Molecular and immunohistochemical study of the inactivation of the p16 gene in primary hepatocellular carcinoma

Obejctive To determine whether p16 gene is involved in the genesis of primary hepatocellular carcinoma (HCC) Methods Twenty five HCC tumor samples with corresponding non tumor liver tissue specimens were examined for p16 gene alterations The identification of deletion of exon 1 and exon 2 in p16 gene was performed using comparative multiplex polymerase chain reaction (PCR) analysis The point mutation of exon 2 in p16 gene was investigated by single strand conformational polymorphism (SSCP) analysis, and the status of p16 gene methylation was screened using a PCR based methylation analysis 35 parafin embedded specimens of HCC with corresponding non tumor liver tissues, including the 25 cases described above for screening p16 gene alterations, were investigated for p16 protein expression using immunohistochemical analysis Results Among 25 cases, 2 homozygous deletions and 1 hemizygous deletion were found in HCC samples No point mutation was identified in the remaining 22 tumor samples without p16 gene deletions Hypermethylation was detected in 24% (6/25) of tumor samples However, the corresponding non tumor liver tissue specimens were always unmethylated at the p16 locus Loss of p16 protein expression occurred in 16 of 35 (45 7%) tumor samples, and all the non tumor liver tissue specimens showed positive p16 staining For the 25 cases examined for p16 gene alterations, the loss of p16 protein expression was observed in all tumors with p16 gene alterations and also in 3 tumors without p16 gene alterations Conclusion Inactivation of the p16 gene may play an important role in the genesis of primary hepatocellular carcinoma

Intimate partner violence(IPV)is simultaneously assumed as a serious crime and a major public health issue,having recurrences as one of its main characteristics and,consequently,re-entries of some alleged offenders in the criminal justice system(CJS).The main goal of this study is to assess if in cases of female victims of IPV,violence decreases after the first entry of the alleged offender in the CJS.A retrospective study was performed based on the analysis of police reports of alleged cases of IPV during a 4-year *** final sample(n=1488)was divided into two groups according to the number of entries in the CJS(single or multiple)followed by a comparative *** suggest that violence decreases after the first entry of alleged offenders in the ***-entries were found in only 15.5%of the cases but they were accountable for 3.3 times more crimes on ***,victims of recidivism presented more injuries and required more medical ***,a small group of alleged offenders seems to be more violent and accountable for most of the IPV crimes registered in the CJS suggesting that regardless of legal sanctions aiming to deter violence,these measures may not be enough for a certain group of *** study sustains the need for a predictive model to quantify the risk of repeated IPV cases within the Portuguese population.

The necrotrophic fungus Rhizoctonia cerealis is the causal agent of devastating diseases of cereal crops including wheat(Triticum aestivum).We present a high-quality genome assembly of *** Rc207,a virulent strain causing wheat sharp *** assembly(56.36 Mb)is composed of 17.87%repeat sequences and 14,433 predicted protein-encoding *** Rc207 genome encodes a large and diverse set of genes involved in pathogenicity,especially rich in those encoding secreted proteins,carbohydrateactive enzymes(CAZymes),peptidases,nucleases,cytochrome P450,and secondary metabolismassociated *** secretory protein-encoding genes,including CAZymes,peroxygenases,dehydrogenases,and cytochrome P450,were up-regulated during fungal infection of *** identified 831 candidate secretory effectors and validated the functions of 10 up-regulated candidate effector *** them,nine were confirmed as necrotrophic pathogen’s effectors promoting fungal *** potential mobile or plastic genomic regions rich in repeat sequences suggest their roles in fungal adaption and virulence-associated genomic *** study provides valuable resources for further comparative and functional genomics on important fungal pathogens,and provides essential tools for development of effective disease control strategies.

