Respiratory antibiotics have been proven clinically beneficial for the treatment of severe lung infections such as Pseudomonas *** a high local concentration of inhaled antibiotics for an extended time in the lung is crucial to ensure an adequate antimicrobial *** this study,we aim to investigate whether an extended exposure of ciprofloxacin(CIP),a model fluoroquinolone drug,in the lung epithelial lining fluid(ELF)could be achieved via a controlled-release formulation *** solutions were intratracheally instilled to the rat lungs at 3 different rates,i.e.,T0h(fast),T2h(medium),and T4h(slow),to mimic different release profiles of inhaled CIP formulations in the ***,the concentration-time profiles of CIP in the plasma and the lung ELF were obtained,respectively,to determine topical exposure index(ELF-Plasma AUC Ratio,EPR).The in silico PBPK model,validated based on the in vivo data,was used to identify the key factors that influence the disposition of CIP in the plasma and *** medium and slow rates groups exhibited much higher EPR than that fast instillation *** ELF AUC of the medium and slow instillation groups were about 200 times higher than their plasma *** contrast,the ELF AUC of the fast instillation group was only about 20 times higher than the plasma *** generated whole-body PBPK rat model,validated by comparison with the in vivo data,revealed that drug pulmonary absorption rate was the key factor that determined pulmonary absorption of *** study suggests that controlled CIP release from inhaled formulations may extend the exposure of CIP in the ELF post pulmonary *** also demonstrates that combining the proposed intratracheal installation model and in silico PBPK model is a useful approach to identify the key factors that influence the absorption and disposition of inhaled medicine.

An efficient and mild synthesis of 2-（guaiazulen-l-yl）furans, starting from easily accessible 1-（3-aryl-2- cyanopropenoyl）guaiazulenes, tributylphosphine and acyl chlorides, is described. The strategy employs the intramolecular Wittig protocol as a key step to append the crucial furan ring, leading to the highly functional furans in good yields.

OBJECTIVES:To investigate the influence of catgut implantation at acupoints on leptin resistance(LR) and insulin resistance(IR) in the simple obesity ***:Rats were made obese with high-fat diets,after which surgical catgut was implanted at Zusanli(ST 36) and Neiting(ST 44) acupoints once a week for 4 weeks(implantation group).Rats from the implantation group were compared with normal rats and unoperated obese rats(control group).Gene expression of the leptin receptor(OB-R) was evaluated using RT-PCR and northern *** and hypothalamus leptin and insulin(INS) levels were determined by *** weight,Lee's index,body fat,serum and hypothalamus leptin and INS levels,and hypothalamic OB-R gene expression were determined before and after ***:Body weights,Lee's index,body fat,and serum leptin and INS levels were significantly higher in obese than in normal *** leptin and INS levels and OB-R gene expression were significantly lower in obese *** implantation at acupoint promoted weight loss and decreased serum leptin and INS *** leptin and INS levels and OB-R gene expression increased ***:Catgut implantation at acupoint adjusts central and peripheral leptin and promotes hypothalamic OB-R gene *** may be an important method for regulation of LR,IR and abnormal endocrinology and metabolism.

Cerebral hypoxia or ischemia results in cell death and cerebral edema, as well as other cellular reactions such as angiogenesis and the reestablishment of functional microvasculature to promote recovery from brain injury. Vascular endothelial growth factor is expressed in the central nervous system after hypoxic/ischemic brain injury, and is involved in the process of brain repair via the regulation of angiogenesis, neurogenesis, neurite outgrowth, and cerebral edema, which all require vascular endothelial growth factor signaling. In this review, we focus on the role of the vascular endothelial growth factor signaling pathway in the response to hypoxic/ischemic brain injury, and discuss potential therapeutic interventions.

Dogwood fruits are a valuable source of active ingredients, such as phenolic compounds, vitamin C, iridoids, flavonoids and anthocyanins. Plant extracts and substances derived from latin Cornus mas L. exhibit not only strong antibacterial but also antioxidant and tonicity properties, effectively preventing the development of inflammation in living organisms. In the present study, we attempted to obtain the innovative, multi-functional plant extract from the fruit of dogwood(Cornus mas L.). During the extraction process a mixture of water, glycerol, and vegetable oil were used as an extractant. The usage of such mixtures of solvents enabled us to extract a variety of active substances, soluble in both water and oils. The obtained extracts were analyzed for their physicochemical and biochemical properties, in order to apply the extract in a body nutrient lotion. The results clearly showed that such extract could be an innovative and multi-functional raw material used in cosmetics industry.
