Development of a stable attenuated double-mutant of tobacco mosaic virus for cross-protection

Tobacco(Nicotiana tabacum)and tomato(Solanum lycopersicum)are two major economic crops in *** mosaic virus(TMV;genus Tobamovirus)is the most prevalent virus infecting both ***,some widely cultivated tobacco and tomato cultivars are susceptible to TMV and there is no effective strategy to control this ***-protection can be a safe and environmentally friendly strategy to prevent viral ***,stable attenuated TMV mutants are *** this study,we found that the substitutions in the replicase p126,arginine at position 196(R^(196))with aspartic acid(D),glutamic acid at position 614(E^(614))with glycine(G),serine at position 643(S^(643))with phenylalanine(F),or D at position 730(D^(730))with S,significantly reduced the virulence and replication of ***,only the mutation of S^(643) to F reduced the RNA silencing suppression activity of TMV p126.A double-mutant TMV-E614G-S643F induced no visible symptom and was genetically stable through six successive passages in tobacco ***,our results showed that TMV-E614G-S643F double-mutant could provide effective protection against the wild-type TMV infection in tobacco and tomato *** study reports a promising mild mutant for cross-protection to control TMV in tobacco and tomato plants.

Timing effect of high temperature exposure on the plasticity of internode and plant architecture in maize

The occurrence of high temperature(HT)in crop production is becoming more frequent and unpredictable with global warming,severely threatening food *** state of an organ’s growth and development is largely determined by the temperature conditions it is exposed to over *** is the main cereal crop,and its stem growth and plant architecture are closely related to lodging resistance,and especially sensitive to ***,systematic research on the timing effect of HT on the sequentially developing internode and stem is currently *** identify the timing effect of HT on the morphology and plasticity of the stem in maize,two hybrids(Zhengdan 958(ZD958),Xianyu 335(XY335))characterized by distinct morphological traits in the stem were exposed to a 7-day HT treatment from the V6 to V17 stages(Vn presents the vegetative stage with n leaves fully expanded)in *** results demonstrated that exposure to HT during V6-V12 accelerated the rapid elongation of *** instance,HT occurring at V7 and V12 specifically promoted the lengths and weights of the 3rd-5th and 9th-11th internodes,***,HT slowed the growth of internodes adjacent to the promoted ***,compared with control,the plant height was significantly increased soon after HT treatment,but the promotion effect became narrower at the subsequent flowering stage,demonstrating a self-adjusting mechanism in the maize plant in response to ***,HT altered the plant architectures,including a rising of the ear position and increase in the ear position ***335 exhibited greater sensitivity in stem development than ZD958 under HT *** findings improve our systematic understanding of the plasticity of internode and plant architecture in response to the timing of HT exposure.

Evolution of Cenozoic sedimentary architecture in Central and Southern South China Sea basins

