Dynamic Changes of Endothelium Derived Factors and Cyclic Nucleotides in Ascites Broilers and Control of L-arginine

A flock of AA breed chickens were reared in peterstme brood-vait chamber using high energy pelleted feed, at 14 days of age, 400 birds were separated into 3 groups randomly as follows:100 birds were exposed to normal ambient temperature (20℃) as control group, 150 birds were exposed to low ambient temperature (11℃) in order to induce ascites as treatment I group, another 100 birds were also exposed to low ambient temperature (11℃) and fed the diets containing 1% L-arginine for ascitic prophylactic treatment as treatment I group. The blood samples were collected on 3, 4, 5, 6, 7 wk, respectively, to measure the contents of plasma endothelin (ET-1), angiotensin Ⅱ(Ang Ⅱ) and cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP). The results indicated that the contents of cAMP, cGMP, AngⅡ and the ratio of cAMP/cGMP in treatment I and ascitic broilers were higher than that of correspondent control group(P<0.01, P<0.05), ET-1 of preascitic broilers were higher than that of control group(P<0. 05), while there were insignificant differences with later ascitic broilers, the contents of cAMP and cGMP in treatment Ⅱ were higher than treatment I and control group(P<0.01, P<0.05), whereas, the ratio of cAMP/ cGMP and the contents of Ang Ⅱ were gradually decreased than that of control group (P<0. 05), the contents of ET-1 were not changed. By further analysis, the increased plasma Ang I at low ambient temperature condition in broilers made endothelium cell secretion of increased ET-1, cAMP, cGMP and decreased NO. So low temperature accelarated ascites syndrome in broilers. Supplemented L-arginine can decrease ET-1, cAMP and cAMP/cGMP. It is concluded that cAMP mediated pulmonary hypertension syndrome in broilers.

Regulation of mouse blastocyst adhesion, outgrowth and secretion of matrix metalloproteinase-2 by cGMP and nitric oxide in vitro

Nitric oxide (NO) is a multifunctional messen-ger molecule produced through oxidation of L-arginine to L-citrulline by enzyme NO synthase (NOS). In the current study, mouse blastocysts were cultured in the different media, and the implantation capacity of blastocyst was evaluated by evaluating the percentage of embryos adhesion and out-growth after culture for 12, 24 or 48 h. Matrix metallopro-teinase-2 (MMP-2) mRNA was detected by RT-PCR, and MMP-2 protein was detected by gelatin zymography. Inhibi-tion of blastocyst adhesion and outgrowth was observed in embryo cultured with 500 mmol/L NOS inhibitor NG-mono- methyl-L-arginine (L-NMMA) alone; however, 100 mmol/L S-nitroso-Nacetylpenicillamine (SNAP), a NO donor, and 20 mmol/L cGMP analogue, 8-Br-cGMP could block this inhibi-tion. The expression and production of MMP-2 in the blas-tocysts were suppressed by L-NMMA, and SNAP or 8-br-cGMP could reverse this suppression. These results suggest that NO induces embryo implantation by cGMP sig-naling pathway.

Studies on Preparation and Characteristics of Monoclonal Antibodies Against Enrofloxacin and Cross-Reactivity of Related Fluoroquinolones

An ester activation method was employed to couple enrofloxacin(ENFX) to the carrier proteins BSA and OVA. The conjugates ENFX-BSA and ENFX-OVA were identified with an UV spectrophotometer and amino acid automation analysis instrument, and resulted in conjugates with 48 ENFX molecules per carrier molecule(BSA). Splenocytes from mice immunized with ENFX-BSA were fused with SP2/0 myeloma cells and hybridomas secreting antibodies against enrofloxacin were selected and cloned. Two stable monoclonal antibodies, 2C5, 5D5 of the subclass IgG2a, were isolated. Using antibody 5D5, an indirect competitive inhibition enzyme-linked immunosorbent assay (Ci-ELISA) was developed for the quantitative detection of enrofloxacin and its metabolites. The IC50 of the standard curve was 21.67 ng mL-1 and the limit of detection for enrofloxacin was 0.13 ng mL-1. This method was sensitive and had a linear range from 0.13 to 10 000 ng mL-1 (r= -0.9782). Monoclonal antibody 5D5 exhibited high relative affinity to enrofloxacin, and the cross-reactivities with ciprofloxacin, marbofloxacin, sarafloxacin and danorfloxacin were 110.8, 27.40, 71.05 and 37.41%, respectively. Three non-fluoroquinolones of cefadroxil, chloramphenicol, sulfadimethoxine were tested and there was no cross-reaction between them.

