OBJECTIVE To investigate the effects of IMM-H004 on permanent focal cerebral ischemia injury and associated cardiopulmonary complications,further elucidating the molecular *** The effects of IMM-H004 were investigated in wild-type(WT) and CKLF1-/-*** effects of IMM-H004 on ischemic stroke injury and its cardiopulmonary complications were determined using 2,3,5-triphenyltetrazolium chloride(TTC) staining,behavior tests,magnetic resonance imaging(MRI)scans,enzyme-linked immunosorbent assay(ELISA),Nissl staining,and histo-pathological *** molecular experiments including immunohistological staining,immunofluorescence staining,quantitative RT-PCR,Western blotting,and co-immunoprecipitation assays were used to elucidate the underlying *** IMM-H004 treatment provided significant protection against ischemic stroke-induced brain injury and associated cardiopulmonary complications,through CKLF1-depedent-anti-inflammation pathway in ***-H004 downregulated the amount of CKLF1 and disturbed the combination between CKLF1 and C-C chemokine receptor type 4,suppressing the inflammatory response and protecting the damaged organs in ischemic *** This preclinical study established efficacy of IMM-H004 as a potential therapeutic medicine for ischemic stroke and associated cardiopulmonary *** protective effects of IMM-H004 may due to its specific mechanism through *** results support further efforts to develop IMM-H004 for human clinical trials in acute cerebral ischemia,especially for patients who are not suitable for reperfusion therapy.

To date,no vaccine or monoclonal antibody(mAb) against *** has been approved for use in *** lab has developed a five-antigen *** vaccine(rFSAV),which is now under efficacy evaluation in a phase Ⅱ clinical *** the current study,using overlapping peptides and antiserum from rFSAV-immunized volunteers,we identified seven B-cell immunodominant epitopes on 4 antigens in rFSAV,including five novel epitopes(Hla48-65,IsdB402-41 9,IsdB432-449,SEB78-95 and MntC7-24).Ten immunodominant-epitope mAbs were generated against these epitopes,and all of them exhibited partial protection in a mouse sepsis *** robust mAb were used together as a mAb cocktail to prevent MRSA252 *** results showed that the mAb cocktail was efficient in combating *** infection and that its protective efficacy correlated with a reduced bacterial burden,and decreased infection pathology,which demonstrates that the mAb cocktail is a promising *** vaccine candidate.

Objective: Diabetes mellitus (DM) is a severe chronic disease that results in high morbidity and mortality. DM causes endothelial injury， such as endothelial inflammation， oxidative stress and dysfunction， as a basis for cardiovascular complications of DM. Therefore， blockage of DM-induced endothelial injury (DEI) is a viable strategy for the prevention of DM-caused adverse outcomes. However， few effective approaches have been developed for the intervention of DEI. Krill oil (KO) has been reported to have anti-inflammatory and anti-oxidative activities， but its effect on DEI is unknown. Hence， the aims of this study were to investigate the effect and molecular mechanism of KO on ***: To investigated the preventive effect of krill oil (KO) on DEI， streptozotocin and high-fat diet-induced mice model of type 2 DM were fed with KO for 6months. RNA sequencing for endothelial cells (ECs) was used to explore the mechanism of KO’s protective function. To clarify the role of NRF2 signaling in KO’s protective function on DEI， KO was given to Nfe2l2 gene-silenced ECs or knockout mice. Molecular docking assay and surface plasmon resonance assay were carried out to reveal the ability of the main components of KO to bind KEAP1. Results: KO significantly alleviated DEI and aortic pathological injury in the wild-type diabetic mice. RNA sequencing revealed that KO dramatically activated nuclear factor erythroid 2-related factor 2 (Nfe2l2/NRF2) antioxidant signaling in high glucose-challenged ECs， the effect of which was further confirmed in the diabetic aortas. Nfe2l2 gene deletion or silencing completely abolished KO’s protection against DEI in vivo and in vitro， demonstrating that NRF2 was required for KO’s action. Further， molecular docking assay and surface plasmon resonance assay identified that KO’s functional component astaxanthin (AST)， but not docosahexaenoic acid and eicosapentaenoic acid， was able to bind the Kelch domain of Kelch like ECH associated protein 1 (KE

Influence of Temperature and Salinity on Germination of Eelgrass (Zostera marina L.) Seeds

Seagrass restoration as part of ocean ecosystem protection has been launched for many years all over the world, but intensive research on this subject in China has just begun in recent years. Seed broadcasting has been widely accepted as the most potentially useful method for seagrass restoration over large areas. We examined the influence of key environmental factors on seed germination to help promote eelgrass bed restoration. Under anoxic conditions, the influence of temperature and salinity on the germination rate of eelgrass (Zostera marina L.) seeds was examined at different combinations of four temperatures (4, 9, 14, and 24℃) and nine salinities (5 to 45, increment of 5). The effect of significant interaction of temperature and salinity on germination rate was observed (ANOVA) (P<0.001). The highest germination rate (83.3 ± 3.5)% was reached in 8 weeks at 14℃ and salinity 5. Higher temperature significantly increased the germination rate at salinity 5 (P<0.001) during the whole observation period except for 24℃, while lower salinity significantly increased the germination rate at 14℃ (P<0.001). Although significant interaction was found between temperature and salinity (P<0.001), the influence of salinity was stronger than that of temperature for the germination of eelgrass seeds. These results provide useful information for the propagation of artificial seedlings for seagrass restoration in China.

