Modifications in aroma characteristics of'Merlot'dry red wines aged in American,French and Slovakian oak barrels with different toasting degrees

Aging in oak barrels is widely used in enology which could bring flavor changes and aromatic complexity to wines.In the present study,the aroma compounds were analyzed from the‘Merlot’dry red wines,which were fermented in two types of fermenters(stainless steel tank and rotated oak barrel)and aged in six types of oak barrels(three geographic origins×two toasting degrees)for different time(0,3,6 and 9 months,respectively).Results showed that 30 volatiles were associated with barrels and increased during oak aging.The fermenters could influence the intensities of the toast,leathery,smoky,fruity,floral and caramel aromas.The concentration of whisky lactone,eugenol,cis-isoeugenol,and the intensities of the toast and spicy aromas were highest in the wines aged in American oak and were lowest in the wines aged in French oak barrels.The concentrations of guaiacol,syringol,trans-isoeugenol,furfural alcohol,vanilla,cis-whisky lactone enabled the medium toasting barrels to be distinguished from the light toasting ones.The compounds originating from the barrels could be used to distinguish the types of different barrels,but the other general grape-derived and fermentation-derived volatiles could not.The fermenters,oak species and toasting degrees of the barrels all had significant effects on the aroma profiles of the aged‘Merlot’dry red wines,but the influence of the geographic origin was not obvious.

The Mechanism of Morin Combined with Celastrol Induces Apoptosis and Inhibits the Growth of Lung Cancer

Morin is a functional flavonoid commonly found in human diet. Compared to being used solely， it is evident that morin can be more effective as a drug adjuvant. However， research on the combined effect and its corresponding mechanism is limited. Here， we found that morin significantly potentiated the inhibitory effects of the natural compound celastrol on the proliferation of lung cancer cells. Morin and celastrol synergistically exhibit marked apoptosis induction in lung cancer cells， accompanied by changes in the abundance of apoptosis-related proteins. Transcriptome analyses revealed that the combination of morin and celastrol had a significant impact on the number of differentially expressed genes in lung cancer cells. Among these genes， BIRC3 was one of the most significantly different ones， which plays a crucial role in the process of tumor resistance to apoptosis. In addition， several genes identified are primarily associated with intracellular signal transduction pathways， specifically the NF-κB signaling pathway. Importantly， the treatment combining morin and celastrol in tumor-bearing mice results in a synergistic effect that significantly suppressed tumor growth. These findings indicate that morin could be a promising functional adjuvant， and the combination of morin and celastrol has potential for the treating lung cancer.

Effects of mesoclimate and microclimate variations mediated by high altitude and row orientation on sucrose metabolism and anthocyanin synthesis in grape berries

Climate change and extreme weather pose significant challenges to the traditional viticulture regions.Emerging high-altitude grape-producing regions with diverse orientations have shown great potential in coping with this challenge.Stable,high-quality wine grape production may be achieved by synchronizing the meso-and microclimate.To clarify the role of high altitude and row orientation in meso-and microclimate and the response of berries to it,we evaluated seven years(2012-2018)of climate data,two years of basic grape(Cabernet Sauvignon,Vitis vinifera L.)quality,and one-year microclimate from veraison to harvest.By comparing two locations(Sidon 2047 m,Sinon 2208 m)in Yunnan Province,China,we found that the average temperature has been stable at approximately 15℃ for seven years,with no extreme weather or,noticeable global warming.The light intensity(LI)in the north-south(NS)was more balanced than the east-west(EW)direction,and the east-west to the south(EW-S)canopy side was almost higher than the other sides.High LI was associated with high photosynthetically active radiation(PAR),ultraviolet(UV),and infrared(IR)light and vice versa.The north-south to the east(NS-E)and east-west to the north(EWN)sides were characterized by lower LI and higher UV and IR light,and higher total anthocyanin content.Most anthocyanin synthesis-related genes,for example,VvF3'H and VvF3'5'H,were highly expressed in NS-E from veraison to maturity.Perhaps UV and IR light induced their expression.This study provides new insights on the role of differently orientated rows in controlling grape quality due to varied light quality.The findings are globally significant,particularly in the context of climate change,and offer fresh insights into berry physiological responses and decision-making for the management of existing vineyards.

