Cloning and characterization of a potato StAN11 gene involved in anthocyanin biosynthesis regulation

Anthocyanins are a class of products of plant secondary metabolism and are responsible for tubers color in potato. The biosynthesis of anthocyanins is a complex biological process, in which multiple genes are involved including structural genes and regulatory genes. In this study, StAN11, a WD4o-repeat gene, was cloned from potato cultivar Chieftain （Solanum tuberosum L.）. StAN11 （HQ5995o6） contained no intron and its open reading frame （ORF） was 1,o29 bp long, encoding a putative protein of 342 amino acids. In order to verify its role in anthocyanin biosynthesis, StAN11 was inserted behind the CaMV-35S promoter of pCMBIA1304 and the recombination vector was introduced into the potato cultivar Desiree plants by Agrobacterium-mediated transfor- mation. The color of transgenic tuber skin was significantly deepened, compared to the wild-type control, which was highly consistent with the accumulation of anthocyanin and expression of StAN11 in transgenic lines tuber skin. Further analysis on the expression of Flavonone-3-hydroxylase （F3H）,Dihydroflavonol reductase （DFR）, Anthocyanidin synthase （ANS）, and F1avonoid 3-O-glucosyl transferase （3GT） in transgenic plants revealed that only DFR was upregulated. This result suggested that StAN11 regulated anthocyanin biosynthesis in potato by controlling DFR expression and accumulation of anthocyanin could be increased through overexpression of .StAN11 in the tubers with the genetic background of anthocyanin biosynthesis.

Muscle architecture of the forelimb of the Golden Pheasant(Chrysolophus pictus)(Aves: Phasianidae)and its implications for functional capacity in flight

Background: Flight is the central avian adaptation in evolution. Wing muscles form an important anatomical basis for avian flight, affecting wing performance and determine modes of flight. However, the roles of distal muscles in adjusting the wing, as well as their functional specializations, remain largely unknown. The importance of muscle fiber architecture has long been recognized. In this study, we provide quantitative anatomical data on the muscle architecture of the forelimb of the Golden Pheasant(Chrysolophus pictus), with an emphasis on brachial,antebrachial and manual ***: The forelimbs of five Golden Pheasants were dissected and detailed measurements of all muscles were made, including muscle mass, muscle belly length, fascicle length. From these values, muscle volume, physiological cross-sectional area(PCSA) and maximum isometric force were ***: General trends such as the distribution of muscle mass, fascicle length and the ratio of tendon length/belly length are revealed. Comparing PCSAs between antebrachial depressors and elevators and between intrinsics of the alular digit and major digit yielded significant differences(p < 0.05). Pronounced development of the antebrachial depressors suggests that ventral rotation of the distal half of the wing is a pivotal factor in shape change and orientation modulation. Large PCSAs in tandem with the force generation capability of the major digit intrinsics may help stabilize the digits while enhancing support of the primary feathers. The architectural properties of the alular digit confirm that alular adjustment is essential to rapid adduction and ***: These observations illustrate the underlying structural basis for the functional capacities of the distal forelimb muscles and may provide additional information useful in further biomechanical and in vivo investigations.

The Clock gene clone and its circadian rhythms in Pelteobagrus vachelli

The Clock gene,a key molecule in circadian systems,is widely distributed in the animal kingdom. We isolated a 936-bp partial c DNA sequence of the C lock gene( Pva- clock) from the darkbarbel catfish P elteobagrus vachelli that exhibited high identity with C lock genes of other species of fish and animals(65%–88%). The putative domains included a basic helix-loop-helix(b HLH) domain and two period-ARNT-single-minded(PAS) domains,which were also similar to those in other species of fish and animals. P va- Clock was primarily expressed in the brain,and was detected in all of the peripheral tissues sampled. Additionally,the pattern of P va- Clock expression over a 24-h period exhibited a circadian rhythm in the brain,liver and intestine,with the acrophase at zeitgeber time 21:35,23:00,and 23:23,respectively. Our results provide insight into the function of the molecular C lock of P. vachelli.

A New Axymyiid Genus with Two New Species from the Middle Jurassic of China (Diptera: Nematocera: Axymyiidae)

The Axymyiidae is one of the small families of the suborder Nematocera within the Diptera. Up to date, three genera and seven species of extant axymyiids have been described from the Holarctic Region, including Canada, China, Hungary, Japan, Russia, and USA, with three genera and three species of fossil taxa described from the Middle Jurassic of Daohugou, northeastern China. A new genus Raraxymyia gen. nov. and two new species, R. parallela and R. proxima gen. et sp. nov. from the Middle Jurassic Jiulongshan Formation of Daohugou Village, Inner Mongolia, China, are herein described based on their well-preserved wings and body characters. A revised key to the genera of axymyiids is provided.

