Unveiling phosphofructokinase as the key target: an innovative strategy combining pull-down assays with optical imaging for snail-killing mechanism exploration

The Pomacea canaliculata, also known as apple snail, is an invasive species in China, posing a serious threat to agricultural production and public health. To prevent its rampant infestation, we developed a highly efficient snail-killing lead compound, 1-(4-chlorophenyl)-3-(pyridin-3-yl)urea (PBQ). However, the killing mechanism of PBQ remains unclear. To elucidate this, two novel compounds, B-PBQ and J-PBQ, were developed through structural modification of PBQ and were used, through both protein pull-down assay and confocal laser scanning microscopy imaging, to identify and confirm that phosphofructokinase(PFK) in the apple snail is the primary target protein of PBQ. Further studies revealed that PBQ successfully inhibits the activity,content, and expression of PFK in apple snail, disrupting the glycolysis pathway and leading to abnormal energy *** disruptions impair the snail’s tolerance to low oxygen, resulting in its efficient eradication. These findings offer a promising direction for developing next-generation PFK-targeted snail-killing pesticides. Furthermore, the strategy of combining pull-down assays with optical imaging used in this work provides a novel approach for the exploration of mechanisms of other pesticide candidates.

Effects of Binaries on Open Cluster Age Determination in Bayesian Inference

We investigate the effects of binaries on the cluster age determination for 561 open clusters in the Galactic disk via the Bayesian statistical *** properties of these star clusters,including age,metallicity,distance modulus,color excess,binary fraction,and rotating star fraction,are derived from color–magnitude-diagrams(CMDs)via isochrone fitting to high-precision Gaia EDR3 *** the simple stellar population of binary and single-star,age differences can be found with the same star cluster.A Bayesian applied regression modeling software,Stan,is employed to explore how much binaries affect the age determination of open *** results present less statistically significant difference between the binary-star simple populations(bs SSPs)and the singlestar simple populations(ss SSPs)for cluster age *** all clusters in our sample,the ages estimated using the bs SSPs models are younger than those estimated using the ss SSPs,with a mean value of~70 ***,it is found that for 52.5%clusters in our sample,ages are relatively sensitive to the presence of binaries,at least~25%*** suggests that in studies of open cluster age determinations,the effects of binary interactions on the whole sample are not prominent,but its effects on some clusters should still be included as an essential ingredient.

Research on Hepatitis B virus Genotypes and Subgenotypes among Bai Nationality in dali, Yunnan Province

To investigate the distribution of hepatitis B virus (HBV) genotypes and subgenotypes among the Bai nationality in dali, a total of 100 serum samples from patients with chronic HBV-infection were collected for the detection of HBV genotypes and subgenotypes by genotype-specific primers and restriction fragment length polymorphism (RLFP), respectively. Among the 100 samples, the proportions of genotype B, C and mixed genotype (B+C) were 41%, 25% and 34%, respectively. All the genotype B strains belonged to subgenotype Ba. In genotype C, 84% were Subgenotype Cs and 12% were subgenotype Ce. The distribution of genotypes B, C and B+C showed no significant difference between male and female patients (P=0.182) and among the age groups of patients (P=0.812). The rates of HBeAg/HBeAg positivity were no significantly different among genotypes B, genotype C and mixed genotype (B+C) (P=0.077/P=0.663). In dali, genotypes B, B+C and C existed among Bai nationality with chronic HBV-infection, and genotype B was the major genotype. Subgenotypes Ba and Cs were the predominant strains in patients with HBV genotype B/C infection. The most prominent characteristic was the higher prevalent rate of mixed genotype (B+C) in patients.

BSEC Method for Unveiling Open Clusters and its Application to Gaia DR3:83 New Clusters

Open clusters(OCs) are common in the Milky Way, but most of them remain undiscovered. There are numerous techniques, including some machine-learning algorithms, available for the exploration of OCs. However, each method has its limitations and therefore, different approaches to discovering OCs hold significant values. We develop a comprehensive approach method to automatically explore the data space and identify potential OC candidates with relatively reliable membership determination. This approach combines the techniques of Hierarchical Density-Based Spatial Clustering of Applications with Noise, Gaussian mixture model, and a novel cluster member identification technique, color excess constraint. The new method exhibits efficiency in detecting OCs while ensuring precise determination of cluster memberships. Because the main feature of this technique is to add an extra constraint(EC) for the members of cluster candidates using the homogeneity of color excess,compared to typical blind search codes, it is called Blind Search-Extra Constraint(BSEC) method. It is successfully applied to the Gaia Data Release 3, and 83 new OCs are found, whose color–magnitude diagrams(CMDs) are fitted well to the isochrones. In addition, this study reports 621 new OC candidates with discernible main sequence or red giant branch. It is shown that BSEC technique can discard some false negatives of previous works, which takes about three percentage of known clusters. It shows that as an EC, the color excess(or twocolor) constraint is useful for removing fake cluster member stars from the clusters that are identified from the positions and proper motions of stars, and getting more precise CMDs, when differential reddening of member stars of a cluster is not large(e.g., ΔE(G_(BP)-G_(RP)) < 0.5 mag). It makes the CMDs of 15% clusters clearer(in particular for the region near turnoff) and therefore is helpful for CMD and stellar population studies. Our result suggests that the color excess con

