AP-1 and SP1 trans-activate the expression of hepatic CYP1A1 and CYP2A6 in the bioactivation of AFB1 in chicken

Dietary exposure to aflatoxin B1(AFB1)is harmful to the health and performance of domestic animals.The hepatic cytochrome P450s(CYPs),CYP1A1 and CYP2A6,are the primary enzymes responsible for the bioactivation of AFB1to the highly toxic exo-AFB1-8,9-epoxide(AFBO)in chicks.However,the transcriptional regulation mechanism of these CYP genes in the liver of chicks in AFB1metabolism remains unknown.Dual-luciferase reporter assay,bioinformatics and site-directed mutation results indicated that specificity protein 1(SP1)and activator protein-1(AP-1)motifs were located in the core region-1,063/-948,-606/-541 of the CYP1A1 promoter as well as-636/-595,-503/-462,-147/-1 of the CYP2A6 promoter.Furthermore,overexpression and decoy oligodeoxynucleotide technologies demonstrated that SP1 and AP-1 were pivotal transcriptional activators regulating the promoter activity of CYP1A1 and CYP2A6.Moreover,bioactivation of AFB1to AFBO could be increased by upregulation of CYP1A1 and CYP2A6 expression,which was trans-activated owing to the upregulation of AP-1,rather than SP1,stimulated by AFB1-induced reactive oxygen species.Additionally,nano-selenium could reduce ROS,downregulate AP-1 expression and then decrease the expression of CYP1A1 and CYP2A6,thus alleviating the toxicity of AFB1.In conclusion,AP-1 and SP1 played important roles in the transactivation of CYP1A1 and CYP2A6 expression and further bioactivated AFB1to AFBO in chicken liver,which could provide novel targets for the remediation of aflatoxicosis in chicks.

Alzheimer's disease(AD)is an irreversible and neurodegenerative disease that slowly impairs memory and neurocognitive function,but the etiology of AD is still unclear.With the explosive growth of electronic health data,the application of artificial inteligence(Al)in the healthcare setting provides excellent potential for exploring etiology and personalized treatment approaches,and improving the disease's diagnostic and prognostic outcome.This paper first briefly introduces Al technologies and applications in medicine,and then presents a comprehensive review of Al in AD.In simple,it includes etiology discovery based on genetic data,computer-aided diagnosis(CAD),computer-aided prognosis(CAP)of AD using multi-modality data(genetic,neuroimaging and linguistic data),and pharmacological or non-pharmacological approaches for treating AD.Later,some popular publicly available AD datasets are introduced,which are important for advancing Al technologies in AD analysis.Finally,core research challenges and future research directions are discussed.

Kinetic analysis of p53 gene network with time delays and PIDD

p53 kinetics plays a key role in regulating cell fate.Based on the p53 gene regulatory network composed by the core regulatory factors ATM,Mdm2,Wipl,and PIDD,the effect of the delays in the process of transcription and translation of Mdm2 and Wipl on the dynamics of p53 is studied theoretically and numerically.The results show that these two time delays can affect the stability of the positive equilibrium.With the increase of delays,the dynamics of p53 presents an oscillating state.Further,we also study the effects of PIDD and chemotherapeutic drug etoposide on the kinetics of p53.The model indicates that(i)PIDD low-level expression does not significantly affect p53 oscillatory behavior,but high-level expression could induce two-phase kinetics of p53;(ii)Too high and too low concentration of etoposide is not conducive to p53 oscillation.These results are in good agreement with experimental findings.Finally,we consider the infuence of internal noise on the system through Binomial r-leap algorithm.Stochastic simulations reveal that high-intensity noise completely destroys p53 dynamics in the deterministic model,whereas low-intensity noise does not alter p53 dynamics.Interestingly,for the stable focus,the internal noise with appropriate intensity can induce quasi-limit cycle oscillations of the system.Our work may provide the useful insights for the development of anticancer therapy.

Exploring the mechanism of Crocus sativus and Rosa rugosa for the treatment of coronary heart disease based on network pharmacology and molecular docking

Coronary atherosclerotic heart disease(CHD)is the main type of cardiovascular disease.The efficacy of Uyghur drug compound Saffron formula in CHD has been clinically proven.However,the underlying mechanism remains unclear.In this study,researchers investigated the active ingredients and mechanism of action of Crocus sativus and Rosa rugosa in the treatment of CHD by network pharmacology and molecular docking techniques,collected target information with the help of TCMSP,GEO,GeneCards,and other databases,constructed protein-protein interaction(PPI)network diagrams by STRING database,performed GO and KEGG pathway enrichment analysis on common targets,and finally molecularly docked the active ingredients with core targets.C.sativus-R.rugosa have a variety of polyphenol compounds,a total of 12 active ingredients,including quercetin and kaempferol,were screened.The first three targets intersected with the core targets of CHD as AKT1,TNF,and IL-1B.Enrichment results of KEGG pathway showed that C.sativus-R.rugosa against CHD involved atherosclerosis pathways.The molecular docking results showed that quercetin and kaempferol were well bound to the core targets,and it was speculated that these components might be the main active ingredients for the treatment of CHD.The potential mechanism of action of C.sativus-R.rugosa for the treatment of coronary heart disease was initially revealed.