Changes in the number and function of dendritic cells(DCs)have been reported to play an important role in endotoxin *** has been reported that expansion of splenic CD11clowCD45RB+DCs occurs in mice injected with sublethal doses of lipopolysaccharide(LPS).However,the function of endotoxic shock-expanded CD11clowCD45RB+DCs has not been *** this work,we show that endotoxic shock promotes the expansion of CD11clowCD45RB+cells with dendritic morphology and the production of low levels of inflammatory cytokines and costimulatory *** expanded cells induce the generation of regulatory T cells(Tregs),show incapability to stimulate T cells,and induce apoptosis of CD4+T cells in *** compared to CD11chiCD45RB-conventional DCs,the expanded cells exert better protection against colitis induction by CD4+CD25-T cells,even though both subpopulations show similar ability to induce Tregs in *** better control of proinflammatory cytokine responses in vivo by the expanded cells is associated with more apoptosis in the Payer’s patches and in colonic tissue-infiltrating ***,the expanded cells can modulate inflammatory T cell responses through multiple *** study facilitates a better understanding how innate immune responses may shape adaptive immunity and immune suppression following LPS-induced acute inflammation.

Multi-source financing for tuberculosis treatment in China: key issues and challenges

Background:The End Tuberculosis(TB)Strategy of the World Health Organization highlights the need for patientcentered care and social protection measures that alleviate the financial hardships faced by many TB *** China,TB treatments are paid for by earmarked government funds,social health insurance,medical assistance for the poor,and out-of-pocket payments from *** part of Phase III of the China-Gates TB project,this paper introduces multi-source financing ofTB treatment in the three provinces of China and analyzes the challenges of moving towards universal coverage and its implications of multi-sectoral engagement for TB *** text:The new financing policies forTB treatment in the three provinces include increased reimbursement forTB outpatient care,linkage ofTB treatment with local poverty alleviation programs,and use of local government funds to cover some costs to reduce out-of-pocket ***,there are several challenges in reducing the financial burdens faced byTB ***,medical costs must be contained by reducing the profit-maximizing behaviors of ***,treatment forTB and multi-drug resistant TB(MDR-TB)is only available at county hospitals and city or provincial hospitals,respectively,and these hospitals have low reimbursement rates and high ***,many patients with TB and MDR-TB are at the edge of poverty,and therefore ineligible for medical assistance,which targets extremely poor *** addition,the local governments of less developed provinces often face fiscal difficulties,making it challenging to use of local government funds to provide financial support for TB *** suggest that stakeholders at multiple sectors should engage in transparent and responsive communications,coordinate policy developments,and integrate resources to improve the integration of social protection ***:The Chinese government is examining the establishment of multi-source financing forTB treatm

Increasing rates of life-threatening infections and decreasing susceptibility to antibiotics urge an effective vaccine targeting Staphylococcus *** we investigate the role of cellular immunity in FnBPA mediated protection in Staphylococcus aureus *** study revealed FnBPA broadly protected mice from seven FnBPA isotypes strains in the sepsis *** immunized B-cell deficient mice were protected against lethal challenge, while T-cell deficient mice were *** mice with FnBPA specific CD4+ T-cells conferred antigen specific *** vitro assays indicated that isolated FnBPA specific splenocytes from immunized mice produced abundant ***-17A deficient mice were not protected from a lethal challenge by FnBPA ***, neutralizing IL-17A, but not IFN-γ,reverses FnBPA-induced protective efficacy in sepsis and skin infection *** findings suggest that IL-17A producing Th17 cells play an essential role in FnBPA vaccine-mediated defense against *** sepsis and skin infection in mice.

Evaluation of harmonious development for economy-environment system in coastal cities: a case study of Tianjin Municipality

With the aim of the harmonious development of economy-environment system in coastal cities in China. an index ,system used to evaluate the economy-environment system is built up in this paper, which includes four aspects： economy, environment, resources, and ocean industry. Based on the analysis on present condition and future trends of economic development in Tianjin and the quantification of various evaluation indices, the aathor applies integrated index valuation model to valuate the harmonious development af economy-environment of Tianjin. The results show that the coordinated degree of economy-environment would drop down in the future, from 0.95（superior level of harmonious development） in 2000 to 0.59（inferior level of harmonious development） in 2015. under the circumstance of the current economic development mode. The level of comprehensive development of Tianjin also presents to descend. Based on the analyzing of status and future trends of environment-economy coordinated development, the paper puts forward the countermeasures such as industry, structure adjustment, increasing the level of environmental protection investment, strengthening the enforcement of en vironmental policies to improve the coordinated development of environment-economy in Tianjin municipality.