Distribution of gamasid mites on small mammals in Yunnan Province, China

An investigation of gamasid mites on the body surface of small mammals was carried out in Yunnan Province of China from 1990 to 2004. The small mammal hosts were captured from 25 counties which represent five geographical subregions, namely Middle Subregion of Hengduan Mountains, Southern Subregion of Hengduan Mountains, Eastern Plateau Subregion of Yunnan, Western Plateau Subregion of Yunnan and Southern Moun- tainous Subregion of Yunnan. The captured 10 803 small mammal hosts belong to nine families, 29 genera and 52 species in four orders （Rodentia, Insectivora, Scandentia and Lagomorpha）. A total of 68 571 gamasid mites were collected from the body surface of the captured small mammal hosts and all the gamasid mites were identified to 10 families, 33 genera and 112 species. This paper lists all the mite species, together with their taxonomic position （genera and families） and their corresponding hosts. Much more mite species were found in the Middle Subregion of Hengduan Mountains than in other geographical subregions. The total individuals of mites and small mammals in the Middle Subregion of Hengduan Mountains are also the most plentiful in the five geographical subregions. Three dominant mite species and three dominant small mammal hosts were determined as the dominant species in the investigated areas of Yunnan Province. The dominant hosts are Rattus flavipectus （which accounts for 34.85% of the total individuals）, Apodemus chevrieri （13.43%） and Rattus norvegicus （10.40%） while the dominant gamasid mite species are Laelaps nuttalli （Hirst, 1915）（27.84%）, Laelaps echidninus （Berlese, 1887）（18.38%） and Laelaps guizhouensis （Gu et Wang, 1981）（14.79%）. The results showed the high species diversity of gamasid mites in Yunnan Province and the uneven distribution feature in different subregions.

The cao vit gibbon is a critically endangered *** approximately 110 individuals remain in degraded karst forest along the China-Vietnam *** forest is unusual gibbon ***,the canopy height of cao vit gibbon habitat is approximately 10 *** on the locomotor behavior of gibbons living in this particular forest type might provide important insight into locomotor stability and variability of *** used 5 min scan samples to record the locomotion mode,support use and canopy strata of gibbons in 3 groups for 2096 h between January 2008 and December *** cao vit gibbon habitat has a lower canopy in comparison to that of other forests inhabited by gibbons,cao vit gibbons displayed a similar overall locomotor pattern to other gibbon species(Symphalangus syndactylus,Hylobates lar and Hylobates agilis)in which brachiation dominate their locomotor *** vit gibbons spent most of their time travelling on inclined branches(2-10 cm)in the middle stratum through the forest *** females appear to more often employ safer modes of locomotion(bridging more often and brachiation less),while adult males choose riskier modes(leaping more and climbing less).As gibbons increased in body weight,as they grew from infant to adult,they tended to use larger *** research documented that locomotor behavior in Hylobatidae is strongly determined by anatomical characters,but cao vit gibbons also show the ability to use various supports,enabling them to survive in karst forest.

CLORKE-SFS:Certificateless One-Round Key Exchange Protocol with Strong Forward Security in Limited Communication Scenarios

Certificateless one-round key exchange(CL-ORKE)protocols enable each participant to share a common key with only one round of communication which greatly saves communication *** protocols can be applied to scenarios with limited communication,such as space *** CL-ORKE protocols have been researched for years,lots of them only consider what secrets can be compromised but ignore the time when the secrets have been *** CL-ORKE protocols,the reveal of the long-term key attacks can be divided into two different attacks according to the time of the long-term key revealed:the attack to weak Forward Security(wFS)and the attack to strong Forward Security(sFS).Many CLKE protocols did not take into account the sFS property or considered sFS as *** this paper,we first propose a new security model for CL-ORKE protocols which considers the sFS property as well as the Ephemeral Key Reveal ***,we give a CL-ORKE protocol which is called ***-SFS is provably secure under the proposed model provided the Elliptic Curve Computational Diffie-Hellman(ECCDH)and the Bilinear Computational Diffie-Hellman problem(BCDH)assumption *** security model and the protocol may give inspiration for constructing oneround key exchange protocols with perfect forward security in certificateless scenarios.

A database of spectral energy distributions of progenitors of core-collapse supernovae

This paper presents a database of the spectroscopic-and photometric-spectral energy distributions(spec-SEDs and phot-SEDs respectively)of the progenitors of core-collapse supernovae(CCSNe).Both binary-and single-star progenitors are included in the *** database covers the initial metallicity(Z)range of 0.0001-0.03,mass range of 8-25 M⊙,binary mass ratio range of 0-1,and orbital period range of 0.1-10000 *** low-resolution spec-SEDs and phot-SEDs of single-and binary-star CCSN progenitors are included in the *** data can be used for studying the basic parameters,e.g.,metallicity,age,and initial and final masses of CCSN *** can also be used for studying the effects of different factors on the determination of parameters of CCSN *** the database is utilized for fitting the SEDs of binary-star CCSN progenitors,it is strongly suggested to determine the metallicity and orbital period in advance,but this is not necessary for single-star progenitors.
