The temporal-spatial expression and functional analysis of three gustatory receptor genes in Solenopsis invicta using sweet and bitter compounds

The insect gustatory system participates in identifying potential food sources and avoiding toxic *** this process,gustatory receptors(GRs)recognize feeding stimulant and deterrent ***,the GRs involved in recognizing stimulant and deterrent compounds in the red imported fire ant,Solenopsis invicta,remain ***,we conducted a study on the genes SinvGR1,SinvGR32b,and SinvGR28a to investigate the roles of GRs in detecting feeding stimulant and deterrent *** this current study,we found that sucrose and fructose are feeding stimulants and the bitter compound quinine is a feeding *** fire ant workers showed significant behavior changes to avoid the bitter taste in feeding stimulant *** transcription quantitative real-time polymerase chain reaction results from developmental stages showed that the SinvGR1,SinvGR32b,and SinvGR28a genes were highly expressed in fire ant ***-specific expression profiles indicated that SinvGR1,SinvGR32b,and SinvGR28a were specifically expressed in the antennae and foreleg tarsi of workers,whereas SinvGR32b gene transcripts were also highly accumulated in the male ***,the silencing of SinvGRI or SinvGR32b alone and the co-silencing of both genes disrupted worker stimulation and feeding on sucrose and *** results also showed that SinvGR28a is required for avoiding quinine,as workers with knockdown of the SinvGR28a gene failed to avoid and fed on *** study first identified stimulant and deterrent compounds of fire ant workers and then the GRs involved in the taste recognition of these *** study could provide potential target gustatory genes for the control of the fire ant.

Induced CTL-S15 gene expression by Bacillus thuringiensis declines susceptibility in Spodoptera exigua

It has been reported that C-type lectins(CTLs),which are pattern recognition receptors of the insect innate immunity response,may compete with Cry toxin for the receptor alkaline phosphatase to decrease its toxicity in ***,to date,which CTLs affect larval susceptibility to Bt in Spodoptera exigua is not *** this study,33 CTL genes were identified from *** on the number of carbohydrate-recognition domains(CRDs)and the domain architectures,they were classified into three groups:(1)nineteen CTL-S(single-CRD),(2)eight immulectin(dual-CRD)and(3)six CTL-X(CRD with other domains).RT-qPCR analysis revealed that expression levels of SeCTL-S15,IML-4 and CTL-X6 were upregulated after challenge with Bt and *** and developmental stage expression analysis showed that only SeCTL-S15 was mainly expressed in the midgut and larva,*** of SeCTL-S15 significantly increased Bt susceptibility,as indicated by reduced survival and larval *** results suggest that CTL-S15 might play a vital role in the low susceptibility of larvae to Bt in *** results provide new insights into CTL function in insects.

The biosynthesis of alarm pheromone in the wheat aphid Rhopalosiphum padi is regulated by hormones via fatty acid metabolism

Aphids are major insect pests in agriculture and forestry *** attacks by natural enemies,many aphids release an alarm pheromone to protect their *** most aphids,the main component of the aphid alarm pheromone(AAP)is the sesquiterpene hydrocarbon(E)-β-farnesene(EβF).However,the mechanisms behind its biosynthesis and regulation remain poorly *** this study,we used the bird cherry–oat aphid Rhopalosiphum padi,which is an important wheat aphid,to investigate the regulatory mechanisms of EβF *** results showed that EβF biosynthesis occurs during the mature embryo period and the molting period of the 1st-and 2nd-instar *** provide the prerequisite material for EβF production and *** on transcriptome sequencing,RNAi analysis,hormone treatments,and quantitative measurements,we found that the biosynthesis of EβF utilizes acetyl coenzyme A produced from fatty acid degradation,which can be suppressed by juvenile hormone but it is promoted by 20-hydroxyecdysone through the modulation of fatty acid *** is the first systemic study on the modulation of EβF production in *** results of our study provide insights into the molecular regulatory mechanisms of AAP biosynthesis,as well as valuable information for designing potential aphid control strategies.

Sex pheromones are considered to play critical roles in partner communication of most parasitic ***,the identification of sex pheromone components remains limited to a few families of parasitoid *** this study,we functionally characterized a candidate sex pheromone component in Microplitis mediator(Hymenoptera:Braconidae),a solitary parasitoid of Noctuidae *** found that the body surface extract from female wasps could significantly stimulate courtship behavior of *** chromatography-electroantennographic detection(GC-EAD)analysis revealed that a candidate semiochemical from extract triggered significant electrophysiological response of antennae of *** performing gas chromatography-mass spectrometer(GC-MS)measurement,GC-EAD active compound was identified as n-octyl acrylate,a candidate sex pheromone component in female *** electroantennogram(EAG)tests,antennae of male wasps showed significantly higher electrophysiological responses to n-octyl acrylate than those of females.Y-tube olfactometer assays indicated that male wasps significantly chose n-octyl acrylate compared with the ***,male wasps showed a remarkable preference for n-octyl acrylate in a simulated field condition behavioral trial;simultaneously,n-octyl acrylate standard could also trigger significant courtship behavior in *** propose that n-octyl acrylate,as a candidate vital sex pheromone component,could be utilized to design behavioral regulators of *** to implement the protection and utilization of natural enemies.

