Integrated multi-omics analysis reveals liver metabolic reprogramming by fish iridovirus and antiviral function of alpha-linolenic acid

Iridovirus poses a substantial threat to global aquaculture due to its high mortality rate;however,the molecular mechanisms underpinning its pathogenesis are not well ***,a multi-omics approach was applied to groupers infected with Singapore grouper iridovirus(SGIV),focusing on the roles of key *** showed that SGIV induced obvious histopathological damage and changes in metabolic enzymes within the ***,SGIV significantly reduced the contents of lipid droplets,triglycerides,cholesterol,and *** analysis indicated that the altered metabolites were enriched in 19 pathways,with a notable down-regulation of lipid metabolites such as glycerophosphates and alpha-linolenic acid(ALA),consistent with disturbed lipid homeostasis in the *** of transcriptomic and metabolomic data revealed that the top enriched pathways were related to cell growth and death and nucleotide,carbohydrate,amino acid,and lipid metabolism,supporting the conclusion that SGIV infection induced liver metabolic *** integrative transcriptomic and proteomic analysis indicated that SGIV infection activated crucial molecular events in a phagosome-immune depression-metabolism dysregulation-necrosis signaling *** note,integrative multi-omics analysis demonstrated the consumption of ALA and linoleic acid(LA)metabolites,and the accumulation of L-glutamic acid(GA),accompanied by alterations in immune,inflammation,and cell death-related *** experimental data showed that ALA,but not GA,suppressed SGIV replication by activating antioxidant and anti-inflammatory responses in the ***,these findings provide a comprehensive resource for understanding host response dynamics during fish iridovirus infection and highlight the antiviral potential of ALA in the prevention and treatment of iridoviral diseases.

Changes of tropical cyclone size in three oceanic basins of the northern hemisphere from 2001 to 2021

In this study the changes of tropical cyclone(TC)size from 2001 to 2021 are analyzed based on linear and quadratic curve fittings of the National Hurricane Center(NHC)/Joint Typhoon Warning Center(JTWC)best track data,based on the radius of maximum wind(RMW)and the average radius of 34-kt wind(AR34),in three oceanic basins of the North Atlantic(NATL),the Western North Pacific(WPAC)and the Eastern North Pacific(EPAC).The computations are done separately for two categories of tropical cyclones:tropical storms(TS)and hurricanes(HT).Size changes of landfalling and non-landfalling TCs are also *** show that there is a great inter-basin variability among the changes in TC *** conclusions include:1)overall,the inner cores of TSs have become larger in all three basins,with the increasing tendencies being significant in the NATL and WAPC,while those of HTs mostly get smaller or remain similar;2)meanwhile,comparatively large inter-basin differences are observed for the TC outer core sizes,and the sizes of landfalling TCs;3)particularly,a significant decrease in landfalling HT outer core size is observed over the EPAC;4)in contrast,significant increases in landfalling TS inner core size are found over the NATL and *** presented analysis results could benefit future research about TC forecasts,storm surge studies,and the cyclone climate and its changes.

Each physical process in a numerical weather prediction(NWP)system may have many different parameterization *** studies have shown that the performance of different physical parameterization schemes varies with the weather situation to be ***,it is necessary to select a suitable combination of physical parameterization schemes according to the variation of weather ***,it is rather difficult to identify an optimal combination among millions of possible parameterization scheme *** study applied a simple genetic algorithm(SGA)to optimizing the combination of parameterization schemes in NWP models for typhoon *** feasibility of SGA was verified with the simulation of Typhoon Mujigae(2015)by using the Weather Research and Forecasting(WRF)model and Typhoon Higos(2020)by using the Coupled Ocean–Atmosphere–Wave–Sediment Transport(COAWST)modeling *** results show that SGA can efficiently obtain the optimal combination of *** Typhoon Mujigae(2015),the optimal combination can be found from the 1,304,576 possible combinations by running only 488 *** results can be obtained for Typhoon Higos(2020).Compared to the default combination proposed by the COAWST model system,the optimal combination scheme significantly improves the simulation of typhoon track and *** study provides a feasible way to search for the optimal combinations of physical parameterization schemes in WRF and COAWST for more accurate typhoon *** can help provide references for future development of NWP models,and for analyzing the coordination and adaptability of different physical process parameterization schemes under specific weather backgrounds.

