Transient chemical-mediated epigenetic modulation confers unrestricted lineage potential on human primed pluripotent stem cells

Human primed pluripotent stem cells are capable of generating all the embryonic ***,their extraembryonic trophectoderm potentials are *** remains unclear how to expand their developmental potential to trophectoderm *** we show that transient treatment with a cocktail of small molecule epigenetic modulators imparts trophectoderm lineage potentials to human primed pluripotent stem cells while preserving their embryonic *** chemically treated cells can generate trophectoderm-like cells and downstream trophoblast stem cells,diverging into syncytiotrophoblast and extravillous trophoblast *** and CUT&Tag analyses reveal that these induced cells share transcriptional profiles with in vivo trophectoderm and cytotrophoblast,and exhibit reduced H3K27me3 modification at gene loci specific to trophoblast lineages compared with primed pluripotent *** exploration highlighted the critical roles of epigenetic modulators HDAC2,EZH1/2,and KDM5s in the activation of trophoblast lineage *** findings demonstrate that transient epigenetic resetting activates unrestricted lineage potential in human primed pluripotent stem cells,and offer new mechanistic insights into human trophoblast lineage specification and in vitro models for studying placental development and related disorders.

We study the influence of screening effect on quantum decoherence for charge qubit and the process of quantum information storage. When the flux produced by the circulating current in SQUID loop is considered, screening effect is formally characterized by a LC resonator. Using large-detuning condition and Fr6hlich transformation in the qubit-cavity-resonator system, we calculate the decoherenee factor for charge qubit and the effective qubit-cavity Hamiltonian. The decoherence factor owns a factorized structure, it shows that screening effect is a resource of decoherence for charge qubit. The effective Hamiltonian shows that the screening effect results in a frequency shift for charge qubit and a modified qubit-cavity coupling constant induced by a LC resonator.

When the nonlinearity of nanomechanical resonator is not negligible, the quantum decoherenee of charge qubit is studied analytically. Using nonlinear Jaynes-Cummings model, one explores the possibility of being quantum data bus for nonlinear nanomechanical resonator, the nonlinearity destroys the dynamical quantum information-storage and maintains the revival of quantum coherence of charge qubit. With the calculation of decoherence factor, we demon- strate the influence of the nonlinearity of nanomechanical resonator on engineered decoherence of charge qubit.

The ketogenic diet(KD) has attracted attention in recent years for its potential anticancer effects. KD is a dietary structure of high fat,moderate protein, and extremely low carbohydrate content. Originally introduced as a treatment for epilepsy, KD has been widely applied in weight loss programs and the management of metabolic diseases. Previous studies have shown that KD can potentially inhibit the growth and spread of cancer by limiting energy supply to tumor cells, thereby inhibiting tumor angiogenesis, reducing oxidative stress in normal cells, and affecting cancer cell signaling and other processes. Moreover, KD has been shown to influence T-cell-mediated immune responses and inflammation by modulating the gut microbiota, enhance the efficacy of standard cancer treatments, and mitigate the complications of chemotherapy. However, controversies and uncertainties remain regarding the specific mechanisms and clinical effects of KD as an adjunctive therapy for cancer. Therefore, this review summarizes the existing research and explores the intricate relationships between KD and cancer treatment.

A new necessary conditions for separability of mixed states in bipartite and multipartite quantum systems of arbitrary dimensions are presented. This method requires only a few local measurements. If a quantum state in a bipartite （or multipartite） system of arbitrary dimension is separable, we show that the trace norm of a constructive matrix is less than or equal to 1.

The optical erasure dynamics in a batch-thermal fixing scheme of holographic storage in photorefractive crystals is investigated theoretically and experimentally. The inter-batch optical erasure time constant τF is introduced to specify the optical erasure to compensated gratings, and measured in a sophisticated experiment. The experimental result shows that τF is much longer than the intra-batch optical erasure time constant τE. The difference between τFand τE is fundamental for enhancing nonvolatile storage density.

Various random models with balanced data that are relevant for analyzing practical test data are described, along with several hypothesis testing and interval estimation problems concerning variance components. In this paper, we mainly consider these problems in general random effect model with balanced data. Exact tests and confidence intervals for a single variance component corresponding to random effect are developed by using generalized p-values and generalized confidence intervals. The resulting procedures are easy to compute and are applicable to small samples. Exact tests and confidence intervals are also established for comparing the random-effects variance components and the sum of random-effects variance components in two independent general random effect models with balanced data. Furthermore, we investigate the statistical properties of the resulting tests. Finally, some simulation results on the type Ⅰ error probability and power of the proposed test are reported. The simulation results indicate that exact test is extremely satisfactory for controlling type Ⅰ error probability.

We perform a comprehensive experimental study on holographic and thermal-fixing characteristics of Zn：Fe：LiNbO3 The measured hologram decay time constants, respectively caused by optical readout and under dark condition, with thermal fixing are 15 times and 75 times longer than that obtained without thermal fixing. This suggests that Zn：Fe：LiNbO3 crystals are suitable for thermal fixing. Multiplexed recordings of 300 holograms using different thermal fixing schemes verify that a proper multiplexing scheme such as track-division thermal-fixing scheme for disc-type holographic storage can effectively compensate for the negative effect of zinc-doping on the dynamic range, thus the storage density is enhanced.