Multiplex PCR Detection of Alleles Responsible for Benzimidazole- Susceptibility or -Resistance in Natural Populations of Haemonchus contortus

A multiplex PCR was developed to detect benzimidazole-resistance (BZ-R) or -susceptibility (BZ-S) in Haemonchus contortus by amplification with 4 primers of a sequence of the GRU-1 gene of β-tubulin of H. contortus making use of sequence information available in Genbank. The method was based on two allele-non-specific primers and two allele- specific primers. F1 (264 bp) and F3 (799 bp) should be produced in BZ-R, F2 (585 bp) and F3 in BZ-S. With this method, we demonstrated that H. contortus BZ-R strain from Australia showed F1 and F3, and the worm BZ-S strain from Shanghai did F2 and F3. Sequence analysis of the isotype 1 gene of β-tubulin of BZ-R from Australia and BZ-S from Shanghai showed the code in residue 200 of the gene was respectively TAC and TTC. The LD50 of albendazole of the Australian BZ- R strain was 0.54 μg mL-1, the Shanghai BZ-S strain was only 0.0023 μg mL-1 by EHA (egg hatch assay). The multiplex PCR could determinate the genotype of single adult worm or several third stage larvae and was performed on at least 50 ng of genomic DNA. BZ-R H. contortus were not detected in Shihezi and Yining of the Xinjiang, Wuhe of the Anhui Province, Nanjing and Xuzhou of the Jiangsu Province. The LD50 of the H. contortus from these locations to albendazole as determined by EHA varied between 0.0023-0.0032 μg mL-1. The result indicated that the multiplex PCR could be used to differentiate BZ-R and BZ-S of H. contortus and that the BZ-R situation of H. contortus was not serious in China.

Alterations of mast cells and TGF-β1 on the silymarin treatment for CCl_4-induced hepatic fibrosis

AIM: Silymarin is a potent antioxidant, antiinflammatory and anti-fibrogenic agent in the liver, which is mediated by alteration of hepatic Kupffer cell function, lipid peroxidation, and collagen production. Especially, in hepatic fibrogenesis, mast cells are expressed in chronic inflammatory conditions, and promote fibroblast growth and stimulate production of the extracellular matrix by hepatic stellate cells. METHODS: We examined the inhibitory mechanism of silymarin on CCI4-induced hepatic cirrhosis in rats. At 4, 8, and 12 wk, liver tissues were examined histopathologically for fibrotic changes produced by silymarin treatment. RESULTS: In the silymarin with CCU-treated group, increase of hepatic stellate cells and TGF-β1 production were lower than in the CCI4-treated group at early stages. Additionally, at the late fibrogenic stage, expressions of TGF-β1 were weaker and especially not expressed in hepatocytes located in peripheral areas. Moreover, the number of mast cell in portal areas gradually increased and was dependent on the fibrogenic stage, but those of CCI4+silymarin-treated group decreased significantly. CONCLUSION: Anti-fibrotic and antiinflammatory effects of silymarin were associated with activation of hepatic stellate cells through the expression of TGF-β1 and stabilization of mast cells. These results suggest that silymarin prevent hepatic fibrosis through suppression of inflammation and hypoxia in the hepatic fibrogenesis.