The protective effect of cyclodextrin on the color quality and stability of Cabernet Sauvignon red wine

The impact of cyclodextrins(CDs)on wine quality and stability remains largely unknown.This study systematically assessed the protective effect of the post-fermentation addition of CDs on color stability of red wine from the viewpoints of color characteristics,copigmentation and phenolic profiles.The grey relational analysis(GRA)and principal component analysis(PCA)methods were employed to dissect the key effective determinants related to color quality.The addition of CDs induced a significant hyperchromic effect of 8.19-25.40%,a significant bathochromic effect and an enhancement of the color intensity.Furthermore,the evolution of anthocyanin forms and the content of monomeric anthocyanins revealed that β-CD is a superior favorable cofactor during wine aging,but for long-term aging,2-HP-β-CD and 2-HP-γ-CD are more beneficial in promoting the formation of polymerized anthocyanins and color stability.This work provides an important reference for the use of CDs to enhance the color quality and stability of red wines.

Natural product chlorogenic acid achieves pharmacological activity and health protection via regulating gut microbiota

Chlorogenic acid (CA) is a natural plant-derived polyphenol compound that is widely present in beverages (coffee and tea)， fruits (blueberry， apple， and mulberry)， and medicinal plants (Lonicera japonica， Eucommia ulmoides， and Cichorium intybus). In recent years， CA has attracted extensive attention due to its various health benefits， such as anti-inflammatory， immunomodulatory， anti-obesity， anti-diabetes mellitus， and neuroprotection activities. Interestingly， the low bioavailability of CA corresponds to high bioactivity， which raises a key scientific question of how low bioavailability CA can achieve high biological activity. In this review， we highlight the gut microbiota as a key target for connecting the two by summarizing a large amount of evidence. Specifically， the composition and abundance of gut microbiota， its metabolites， intestinal immunity， barrier function and so on were all changed under different pathological conditions such as inflammatory bowel disease， liver disease， kidney disease， cardiovascular disease， diabetes mellitus， obesity， etc. Conversely， these changes could be reversed by CA. In a word， CA could achieve pharmacological activity as well as health-protective effects by modulating intestinal immune and barrier function， gut microbiota composition， gut microbiota metabolites， and signaling pathways. This review provides new targets for the prevention and treatment of diseases by CA. Meanwhile， regulating the composition of the gut microbiota through natural products may be a potential strategy to achieve health protection.

Microbial production of value-added products is a promising alternative to plant-and chemical-based routes[1].However,only few of interested chemicals are producing at bulk scale with microbial cell factories(eg.artemisinic acid)while most are staying at bench level.To enable the microbes toward efficient bioproduction,extensive endeavors have been expended on enzyme exploration and pathway decoration,leaving the role of cofactors,especially the rare cofactors like SAM,FMN(H_(2))and FAD(H_(2)),largely unconsidered[2,3].

Fine-grained classification of grape leaves via a pyramid residual convolution neural network

The value of grape cultivars varies.The use of a mixture of cultivars can negate the benefits of improved cultivars and hamper the protection of genetic resources and the identification of new hybrid cultivars.Classifying cultivars based on their leaves is therefore highly practical.Transplanted grape seedlings take years to bear fruit,but leaves mature in months.Foliar morphology differs among cultivars,so identifying cultivars based on leaves is feasible.Different cultivars,however,can be bred from the same parents,so the leaves of some cultivars can have similar morphologies.In this work,a pyramid residual convolution neural network was developed to classify images of eleven grape cultivars.The model extracts multi-scale feature maps of the leaf images through the convolution layer and enters them into three residual convolution neural networks.Features are fused by adding the value of the convolution kernel feature matrix to enhance the attention on the edge and center regions of the leaves and classify the images.The results indicated that the average accuracy of the model was 92.26%for the proposed leaf dataset.The proposed model is superior to previous models and provides a reliable method for the fine-grained classification and identification of plant cultivars.

