Erythropoiesis is a process during which multipotential hematopoietic stem cells proliferate, differentiate and eventually form mature erythrocytes. Interestingly, unlike most cell types, an important feature of erythropoiesis is that following each mitosis the daughter cells are morphologically and functionally different from the parent cell from which they are derived, demonstrating the need to study erythropoiesis in a stage-specific manner. This has been impossible until recently due to lack of methods for isolating erythroid cells at each distinct developmental stage. This review summarizes recent advances in the development of methods for isolating both murine and human erythroid cells and their applications. These methods provide powerful means for studying normal and impaired erythropoiesis associated with hematological disorders.

Two new species of free-living marine nematodes of the genera Hopperia and Nemanema are described from the sublittoral sediment of the East China Sea. Hopperia macramphida sp. nov. is characterized by having large amphidial fovea with five turns; arcuate spicules with enlarged proximal end and central internal lamella extending one third of spicule length from proximal end; gubernaculum with bent apophyses, six papilliform precloacal supplements; and conico-cylindrical tail with swollen tip and terminal setae. Nernanema minutum sp, nov. is characterized by relatively smaller body size in the genus; large oval amphidial fovea; conical tail; slender spicules with alae and hooked proximal end; ring-like gubernaculum; and papilliform precloacal supplements with three short setae. An updated dichotomous key is provided for the 17 species of Hopperia.

Somatic Embryogenesis Receptor Kinases Control Root Development Mainly via Brassinosteroid-Independent Actions in Arabidopsis thaliana

Brassinosteroids （BRs）, a group of plant steroidal hormones, play critical roles in many aspects of plant growth and development. Previous studies showed that BRI1-mediated BR signaling regulates cell division and differentiation during Arabidopsis root development via interplaying with auxin and other phytohormones. Arabidopsis somatic embryogenesis receptor-like kinases （SERKs）, as co-receptors of BRI1, were found to play a fundamental role in an early activation step of BR signaling pathway. Here we report a novel function of SERKs in regulating Arabidopsis root development. Genetic analyses indicated that SERKs control root growth mainly via a BR-independent pathway. Although BR signaling pathway is completely disrupted in the serkl bakl bkkl triple mutant, the root growth of the triple mutant is much severely damaged than the BR deficiency or signaling null mutants. More detailed analyses indicated that the triple mutant exhibited drastically reduced expression of a number of genes critical to polar auxin transport, cell cycle, endodermis development and root meristem differentiation, which were not observed in null BR biosynthesis mutant cpd and null BR signaling mutant bril-701.

Genetic diversity analysis with ISSR PCR on green algae Chlorella vulgaris and Chlorella pyrenoidosa

In the present study, genetic polymorphism and diversity in unicellular clones of Chlorella vulgaris Beijerinck and Chlorella pyrenoidosa Chick were studied with Inter Simple Sequence Repeats PCR （ISSR PCR）. Samples including four clones of C. vulgaris and three clones of C. pyrenoidosa were purified by single-clone-choice method. For four C. vulgaris unicellular clones, the total number of the bands scored for 18 primers was 298; and the number of the polymorphic bands was 118, of which 39.6% were polymorphic. The size of PCR products ranged from 200 to 2 500 bp. The total number of bands scored for 18 primers, the number of polymorphic bands and the percentage of three C. pyrenoidosa unicellular clones was 194.83 and 30.8%, respectively. POPGENE analysis show that the average Nei genetic diversity （h^＊） and Shannon index of diversity （I^＊） in the four C. vulgaris unicellular clones was 0.2181 and 0.3208, respectively, which is slightly higher than those of the three C. pyrenoidosa unicellular clones （0.190 3 and 0.274 8）, which agreed with the percentage of polymorphic bands in the mixed samples of the two species. The results suggest that ISSR is a useful method to Chlorella for intra-species genetic analysis.