Pre-mating nitenpyram exposure in male mice leads to depression-like behavior in offspring by affecting tryptophan metabolism in gut microbiota

Several studies have confirmed that the health status of the paternal affects the health of the offspring,however,it remains unknown whether paternal exposure to pesticides affect the offspring ***,we used untargeted metabolomics and 16S rRNA sequencing technology,combined with tail suspension test and RT-qPCR to explore the effects of paternal exposure to nitenpyram on the neurotoxicity of *** results found that the paternal exposure to nitenpyram led to the offspring’s depressive-like behaviors,accompanied by the reduction of tryptophan content and the disorder of microbial abundance in the gut of the ***,we determined the expression of tryptophan metabolism-related genes tryptophanase(tnaA)and tryptophan hydroxylase 1(TpH1)in gut bacteria and colonic *** found that tryptophan is metabolized to indoles rather than being absorbed into colonocytes,which coursed the reduce of tryptophan availability after nitenpyram *** conclusion,our study deepens our understanding of the intergenerational toxic effects of pesticides.

Sensitivity and resistance risk analysis of Didymella bryoniae populations to fluopyram

Fluopyram is an succinate dehydrogenase inhibitors(SDHI)fungicide that has been registered in China to control gummy stem blight(GSB)in watermelons for many ***,whether the field pathogens of GSB are still sensitive to fluopyram or not is ***,we collected 69 Didymella bryoniae isolates from the fields that usually use fluopyram to control GSB to determine the sensitivity *** EC_(50)(50%inhibition effect)values of fluopyram against *** ranged from 0.0691 to 0.3503μg mL^(–1) and the variation factor was *** mean EC_(50) value was(0.1579±0.0669)μg mL^(–1) and the curve of sensitivity was *** resistant strains were found in the isolates,which means that the pathogens were still sensitive to *** minimal inhibition concentration(MIC)of fluopyram against *** was 3μg mL^(–1).Four low-resistant mutants and two medium-resistant mutants were obtained using fungicide taming and the resistance of mutants could be inherited *** growth rate of mutants decreased significantly compared with that of wild-type strains while the biomass of most mutants was similar to that of wild-type *** sensitivity of most resistant mutants to various stresses was increased compared with that of wild-type *** virulence of mutants receded except for low-resistant mutant XN51FR-1,which had the same lesion area as XN51 on the watermelon *** results indicated that the fitness of resistant mutants was decreased compared with that of wild-type *** cross-resistance assay indicated that fluopyram-resistant mutants were positive cross-resistant to all six SDHI fungicides in this test but were still sensitive to fluazinam and *** the resistance risk of *** to fluopyram was *** addition,we found that the SdhB gene of low-resistant mutant XN30FR-1 had three new point mutations at positions K258N,A259P,and ***-resistant mutant XN52FR-1 showed a mutation at position H277N an

A viral movement protein targets host catalases for 26S proteasome-mediated degradation to facilitate viral infection and aphid transmission in wheat

The infection of host plants by many different viruses causes reactive oxygen species(Ros)accumulation and yellowing symptoms,but the mechanisms through which plant viruses counteract RoS-mediated immunity to facilitate infection and symptom development have not been fully *** plant viruses are transmitted by insect vectors in the field,but the molecular mechanisms underlying virus-host-insect interactions are *** this study,we investigated the interactions among wheat,barley yellow dwarf virus(BYDV),and its aphid vector and found that the BYDV movement protein(MP)interacts with both wheat catalases(CATs)and the 26S proteasomeubiquitin receptor non-ATPase regulatorysubunit2homolog(PSMD2)to facilitate the 26S proteasome-mediateddegradation of CATs,promotingviral infection,disease symptom development,and aphid *** of the BYDV MP gene in wheat enhanced the degradation of CATs,which leading to increased accumulation of ROS and thereby enhanced viral ***,transgenic wheat lines overexpressing BYDV MP showed significantly reduced proliferation of wingless aphids and an increased number of winged *** with this observation,silencing of CAT genes also enhanced viral accumulation and reduced the proliferation of wingless aphids but increased the occurrence of winged *** contrast,transgenic wheat plants overexpressing TaCAT1 exhibited the opposite changes and showed increases in grain size and weight upon infection with *** assays demonstrated that BYDV MP interacts with PSMD2 and promotes 26S proteasome-mediated degradation of TaCAT1 likely in a ubiquitination-independent ***,our study reveals a molecular mechanism by which a plant virus manipulates the Ros production system of host plants to facilitate viral infection and transmission,shedding new light on the sophisticated interactions among viruses,host plants,and insect vectors.