Melatonin is an endogenous monoamine hormone secreted by the pineal gland.Melatonergic signaling has been shown to play a role in circadian rhythm regulation,lipid and glucose metabolism,and obesity,and it has anti-inflammatory and antioxidant properties.The melatonin receptor 1B gene(MTNR1B)is expressed in,among other tissues,the brain and pancreatic beta cells,and risk variants for T2D have been reported as impairing early insulin secretion and increasing fasting glucose levels.^(1) Variants in the MTNR1B gene also have been reported in patients with depression(MDD).

Bibliometric analysis of research challenges and trends in urban underground space

The utilization and development of urban underground space play a crucial role in optimizing the layout of civic architecture and enhancing the urban ecological environment,which contributes toward increasing the overall carrying capacity and promoting sustainable development in megacities.To delve into the research progress of urban underground space,knowledge maps were created using the information visualization software VOSviewer.The literature was systematically extracted from three prominent databases,namely,Web of Science,Scopus,and China National Knowledge Infrastructure.According to the bibliometric analysis of the co-citation and core words co-occurrence,the trends and challenges of research on urban underground space were identified.As highlighted by the results obtained,it still remains highly challenging to achieve interdisciplinary collaboration in urban underground space research;the research trends of urban underground space consist of collaborative planning and whole life cycle sustainable development,multisource geological data informatization and resource evaluation,infrastructure design optimization,and intelligent construction.The knowledge map,drawn using bibliometric methods,offers a quantitative analysis of literature retrieval across various levels.It is recognized as an essential tool for exploring and identifying challenges and trends in urban underground space.

“OMINO”: A Modern Approach to Postural Standing Reeducation through Breathing Techniques

Postural problems are very common, which is why plenty of Techniques have been created to solve them: Mezieres Technique (including muscle chains), RPG (Global Postural Re-Education, including pelvic diaphragm), Pilates Method (including core muscles), Bobath Concept (including COG, center of gravity) and Yoga. In this work, I show a new way of treating and solving postural problems. I am basing this physiotherapy approach on a new Image I created. Every patient can use it for every correction and change in posture. I called the new Image: “OMINO (little man)”. “Postural Card” could be a good work to make “OMINO” possible. “OMINO” is immediately understandable and, using “OMINO POCKET INSTRUCTIONS”, it can be easily done. Contribution of paper: • Trunk is a vital element for any kind of rehabilitation;• “core stability” is the key to activating the trunk. • Doing “OMINO” means working on “core stability”;• “OMINO” makes rehabilitation simple for the patient.

Embedded system design is the core course of the telecommunication major in engineering universities,which combines software and hardware through embedded development boards.Aiming at the problems existing in traditional teaching,this paper proposes curriculum teaching reform based on the outcome-based education(OBE)concept,including determining course objectives,reforming teaching modes and methods,and improving the curriculum assessment and evaluation system.After two semesters of practice,this method not only enhances students’learning initiative but also improves teaching quality.

Advances in selective catalytic oxidation of ammonia (NH_(3)-SCO): A review of catalyst structure-activity relationship and design principles

NH_(3) in ambient air directly leads to an increase in the aerosol content in the air. These substances lead to the formation of haze to various environmental problems after atmospheric circulation and diffusion. Controlling NH_(3) emissions caused by ammonia escaping from mobile and industrial sources can effectively reduce the NH_(3) content in ambient air. Among the various NH_(3) removal methods, the selective catalytic oxygen method (NH_(3)-SCO) is committed to oxidizing NH_(3) to environmentally harmless H_(2)O and N_(2);therefore, it is the most valuable and ideal ammonia removal method. In this review, the characteristics of loaded and core-shell catalysts in NH_(3)-SCO have been reviewed in the context of catalyst structure-activity relationships, and the H_(2)O resistance and SO2 resistance of the catalysts are discussed in the context of practical application conditions. Then the effects of the valence state of the active center, oxygen species on the catalyst surface, dispersion of the active center and acidic sites on the catalyst performance are discussed comprehensively. Finally, the shortcomings of the existing catalysts are summarized and the catalyst development is discussed based on the existing studies.

Chronic heart failure(HF)is a clinical syndrome with high morbidity and mor-tality worldwide.Cardiac rehabilitation(CR)is a medically supervised program designed to maintain or improve cardiovascular health of people living with HF,recommended by both American and European guidelines.A CR program con-sists of a multispecialty group including physicians,nurses,physiotherapists,trainers,nutritionists,and psychologists with the common purpose of improving functional capacity and quality of life of chronic HF patients.Physical activity,lifestyle,and psychological support are core components of a successful CR program.CR has been shown to be beneficial in all ejection fraction categories in HF and most patients,who are stable under medication,are capable of participating.An individualized exercise prescription should be developed on the basis of a baseline evaluation in all patients.The main modalities of exercise training are aerobic exercise and muscle strength training of different intensity and frequency.It is important to set the appropriate clinical outcomes from the beginning,in order to assess the effectiveness of a CR program.There are still significant limitations that prevent patients from participating in these programs and need to be solved.A significant limitation is the generally low quality of research in CR and the presence of negative trials,such as the rehabilitation after myocardial infarction trial,where comprehensive rehabilitation following myocardial infraction had no important effect on mortality,morbidity,risk factors,or health-related quality of life or activity.In the present editorial,we present all the updated knowledge and recommendations in CR programs.
