Pinus is an eco.omically and eco.ogically important genus whose members are dominant co.ponents globally in low-latitude mountainous and mid-latitude temperate *** species richness is currently co.centrated in subtropical mid-low latitudes of the Northern Hemisphere,differing from the latitudinal diversity gradient mostly reco.nized in woody *** the present pattern was developing in Earth's past is still poorly studied,particularly in eastern ***,a new fossil species,Pinus shengxianica *** described based on a fossil seed co.e from the Late Miocene Shengxian Formation in Zhejiang,southeast China.A co.occurring co.e is reco.nized as a known fossil species,Pinus speciosa *** co.parison of extant and fossil members of Pinus suggests *** shares a striking co.e similarity to Pinus merkusii and Pinus latteri(subsection Pinus)from tropical Southeast Asia in having annular bulges around the umbo on the *** morphological resemblance indicates these two extant low-latitude pines probably possess a close affinity with the present newly-disco.ered *** and originated from East Asian mid-low latitude ancestors during this generic re-diversification in the *** scenario is co.sistent with the evolutionary trajectory reflected by the pine fossil history and molecular data,marking the Miocene as a key period for the origin and evolution of most extant pines *** co.occurrences of diverse co.ifers and broadleaved angiosperms preferring diverse niches demonstrate Late Miocene eastern Zhejiang was one of the hot spots for co.iferophyte diversity and hosted a needled-broadleaved mixed forest with co.plex vegetation structure and an altitudinal zonation.

Identification and Expression Analysis of Abscisic Acid Signal Transduction Genes in Hemp seeds

Abscisic acid(ABA)is involved in regulating diverse biological processes,but its signal transduction genes and roles in hemp seed germination are not well ***,the ABA signaling pathway members,PYL,PP2C and SnRK2 gene families,were identified from the hemp reference genome,including 7 CsPYL(pyrab-actin resistance1-like,ABA receptor),8 CsPP2CA(group A protein phosphatase 2c),and 7 CsSnRK2(sucrose nonfermenting1-related protein kinase 2).The co.tent of ABA in hemp seeds in germination stage is lower than that in non-germination *** ABA(1 or 10μM)treatment had a significant regulatory effect on the selected PYL,PP2C,SnRK2 gene ***3 and CsHAI1 were most significantly affected by exogenous ABA *** two-hybrid experiments were performed to reveal that CsPYL5,CsSnRK2.2,and CsSnRK2.3 co.ld interact with CsPP2CA7 and demonstrate that this interaction was *** results indicated that CsPYL5,CsSnRK2.2,CsSnRK2.3 and CsPP2CA7 might involve in the ABA signaling transduction pathway of hemp seeds during the hemp seed germination *** study suggested that novel genetic views can be brought into investigation of ABA signaling pathway in hemp seeds and lay the foundation for further exploration of the mechanism of hemp seed germination.

Effects of Near-surface Low Temperature on Freezing Injury of ‘Tunisian Soft-seed’ Pomegranate

[Objective]The paper was to explore the influence of near-surface low temperature on cultivation of soft-seed pomegranate,and to provide guidance for planting location of soft-seed pomegranate.[Method]Taking 10 soft-seed pomegranate planting plots under different site co.ditions as the research objects,the near-surface low temperature of 45-50 cm was dynamically monitored from December 1,2018 to February 20,2019,and co.parative analysis was made based on the local meteorological data over the same period.[Result]The near-surface low temperature of each temperature monitoring point was lower than the local meteorological data,which were all in the range of low temperature causing freezing in-jury of pomegranate trees,but the degree of freezing injury was *** variation of near-surface low temperature was positively co.related with the altitude of terrain,but negatively co.related with the difference of topography and *** the local topography and landform were similar,the accumulation time of near-surface low temperature was negatively co.related with the altitude of terrain,while the duration of low tem-perature directly affected the degree of freezing injury.[co.clusion]The development of soft-seed pomegranate cultivation in Tunisia along Huang Mangling region in Henan Province refers to the local meteorological ***,it is also necessary to pay attention to the impact of regional microclimate environment,especially early monitoring of near-surface temperature to select suitable site and natural co.ditions.

Efficient genotype-independent co.ton genetic transformation and genome editing

co.ton(Gossypium spp.)is one of the most important fiber crops *** the last two decades,transgenesis and genome editing have played important roles in co.ton ***,genotype dependence is one of the key bottlenecks in generating transgenic and gene-edited co.ton plants through either particle bombardment or Agrobacterium-mediated ***,we developed a shoot apical meristem(SAM)cell-mediated transformation system(SAMT)that allowed the transformation of recalcitrant co.ton genotypes including widely grown upland co.ton(Gossypium hirsutum),Sea island co.ton(Gossypium barbadense),and Asiatic co.ton(Gossypium arboreum).Through SAMT,we successfully introduced two foreign genes,GFP and RUBY,into SAM cells of some recalcitrant co.ton *** 2–3 months,transgenic adventitious shoots generated from the axillary meristem zone co.ld be reco.ered and grown into whole co.ton *** GFP fluorescent signal and betalain accumulation co.ld be observed in various tissues in GFP-and RUBY-positive plants,as well as in their progenies,indicating that the transgenes were stably integrated into the genome and transmitted to the next ***,using SAMT,we successfully generated edited co.ton plants with inheritable targeted mutagenesis in the GhPGF and GhRCD1 genes through CRISPR/Cas9-mediated genome *** summary,the established SAMT transformation system here in this study bypasses the embryogenesis process during tissue culture in a co.ventional transformation procedure and significantly accelerates the generation of transgenic and gene-edited plants for genetic improvement of recalcitrant co.ton varieties.

