Etanercept-synthesizing adipose-derived stem cell secretome:A promising therapeutic option for inflammatory bowel disease

BACKGROUND Inflammatory bowel disease(IBD)is a chronic inflammatory condition of the gastrointestinal tract,with tumor necrosis factor(TNF)-αplaying a key role in its pathogenesis.Etanercept,a decoy receptor for TNF,is used to treat inflammatory conditions.The secretome derived from adipose-derived stem cells(ASCs)has anti-inflammatory effects,making it a promising therapeutic option for IBD.AIM To investigate the anti-inflammatory effects of the secretome obtained from ASCs synthesizing etanercept on colon cells and in a dextran sulfate sodium(DSS)-induced IBD mouse model.METHODS ASCs were transfected with etanercept-encoding mini-circle plasmids to create etanercept-producing cells.The secretory material from these cells was then tested for anti-inflammatory effects both in vitro and in a DSS-induced IBD mouse model.RESULTS This study revealed promising results indicating that the group treated with the secretome derived from etanercept-synthesizing ASCs[Etanercept-Secretome(Et-Sec)group]had significantly lower expression levels of inflammatory mediators,such as interleukin-6,Monocyte Chemoattractant Protein-1,and TNF-α,when compared to the control secretome(Ct-Sec).Moreover,the Et-Sec group exhibited a marked therapeutic effect in terms of preserving the architecture of intestinal tissue compared to the Ct-Sec.CONCLUSION These results suggest that the secretome derived from ASCs that synthesize etanercept has potential as a therapeutic agent for the treatment of IBD,potentially enhancing treatment efficacy by merging the anti-inflam-matory qualities of the ASC secretome with etanercept's targeted approach to better address the multifaceted pathophysiology of IBD.

The development of intestinal anastomosis techniques,including hand suturing,stapling,and compression anastomoses,has been a significant advancement in surgical practice.These methods aim to prevent leakage and minimize tissue fibrosis,which can lead to stricture formation.The healing process involves various phases:hemostasis and inflammation,proliferation,and remodeling.Mechanical staplers and sutures can cause inflammation and fibrosis due to the release of profibrotic chemokines.Compression anastomosis devices,including those made of nickel-titanium alloy,offer a minimally invasive option for various surgical challenges and have shown safety and efficacy.However,despite advancements,anastomotic techniques are evaluated based on leakage risk,with complications being a primary concern.Newer devices like Magnamosis use magnetic rings for compression anastomosis,demonstrating greater strength and patency compared to stapling.Magnetic technology is also being explored for other medical treatments.While there are promising results,particularly in animal models,the realworld application in humans is limited,and further research is needed to assess their safety and practicality.

Prostate cancer(PCa)is the second most common malignancy among men globally.The Fu-Zheng-Yi-Liu(FZYL)Formula has been widely utilized in the treatment of PCa.This study investigates whether the FZYL Formula can inhibit PCa by tar-geting the TAMs/CCL5 pathway.We conducted in vitro co-cultures and in vivo co-injections of PCa cells and TAMs to mimic their in-teraction.Results showed that the FZYL Formula significantly reduced the proliferation,colony formation,subpopulations of PCSCs,and sphere-formation efficacy of PCa cells,even in the presence of TAM co-culture.Additionally,the Formula markedly decreased the migration,invasion,and epithelial-mesenchymal transition(EMT)of PCa cells induced by TAMs.The FZYL Formula also reversed M2 phenotype polarization in TAMs and dose-dependently reduced their CCL5 expression and secretion,with minimal cytotoxicity observed.Mechanistic studies confirmed that the TAMs/CCL5 axis is a critical target of the FZYL Formula,as the addition of exogen-ous CCL5 partially reversed the formula’s inhibitory effects on PCSCs self-renewal in the co-culture system.Importantly,the Formula also significantly inhibited the growth of PCa xenografts,bone metastasis,and PCSCs activity in vivo by targeting the TAMs/CCL5 pathway.Overall,this study not only elucidates the immunomodulatory mechanism of the FZYL Formula in PCa therapy but also highlights the TAMs/CCL5 axis as a promising therapeutic target.

