Unveiling phosphofructokinase as the key target: an innovative strategy combining pull-down assays with optical imaging for snail-killing mechanism exploration

The Pomacea canaliculata, also known as apple snail, is an invasive species in China, posing a serious threat to agricultural production and public health. To prevent its rampant infestation, we developed a highly efficient snail-killing lead compound, 1-(4-chlorophenyl)-3-(pyridin-3-yl)urea (PBQ). However, the killing mechanism of PBQ remains unclear. To elucidate this, two novel compounds, B-PBQ and J-PBQ, were developed through structural modification of PBQ and were used, through both protein pull-down assay and confocal laser scanning microscopy imaging, to identify and confirm that phosphofructokinase(PFK) in the apple snail is the primary target protein of PBQ. Further studies revealed that PBQ successfully inhibits the activity,content, and expression of PFK in apple snail, disrupting the glycolysis pathway and leading to abnormal energy *** disruptions impair the snail’s tolerance to low oxygen, resulting in its efficient eradication. These findings offer a promising direction for developing next-generation PFK-targeted snail-killing pesticides. Furthermore, the strategy of combining pull-down assays with optical imaging used in this work provides a novel approach for the exploration of mechanisms of other pesticide candidates.

THE PREVALENCE OF Sarcocystis suihominis IN THE NATIVES OF YUNNAN PROVINCE, CHINA

Sarcocystis has an obligatory two-host life cycle. An intennediate host acquires infection by ingesting sporocysts or oocysts from the faeces of an infected definitive host. A definitive host becomes infected with Sarcocystis by eating the sarcocyst-infected flesh of the intermediate host. For Sarcocystis suihominis, humans, chimpanzees (Pan troglodytes), rhesus monkeys (Macaca mulatta) and cynomolgus monkeys (Macaca irus) are definitive hosts and the pig is its intermediate host. The infection situation of humans in other regions of the world is unknown. Zuo et al. (1983) first reported two cases with natural S. suihominis infection in Yunnan Province, China1.

Malaria Transmission Potential in the Three Gorges Reservoir of the Yangtze River, China

Objective To define and evaluate the malaria transmission potential in the Yangtze River, following construction of the Three Gorges Reservoir. Methods Six villages, namely, Kaixian, Fengjie, Wanzhou, Fuling, Yubei, and Zigui were selected for investigating the malaria transmission potential in the reservoir. Transmission potential was estimated by mathematical modeling and evaluation of the local malaria situation. Factors that influenced the transmission potential were analyzed using Poisson regression analysis in combination with Grey Systematic Theory （based on evaluation by the Delphi method）. Results Indirect fluorescent antibody data and the incidence of malaria in the local population were consistent with the malaria transmission potential calculated for the area. Multivariate Poisson regression analysis showed a statistically significant association between the riparian zone and the man‐biting rate. Conclusion The risk of a malaria epidemic can be forecasted using the malaria transmission potential parameters investigated here which was closely correlated with the riparian zone.

Identification and investigation of Calodium hepaticum in rodents and insectivores from Wuhan section of the Yangtze River in China

Coexistence of multiple coronaviruses in several bat colonies in an abandoned mineshaft

Since the 2002–2003 severe acute respiratory syndrome(SARS) outbreak prompted a search for the natural reservoir of the SARS coronavirus, numerous alpha- and betacoronaviruses have been discovered in bats around the world. Bats are likely the natural reservoir of alpha- and betacoronaviruses, and due to the rich diversity and global distribution of bats, the number of bat coronaviruses will likely increase. We conducted a surveillance of coronaviruses in bats in an abandoned mineshaft in Mojiang County, Yunnan Province, China, from 2012–2013. Six bat species were frequently detected in the cave: Rhinolophus sinicus, Rhinolophus affinis, Hipposideros pomona, Miniopterus schreibersii, Miniopterus fuliginosus, and Miniopterus fuscus. By sequencing PCR products of the coronavirus RNA-dependent RNA polymerase gene(Rd Rp), we found a high frequency of infection by a diverse group of coronaviruses in different bat species in the mineshaft. Sequenced partial Rd Rp fragments had 80%–99% nucleic acid sequence identity with well-characterized Alphacoronavirus species, including Bt CoV HKU2, Bt CoV HKU8, and Bt CoV1,and unassigned species Bt CoV HKU7 and Bt CoV HKU10. Additionally, the surveillance identified two unclassified betacoronaviruses, one new strain of SARS-like coronavirus, and one potentially new betacoronavirus species. Furthermore, coronavirus co-infection was detected in all six bat species, a phenomenon that fosters recombination and promotes the emergence of novel virus strains. Our findings highlight the importance of bats as natural reservoirs of coronaviruses and the potentially zoonotic source of viral pathogens.

