providing insights into the behavior of nonlinear harmonics.At a low driving frequency,i.e.13.56 MHz,no remarkable nonlinear standing waves were identified and the bulk ohmic electron power absorption was observed to be negligible.The harmonic magnetic field/current was found to increase dramatically with the driving frequency,due to decreased sheath reactance and more remarkable nonlinear standing waves at a higher driving frequency,leading to the enhancements of the ohmic heating and the plasma density in the bulk,specifically at the electrode center.At a high driving frequency,i.e.60 MHz,the high-order harmonic current density and the corresponding ohmic electron power absorption exhibited a similar node structure,with the main peak on axis,and one or more minor peaks between the electrode center and the edge,contributing to the center-high profile of the plasma density.

To the Editor:Coronary computed tomographic angiography(CCTA)has been considered as one of the most important noninvasive imaging modalities in diagnosing coronary artery disease(CAD).[1]Modern scanner of CCTA can provide precise coronary atherosclerotic plaque information,showing improved diagnostic accuracy and sensitivity for identifying obstructive CAD with a preferable temporal and spatial resolution.Several studies have demonstrated the prognosis value of CCTA for the prediction of future adverse CAD events.
