Emerging structures and dynamic mechanisms ofγ-secretase for Alzheimer’s disease

γ-Secretase,called“the proteasome of the membrane,”is a membrane-embedded protease complex that cleaves 150+peptide substrates with central roles in biology and medicine,including amyloid precursor protein and the Notch family of cell-surface *** inγ-secretase and amyloid precursor protein lead to early-onset familial Alzheimer’s disease.γ-Secretase has thus served as a critical drug target for treating familial Alzheimer’s disease and the more common late-onset Alzheimer’s disease as ***,critical gaps remain in understanding the mechanisms of processive proteolysis of substrates,the effects of familial Alzheimer’s disease mutations,and allosteric modulation of substrate cleavage byγ-*** this review,we focus on recent studies of structural dynamic mechanisms ofγ-*** mechanisms,including the“Fit-Stay-Trim,”“Sliding-Unwinding,”and“Tilting-Unwinding,”have been proposed for substrate proteolysis of amyloid precursor protein byγ-secretase based on all-atom molecular dynamics *** an incorrect registry of the Notch1 substrate was identified in the cryo-electron microscopy structure of Notch1-boundγ-secretase,molecular dynamics simulations on a resolved model of Notch1-boundγ-secretase that was reconstructed using the amyloid precursor protein-boundγ-secretase as a template successfully capturedγ-secretase activation for proper cleavages of both wildtype and mutant Notch,being consistent with biochemical experimental *** approach could be potentially applied to decipher the processing mechanisms of various substrates byγ-*** addition,controversy over the effects of familial Alzheimer’s disease mutations,particularly the issue of whether they stabilize or destabilizeγ-secretase-substrate complexes,is ***,an outlook is provided for future studies ofγ-secretase,including pathways of substrate binding and product release,effects of modulators on familial Alzheimer’s disease mutations

Performance Analysis of a Coupled System based on Organic Rankine Cycle and Double Effect Absorption Refrigeration for Waste Heat Recovery in Data Center

During data center operation,it generates a significant volume of low-grade waste *** recover waste heat,a coupled system including solar collector,double effect absorption refrigeration and organic Rankine cycle is *** system performance is analyzed in *** the organic Rankine cycle,five organic working fluids(R245fa,R245ca,R123,R11,and R113) are selected.R245fa,R113 and R245ca obtain the maximum net power output,thermal efficiency and exergy efficiency,*** the double effect absorption refrigeration system,the evaporation temperature,condensation temperature,and generation pressure affect the COP and exergy *** the generator pressure is unchanged,the COP increases with increasing evaporation temperature and decreasing condensation *** the COP reaches 1.3,the COP slightly decreases as the evaporation temperature or condensation temperature ***,the exergy efficiency of refrigeration systems exhibits the same trend as the COP,and the exergy efficiency maximum value appears at approximately 0.32.A new performance indicator,rPUE,was defined to evaluate the data center power utilization *** flow distribution ratio and heat source temperature were optimized with multi-objective *** the mass flow distribution rate is 0.6 and the heat source temperature is 441.5 K,rPUE and the total unit production costs of the system obtain the optimal solution.

Hypidone hydrochloride(YL-0919)ameliorates functional deficits after traumatic brain injury in mice by activating the sigma-1 receptor for antioxidation

Traumatic brain injury involves complex pathophysiological mechanisms,among which oxidative stress significantly contributes to the occurrence of secondary *** this study,we evaluated hypidone hydrochloride(YL-0919),a self-developed antidepressant with selective sigma-1 receptor agonist properties,and its associated mechanisms and targets in traumatic brain *** experiments to assess functional deficits were followed by assessment of neuronal damage through histological analyses and examination of blood-brain barrier permeability and brain ***,we investigated the antioxidative effects of YL-0919 by assessing the levels of traditional markers of oxidative stress in vivo in mice and in vitro in HT22 ***,the targeted action of YL-0919 was verified by employing a sigma-1 receptor antagonist(BD-1047).Our findings demonstrated that YL-0919 markedly improved deficits in motor function and spatial cognition on day 3 post traumatic brain injury,while also decreasing neuronal mortality and reversing blood-brain barrier disruption and brain ***,YL-0919 effectively combated oxidative stress both in vivo and in *** protective effects of YL-0919 were partially inhibited by *** results indicated that YL-0919 relieved impairments in motor and spatial cognition by restraining oxidative stress,a neuroprotective effect that was partially reversed by the sigma-1 receptor antagonist ***-0919 may have potential as a new treatment for traumatic brain injury.