What is spinal cord injury:Spinal cord injury(SCI)is the damage to the structure of the bundles of cells and nerves that communicate signals from the brain to the body and extremities.The pathology of SCI includes both primary and secondary injuries(Morales et al.,2016).Physical forces such as compression,shearing,contusion,and tearing are major causes of primary injury in SCI.There are two main processes in primary injury:acute and subacute.The acute phase includes traumatic disruption of axons and hemorrhage of the blood vessels around the spinal cord.Hemorrhagic injury to the vessels can lead to increased edema within the neural and cord tissues,susceptibility to infiltration by microglia and astrocytes,excitotoxicity,and demyelination.Similarly,disruption of the blood-spinal cord barrier results in the release of inflammatory cytokines from specific cells and vessels.

Aim:To investigate the ultrastructural localization of atrial natriuretic peptide(ANP)-synthesizing cells and the relationship between ANP-synthesizing cellsand microvessels in rat gastric mucosa.Methods:Immunohistochemistry tech-niques and postembedding immunoelectron microscopy techniques were used tovalidate the findings regarding the expression of ANP-synthesizing cells and theultrastructural localization of ANP-synthesizing cells in the gastric mucosa.His-tochemistry techniques and the tannic acid-ferric chloride method(TA-Fe stain-ing method)were used to reveal microvessel density and the distribution of ANP-synthesizing cells in different regions of the stomach.Results:Cells expressingANP were localized and ANP-synthesizing cells were identified as enterochroma-ffin(EC)cells in the gastric mucosa.ANP-synthesizing cells existed in differentregions of the stomach.The percentage ANP-synthesizing cells in the mucosawas greatest in the fundus(46.7%e5.3%),intermediate in the antrum(40.1%±4.5%),and least in the body(21.6%±3.6%).There was a positive relationshipbetween the percentage of ANP-synthesizing cells and the density of microvesselsin the antral mucosa,but not in the fundus or body mucosa.Conclusion:ANP issynthesized by EC cells in rat gastric mucosa,and ANP-synthesizing cells aremost dense in the gastric fundus.ANP may act not only as a regional autocrineand/or paracrine regulator,but also as an endocrine regulatory peptide in thegastrointestinal tract.

Sinus venosus defect(SVD) accounts for 4%–11% of all atrial septal defects(ASD) and may present with dyspnea, heart failure(HF), palpitations, or angina.^[1] The malformation typically involves an interatrial communication due to a deficiency in the common wall connecting the right pulmonary veins and the superior vena cava(SVC) and often accompanies anomalous pulmonary venous connection of some or all of the pulmonary veins.

AIM: To study the association between APOE polymorphisms and cerebral infarction through a case-control study among the Chinese Han population. METHODS: First-ever cerebral infarction patients (n=226) whose ages ranged from 40 to 60 years old were recruited from Department of Neurology, Zhongshan Hospital, Shanghai, and Zhejiang Chinese Traditional medicine Hospital, Zhejiang, China. Unrelated healthy controls (n=201) were selected from the general population in the same area with similar age and sex distribution. APOE was amplified by one-stage PCR using the forward primer: 5’-GGC ACG GCT GTC CAA GGA GCT-3’ and reverse primer: 5’-GAT GGC GCTGAG GCC GCG CT-3’. The PCR product was digested directly with 5 U of CfoI and separated by a 20 % polyacryla- mide (acrylamide: bis-acrylamide=29:1) nondenaturing gel. RESULTS: Both cerebral infarction patient and control groups were in Hardy-Weinberg equilibrium. The allele frequency of APOE*2, APOE*3, and APOE*4 was 4.6 %, 81.9 %, and 13.5 % respectively in the patients with cerebral infarction; 5.7 %, 87.3 %, and 7.0 % respectively in the healthy control group. Compared with APOE3/3 subjects, APOE4/4 carriers had a 2.1-fold risk of cerebral infarction (odds ratio 2.1, 95 % confidence limits 1.3 to 3.4). The allele frequency of APOE*4 in the cerebral infarction patient group was significantly higher than that in the control group (13.5 % vs 7.0 %; P=0.002). CONCLUSION: APOE 4 is a risk factor for cerebral infarction among the Chinese Han population.

