Proteomic characterization of four subtypes of M2 macrophages derived from human THP-1 cells

Macrophages are widely distributed immune cells that contribute to tissue homeostasis.Human THP-1 cells have been widely used in various macrophage-associated studies,especially those involving pro-inflammatory M1 and anti-inflammatory M2 phenotypes.However,the molecular characterization of four M2 subtypes(M2a,M2b,M2c,and M2d)derived from THP-1has not been fully investigated.In this study,we systematically analyzed the protein expression profiles of human THP-1-derived macrophages(M0,M1,M2a,M2b,M2c,and M2d)using quantitative proteomics approaches.The commonly and specially regulated proteins of the four M2 subtypes and their potential biological functions were further investigated.The results showed that M2a and M2b,and M2c and M2d have very similar protein expression profiles.These data could serve as an important resource for studies of macrophages using THP-1 cells,and provide a reference to distinguish different M2 subtypes in macrophage-associated diseases for subsequent clinical research.

Cancer immunotherapy can effectively inhibit cancer progression by activating the autoimmune system, with low toxicity and high effectiveness. Some of cancer immunotherapy had positive effects on clinical cancer treatment. However, cancer immunotherapy is still restricted by cancer heterogeneity, immune cell disability, tumor immunosuppressive microenvironment and systemic immune toxicity. Cell membrane-coated nanoparticles(CMCNs) inherit abundant source cell-relevant functions, including “self” markers, cross-talking with the immune system, biological targeting, and homing to specific regions. These enable them to possess preferred characteristics, including better biological compatibility, weak immunogenicity, immune escaping, a prolonged circulation, and tumor targeting.Therefore, they are applied to precisely deliver drugs and promote the effect of cancer immunotherapy.In the review, we summarize the latest researches of biomimetic CMCNs for cancer immunotherapy,outline the existing specific cancer immune therapies, explore the unique functions and molecular mechanisms of various cell membrane-coated nanoparticles, and analyze the challenges which CMCNs face in clinical translation.

The degradation of tilmicosin(TLM),a semi-synthetic 16-membered macrolide antibiotic,has been receiving increasing attention.Conventionally,there are three tilmicosin degradation methods,and among them microbial degradation is considered the best due to its high efficiency,eco-friendliness,and low cost.Coincidently,we found a new strain,Glutamicibacter nicotianae sp.AT6,capable of degrading high-concentration TLM at 100 mg/L with a 97%removal efficiency.The role of tryptone was as well investigated,and the results revealed that the loading of tryptone had a significant influence on TLM removals.The toxicity assessment indicated that strain AT6 could efficiently convert TLM into less-toxic substances.Based on the identified intermediates,the degradation of TLM by AT6 processing through two distinct pathways was then proposed.

Three-dimensional(3D)ordered porous carbon is generally believed to be a promising electromagnetic wave(EMW)absorbing material.However,most research works targeted performance improvement of 3D ordered porous carbon,and the specific attenuation mechanism is still ambiguous.Therefore,in this work,a novel ultra-light egg-derived porous carbon foam(EDCF)structure has been successfully constructed by a simple carbonization combined with the silica microsphere template-etching process.Based on an equivalent substitute strategy,the influence of pore volume and specific surface area on the electromagnetic parameters and EMW absorption properties of the EDCF products was confirmed respectively by adjusting the addition content and diameter of silica microspheres.As a primary attenuation mode,the dielectric loss originates from the comprehensive effect of conduction loss and polarization loss in S-band and C band,and the value is dominated by polarization loss in X band and Ku band,which is obviously greater than that of conduction loss.Furthermore,in all samples,the largest effective absorption bandwidth of EDCF-3 is 7.12 GHz under the thickness of 2.13 mm with the filling content of approximately 5 wt%,covering the whole Ku band.Meanwhile,the EDCF-7 sample with optimized pore volume and specific surface area achieves minimum reflection loss(RL_(min))of−58.08 dB at 16.86 GHz while the thickness is 1.27 mm.The outstanding research results not only provide a novel insight into enhancement of EMW absorption properties but also clarify the dominant dissipation mechanism for the porous carbon-based absorber from the perspective of objective experiments.

In this paper,the authors extend the Roper-Suffridge operator on the generalized Hartogs domains.They mainly research the properties of the extended operator.By the characteristics of Hartogs domains and the geometric properties of subclasses of spirallike mappings,they obtain the extended Roper-Suffridge operator preserving almost starlikeness of complex orderλ,almost spirallikeness of typeβand orderα,parabolic spirallikeness of typeβand orderρon the Hartogs domains in different conditions.They conclude that the corresponding extension operator preserves the same geometric invariance on the unit ball B^(n) in C^(n).The conclusions provide a new approach to study these geometric mappings in C^(n).

Instance segmentation is an important task in computer vision. In order to enhance the multilevel features expression ability of the segmentation networks, a novel module is proposed in this paper. Firstly,we design a weighted bi-directional feature fusion way by computing the weight distribution function of bi-directional feature pyramid network. Secondly, we propose a joint attention mechanism to effectively filter different levels of feature information by adopting serial and parallel ways to combine the channel attention and spatial attention modules. At the same time, the module uses dynamic convolution to stabilize the calculation speed while improve the 6.7% mean average precision of segmentation.The experiments on the COCO dataset demonstrate that the module can effectively improve the performance of the existing instance segmentation networks.

The effectiveness of modeling contextual information has been empirically shown in numerous computer vision tasks. In this paper, we propose a simple yet efficient augmented fully convolutional network(AugFCN) by aggregating content-and position-based object contexts for semantic segmentation.Specifically, motivated because each deep feature map is a global, class-wise representation of the input,we first propose an augmented nonlocal interaction(AugNI) to aggregate the global content-based contexts through all feature map interactions. Compared to classical position-wise approaches, AugNI is more efficient. Moreover, to eliminate permutation equivariance and maintain translation equivariance, a learnable,relative position embedding branch is then supportably installed in AugNI to capture the global positionbased contexts. AugFCN is built on a fully convolutional network as the backbone by deploying AugNI before the segmentation head network. Experimental results on two challenging benchmarks verify that AugFCN can achieve a competitive 45.38% mIoU(standard mean intersection over union) and 81.9% mIoU on the ADE20K val set and Cityscapes test set, respectively, with little computational overhead. Additionally, the results of the joint implementation of AugNI and existing context modeling schemes show that AugFCN leads to continuous segmentation improvements in state-of-the-art context modeling. We finally achieve a top performance of 45.43% mIoU on the ADE20K val set and 83.0% mIoU on the Cityscapes test set.

In this paper,the distributed optimization problem is investigated for a class of general nonlinear model-free multi-agent systems.The dynamical model of each agent is unknown and only the input/output data are available.A model-free adaptive control method is employed,by which the original unknown nonlinear system is equivalently converted into a dynamic linearized model.An event-triggered consensus scheme is developed to guarantee that the consensus error of the outputs of all agents is convergent.Then,by means of the distributed gradient descent method,a novel event-triggered model-free adaptive distributed optimization algorithm is put forward.Sufficient conditions are established to ensure the consensus and optimality of the addressed system.Finally,simulation results are provided to validate the effectiveness of the proposed approach.