PCR Amplication and Sequencing of ITS-2 rDNA of Leucocytozoon caulleryi

The second internal transcribed spacer (ITS-2) of Leucocytozoon caulleryi was amplified by PCR using a pair of conserved primers and cloned into the T-T windows of plasmid pGEM-T easy vector. The inserts were successfully sequenced and the results revealed that the ITS-2 plus flanking sequence (ITS2+) was composed of 270 nucleotides. and the ITS-2 was 113 bp in length. The ITS-2 of L. caulleryi was analysed by NCBI Blast, and the degree of homology of the ITS-2 of L. caulleryi was compared with that of Eimeria tenella , Candida tropicalis and Saccharomyces kluyveri and others by wDNASIS. The results showed that the ITS-2 of L. caulleryi is characteristic and has the greatest similarity with E. tenella (21.2%).

Changes of the Levels of Blood NO and TNF-αConcentrations in Water Buffaloes and Goats Infected with Fasciola hepatica

Changes of concentrations were studied in water buffaloes and goats infected with Fasciola he-patica on blood NO(nitric oxide) and TNF-α(tumor necrosis factor-α). Twenty healthy male castrated water buffaloes of 2 - 3 years old and weighing 300 - 500 kg as well as six goats were confirmed free of fasciolosis by fecal examination and Dot-ELISA. Two studies were conducted using the water buffaloes. In the first experiment, 8 water buffaloes were randomly divided into control group (n = 3) and infection group (n = 5). Each buffalo in the infected group received orally 60 metacercariae of F. hepatica per day for 20 days (total 1 200 metacercariae) to produce a chronic infection. In the second experiment, 12 water buffaloes were randomly divided into infected (n=9) and control group (n = 3). Each buffalo in the infected group was given a single oral dose of 1 600 metacercariae to produce an acute infection. The 6 goats were randomly divided into two infected groups and a control group. The sheep in two infections received a single oral dose of 200 and 500 metacercariae respectively, the control group remained uninfected. Blood NO and TNF-a concentrations of the test animals were measured by a reductive enzyme assay and RIA, respectively. Blood NO concentration in both acutely and chronically infected water buffaloes progressively increased from week 3 post-infection and was significantly greater than that of the control group (P<0. 05) at the 5th week (acute infection) and 7th week (chronic infection), and remaining at higher concentration for the remaining period of the studies. Blood TNF-αconcentrations in both chronically and acutely infected water buffaloes also increased after infection. In the goat experiment, plasma NO concentrations in both infection groups increased from week 3 after infection, and remained higher than that of the control group until the end of the experiment. The Effects of Exogenous Nitric Oxide on Eimeria tenella Oocysts

The effects of exogenous NO on Eimeria tenella oocysts were studied. The E. tenella oocysts freshly isolated from feces of experimental chickens infected with E. tenella and sporulated E. tenella oocysts were treated in vitro by the well-known NO-donors, SNP, SPER/NO and GSNO. The results showed that among the 3 NO donors, only GSNO had significant effects on the percentage of sporulated oocysts, the inhibiting rate of purified oocysts was from 93 to 97%, and reached 100% on the oocysts in the feces of chickens with E. tenella infection. GSNO also significantly decreased the virulence of E. tenella oocysts, and the oocysts treated by 2 - 8 mmol L-1 GSNO almost lost their pathogenicity to chickens.

Study on Changes of Plasma Endothelin-1 in Sheep with Experimental Colonic Obstruction

The present study was designed to determine the relationship between changes of plasma endothelin 1 concentrations and development of colonic obstruction in *** were bound in three sheep with sterilized plastic tubes to make pathological model of colonic *** were collected before and after modeling operation on both controls and *** 1 concentrations were measured by *** ET llevels decreased continually after the modeling operation in the models,it dropped from a basal value of 60 93±18 66 to 34 11±8 22 and 33 54±11 12 pg/mL(P<0 05) on the 3rd and 5 th day,respectively,which were also lower than controls 64 16±12 93 pg/mL(P<0 05).It suggests that colonic obstruction may induce a decrease in plasma endothelin 1 concentrations in sleep.