Seed bank and regeneration dynamics of Emmenopterys henryi population on the western side of Wuyi Mountain,South China

Emmenopterys henryi Oliv. (Rubiaceae) is an endangered tree species that is native to China. The wild populations of E. henryi have declined rapidly because of its poor natural regeneration, but the actual regeneration processes are not yet understood. In field tracking surveys and experiments in the Wuyishan Nature Reserve in Southeast China to determine the most important stage that affects the regeneration process, seed bank characteristics, seed germination, and seedling growth dynamics were studied in the typical habitats of E. henryi, Phyllostachys pubescens and broad-leaved forests. Results showed that in both P. pubescens and broad-leaved forests, more than 70% of the E. henryi seeds were distributed in moss and litter layers, and few were found in the soil beneath them. However, seed germination in the soil layer was significantly higher than in the moss and litter layers. Seed density, overall seed quality, and germination rate in the broad-leaved forest were significantly higher than in P. pubescens forest. Seed germination was highest in the microsites around the edge of the crown projection area of E. henryi mother trees. The order of survival rate of the seedlings on different ground surfaces was soil > moss > litter. In both habitats, the average seed density was 24.9 seeds m(-2), and the total germination rate was less than 3.5 parts per thousand. However, seedlings developed from only 1% of the germinated seeds, indicating that the seed germination is the most important stage in the natural regeneration of E. henryi.

Three well-preserved fossil Pelecinids from the Late Mesozoic Yixian Formation,Liaoning Province,China are described and assigned to two new species,Eopelecinus huangi *** Eopelecinus tumidus *** Eopelecinus Zhang,Rasnitsyn and Zhang,*** of now,17 species have been included in this genus,which is the most diverse in the Pelecinidae *** new information and characters obtained from these new specimens,the diagnosis of Eopelecinus Zhang, Rasnitsyn and Zhang,2002 is emended.

Physiological and metabolic processes of ectotherms are markedly influenced by ambient temperature. Previous studies have shown that the abdominal black-speckled area becomes larger with increased elevation in plateau Phrynocephalus, however, no studies have verified the hypothesis that this variation is correlated with the lizard＇s thermoregulation. In this study, infrared thermal imaging technology was first used to study the skin temperature variation of torsos, heads, limbs and tails of a cold-climate agamid lizard, Phrynocephalus guinanensis. The heating rates of the central abdominal black-speckled skin area and peripheral non-black-speckled skin area under solar radiation were compared. Our results showed that the heating rates of limbs and tails were relatively faster than the torsos, as heating time was extended, rates gradually slowed before stabilizing under solar radiation. Under the environment without solar radiation, the cooling rates of limbs and tails were also relatively faster than the torsos of lizards, the rates slowed down and finally became stable as the cooling time was extended. We also found that the heating rate of the abdominal black-speckled skin area was faster than the nearby non-black-speckled skin area. These results increased our insights into the functional significance of these phenotypic traits and help explain their covariation with the thermal environment in these cold-climate agamid lizards.

Dendrobium species and their corresponding medicinal slices have been extensively used as traditional Chinese medicine(TCM) in many Asian countries. However, it is extremely difficult to identify Dendrobium species based on their morphological and chemical features. In this study, the plastomes of D. huoshanense were used as a model system to investigate the hypothesis that plastomic mutational hotspot regions could provide a useful single nucleotide variants(SNVs) resource for authentication studies. We surveyed the plastomes of 17 Dendrobium species, including the newly sequenced plastome of D. huoshanense. A total of 19 SNVs that could be used for the authentication of D. huoshanense were detected. On the basis of this comprehensive comparison, we identified the four most informative hotspot regions in the Dendrobium plastome that encompass ccsA to ndhF, matK to 3′trnG, rpoB to psbD, and trnT to rbcL. Furthermore, to established a simple and accurate method for the authentication of D. huoshanense and its medicinal slices, a total of 127 samples from 20 Dendrobium species including their corresponding medicinal slices(Fengdous) were used in this study. Our results suggest that D. huoshanense and its medicinal slices can be rapidly and unequivocally identified using this method that combines real-time PCR with the amplification refractory mutation system(ARMS).

Anthocyanin accumulation is a common phenomenon seen in plants under environmental stress. In this study,we identified a new allele of ROOT HAIR DEFECTIVE3（RHD3）showing an anthocyanin overaccumulation phenotype under nitrogen starvation conditions. It is known that ethylene negatively regulates light- and sucrose-induced anthocyanin biosynthesis. We hypothesized that RHD3 achieves its negative effect on anthocyanin biosynthesis via an ethylene-regulating pathway. In support of this, similar to rhd3 mutants, the Arabidopsis ethylene signaling mutants etr1, ein2, and ein3/eil1 showed an anthocyanin overaccumulation phenotype under nitrogen starvation conditions. The ethylene precursor ACC strongly suppressed anthocyanin accumulation, dependent on ETR1, EIN2, EIN3/EIL1, and, partially, RHD3. In addition,inactivating RHD3 partially reversed the suppressive effect of ETO1 inactivation-evoked endogenous ethylene production on anthocyanin accumulation. The expression of nitrogen starvation-induced anthocyanin biosynthesis genes was negatively regulated by RHD3, but ethylene response genes were Researchpositively regulated by RHD3. Wild-type seedlings overexpressing RHD3 showed similar phenotypes to rhd3 mutants,indicating the existence of a fine-tuned relationship between gene expression and function. RHD3 was initially identified as a gene involved in root hair development. This study uncovered a new physiological function of RHD3 in nitrogen starvationinduced anthocyanin accumulation and ethylene homeostasis.