Research progress of protein haze in white wines

Protein haze was one of the main causes of the instability of white wines. Proteins that caused haze or precipitation in white wines mainly came from grape fruits, and their compositions and contents were affected by many factors such as fruit diseases, harvesting methods and water stress. Unstable wine proteins were usually pathogenesis-related(PR)proteins of grapes, mainly chitinases and thaumatin-like proteins(TLPs), which had lower isoelectric point(pI)and smaller molecular weight, and were highly resistant to the low pH values of wines and the protease hydrolysis during fermentation. At present, the technology of protein stabilization and clarification in white wines mainly included bentonite fining, heat treatment, enzymatic hydrolysis, polysaccharide treatment and ultrafiltration methods. Among them, the most commonly used method was bentonite treatment. In this paper, the research progresses of the origin, mechanism and influencing factors of the unstable proteins in white wines were summarized, and the applications, advantages and disadvantages of various clarification techniques were also concluded, in order to provide some support for the theoretical and technological research of the protein stability in white wines in the future.

An Adaptive Parameter-Free Optimal Number of Market Segments Estimation Algorithm Based on a New Internal Validity Index

An appropriate optimal number of market segments(ONS)estimation is essential for an enterprise to achieve successful market segmentation,but at present,there is a serious lack of attention to this issue in market segmentation.In our study,an independent adaptive ONS estimation method BWCON-NSDK-means++is proposed by integrating a newinternal validity index(IVI)Between-Within-Connectivity(BWCON)and a newstable clustering algorithmNatural-SDK-means++(NSDK-means++)in a novel way.First,to complete the evaluation dimensions of the existing IVIs,we designed a connectivity formula based on the neighbor relationship and proposed the BWCON by integrating the connectivity with other two commonly considered measures of compactness and separation.Then,considering the stability,number of parameters and clustering performance,we proposed the NSDK-means++to participate in the integrationwhere the natural neighbor was used to optimize the initial cluster centers(ICCs)determination strategy in the SDK-means++.At last,to ensure the objectivity of the estimatedONS,we designed a BWCON-based ONS estimation framework that does not require the user to set any parameters in advance and integrated the NSDK-means++into this framework forming a practical ONS estimation tool BWCON-NSDK-means++.The final experimental results showthat the proposed BWCONand NSDK-means++are significantlymore suitable than their respective existing models to participate in the integration for determining theONS,and the proposed BWCON-NSDK-means++is demonstrably superior to the BWCON-KMA,BWCONMBK,BWCON-KM++,BWCON-RKM++,BWCON-SDKM++,BWCON-Single linkage,BWCON-Complete linkage,BWCON-Average linkage and BWCON-Ward linkage in terms of the ONS estimation.Moreover,as an independentmarket segmentation tool,the BWCON-NSDK-means++also outperforms the existing models with respect to the inter-market differentiation and sub-market size.

VabHLH137 promotes proanthocyanidin and anthocyanin biosynthesis and enhances resistance to Colletotrichum gloeosporioides in grapevine

Proanthocyanidins(PAs)and anthocyanins are involved in the response of plants to various environmental stresses.However,the mechanism behind defense-induced PA biosynthetic regulation is still not completely elucidated,also in grapevine.This study performed a transcriptome sequencing analysis of grape berries infected with Colletotrichum gloeosporioides to highlight the induction of the VabHLH137 factor from the basic helix-loop-helix(bHLH)XII subfamily by the fungus,which appeared to be significantly co-expressedwith PA-related genes.The functional analysis of VabHLH137 overexpression and knockdownin transgenic grape calli showed that it positively regulated PA and anthocyanin biosynthesis.Moreover,VabHLH137 overexpression in the grape calli significantly increased resistance to C.gloeosporioides.A yeast one-hybrid and electrophoretic mobility shift assay revealed that VabHLH137 directly bound to the VaLAR2 promoter,enhancing its activity and interacting with VaMYBPAR,a transcriptional activator of PA biosynthesis.Furthermore,transient experiments showed that although the VabHLH137+VaMYBPAR complex activated VaLAR2 expression,it failed to further enhance VaLAR2 expression compared to VaMYBPAR alone.The findings indicated that VabHLH137 enhanced PA biosynthesis by activating of VaLAR2 expression,providing new insight into the transcriptional regulation of defense-induced PA biosynthesis in grapevine.