Arecoline inhibits catecholamine release from perfused rat adrenal gland

Aim:To study the effect of arecoline,an alkaloid isolated from Areca catechu,onthe secretion of catecholamines (CA) evoked by cholinergic agonists and themembrane depolarizer from isolated perfused rat adrenal gland.Methods:Adrenalglands were isolated from male Sprague-Dawley rats.The adrenal glands wereperfused with Krebs bicarbonate solution by means of a peristaltic pump.The CAcontent of the perfusate was measured directly using the fluorometric method.Results:Arecoline (0.1-1.0 mmol/L) perfused into an adrenal vein for 60 min pro-duced dose-and time-dependent inhibition in CA secretory responses evoked byacetylcholine (ACh) (5.32 mmol/L),1.1-dimethyl-4-phenyl piperazinium iodide(DMPP) (100μmol/L for 2 min) and 3-(m-choloro-phenyl-carbamoyl-oxy)-2-butynyltrimethyl ammonium chloride (McN-A-343)(100μmol/L for 2 min).However,lowerdoses of arecoline did not affect CA secretion of high K~+(56 mmol/L);higherdoses greatly reduced CA secretion of high K~+.Arecoline also failed to affectbasal catecholamine output.Furthermore,in adrenal glands loaded with arecoline(0.3 mmol/L),CA secretory response evoked by Bay-K-8644 (10μmol/L),an activa-tor of L-type Ca2+channels,was markedly inhibited,whereas CA secretion bycyclopiazonic acid(10μmol/L),an inhibitor of cytoplasmic Ca2+-ATPase,was notaffected.Nicotine (30μmol/L),which was perfused into the adrenal gland for 60min,however,initially enhanced ACh-evoked CA secretory responses.As timeelapsed,these responses became more inhibited,whereas the initially enhancedhigh K~+-evoked CA release diminished.CA secretion evoked by DMPP and McN-A-343 was significantly depressed in the presence of nicotine.Conclusion:Arecoline dose-dependently inhibits CA secretion from isolated perfused rat ad-renal gland evoked by activation of cholinergic receptors.At lower doses arecolinedoes not inhibit CA secretion through membrane depolarization,but at largerdoses it does.This inhibitory effect of arecoline may be mediated by blocking thecalcium influx into the rat adrenal medullary chromaffin ce

Biomarkers of neurodegenerative disorders: How good are they?

Biomarkers are very important indicators of normal and abnormal biological processes. Specific changes in pathologies, biochemistries and genetics can give us comprehensive information regarding the nature of any particular disease. A good biomarker should be precise and reliable, distinguishable between normal and interested disease, and differential between different diseases. It is believed that biomarkers have great potential in predicting chances for diseases, aiding in early diagnosis, and setting standards for the development of new remedies to treat diseases. New technologies have enabled scientists to identify biomarkers of several different neurodegenerative diseases. The followings, for instance, are only a few of the many new biomarkers that have been recently identified: the phosphorylated tau protein and aggregated β-amyloid peptide for Alzheimer’s disease (AD), α-synuclein contained Lewy bodies and altered dopamine transporter (DAT) imaging for Parkinson’s disease (PD), SOD mutations for familial amyotrophic lateral sclerosis (ALS), and CAG repeats resulted from Huntington’s gene mutations in Huntington’s disease (HD). This article will focus on the most-recent findings of biomarkers belonging to the four mentioned neurodegenerative diseases.

Copper,as an essential trace nutrient,plays a crucial role in biological processes such as mitochondrial respiration,antioxidant stress response,and the synthesis of biomolecules.Typically,cellular copper concentrations are maintained at very low levels,a pattern also observed in cancer cells to prevent adverse consequences of copper overload,such as cuproptosis.This involves copper dependency,accumulation of lipidated proteins,and a reduction in Fe-S cluster proteins[1].Various neurodegenerative diseases are associated with imbalances in copper homeostasis.