Leaf choice in black-and-white snub-nosed monkeys Rhinopithecus bieti is related to the physical and chemical properties of leaves

To understand the effects of the chemical and physical properties of plant leaves on food choice in Rhinopithecus bieti, we collected mature leaves of nine food and five non-food plant species at the southernmost part of the species＇s range. Chemical properties such as fat, ash, crude protein （CP）, total phenolics （TP）, and fiber content including neutral detergent fiber （NDF）, acid detergent fiber （ADF）, lignin, celluloses and hemicelluloses, and physical toughness were measured. R. bieti tended to choose leaves with lower fiber content, higher ash, a higher ratio of CP/ADF, and lower toughness. No difference was found for fat, crude protein, total phenolics, hemicelluloses and lignin between food and non-food leaves. Even though the ratio of CP/ADF is generally regarded as a good indicator for colobine food choice, the difference in the ratio of CP/ADF between food and non-food leaves was only the result of differences in ADE Since positive correlations were found between ADF and tough- ness from all leaves （both food and non-food species）, and toughness of leaves was likely easier for R. bieti to sense than fiber content via mastication, the toughness of leaves may function as a predictor of food choice in this species [Current Zoology 56 （6） 643-49, 2010].

Hepatitis E virus(HEV)is the prototype of the family Hepeviridae and the causative agent of common acute viral *** diverse HEV-related viruses have been detected in a variety of mammals and some of them may have zoonotic *** this study,we tested 278 specimens collected from seven wild small mammal species in Yunnan province,China,for the presence and prevalence of orthohepevirus by broad-spectrum reverse transcription(RT)-***-related sequences were detected in two rodent species,including Chevrier’s field mouse(Apodemus chevrieri,family Muridae)and Père David’s vole(Eothenomys melanogaster,family Cricetidae),with the infection rates of 29.20%(59/202)and 7.27%(4/55),*** four representative full-length genomes were generated:two each from Chevrier’s field mouse(named Rd HEVAc14 and Rd HEVAc86)and Père David’s vole(Rd HEVEm40 and Rd HEVEm67).Phylogenetic analyses and pairwise distance comparisons of whole genome sequences and amino acid sequences of the gene coding regions showed that orthohepeviruses identified in Chinese Chevrier’s field mouse and Père David’s vole belonged to the species Orthohepevirus C but were highly divergent from the two assigned genotypes:HEV-C1 derived from rat and shrew,and HEV-C2 derived from ferret and possibly *** real-time RT-PCR demonstrated that these newly discovered orthohepeviruses had hepatic *** summary,our work discovered two putative novel genotypes orthohepeviruses preliminarily named HEVC3 and HEV-C4 within the species Orthohepevirus C,which expands our understanding of orthohepevirus infection in the order Rodentia and gives new insights into the origin,evolution,and host range of orthohepevirus.

Comparative Genomic Analysis of Gene Variations of Two Chinese Yersinia pestis Isolates from Vaccine Strain EV76

Objective To investigate genomic variations of two Chinese Yersinia pestis isolates that were isolated from different plague foci obtained from vaccine strain EV76 from the Yunnan province of China. Methods A microarray containing 12 000 probes covering the entire genome of seven Yersinie pestis and two Yersinia pseudotuberculosis strains, was used. PCR assays were performed to confirm microarray results. Results The gene variations detected included the absence of five genes related to the synthesis of betaine in both EV76 and another sequenced attenuated strain, KIM D27. Several genes related to phage-related membrane proteins were found to be absent in the Antiqua biovar Yunnan strain, 485, which was isolated from a rodent plague foci. Conclusion These findings provide initial insight into the distinct strains isolated from natural foci, within their genomic context, including Yunnan Y. pestis strains. This information will be used therefore to establish subsequent comparisons of these sequences with published complete genomes of other strains.

Echinococcus Granulosus: Suitable in vitro Protoscolices Culture Density

The present study is to determine the suitable protoscolices （PSCs） density for long-time culturing in vitro. The PSCs were divided into eight groups with different densities and the viability tests were carried out with 0.1% methylene blue staining. Then the infection ability of cultured PSCs was assessed by the mean cyst weight of mice inoculated intraperitoneally with PSCs after 8 months post-infection.