Transplantation of placenta-derived mesenchymal stem cell-induced neural stem cells to treat spinal co.d injury

Because of their strong proliferative capacity and multi-potency, placenta-derived mesenchymal stem cells have gained interest as a cell source in the field of nerve damage repair. In the present study, human placenta-derived mesenchymal stem ceils were induced to differentiate into neural stem cells, which were then transplanted into the spinal co.d after local spinal co.d injury in rats. The motor functional reco.ery and pathological changes in the injured spinal co.d were observed for 3 successive weeks. The results showed that human placenta-derived mesenchymal stem cells can differentiate into neuron-like cells and that induced neural stem cells co.tribute to the restoration of injured spinal co.d without causing transplant rejection. Thus, these cells promote the reco.ery of motor and sensory functions in a rat model of spinal co.d injury. Therefore, human placenta-derived mesenchymal stem cells may be useful as seed cells during the repair of spinal co.d injury.

Organelle genome assembly unco.ers the dynamic genome reorganization and cytoplasmic male sterility associated genes in tomato

To identify cytoplasmic male sterility(CMS)-associated genes in tomato,we determined the genome sequences of mitochondria and chloroplasts in three CMS tomato lines derived from independent asymmetric cell fusions,their nuclear and cytoplasmic donors,and male fertile weedy cultivated tomato and wild *** structures of the CMS mitochondrial genomes were highly divergent from those of the nuclear and cytoplasmic donors,and genes of the donors were mixed up in these *** the other hand,the structures of CMS chloroplast genomes were moderately co.served across the donors,but CMS chloroplast genes were unexpectedly likely derived from the nuclear *** analysis of the structures and co.tents of organelle genes and transcriptome analysis identified three genes that were uniquely present in the CMS lines,but not in the donor or fertile ***-sequencing analysis indicated that these three genes transcriptionally expressed in anther,and identified different RNA editing levels in one gene,orf265,that was partially similar to ATP synthase subunit 8,between fertile and sterile *** orf265 was a highly potential candidate for CMS-associated *** study suggests that organelle reorganization mechanisms after cell fusion events differ between mitochondria and chloroplasts,and provides insight into the development of new F1 hybrid breeding programs employing the CMS system in tomato.

Germplasm resource innovation is a crucial factor for cultivar development,particularly within the co.text of hybrid rice breeding based on the three-line *** 9311A,a cytoplasmic male sterile(CMS)line,has been successfully cultivated using rice restoration materials and extensively employed as a female parent in hybrid breeding program in *** line was developed by crossing the CMS line Zhong 9A with a two-line restorer line 93-11,with the intention of eliminating the restoring ability of 93-11 while retaining the sterility gene WA352c from Zhong *** 9311A effectively amalgamates the most favorable agronomic traits from both parental *** this study,the relationship between phenotypic characteristics and the known functional genes of Quan 9311A were analyzed using the rice genome navigation technology based on whole-genome *** findings revealed that Quan 9311A harbors multiple superior alleles from both 93-11 and Zhong 9A,providing exceptional agronomic traits that are unavailable in earlier CMS *** the removal of the fertility restorer gene Rf3 from 93-11,numerous chromosomal segments from 93-11 persist in the Quan 9311A ***,the hybrid rice Quanyousimiao(QYSM)and the restorer line Wushansimiao(WSSM)were used as examples to illustrate the important role of Quan 9311A as the female parent in *** was found that QYSM carries a great number of superior alleles,which acco.nts for its high grain yield and wide *** insights not only advanced the utilization of hybrid rice pairing groups but also provided guidance for future breeding *** study introduced innovative co.cepts to further integrate genomics with traditional breeding ***,Quan 9311A signified a significant milestone in rice breeding technology,opening up novel avenues for hybrid rice development.

[Objective] This study aimed to screen out floribunda rose cultivars resistant to high temperature in tropical areas. [Method] A total of 8 floribunda rose cul- tivars （Carefree Wonder, Scloss Mannheim, Bright Smile, Goldmarie, Betty Prior, Red Riding Hood Fairy Tale, Golden Scepter and Iceberg） were selected as the research objects, and their growth, adaptability and disease resistance in Sanya area were co.pared. [Result] Carefree Wonder, Scloss Mannheim, Iceberg and Bright Smile were suitable to be planted in Sanya area. Among them, Carefree Wonder, Scloss Mannheim and Iceberg were characterized with fewer diseases, bright co.ors, heat resistance and strong stress resistance, and they co.ld be planted as ornamental flowers in Sanya area. [co.clusion] This study will provide a reference for the selection of ornamental flowers in Sanya area.

Performance of elastic ball screen-cleaning device based on DEM-MBD co.pling simulation

In seed breeding, the vibrating screen used for pre-sowing grading parental seeds often has the problem of poor self-purification rate. Using elastic balls to clean the plugging seeds and ensure the self-purification of the screen is a viable technical solution. To clarify and improve the operating performance of the elastic ball particles, a DEM-MBD co.pling simulation model of the elastic ball screen-cleaning device was established in this study, and a numerical simulation analysis of the co.plex motion process was carried out. The mechanism of action of the elastic ball was explained more co.pletely from the aspects of excitation force and energy transfer. Through the Plackett-Burman test, it was determined that the vibration frequency has the most significant effect on the excitation force. Multiple linear regression analysis was performed on each factor and the mathematical relationship equation. It was found that when the maximum excitation force was greater than 7 N and the average excitation force was greater than 2 N, the self-purification rate was greater than 95%. The self-purification rate was then increased to 99.81% by variable frequency tests. The variable frequency achieved better self-purification effect.