The origin of T cells in the teleost's brain is *** viewing the central nervous system(CNS)as immune privileged has been widely accepted,previous studies suggest that T cells residing in the thymus but not in the spleen of the teleost play an essential role in communicating with the peripheral ***,we identified nine T cell subpopulations in the thymus and spleen of orange-spotted grouper(Epinephelus coioices)through single-cell RNA-sequencing *** viral CNS infection with red-spotted grouper nervous necrosis virus(RGNNV),the number of slc43a2^(+)T cells synchronously increased in the spleen and *** the infection tests in asplenic zebrafish(tlx1^▲zebrafish model),no increase in the number of slc43a2^(+)T cells was observed in the ***-cell transcriptomic analysis indicated that slc43a2^(+)T cells mature and functionally differentiate within the spleen and then migrate into the brain to trigger an immune *** study suggests a novel route for T cell migration from the spleen to the brain during viral infection in fish.

Tilapia Head Glycolipid Alleviates Indomethacin-Induced Gastric Ulcer via Regulating Oxidative Stress and Inflammation Through COX/PGE2 Signaling Pathway in Adult Rats

The aim of this experiment was to investigate the ameliorative effect and molecular mechanism of tilapia head glycolipid(TH-GL)on indomethacin(IDM)-induced gastric ulcer in male Sprague Dawley(SD)*** gastric ulcer model was established by oral administration of 30mgkg^(-1) IDM after 7 days of TH-GL or omeprazole(OME)administration in *** the macroscopic gastric injury symptoms,gastric mucosa protective factor cyclooxygenase 1(COX-1),cyclooxygenase 2(COX-2),prostaglandin E_(2)(PGE_(2)),the levels of oxidative stress,and inflammatory cytokine expression levels in the rats were *** experimental results showed that multiple ulcers appeared on the gastric surface of the rats in the model *** to the model group,TH-GL significantly alleviated gastric ulcers and reduced the gastric damage index in *** addition,TH-GL significantly promoted the expression of constitutive enzyme COX-1 while inhibited the expression of inducible enzyme COX-2,and make PGE2 maintain at normal ***-GL also inhibited oxidative stress and inflammatory responses,increased superoxide dismutase(SOD)activity and glutathione(GSH)content,decreased the level of malondialdehyde(MDA)and the content of pro-inflammatory *** conclusion,these results suggested that TH-GL could maintain the expression levels of COX-1 and PGE2 while inhibit the expression of COX-2 in the gastric of rat and then prevent IDM-induced gastric ulcer,which may be related to the regulation of oxidative stress and inflammatory ***,TH-GL might be a new option for the prevention of gastric diseases induced by IDM.

Growth History of Complex Fault System and Syn- to Post-rift Transition in the Wushi East Subsag， Beibuwan Basin

The Wushi sag is located in the central part of the Beibuwan Basin， northern south China Sea. Above the thick oily shale unit in the Wushi East Subsag， a suite of terrace-like extensional décollement fault systems exists， which， in conjunction with the No.7 boundary fault， control a complex fault system. This study integrates the high-resolution 3D seismic data and quantitative analysis of fault displacement to analyze the growth history of the complex fault system and to establish a model of fault system evolution in the Wushi East Subsag. The findings reveal two phases of fault growth northward migration during the rifting process. The first phase occurred between the rift I phase and the décollement I phase， witnessing fault activity migrating northward from the No.7 boundary fault to the décollement faults (Fd and Fe) in the hangingwall. The second phase occurred between the décollement I phase and the décollement II phase， with fault activity migrating further northward from Fd and Fe to Fa， Fb and Fd. By the end of the Oligocene， basin inversion controlled the syn- to post-rift transition of the Wushi East Subsag. This study bears significant implications for hydrocarbon exploration and for understanding the mechanism underlying the syn- to post-rift process in the north continental marginal basins of the south China Sea.