Juvenile hormone induces reproduction via miR-1175-3p in the red imported fire ant, Solenopsis invicta

Juvenile hormone(JH)acts in the regulation of caste differentiation between queens and workers(i.e.,with or without reproductive capacity)during vitellin synthesis and oogenesis in social ***,the regulatory mechanisms have not yet been ***,we identified a highly expressed microRNA(miRNA),miR-1175-3p,in the red imported fire ant,Solenopsis *** found that miR-1175-3p is prominently present in the fat bodies and ovaries of ***,miR-1175-3p interacts with its target gene,broad-complex core(Br-C),in the fat *** utilizing miR-1175-3p agomir,we successfully suppressed the expression of the Br-C protein in queens,resulting in reduced vitellogenin expression,fewer eggs,and poorly developed ***,decreasing miR-1175-3p levels led to the increased expression of Br-C and vitellogenin in workers,triggering the“re-development”of the ***,when queens were fed with JH,the expression of miR-1175-3p decreased,whereas the expression of vitellogenin-2 and vitellogenin-3 ***,the suppression of fertility in queens caused by treatment with agomir miR-1175-3p was completely rescued by the increased vitellogenin expression induced by being fed with *** results suggest the critical role of miR-1175-3p in JH-regulated reproduction,shedding light on the molecular mechanism underlying miRNA-mediated fecundity in social insects and providing a novel strategy for managing ***.

Both host and diet shape bacterial communities of predatory mites

Microbial communities,derived from food,ambient,and inner,can affect host ecological adaption and *** with most phytophagous arthropods,predators may have more opportunities to develop specific microbiota depending on the level of prey *** explore how diet sources affect host microbial communities and vary across predator species,we considered 3 types of predators from Phytoseiidae(Acari:Mesostigmata):polyphagous(Amblyseius orientalis Ehara,Neoseiulus barkeri Hughes,and Amblyseius swirski Athias-Henrio),oligophagous(Neoseiulus californicus McGregor),and monophagous(Phytoseiulus persimilis Athias-Henriot)predatory *** polyphagous species were fed on 2 types of diets,natural prey and alternative *** using 16S rRNA sequencing,we found that diet was the main source of microbiota in predatory mites,while there was no clear pattern affected by prey *** 3 polyphagous predators,host species had a larger impact than prey on microbial *** *** or *** which showed consistency in their microbiota,prey switching significantly affectedβ-diversity of bacterial composition in ***,with 56%of the microbial *** short,our results confirmed the substantial influence of diet on host microbial construction in predatory species,and highlighted species differences in shaping the microbiota which are not necessarily related to prey specialization.

Synergistic effects of the entomopathogenic fungus Isaria javanica and low doses of dinotefuran on the efficient control of the rice pest Sogatella furcifera

The rice planthopper,Sogatella furcifera,is a piercing-sucking insect pest of rice,Oryza *** is responsible for significant crop yield losses,and has developed moderate to high resistance to several commonly used chemical *** investigated the effects of the insect fungal pathogen Isaria javanica,alone and in combination with the chemical insecticide dinotefuran,on *** under both laboratory and field *** results show that *** displays high infection efficiency and mortality for different stages of ***,reducing adult survival,female oviposition and ovary *** bioassays showed that the combined use of *** with a low dose(4-16 mg L^(-1))of dinotefuran resulted in higher mortality in *** than the use of *** or dinotefuran *** combined treatment also had more significant effects on several host enzymes,including superoxide dismutase,catalase,peroxidase,and prophenol oxidase *** field trials,*** effectively suppressed populations of rice planthoppers to low levels(22-64%of the level in untreated plots).Additional field experiments showed synergistic effects,i.e.,enhanced efficiency,for the control of *** populations using the combination of a low dose of ***(1×10^(4) conidia mL^(-1))and a low dose of dinotefuran(~4.8-19.2%of normal field use levels),with control effects of>90%and a population level under 50 insects per 100 hills at 3-14 days *** findings indicate that the entomogenous fungus *** offers an attractive biological control addition as part of the integrated pest management(IPM)practices for the control of rice plant pests.