Transgenesis and genome editing in birds are based on a unique germline transmission system using primordial germ cells(PGCs), which is quite different from the mammalian transgenic and genome editing system. PGCs are progenitor cells of gametes that can deliver genetic information to the next generation. Since avian PGCs were first discovered in nineteenth century, there have been numerous efforts to reveal their origin, specification, and unique migration pattern, and to improve germline transmission efficiency. Recent advances in the isolation and in vitro culture of avian PGCs with genetic manipulation and genome editing tools enable the development of valuable avian models that were unavailable before. However, many challenges remain in the production of transgenic and genome-edited birds,including the precise control of germline transmission, introduction of exogenous genes, and genome editing in ***, establishing reliable germline-competent PGCs and applying precise genome editing systems are critical current issues in the production of avian models. Here, we introduce a historical overview of avian PGCs and their application, including improved techniques and methodologies in the production of transgenic and genome-edited birds, and we discuss the future potential applications of transgenic and genome-edited birds to provide opportunities and benefits for humans.

Two new species of the genus Wieseria (Nematoda:Enoplaida:Oxystominidae) from the Jiaozhou Bay

Two new species of free-living marine nematodes of the genus Wieseria are described from the sublittoral sediment of the Jiaozhou Bay in Qingdao. Wieseria sinica sp. nov. is characterized by having short labial and cephalic setae in a backward direction; oblong amphidial foveas with double contour; arcuate spicules with alea; ring-like guberculum; and clavated tail. Wieseria tenuisa sp. nov. is characterized by its very large ＂a＂ value （133.6）, long labial setae in a forward direction; spicules with alea and hooked proximal end; rodlike gubernaculums; and relatively longer clavated tail （7.9 a.b.d, long）. It differs from Wieseria sinica sp. nov. mainly by the longer labial setae, slightly straight spicules with hooked proximal end, and its thinner body. Amended dichotomous key to male species of Wieseria is provided.

Heavy metal accumulation of urban domestic rubbish compost in turfgrass by EDTA chelating

Seven kinds of heavy metal accumulation of rubbish compost in turfgrass by EDTA chelating were investigated. With EDTA application, heavy metal accumulation by two species of turfgrass was increased significantly. The enrichment coefficients of Lolium perenne L（L） and Festuca arundinacea L（F） to Cr reached 9.4.5 and 6.15 respectively. In the range of EDTA dosages given, heavy metal accumulation in turfgrass increased with increasing EDTA level. There were significant differences in remediation of different metals by applying EDTA. L had high ability to accumulate Cr, Cd, Ni and Zn, showing better remediation to heavy metals of rubbish compost. In contrast, F showed high ability to accumulate Cr, Ni, Cu and Zn. Low EDTA level increased aboveground net primary production （ANP） of turfgrass, but EDTA would considerably inhibit it when EDTA was higher than 20 mmol/kg. The results demonstrated that the optimum dosage of EDTA for remediating heavy metals in rubbish compost by turfgrass was between 10 mmol/kg and 20 mmol/kg.

Background： Primordial germ cells（PGCs）, the precursors of functional gametes, have distinct characteristics and exhibit several unique molecular mechanisms to maintain pluripotency and germness in comparison to somatic cells. They express germ cel-specific RNA binding proteins（RBPs） by modulating tissue-specific cis-and trans-regulatory elements. Studies on gene structures of chicken vasa homologue（CVH）, a chicken RNA binding protein, involved in temporal and spatial regulation are thus important not only for understanding the molecular mechanisms that regulate germ cel fate, but also for practical applications of primordial germ cells. However, very limited studies are available on regulatory elements that control germ cel-specific expression in chicken. Therefore, we investigated the intricate regulatory mechanism（s） that governs transcriptional control of ***： We constructed green fluorescence protein（GFP） or luciferase reporter vectors containing the various 5′ flanking regions of CVH gene. From the 5′ deletion and fragmented assays in chicken PGCs, we have identified a CVH promoter that locates at-316 to ＋275 base pair fragment with the highest luciferase activity. Additional y, we confirmed for the first time that the 5′ untranslated region（UTR） containing intron 1 is required for promoter activity of the CVH gene in chicken PGCs. Furthermore, using a transcription factor binding prediction, transcriptome analysis and siR NA-mediated knockdown,we have identified that a set of transcription factors play a role in the PGC-specific CVH gene ***： These results demonstrate that cis-elements and transcription factors localizing in the 5′ flanking region including the 5′ UTR and an intron are important for transcriptional regulation of the CVH gene in chicken PGCs.
