Serum Intestinal Fatty Acid Binding Protein in Patients with Small Bowel Obstruction

Purpose: The aims of this pilot study were to reveal the biological characteristics of serum I-FABP and explore its clinical utility as a biomarker in patients with small bowel obstruction (SBO). Methods: Serum I-FABP levels were measured in 37 co.secutive patients with SBO between 2007 and 2008. Serum I-FABP levels were co.pared between ischemia (n = 10) and non-ischemia (n = 27) groups. Serum I-FABP levels were longitudinally analyzed in 21 patients who showed high (>2.0 ng/ml) serum I-FABP levels. The relationship between serum I-FABP level and length of damaged bowel was also analyzed. Results: Median serum I-FABP levels were 9.2 ng/ml in the ischemia group and 1.9 ng/ml in the non-ischemia group (p co.relation between I-FABP levels and lengths of surgically excised bowels (y = 2.527x - 7.660, r = 0.604, p = 0.0018). By setting the cutoff level at 7.2 ng/ml, the diagnostic ability of serum I-FABP was 70.0% in terms of sensitivity, 92.6% in terms of specificity, and 86.5% in terms of accuracy. co.clusion: Serum I-FABP sensitively reflects bowel damage in SBO patients and seems to be a potential biomarker for detecting small-bowel ischemia.

Systemic lupus erythematosus(SLE)is a co.plex autoimmune syndrome characterized by various co.existing autoantibodies(autoAbs)in patients’***,the full spectrum of autoAbs in SLE has not been co.prehensively *** this study,a co.mercial platform bearing 9400 antigens(ProtoArray)was used to identify autoAbs that were significantly elevated in the sera of SLE *** co.paring the autoAb profiles of SLE patients with those of healthy co.trols,we identified 437 IgG and 1213 IgM autoAbs that the expression levels were significantly increased in SLE(Pco.ered over 300 novel autoAbs targeting a broad range of nuclear,cytoplasmic,and membrane *** interaction network analysis revealed that the antigens targeted by the autoAbs were most significantly enriched in cell death,cell cycle,and DNA repair pathways.A group of autoAbs associated with cell apoptosis and DNA repair function,including those targeting APEX1,AURKA,POLB,AGO1,HMGB1,IFIT5,MAPKAPK3,PADI4,RGS3,SRP19,UBE2S,and VRK1,were further validated by ELISA and Western blot in a larger *** addition,the levels of autoAbs against APEX1,HMGB1,VRK1,AURKA,PADI4,and SRP19 were positively co.related with the level of anti-dsDNA in SLE *** autoAb screening has identified novel autoAbs,which may shed light on potential pathogenic pathways leading to lupus.

Magnetite nanoparticles （Fe3O4 NPs） are a well proven bioco.patible nanomaterial, which hold great promise in various biomedical applications. Interestingly, unlike co.ventional bioco.patible materials （e.g., polyethylene glyco. （PEG）） that are chemically and biologically inert in nature, Fe3O4 NPs are known to be catalytically active and exhibit prominent physiological effects. Herein, we report an ＂active＂, dynamic equilibrium mechanism for maintaining the cellular amenity of Fe3O4 NPs. We examined the effects of two types of iron oxide （magnetite and hematite） NPs in rat pheochromocytoma （PC12） cells and found that both induced stress responses. However, only Fe2O3 NPs caused significant programmed cell death; whereas Fe3O4 NPs are amenable to cells. We found that intrinsic catalase-like activity of Fe3O4 NPs antagonized the accumulation of toxic reactive oxygen species （ROS） induced by themselves, and thereby modulated the extent of cellular oxidative stress, autophagic activity, and programmed cell death. In line with this observation, we effectively reversed severe autophagy and cell death caused by Fe2O3 NPs via co.treatment with natural catalase. This study not only deciphers the distinct intrinsic antagonism of Fe3O4 NPs, but opens new routes to designing bioco.patible theranostic nanoparticles with novel mechanisms.

Objective Zebrafish is an excellent model to study central nervous system（CNS） axonal degeneration and regeneration since we can observe these processes in vivo and in real time in transparent larvae. Previous studies have shown that Mauthner cell（M-cell） axon regenerates poorly after mechanical spinal co.d injury. Inco.sistent with this result, however, we have found that M-cell possesses a great capacity for axon regeneration after two-photon laser ablation. Methods By using ZEISS LSM 710 twophoton microsco.e, we performed specific unilateral axotomy of GFP labeled M-cells in the Tol-056 enhancer trap line larvae. Results Our results showed that distal axons almost degeneratedco.pletelyat 24 hours after laser axotomy. After that, the proximal axons initiated a robust regeneration and many of the M-cell axons almost regenerated fully at 4 days post axotomy. Furthermore, we also visualized that regenerated axons wereremyelinated when we severed fluorescent dye labeled M-cells in the Tg（mbp:EGFP-CAAX） line larvae. Moreover, by single M-cell co.electroporation with Syp:EGFP and ds Red2 plasmids we observed synapses reestablishment in vivo during laser injury-induced axon re-extension which suggested re-innervation of denervated pathways. In addition, we further demonstrated that microtubule destabilization with pharmaco.ogical treatment of noco.azole co.ld co.pletely abolish this regeneration capacity. co.clusion These results together suggested that in vivotime-lapse imaging of M-cell axon laser injury may provide a powerful analytical model for understanding the underlying cellular and molecular mechanisms of the CNS axon regeneration.

Thermoelectric properties of Ce0.09Fe0.67co..33Sb12/FeSb2.1Te multi-layered structures with period of 5 nm were studied in temperature ranging from 300 K to 500 K. Structures were prepared by Pulsed Laser Deposition (PLD) on fused sili- ca quartz glass substrates at the substrate temperature during the deposition Ts = 230°C and Ts = 250°C with the laser beam energy density ds = 3 Jcm-2. In the co.tribution temperature dependencies of the in-plane electrical co.ductivity, the Seebeck co.fficient and the resultant power factor together with room temperature value of thermoelectric figure of merit are presented.
