Development of T. aestivum ***. californicum Alien Chromosome Lines and Assignment of Homoeologous Groups of Hordeum californicum Chromosomes

Hordeum californicum （2n = 2x = 14, HH） is resistant to several wheat diseases and tolerant to lower nitrogen. In this study, a molecular karyotype of H. californicum chromosomes in the Triticum aestivum L. cv. Chinese Spring （CS）-H. californicum amphidiploid （2n = 6x = 56, AABBDDHH） was established. By genomic in situ hybridization （GISH） and multicolor fluorescent in situ hybridization （FISH） using repetitive DNA clones （pTa71, pTa794 and pSc119.2） as probes, the H. californicum chromosomes could be differentiated from each other and from the wheat chromosomes unequivocally. Based on molecular karyotype and marker analyses, 12 wheat--alien chromosome lines, including four disomic addition lines （DAH1, DAH3, DAH5 and DAH6）, five telosomic addition lines （MtH7L, MtHIS, MtH1L, DtH6S and DtH6L）, one multiple addition line involving H. californicum chromosome H2, one disomic substitution line （DSH4） and one translocation line （TH7S/1BL）, were identified from the progenies derived from the crosses of CS-H. californicum amphidiploid with common wheat varieties. A total of 482 EST （expressed sequence tag） or SSR （simple sequence repeat） markers specific for individual H. californicum chromosomes were identified, and 47, 50, 45, 49, 21, 51 and 40 markers were assigned to chromosomes H1, H2, H3, H4, H5, H6 and H7, respectively. According to the chromosome allocation of these markers, chromosomes H2, H3, H4, H5, and H7 of H. californicum have relationship with wheat homoeologous groups 5, 2, 6, 3, and 1, and hence could be designated as 5Hc, 2He, 6Hc, 3Hc and 1Hc, respectively. The chromosomes H1 and H6 were designated as 7Hc and 4Hc, respectively, by referring to SSR markers located on rye chromosomes.

Twenty vegetable amaranth genotypes were evaluated for total antioxidant capacity, antioxidant leaf pigments, vitamins, and selection of suitable genotypes for extraction of juice in a randomized complete block design（RCBD） with three replications. Vegetable amaranth was rich in chlorophyll, β-cyanins, β-xanthins, betalains, carotene, ascorbic acid and total antioxidant. The genotypes VA14, VA16, VA18, VA15, and VA20 could be selected as amaranth vegtable varieties with high yields and abundance antioxidant leaf pigments and vitamins to produce juice. The genotypes VA13 and VA19 had above-average foliage yield and high antioxidant profiles while the genotypes VA2, VA3, VA9, VA11, VA12, and VA17 had a high antioxidant profiles and below-average foliage yield. These genotypes could be used as a donor parent for integration of potential high antioxidant profiles genes into other genotypes. The correlation study revealed a strong positive association among all the antioxidant leaf pigments, total antioxidant capacity and foliage yield. Selection based on total antioxidant capacity, antioxidant leaf pigments could economically viable to improve the yield potential of vegetable amaranth genotypes. Total carotene and ascorbic acid exhibited insignificant genotypic correlation with all the traits except total antioxidant capacity. This indicates that selection for antioxidant vitamins might be possible without compromising yield loss.

Metastasis is the main cause of cancer-associated mortality. During this complicated process, some cancer cells, also called circulating tumor cells （CTCs）, detach from primary sites, enter bloodstream and extravasate at metastatic site. Thrombocytosis is frequently observed in patients with metastatic cancers suggesting the important role of platelets in metastasis. Therefore this review focuses on how platelets facilitate the generation of CTCs, protect them from various host attacks, such as immune assaults, apoptosis and shear stress, and regulate CTCs intravasation/extravasation. Platelet-derived cytokines and receptors are involved in this cascade. Identification the mechanisms underlie platelet-CTCs interactions could lead to the development of new platelet-targeted therapeutic strategy to reduce metastasis.