Genes encoding early signaling events in pathogen defense often are identified only by their phenotype. Such genes involved in barley-powdery mildew interactions include Mla, specifying race-specific resistance; Rarl （Required for Mla12-specified resistance1）, and Roml （Restoration of Mla-specified resistancel）. The HSP90-SGT1-RAR1 complex appears to function as chaperone in MLA-specified resistance, however, much remains to be discovered regarding the precise signaling underlying plant immunity. Genetic analyses of fast-neutron mutants derived from CI 16151 （Mla6） uncovered a novel locus, designated Rar3 （R_equired for Mla6-specified resitance3）. Rar3 segregates independent of Mla6 and Rarl, and rar3 mutants are susceptible to Blumeria graminis f. sp. hordei （Bgh） isolate 5874 （A VRar）, whereas, wild-type progenitor plants are resistant. comparative expression analyses of the rar3 mutant vs. its wild-type progenitor were conducted via Barleyl GeneChip and GAIIx paired-end RNA-Seq. Whereas Rarl affects transcription of relatively few genes; Rar3 appears to influence thousands, notably in genes controlling ATP binding, catalytic activity, transcription, and phosphorylation; possibly membrane bound or in the nucleus, eQTL analysis of a segregating doubled haploid population identified over two-thousand genes as being regulated by Mla （q value/FDR=0.00001）, a subset of which are significant in Rar3 interactions. The intersection of datasets derived from mla-loss-of-function mutants, Mla-associated eQTL, and rar3-mediated transcriptome reprogramming are narrowing the focus on essential genes required for Mla-specified immunity.

DNA Copy Number Aberrations in Breast Cancer by Array comparative Genomic Hybridization

Array comparative genomic hybridization (CGH) has been popularly used for analyzing DNA copy number variations in diseases like cancer. In this study, we investigated 82 sporadic samples from 49 breast cancer patients using 1-Mb resolution bacterial artificial chromosome CGH arrays. A number of highly frequent genomic aberrations were discovered, which may act as "drivers" of tumor progression. Meanwhile, the genomic profiles of four "normal" breast tissue samples taken at least 2 cm away from the primary tumor sites were also found to have some genomic aberrations that recurred with high frequency in the primary tumors, which may have important implications for clinical therapy. Additionally, we performed class comparison and class prediction for various clinicopathological parameters, and a list of characteristic genomic aberrations associated with different clinicopathological phenotypes was compiled. Our study provides clues for further investigations of the underlying mechanisms of breast carcinogenesis.

This study aimed to determine the medicinal significance of traditionally used Aloe plants by testing their crude extracts and Anthraquinone fractionates for antimicrobial activity against standard stock organisms. The significance of this study is to promote the discovery of new alternative to drugs currently being used. The gel and leaf cover of these four plants were extracted using cold ethanol extraction method and Anthraquinones were fractionated from the leaf cover using the liquid-liquid extraction technique. The extracts obtained were then screened in different concentrations for their antimicrobial activity against stock organisms Staphylococcus aureus (ATCC1 6538) and Escherchia coli (ATCC 25922) to access the activity of the aloe species against gram positive and gram negative bacteria. It was determined that the crude extracts of the gel and leaf part of the four aloe species had antimicrobial activity against gram positive (Staphylococcus aureus) and gram negative (Escherchia coli) bacteria and Escherchia coli showed the least sensitivity towards the four plant extracts while Staphylococcus aureus was highly susceptible.

Biocontrol Endotherapy with Trichoderma spp. and Bacillus amyloliquefaciens against Phytophthora spp.： A comparative Study with Phosphite Treatment on Quercus robur and Fagus sylvatica

Phytophthora species are particularly aggressive plant pathogens and are often associated with the decline of many tree species, including oak and beech. Several fungi and bacteria species are known as potential antagonists usable as biological control agents. Phosphonate （H3PO3）, commonly branded as phosphite, has also been used in the past years to protect trees against invasive Phytophthora spp.. This study aimed at comparing the effects of selected antagonist microorganisms and phosphonate, when applied by microinjection or leaf treatment. Antagonistic species were first selected for their high inhibitory activity against problematic Phytophthora species, such as Phytophthora cactorum, P. quercina and P. plurivora attacking Quercus robur and Fagus sylvatica in Polish forests. Three endophytic species Trichoderma atroviride （two strains）, T. harzianum and Bacillus amyloliquefaciens showed a high control activity, and their efficacy was then assessed in comparison with a phosphonate treatment. Two application methods were experimented in this study： injection of a solution of spores or phosphonate into the sap vessels of beech or a foliar treatment on oak. Phosphonate and two strains of Trichoderma significantly reduced the necrotic area on oak leaves inoculated with P. plurivora and one strain of T. atroviride significantly reduced necrotic areas on beech branches. These results are therefore promising of a novel way to control Phytophthora spp. in forest stands and nurseries.
