In this present time,Human Activity Recognition(HAR)has been of considerable aid in the case of health monitoring and recovery.The exploitation of machine learning with an intelligent agent in the area of health informatics gathered using HAR augments the decision-making quality and significance.Although many research works conducted on Smart Healthcare Monitoring,there remain a certain number of pitfalls such as time,overhead,and falsification involved during analysis.Therefore,this paper proposes a Statistical Partial Regression and Support Vector Intelligent Agent Learning(SPR-SVIAL)for Smart Healthcare Monitoring.At first,the Statistical Partial Regression Feature Extraction model is used for data preprocessing along with the dimensionality-reduced features extraction process.Here,the input dataset the continuous beat-to-beat heart data,triaxial accelerometer data,and psychological characteristics were acquired from IoT wearable devices.To attain highly accurate Smart Healthcare Monitoring with less time,Partial Least Square helps extract the dimensionality-reduced features.After that,with these resulting features,SVIAL is proposed for Smart Healthcare Monitoring with the help of Machine Learning and Intelligent Agents to minimize both analysis falsification and overhead.Experimental evaluation is carried out for factors such as time,overhead,and false positive rate accuracy concerning several instances.The quantitatively analyzed results indicate the better performance of our proposed SPR-SVIAL method when compared with two state-of-the-art methods.

Threshold of climate extremes that impact vegetation productivity over the Tibetan Plateau

Vegetation growth is adversely impacted by multiple climate extremes related to the water and thermal stress over the Tibetan Plateau(TP).However,it remains unknown at which stress level these climate extremes can trigger the abrupt shifts of vegetation response to climate extremes and result in the maximum vegetation response across TP.To fill this knowledge gap,we combined the hydrometeorological data and the satellite-derived vegetation index to detect two critical thresholds that determine the response of vegetation productivity to droughts,high-temperature extremes,and low-temperature extremes,respectively,during 2001-2018.Our results show that the response of vegetation productivity to droughts rapidly increases once crossing -1.41±0.6 standard deviation(σ)below the normal conditions of soil moisture.When crossing-2.98σ±0.9σ,vegetation productivity is maximum damaged by droughts.High-temperature extremes,which have the two thresholds of 1.34σ±0.4σand 2.31σ±0.4σover TP,are suggested to trigger the strong response of vegetation productivity at a milder stress level than low-temperature extremes(two thresholds:-1.44σ±0.5σand-2.53σ±0.8σ).Moreover,we found the compounded effects of soil moisture deficit in reducing the threshold values of both high-and low-temperature extremes.Based on the derived thresholds of climate extremes that impact vegetation productivity,Earth System Models project that southwestern TP and part of the northeastern TP will become the hotspots with a high exposure risk to climate extremes by 2100.This study deciphers the high-impact extreme climates using two important thresholds across TP,which advances the understanding of the vegetation response to different climate extremes and provides a paradigm for assessing the impacts of climate extremes on regional ecosystems.

Endochondral ossification of hindlimbs in embryonic development of Japanese Quail(Coturnix japonica)

The endochondral ossification of hindlimb is essential to a bird’s ability to stand,walk and fly.Most hindlimb is ossified in the embryos before hatching in precocial birds.However,the molecular mechanisms of hindlimb ossification in birds is still unclear.Therefore,we tried to examine the process of hindlimb ossification and its molecular regulation by using an animal model—Japanese Quail(Coturnix japonica).We selected four critical stages(Embryo Day:E6,E8,E12 and E16) of skeletal development of embryonic quails for hindlimb skeleton staining to show the process of endochondral ossification and to examine the molecular regulation of endochondral osteogenesis by RNA-Seq analysis.The results showed that ossification became increased with embryonic development and most hindlimb was ossified before hatching.RNA-Seq analysis revealed that various signaling pathways were involved with endochondral ossification with thyroid hormone signaling and WNT signaling pathway particularly enriched.Moreover,the expression levels of 42 genes were continuously upregulated and 14 genes were continuously downregulated from E6 to E16.The present study might provide new insights into complex molecular mechanisms in regulation of endochondral ossification.

Upgraded durian genome reveals the role of chromosome reshuffling during ancestral karyotype evolution,lignin biosynthesis regulation,and stress tolerance