The blunt snout bream(Megalobrama amblycephala)is a major aquaculture species in the Chinese freshwater polyculture *** this study,complete diallel crossing of three strains(i.e.,Liangzi[LZ],Poyang[PY]and Yuni[YN])was used to evaluate the combining ability and heterosis effect of intraspecific crossbreeding on the growth performance(i.e.,body weight,total length,body length,body height,and Fulton’s coefficient of condition).The offspring produced from the three strains were reared in a communal pond for 20 months,and nine microsatellites were used to assess their *** the749 offspring,708(94.53%)could be assigned directly to a single parental *** differences were observed in body weight(BW)among the different combinations,indicating that the combining ability of BW from each strain was significantly *** general combining ability(GCA)of BW from the sire was much higher than that from the *** GCA for the dam and sire of the LZ and YN were the largest and the smallest,*** special combining ability of YN$9 PY#was the largest,and YN$9 PY#showed significantly positive heterosis effects on all F1growth traits,except for Fulton’s condition coefficient(K)(P\0.05).In addition,significant positive linear correlations were found between the mean BW of corresponding progeny and the H E of the parental generation(y=600.7x-29.472,r=0.8651,P=0.0026),H O(y=1206.9x-518.14,r=0.7436,P=0.0216),and the polymorphism information content(y=1021.3x-331.31,r=0.8245,P=0.0063)as determined by a correlative *** findings obtained from the study will be important for future considerations of *** stock improvement programs.

The aquaculture of yellowtail kingfish(Seriola lalandi)has expanded around the globe,including *** resource of Chinese *** urgently needs to be assessed for improving ***,we collected wild *** samples from the Bohai Sea,China and evaluated its genetic diversity based on 17,690 nuclear loci.A population from the Southern Ocean,Australia was used for *** analyses showed that the Chinese and Australian *** formed two completely distinct clusters and there was no genetic introgression from the Australian *** into the Chinese *** *** genetic diversity is slightly lower,but comparable in Chinese versus Australian yellowtail kingfish(Chinese population,expected heterozygosity:0.19,observed heterozygosity:0.19,nucleotide diversity:0.19±0.09;Australian population,expected heterozygosity:0.23,observed heterozygosity:0.22,nucleotide diversity:0.22±0.11).Overall,our results indicated that Chinese *** could be a potential subject for genetic breeding *** also investigated morphological characters and developed molecular markers for population *** of meristic characters between the Chinese population and the Australian population revealed that main shape difference were in the number of dorsal spines,dorsal fin rays and the number of upper gill *** morphology based on eight landmarks also revealed significant difference between the two populations including the distance between the tip of snout to origin of pelvic fin and the distance between the tip of snout to origin of pectoral *** morphological characters can be used for easy identification of the Chinese ***.

Competitive Expression of Endogenous Wheat CENH3 May Lead to Suppression of Alien ZmCENH3 in Transgenic Wheat × Maize Hybrids

Uniparental chromosome elimination in wheat × maize hybrid embryos is widely used in double haploid production of wheat. Several explanations have been proposed for this phenomenon, one of which is that the lack of cross-species CENH3 incorporation may act as a barrier to interspecies hybridization. However, it is unknown if this mechanism applies universally. To study the role of CENH3 in maize chromosome elimination of wheat x maize hybrid embryos,？cos, maize ZmCENH3 and wheat aTaCENH3-B driven by the constitutive CaMV35S promoter were transformed into wheat variety Yangmai 158. Five transgenic lines for ZmCENH3 and six transgenic lines for ctTaCENH3-B were identified. RT-PCP analysis showed that the transgene could be transcribed at a low level in all ZmCENH3 transgenic lines, whereas transcription of endogenous wheat CENH3 was significantly up-regulated. Interestingly, the expression levels of both wheat CENH3 and ZmCENH3 in the ZmCENH3 transgenic wheat × maize hybrid embryos were higher than those in the non-transformed Yangmai 158 × maize hybrid embryos. This indicates that the alien ZmCENH3 in wheat may induce competitive expression of endogenous wheat CENH3, leading to suppression of ZmCENH3 over-expression. Eliminations of maize chromosomes in hybrid embryos of ZmCENH3 transgenic wheat ×maize and Yangmai 158 x maize were compared by observations on micronuelei presence, by marker analysis using maize SSRs （simple sequence repeats）, and by FISH （fluorescence in situ hybridization） using 45S rDNA as a probe. The results indicate that maize chromosome elimination events in the two crosses are not sigmficantly different. Fusion protein ZmCENH3- YFP could not be detected in ZmCENH3 transgenic wheat by either Western blotting or immnunostaining, whereas accumulation and loading of the αTaCENH3-B-GFP fusion protein was normal in aTaCENH3-B transgenic lines.