Enhancing output performance of triboelectric nanogenerator by increasing charge storage capacity of electrodes

ABSTRACT:The practical application of rotating triboelectric nanogenerators is often limited by the wear of high-friction surface materials and low surface charge *** addition to the charge pump replenishment strategy,suppressing charge decay is also crucial for increasing surface charge ***,we present a high performance rotary triboelectric nanogenerator(HPR-TENG)based on a coplanar charge pumping strategy and polyvinyl chloride(PVC)*** has been demonstrated that applying PVC film to the surface of the storage electrode of the main TENG(M-TENG)significantly enhances the M-TENG’s output ***,the HPR-TENG with three layers of PVC film pasted achieved the best output performance,with a peak-topeak output voltage of 2828 V,a peak-to-peak output current of 327μA and a charge transfer of 0.81μC at 500 *** addition,the output improvement effects of different materials are *** TENG with 3 layers of PVC film pasted on it has a maximum output power of 748 mW at a load resistance of 4×10^(6)Ω.HPR-TENG’s output performance remains consistent after 100,000 cycles,which shows excellent *** excellent electrical performance of the HPR-TENG can be used as the energy supply for the tip high-voltage breakdown sensor system,which can achieve 14 breakdowns in 10 *** to its extraordinary electrical performance,HPRTENG can not only serve as an energy supply for cutting-edge high-voltage breakdown sensor systems,but also has the potential to serve as an energy supply for high-pressure sterilization,high-pressure vacuum and water electrolysis.

Synthesis of crystalline γ-Al_2O_3 with high purity

Crystallineγ-AlO(OH)was synthesized by the precipitation of sodium aluminate and oxalic acids in aqueous solution. And thenγ-AlO(OH)was successfully transferred toγ-Al2O3 after subsequent high temperature heat *** effects of reaction conditions on formation ofγ-AlO(OH)andγ-Al2O3 were further investigated in *** XRD analysis shows that the complete formation of crystallineγ-Al2O3 is at pH 8-9,reaction temperature of 93-96℃and calcination temperature of higher than 400℃.The product ofγ-Al2O3 contains impurity,including iron,calcium and silicon ion with a low content of about 0.01%and has large specific surface area and high pore volume of 269.9 m2/g and 0.57 mL/g,which can be applied in catalysts and catalyst supports.

High strength and high ductility Mg-Gd-Y-Zn-Zr alloys obtained by controlling texture and dynamic precipitation through LPSO phase structure of different initial morphologies

This paper investigated the effects of three different initial phase structures on the microstructure evolution and tensile properties of extruded Mg-Gd-Y-Zn-Zr alloy, using an extrusion ratio of 3.6. These three initial phase structures were obtained by heat treatment, which is narrow spacing long-period stacking ordered phase(LPSO) phase structure alloy(EN alloy), wide spacing LPSO phase structure alloy(EW alloy), and narrow spacing LPSO phase and β phase overlapping phase structure alloy(EO alloy). The dynamic recrystallization(DRX) and dynamic precipitation behavior of extruded alloys with different initial structures, as well as their strengthening and ductility mechanisms were studied in detail. After hot extrusion with a low extrusion ratio, the alloy exhibits a bimodal structure composed of undynamic recrystallization(UN-DRX) grains and dynamic recrystallization(DRX) grains with strong textures. The narrow-spacing LPSO phase structure inhibits DRX and dynamic precipitation, while both the wide-spacing LPSO phase structure and the overlapping phase structure alloys promote DRX and dynamic precipitation. The strength improvement is mainly due to the strong texture and internal dislocation pinning of the undynamic recrystallization zone(UN-DRX) and the high strengthening effect of the narrow spacing LPSO phase. Although the promotion of DRX improves grain boundary strengthening effect, it cannot make up for reducing the UN-DRXed grain strengthening effect. A lower volume fraction of β dynamic precipitation phase is beneficial for improving the ductility of the alloy.

Preparation and photoluminescence enhancement of Li^+ and Eu^(3+) co-doped YPO_4 hollow microspheres

Li＋ and Eu3＋ co-doped YPO4 hollow microspheres were successfully synthesized by a sacrificial template method using polystyrene （PS） as template. Techniques of X-ray diffraction （XRD）, scanning electron microscopy （SEM）, as well as transmission electron microscopy （TEM） were employed to characterize the as-synthesized sample. Furthermore, the photoluminescence （PL） characterization of the Li＋ and Eu3＋ co-doped YPO4 microsphere was carried out and the effects of the doping concentration of Li＋ and Eu3＋ active center concentration as well as calcination temperature on the PL properties were studied in detail. The results showed that the incorporation of Li＋ ions into the YPOa：Eu3＋ lattice could induce a remarkable improvement of the PL intensity. The highest emission intensity was observed with the compound of 5%Li＋ and 5%Eu3＋ co-doped YPO4, whose brightness was increased by a factor of more than 2.2 in comparison with that of the YPO4：5%Eu3＋.

Oxidation Resistance of Form-stable High-temperature Phase Change Thermal Energy Storage Materials Doped by Impregnated Graphite

We adopted the solution impregnation route with aluminum dihydrogen phosphate solution as liquid medium for effective surface modification on graphite *** mass ratio of graphite to Al（H2PO4）3changed from 0.5:1 to 4:1,and the impregnation time changed from 1 to *** typical composite phase change thermal storage materials doped with the as-treated graphite were fabricated using form-stable *** investigate the oxidation and anti-oxidation behavior of the impregnated graphite at high temperatures,the samples were put into a muffle furnace for a cyclic heat *** on SEM,EDS,DSC techniques,analyses on the impregnated technique suggested an optimized processing conditions of a 3 h impregnation time with the ratio of graphite:Al（H2PO4）3as 1:3 for graphite impregnation *** investigations on high-temperature phase change heat storage materials doped by the treated graphite suggested excellent oxidation resistance and thermal cycling performance.