The Central and Southern South China Sea(CSSCS) has a complex tectonic dynamic background and abundant oil and gas resources, which has always been a hot topic of academic and industrial ***, systematic analyses are still lacking regarding its sediment filling structure and evolution, mostly due to limited borehole penetration and poor quality of seismic reflection data for deeply buried sequences. No consensus has been reached yet on the sedimentary infilling processes, which impeded the reconstruction of the palaeogeography of Southeast Asia and the oil-and-gas exploration undertakings. Here, we illustrate the Cenozoic sedimentary evolution of the CSSCS region by synthesizing relevant data from previous literature and our own observations and displaying the evolution of depositional systems in sequential reconstructions. Besides, the controlling factors of preferred sedimentary scenarios in the CSSCS incorporate the latest interpretations of the spreading of South China Sea(SCS) as well as the demise of the hypothetical Proto-South China Sea(PSCS). The results show that there are three types of sedimentary basins in the CSSCS(foreland,strike-slip, and rift basins) with different sedimentary filling structures. The foreland basins formed a depositional pattern of ‘transition from deep water to shallow water environments', dominated by deep-water depositional systems which were formed before the Early Oligocene with submarine fans developed. Later,the foreland basins were gradually dominated by shallow-water depositional systems with deltas and shallow marine facies. The strike-slip basins showed the depositional architecture of ‘transition from lake to marine environments', i.e. the basins were dominated by lacustrine deposits during the Eocene and evolved into the marine depositional environment since Oligocene with delta developed in the western part of the basin. The nuclear event risk(NER)is an important and disputed factor that should be reasonably considered when planning the pathway of nuclear power development(NPD)to assess the benefits and risks of developing nuclear power more *** paper aims to explore the impact of nuclear events on NPD pathway *** influence of nuclear events is quantified as a monetary risk component,and an optimization model that incorporates the NER in the objective function is *** optimize the pathway of NPD in the lowcarbon transition course of power supply structure evolution,a simulation model is built to deduce alternative NPD pathways and corresponding power supply evolution scenarios under the constraint of an exogenously assigned carbon emission pathway(CEP);moreover,a method is proposed to describe the CEP by superimposing the maximum carbon emission space and each carbon emission reduction(CER)component,and various CER components are clustered considering the emission reduction characteristics and resource endowments of different power generation technologies.A case study is conducted to explore the impact of NER and its risk valuation uncertainty on NPD pathway *** method presented in this paper allows the impact of nuclear events on NPD pathway planning to be quantified and improves the level of coordinated optimization of benefits and risks.

Improving IEC thermal model for oil natural air natural transformers using optimised parameters based on dynamic simulation

Accurate assessment of hot-spot temperature is essential for the safe operation of power *** dynamic thermal models cannot estimate hot-spot temperature accurately since some input parameters are roughly determined by transformer capacity and cooling mode while ignoring the effect of winding structure,tank dimensions,and material physical *** improve the accuracy of temperature assessment,empirical parameters of the IEC thermal model including thermal constants,winding and oil exponents are optimised with the help of numerical simulation in this *** on energy conservation and heat transfer theory,a computational fluid dynamic(CFD)model of a transformer in oil natural air natural(ONAN)cooling mode is *** CFD model simulates the entity's structure,sizes,and multi-stage heat dissipation processes realistically,so it can more precisely calculate the dynamic hot-spot *** to the simulated temperature curves at different operating conditions,the thermal constants and oil exponent are estimated using non-linear regression,and the winding exponent is optimised using linear regression.A case study is conducted on an ONAN *** shows the improved IEC model with optimised parameters can more accurately evaluate hot-spot temperature,and the absolute error is decreased by 2.4 K(38.7%)compared with traditional thermal models.

In this paper,an intelligent control method applying on numerical virtual flight is *** proposed algorithm is verified and evaluated by combining with the case of the basic finner projectile model and shows a good application ***,a numerical virtual flight simulation model based on overlapping dynamic mesh technology is *** order to verify the accuracy of the dynamic grid technology and the calculation of unsteady flow,a numerical simulation of the basic finner projectile without control is carried *** simulation results are in good agreement with the experiment data which shows that the algorithm used in this paper can also be used in the design and evaluation of the intelligent controller in the numerical virtual flight ***,combined with the real-time control requirements of aerodynamic,attitude and displacement parameters of the projectile during the flight process,the numerical simulations of the basic finner projectile’s pitch channel are carried out under the traditional PID(Proportional-Integral-Derivative)control strategy and the intelligent PID control strategy *** intelligent PID controller based on BP(Back Propagation)neural network can realize online learning and self-optimization of control parameters according to the acquired real-time flight *** with the traditional PID controller,the concerned control variable overshoot,rise time,transition time and steady state error and other performance indicators have been greatly improved,and the higher the learning efficiency or the inertia coefficient,the faster the system,the larger the overshoot,and the smaller the stability *** intelligent control method applying on numerical virtual flight is capable of solving the complicated unsteady motion and flow with the intelligent PID control strategy and has a strong promotion to engineering application.
