An information theoretic method to identify combinations of genomic alterations that promote glioblastoma

The last 40 years have witnessed how p53 rose from a viral binding protein to a central factor in both stress responses and tumor *** exquisite regulation of p53 functions is of vital importance for cell fate *** the multiple layers of mechanisms controlling p53 function,posttranslational modifications (PTMs) represent an efficient and precise *** p53 PTMs include phosphorylation,ubiquitination,acetylation,and ***,other PTMs like sumoylation,neddylation,O-GlcNAcylation,adenosine diphosphate (ADP)-ribosylation,hydroxylation,and p-hydroxybutyrylation are also shown to play various roles in p53 *** independent action or interaction,PTMs affect p53 stability,conformation,localization,and binding *** of the PTM-related pathway is among the major causes of p53-associated developmental disorders or diseases,especially in *** review focuses on the roles of different p53 modification types and shows how these modifications are orchestrated to produce various outcomes by modulating p53 activities or targeted to treat different diseases caused by p53 dysregulation.

PREVALENCE OF AGE-RELATED CATARACT IN GANZI AND SHANGHAI

It has beeen reported that the prevalence of cataract is much higher in plateau than that in plain regions. But the data are incomplete and can not be compared because of different examination methods and different diagnostic criteria. A small group of ophthalmologists was sent to Ganzi, Sichuan Province at the altitude of 3.393 m, and to Qinpu in the suburb of Shanghai which is at the sea level. In Ganzi 3 905 (91.97%) people including 822 Hans and 3 083 Zang and in Qinpu 3 101 (93.20%) were studied. Age-related cataract was diagnosed when there are cortical or nuclear, or mixed opacities; and aphakia due to previous extraction of age-related cataract. Direct method was used to standardize the data. The prevalence of age-related cataract in Ganzi is about 82% higher than that in Qinpu in all age groups; from age 16-60,the prevalence of age-related cataract in Ganzi is about 180% (in Hans) and 160% (in Zangs) higher than that in Qinpu. There is no significant difference in the prevalence of age-related cataract between the ethnic groups in Ganzi. In Ganzi, the sunshine hours per year are 1.28 times that in Qinpu, and the global radiation in Ganzi is 1.47 times that in Qinpu. The higher prevalence of cataract in Ganzi than in Qinpu may be due to the greater exposure to the sunshine on the Tibetan plateau. The differences in prevalence between males and females and between indoor and outdoor work are discussed.

Vibration-controlled Transient Elastography for Assessment of Liver Fibrosis at a USA Academic Medical Center

Background and Aims:Vibration-controlled transient elastography(VCTE)is a noninvasive tool that uses liver stiffness measurement(LSM)to assess *** real-life data during everyday clinical practice in the USA are lacking,we describe the patterns of use and diagnostic performance of VCTE in patients at an academic medical center in New York ***:Patients who received VCTE scans were included if liver biopsy was performed within 1 *** performance of VCTE in differentiating dichotomized fibrosis stages was assessed via area under the receiver operating characteristics(AUROC).Fibrosis stage determined from VCTE LSM was compared to liver ***:Of 109 patients,49 had nonalcoholic fatty liver disease,16 chronic hepatitis C,15 congestive hepatopathy,and 22 at least two *** was 0.90 for differentiating cirrhosis(stage 4)with a positive predictive value(PPV)range of 0.28 to 0.45 and negative predictive value range of 0.96 to *** 31(32%)patients,VCTE fibrosis stage was at least two stages higher than liver biopsy fibrosis *** of thirty-five pa-tients considered to have cirrhosis by VCTE had stage 0 to 2 and 12 stage 3 fibrosis on liver ***:VCTE has reasonable diagnostic accuracy and is reliable at ruling out ***,because of its low PPV,caution must be exercised when used to diagnose cirrhosis,as misdiagnosis can lead to unnecessary health care *** routine practice,VTCE is also sometimes performed for disease etiologies for which it has not been robustly validated.

The induction of brown-like adipocyte development in white adipose tissue （WAT） confers numerous metabolic benefits by decreasing adiposity and increasing energy expenditure. Therefore, WAT browning has gained considerable attention for its potential to reverse obesity and its associated co-morbidities. However, this perspective has been tainted by recent studies identifying the detrimental effects of inducing WAT browning. This review aims to highlight the adverse outcomes of both overactive and underactive browning activity, the harmful side effects of browning agents, as well as the molecular brake-switch system that has been proposed to regulate this process. Developing novel strategies that both sustain the metabolic improvements of WAT browning and attenuate the related adverse side effects is therefore essential for unlocking the therapeutic potential of browning agents in the treatment of meta- bolic diseases.

Coronavirus Disease 2019 and Liver Injury:A Retrospective Analysis of Hospitalized Patients in New York City

Background and Aims:Coronavirus disease 2019(COVID-19)is a global threat,affecting more than 100 million people and causing over 2 million *** laboratory test abnormalities are an extrapulmonary manifestation of COVID-19,yet characterization of hepatic injury is *** objective was to further characterize and identify causes of liver injury in patients with ***:We conducted a retrospective cohort study of 551 patients hospitalized with COVID-19 at NewYork-Presbyterian Hospital/Columbia University Irving Medical Center between March 1,2020 and May 31,*** analyzed patient demographics,liver laboratory test results,vital signs,other relevant test results,and clinical outcomes(mortality and intensive care unit admission).Results:Abnormal liver laboratory tests were common on hospital admission for COVID-19 and the incidence increased during *** those with elevated serum alanine aminotransferase and/or alkaline phosphatase activities on admission,58.2%had a cholestatic injury pattern,35.2%mixed,and 6.6%*** liver disease was not associated with outcome;however,abnormal direct bilirubin or albumin on admission were associated with intensive care unit stay and *** average,patients who died had greater magnitudes of abnormalities in all liver laboratory tests than those who *** hepatitis was a mechanism of severe hepatocellular injury in some ***:Liver laboratory test abnormalities are common in hospitalized patients with COVID-19,and some are associated with increased odds of intensive care unit stay or *** hepatocellular injury is likely attributable to secondary effects such as systemic inflammatory response syndrome,sepsis,and ischemic hepatitis.