Durian(Durio zibethinus)is a tropical fruit that has a unique flavor and aroma.It occupies a significant phylogenetic position within the Malvaceae family.Extant core-eudicot plants are reported to share seven ancestral karyotypes that have undergone reshuffling,resulting in an abundant genomic diversity.However,the ancestral karyotypes of the Malvaceae family,as well as the evolution trajectory leading to the28 chromosomes in durian,remain poorly understood.Here,we report the high-quality assembly of the durian genome with comprehensive comparative genomic analyses.By analyzing the collinear blocks between cacao and durian,we inferred 11 Malvaceae ancestral karyotypes.These blocks were present in a single-copy form in cacao and mainly in triplicates in durian,possibly resulting from a recent whole genome triplication(WGT)event that led to hexaploidization of the durian genome around 20(17–24)million years ago.A large proportion of the duplicated genes in durian,such as those involved in the lignin biosynthesis module for phenylpropane biosynthesis,are derived directly from whole genome duplication,which makes it an important force in reshaping its genomic architecture.Transcriptome studies have revealed that genes involved in feruloyl-Co A formations were highly preferentially expressed in fruit peels,indicating that the thorns produced on durian fruit may comprise guaiacyl and syringyl lignins.Among all the analyzed transcription factors(TFs),members of the heat shock factor family(HSF)were the most significantly upregulated under heat stress.All subfamilies of genes encoding heat shock proteins(HSPs)in the durian genome appear to have undergone expansion.The potential interactions between HSF Dzi05.397 and HSPs were examined and experimentally verified.Our study provides a high-quality durian genome and reveals the reshuffling mechanism of ancestral Malvaceae chromosomes to produce the durian genome.We also provide insights into the mechanism underlying lignin biosynthesis and heat stress tolerance.

Comparative analysis of the microbiome of sympatric wintering Bean Geese,Domestic Ducks,humans,and soil at Shengjin Lake of China reveals potential public risk to human health

The gut microbiota of migratory waterbirds is affected by various complex factors,including cross-species transmission,which increases the risk of pathogen spreading among sympatric animals and poses a potential public health risk to humans.In this study,we investigated the microbial communities of wintering Bean Geese(Anser fabalis),Domestic Ducks(A.platyrhynchos domesticus),humans,and soil using high-throughput sequencing of the 16S rRNA gene region in Shengjin Lake,China.In total,6,046,677 clean reads were obtained,representing 41,119 operational taxonomic units(OTUs)across the four groups.The dominant microbial phyla were the Proteobacteria,Firmicutes,Bacteroidota,and Actinobacteriota.The Sorensen similarity index and alpha and beta diversity results showed that the gut microbial communities of Bean Geese and Domestic Ducks were more similar to those of the other pairs.Network analysis revealed that Faecalibacterium prausnitzii,Pseudomonas fragi,and Bradyrhizobium elkanii were hubs of the three major modules.Fourteen common microbiomes were iden-tified in Bean Geese,Domestic Ducks,humans,and soil in Shengjin Lake.A total of 96 potential pathogens were identified among the four groups,with 20 specific potentially pathogenic microbiomes found in the gut of Bean Geese.Some of these pathogens are responsible for significant financial losses in the poultry industry and pose risks to human health.Klebsiella pneumoniae,Morganella morganii,Escherichia coli,and Ralstonia insidiosa are potential core pathogens found in the four groups at Shengjin Lake that can cause diseases in humans and an-imals and facilitate cross-species transmission through various media.Therefore,humans are at risk of con-tracting these pathogens from migratory birds because of their frequent contact with domestic poultry.However,further studies are required to explore the potential pathogenic species and transmission pathways among sympatric wintering Bean Geese,Domestic Ducks,humans,and soil.

Strigolactones modulate cotton fiber elongation and secondary cell wall thickening

Cotton is one of the most important economic crops in the world,and it is a major source of fiber in the textile industry.Strigolactones(SLs)are a class of carotenoid-derived plant hormones involved in many processes of plant growth and development,although the functions of SL in fiber development remain largely unknown.Here,we found that the endogenous SLs were significantly higher in fibers at 20 days post-anthesis(DPA).Exogenous SLs significantly increased fiber length and cell wall thickness.Furthermore,we cloned three key SL biosynthetic genes,namely GhD27,GhMAX3,and GhMAX4,which were highly expressed in fibers,and subcellular localization analyses revealed that GhD27,GhMAX3,and GhMAX4 were localized in the chloroplast.The exogenous expression of GhD27,GhMAX3,and GhMAX4 complemented the physiological phenotypes of d27,max3,and max4 mutations in Arabidopsis,respectively.Knockdown of GhD27,GhMAX3,and GhMAX4 in cotton resulted in increased numbers of axillary buds and leaves,reduced fiber length,and significantly reduced fiber thickness.These findings revealed that SLs participate in plant growth,fiber elongation,and secondary cell wall formation in cotton.These results provide new and effective genetic resources for improving cotton fiber yield and plant architecture.