Natural radionuclides are powerful tools for understanding the sources and fate of suspended particulate matter(SPM).Particulate matter with different particle sizes behaves differently with respect to adsorption and *** analyzed the activi-ties and distribution characteristics of multiple natural radionuclides(238U,226Ra,40K,228Ra,7Be and 210Pbex)on size-fractionated SPM at the Lijin Hydrographic Station(Huanghe or Yellow River)every month over a one-year *** showed that medium silt(16–32µm)was the main *** expected,the activity of each radionuclide decreased with an increase of particle *** examined the sources of SPM with different particle sizes using activity ratios of 226Ra/238U,228Ra/226Ra,40K/238U and 7Be/210Pbex,and concluded that SPM with different particle sizes originated from different *** results indicate that fine SPM(32µm)was mainly derived from resuspension of riverbed *** high runoff periods,the concentration of SPM increased significantly,and the pro-portion of fine particles originating upstream *** occurring radioactive isotopes,especially on size-fractionated par-ticles,are therefore seen as useful tracers to understand the sources and behaviors of riverine particles transported from land to sea.

The inland saltwater lakes harbor exceptional ***,two new species of solitary sessile peritrich ciliates were isolated from Qinghai Lake,the largest inland saltwater lake in *** morphology,ciliature,silverline system,and molecular phylogeny were investigated based on live observation,silver staining,and analysis of the small subunit ribosomal DNA(SSU rDNA).Vorticella paraglobosa *** characterized mainly by its obconical or elongate bell-shaped zooid,C-shaped macronucleus,single ventrally located contractile vacuole,two-rowed infundibular polykinety 3,and 28-38 silverlines between peristome and aboral tro-chal band and 10-15 between aboral trochal band and *** cotyliformis *** from its congeners mainly by its double-layered peristomial lip,cup-shaped zooid,J-shaped macronucleus,single ventrally positioned contractile vacuole,three-rowed infundibular polykinety 3,and 70-85 silverlines between peristome and aboral trochal band and 21-25 between aboral trochal band and *** SSU rDNA sequences of the two new species were obtained,and the subsequent molecular phylogenetic analysis supported their taxonomic classification.

Backgroundand Originality Content,Cembranoids are a structurally diverse class of diterpenoids commonly occurring in marine and terrestrial organisms.(+)-Cembrene was the first reported cembranoid diterpenoid discovered in 1962,l and more than 1400 cembranoid diterpenoids have been identified in the last six decades,[2]Cembranoids are characterized by a 14-membered carbocyclic ring,an isopropyl group and three methyl groups,which form their core carbon *** processes such as oxidation,oxidative rearrangements and transannular cyclizations occur in organisms,leading to the formation of distinctive specialized metabolites such as epoxide,lactone,ester,furan,pyrane and other ringbased cembranoids.[2-3]

Modulating electronic structure of metal nanoparticles via metal–support interaction has attracted intense interest in the field of catalytic ***,the roles of supporting substrates in regulating catalytic properties of nanozymes remain *** this study,we find that the use of graphdiyne oxide(GDYO)as the substrate for self-terminating growth of Ru nanoparticles(Ru@GDYO)endows the peroxidase-like activity of Ru nanoparticles with intrinsic physiological pH preference and natural horseradish peroxidase(HRP)comparable *** nanoparticles electrolessly deposited onto GDYO possess a partially oxidized electronic structure owing to limited charge transfer between Ru and GDYO,contributing to the intrinsic physiological pH preference of the peroxidase-mimicking *** importantly,the substrate GDYO plays an influential factor in enhancing catalytic activity,that is,the activity of Ru@GDYO is much higher than that of Ru nanoparticles deposited on other carbon substrates including graphene oxides and *** demonstrate the application of Ru@GDYO nanozyme in neutral solutions,we employ Ru@GDYO with nicotinamide adenine dinucleotide(NAD+)-dependent dehydrogenases in physiological conditions to realize a sustainable cascade reaction by means of forming continuous NAD^(+)/dihydronicotiamide adenine dinucleotide(NADH)*** finding represents a promising perspective on designing high-performance peroxidase-mimicking nanozymes with broader applicability,raising fundamental understanding of structure–activity relationship,and investigating new applications of nanozymes in biological systems.