Update on hepatocellular carcinoma: Pathologists' review

Histopathologic diversity and several distinct histologic subtypes of hepatocellular carcinoma(HCC) are well-recognized. Recent advances in molecular pathology and growing knowledge about the biology associated with distinct histologic features and immuno-profile in HCC allowed pathologists to update classifications. Improving sub-classification will allow for more clinically relevant diagnoses and may allow for stratification into biologically meaningful subgroups. Therefore, immuno-histochemical and molecular testing are not only diagnostically useful, but also are being incorporated as crucial components in predicting prognosis of the patients with HCC. Possibilities of targeted therapy are being explored in HCC, and it will be important for pathologists to provide any data that may be valuable from a theranostic perspective. Herein, we review and provide updates regarding the pathologic sub-classification of *** diagnostic approach and the role of biomarkers as prognosticators are reviewed. Further, the histopathology of four particular subtypes of HCC:Steatohepatitic, clear cell, fibrolamellar and scirrhous-and their clinical relevance, and the recent consensus on combined HCC-cholangiocarcinoma is summarized. Finally, emerging novel biomarkers and new approaches to HCC stratification are reviewed.

Colorectal cancer in patients under 50 years of age:A retrospective analysis of two institutions' experience

AIM:To investigate the epidemiological characteristics of colorectal cancer(CRC)in patients under 50 years of age across two ***:Records of patients under age 50 years of age who had CRC surgery over a 16 year period were assessed at two *** following documents where reviewed:admission notes,operative notes,and discharge *** main study variables included:age,presenting symptoms,family history,tumor location,operation,stage/differentiation of disease,and post operative *** of disease was classified according to the American Joint Committee on Cancer TNM staging system:tumor depth;node status;and ***:CRC was found in 180 patients under age50 years(87 females,93 males;mean age 41.4±6.2years).Young patients accounted for 11.2%of cases during a 6 year period for which the full data set *** percent had a 1stdegree and 12%a 2nd degree family CRC *** all patients(94%)were symptomatic at diagnosis;common symptoms included:bleeding(59%),obstruction(9%),and abdominal/rectal pain(35%).Evaluation was often delayed and bleeding frequently attributed to *** stage CRC(Stage 3 or 4)was noted in 53%of *** tumors were distal to the splenic flexure(77%)and 39%involved the *** patients(95%)had segmental resections;6 patients had subtotal/total *** differentiated tumors were noted in 12%and mucinous lesions in 19%of patients of which most had Stage 3 or 4 ***-two patients(13%)developed recurrence and/or progression of disease to *** patients(ages 42,42and 49 years)went on to develop metachronous primary colon cancers within 3 to 4 years of their initial ***:CRC was common in young patients with no family *** patients with symptoms merit a timely evaluation to avoid presentation with late stage CRC.

Biological Markers on Human Pregnancy

This article reviews a selected set of recently described pregnancy-associated proteins which possess potential for both signaling pregnancy onset and monitoring its course. These molecules are compared and contrasted with human chorionic gonadotropin, the first pregnancy-associated protein to be discovered, and the standard biomarker of pregnancy to which all others must still be referenced. Recent advances in hCG research have focused on the structural determination and diagnostic significance of the subunits and fragments of the hCG molecule, particularly in urine. An outline of the potential utility of this approach is also presented.

Antidepressant foods:An evidence-based nutrient profiling system for depression

AIM To investigate which foods are the most nutrient dense sources of nutrients demonstrated by the scientific literature to play a role in the prevention and promotion of recovery from depressive disorders. METHODS A systematic literature review was conducted to derive a list of Antidepressant Nutrients from the 34 nutrients known to be essential for humans using level of evidence criteria. Nutritional data was extracted for a subset of foods with a high content of at least 1 Antidepressant Nutrient using a USDA database. These foods were analyzed for Antidepressant Nutrient density resulting in an Antidepressant Food Score(AFS). Plant and animal foods were analyzed separately. RESULTS Twelve Antidepressant Nutrients relate to the prevention and treatment of depressive disorders: Folate, iron, longchain omega-3 fatty acids(EPA and DHA), magnesium, potassium, selenium, thiamine, vitamin A, vitamin B6, vitamin B12, vitamin C, and zinc. The highest scoring foods were bivalves such as oysters and mussels, various seafoods, and organ meats for animal foods. The highest scoring plant foods were leafy greens, lettuces, peppers, and cruciferous vegetables. CONCLUSION The AFS is based on a nutrient profiling system devised to identify foods with the highest nutrient density of nutrients with clinical evidence to support theirrole in depressive disorders. This list of foods and food categories with the highest density of the 12 Antidepressant Nutrients, the Antidepressant Foods, should be considered by researchers in the design of future intervention studies and clinicians as dietary options to support prevention and recovery from depression disorders.