Targeting the RAS/RAF/MAPK pathway for cancer therapy: from mechanism to clinical studies

Metastatic dissemination of solid tumors,a leading cause of cancer-related mortality,underscores the urgent need for enhanced insights into the molecular and cellular mechanisms underlying metastasis,chemoresistance,and the mechanistic backgrounds of individuals whose cancers are prone to migration.The most prevalent signaling cascade governed by multi-kinase inhibitors is the mitogen-activated protein kinase(MAPK)pathway,encompassing the RAS–RAF–MAPK kinase(MEK)–extracellular signal-related kinase(ERK)pathway.RAF kinase is a primary mediator of the MAPK pathway,responsible for the sequential activation of downstream targets,such as MEK and the transcription factor ERK,which control numerous cellular and physiological processes,including organism development,cell cycle control,cell proliferation and differentiation,cell survival,and death.Defects in this signaling cascade are associated with diseases such as cancer.RAF inhibitors(RAFi)combined with MEK blockers represent an FDAapproved therapeutic strategy for numerous RAF-mutant cancers,including melanoma,non-small cell lung carcinoma,and thyroid cancer.However,the development of therapy resistance by cancer cells remains an important barrier.Autophagy,an intracellular lysosome-dependent catabolic recycling process,plays a critical role in the development of RAFi resistance in cancer.Thus,targeting RAF and autophagy could be novel treatment strategies for RAF-mutant cancers.In this review,we delve deeper into the mechanistic insights surrounding RAF kinase signaling in tumorigenesis and RAFi-resistance.Furthermore,we explore and discuss the ongoing development of next-generation RAF inhibitors with enhanced therapeutic profiles.Additionally,this review sheds light on the functional interplay between RAF-targeted therapies and autophagy in cancer.

NFXL1 functions as a transcriptional activator required for thermotolerance at reproductive stage in Arabidopsis

Plants are highly susceptible to abiotic stresses,particularly heat stress during the reproductive stage.However,the specific molecular mechanisms underlying this sensitivity remain largely unknown.In the current study,we demonstrate that the Nuclear Transcription Factor,X-box Binding Protein 1-Like 1(NFXL1),directly regulates the expression of DEHYDRATION-RESPONSIVE ELEMENT-BINDING PROTEIN 2A(DREB2A),which is crucial for reproductive thermotolerance in Arabidopsis.NFXL1 is upregulated by heat stress,and its mutation leads to a reduction in silique length(seed number)under heat stress conditions.RNA-Seq analysis reveals that NFXL1 has a global impact on the expression of heat stress responsive genes,including DREB2A,Heat Shock Factor A3(HSFA3)and Heat Shock Protein 17.6(HSP17.6)in flower buds.Interestingly,NFXL1 is enriched in the promoter region of DREB2A,but not of either HSFA3 or HSP17.6.Further experiments using electrophoretic mobility shift assay have confirmed that NFXL1 directly binds to the DNA fragment derived from the DREB2A promoter.Moreover,effector-reporter assays have shown that NFXL1 activates the DREB2A promoter.The DREB2A mutants are also heat stress sensitive at the reproductive stage,and DEREB2A is epistatic to NFXL1 in regulating thermotolerance in flower buds.It is known that HSFA3,a direct target of DREB2A,regulates the expression of heat shock proteins genes under heat stress conditions.Thus,our findings establish NFXL1 as a critical upstream regulator of DREB2A in the transcriptional cassette responsible for heat stress responses required for reproductive thermotolerance in Arabidopsis.

Selective extinction and habitat nestedness are the main drivers of lizard nestedness in the Zhoushan Archipelago

Nested subset pattern(nestedness)is an important part of the theoretical framework of island biogeography and community ecology.However,most previous studies often used nestedness metrics or randomization algorithms that are vulnerable to type I error.In this study,we inves-tigated the nestedness of lizard assemblages on 37 islands in the Zhoushan Archipelago,China.We used the line-transect method to survey species occurrence,abundance,and habitat types of lizards on 37 islands during 2 breeding seasons in 2021 and 2022.We applied the nested metric WNODF and the conservative rc null model to control for type I error and quantify the significance of nestedness.Spearman rank cor relations were used to evaluate the role of 4 habitat variables(island area,2 isolation indices,and habitat diversity)and 4 ecological traits(body size,geographic range size,clutch size,and minimum area requirement)in generating nestedness.The results of WNODF analyses showed that lizard assemblages were significantly nested.The habitat-by-site matrix estimated by the program NODF was also significantly nested,support-ing the habitat nestedness hypothesis.The nestedness of lizard assemblages were significantly correlated with island area,habitat diversity.clutch size,and minimum area requirement.Overall,our results suggest that selective extinction and habitat nestedness were the main drivers of lizard nestedness in our system.In contrast,the nestedness of lizard assemblages was not due to passive sampling or selective colonization.To maximize the number of species preserved,our results indicate that we should protect both large islands with diverse habitats and species with largearea requirement and clutch size.

Osteoporosis(OP)is an aging-associated condition that significantly affects the quality of life in aging humans and has few treatment options.As an important marker,recombinant immunoglobulin superfamily containing leucinerich repeat(ISLR)could be used as a gene therapy for OP.We herein used alkaline phosphatase(ALP)and alizarin red S(ARS)staining to evaluate osteogenic differentiation potential.
