The bacterial abundance and production in the East China Sea: seasonal variations and relationships with the phytoplankton biomass and production

The East China Sea is a productive marginal sea with a wide continental shelf and plays an important role in absorbing atmospheric carbon dioxide and transferring terrigenous organic matter to the open ocean. To investigate the roles of heterotrophic bacteria in the biogeochemical dynamics in the East China Sea, bacterial biomasses （BB） and productions （BP） were measured in four cruises. The spatial distributions of the BB and the BP were highly season-dependent. Affected by the Changjiang River discharge, the BB and the BP were high in shelf waters （bottom depth not deeper than 50 m） and generally decreased offshore in August 2009. In December 2009 to lanuary 2010, and November to December 2010, the BB and the BP were high in waters with medium bottom depth. The onshore-offshore decreasing trends of the BB and the BP also existed in May-June 2011, when the BB was significantly higher than in other cruises in shelf break waters （bottom depth deeper than 50 m but not deeper than 200 m）. The results of generalized additive models （GAM） suggest that the BB increased with the temperature at a range of 8-20~C, increased with the chlorophyll concentration at a range of 0.02-3.00 mg/m3 and then declining, and decreased with the salinity from 28 to 35. The relationship between the temperature and the log-transformed bacterial specific growth rate （SGR） was linear. The estimated temperature coefficient （Q10） of the SGR was similar with that of the phytoplankton growth. The SGR also increased with the chlorophyll concentration. The ratio of the bacterial to phytoplankton production ranged from less than 0.01 to 0.40, being significantly higher in November December 2010 than in May-June 2011. Calculated from the bacterial production and growth efficiency, the bacterial respiration consumed, on average, 59%, 72% and 23% of the primary production in August 2009, November-December 2010, and May-/une 2011, respectively.

Zooplankton spatial and diurnal variations in the Changjiang River estuary before operation of the Three Gorges Dam

Estuarine plankton communities can serve as indicators of ecosystem modification in response to anthropogenic influences. The main objectives of this study were to describe the spatial distribution and diurnal variability in zooplankton abundance and biomass over almost entire salinity gradient of the Changjiang (Yangtze) River estuary and to provide a background reference for future studies. To accomplish this, data were collected from 29 stations in the estuary from May 19 to 26, 2003, including two anchor stations. The spatial and diurnal variations in zooplankton characteristics, i.e. abundance, biomass, and gross taxonomic composition, were examined. Generally, both the abundance and biomass gradually increased seaward and presented distinct spatial variations. In addition, the spatial data revealed a significant correlation between abundance and biomass; however, there was no significant correlation between abundance and biomass for the diurnal data. Although the zooplankton composition indicated distinct spatial differences in terms of dominant groups, copepods accounted for >50% of the total zooplankton abundance in most regions and times. Three zooplankton assemblages were recognized through hierarchical cluster analysis. These assemblages existed along the salinity gradient from fresh water to seawater, and their positions coincided with those of the three principal water masses in the estuary. The assemblages were classified as: (1) true estuarine, (2) estuarine and marine, and (3) euryhaline marine, which were characterized by the copepods Sinocalanus dorrii, Labidocera euchaeta, and Calanus sinicus, respectively. Both spatial and diurnal data indicated that there was no significant correlation between zooplankton abundance/biomass and depth-integrated phytoplankton abundance.

Seasonal variations of phytoplankton phosphorus stress in the Yellow Sea Cold Water Mass

The Yellow Sea is located between the China Mainland and the Korean Peninsula, representing a typical shallow epicontinental sea. The Yellow Sea Cold Water Mass（YSCWM） is one of the most important physical features in the Yellow Sea. The characteristics of vertical profiles and seasonal variations of biogenic elements in the YSCWM may lead the variations of nutrient availability（e.g., phosphorus） and phosphorus stress of phytoplankton. In this study, the authors surveyed the seasonal variations of phytoplankton phosphorus stress with emphasis on the effect of the YSCWM during the four cruises in April and October 2006, March and August 2007. Using both bulk and single-cell alkaline phosphatase activity（APA） assays, this study evaluated phosphorus status of phytoplankton community, succession of phytoplankton community and ecophysiological responses of phytoplankton to phosphorus in the typical region of the YSCWM. With the occurrence of the YSCWM, especially the variations of concentration of dissolved inorganic phosphorus（DIP）, the results of bulk APA appeared corresponding seasonal variations. Along Transects A and B, the mean APA in August was the highest, and that in March was the lowest. According to the ELF-labeled assay＇s results, seasonal variations of the ELF-labeled percentages within dominant species indicated that diatoms were dominant in March, April and October, while dinoflagellates were dominant in August. During the four cruises, the ELF-labeled percentages of diatoms except Paralia sulcata showed that diatoms were not phosphorus deficient in April 2006 at all, but suffered from severe phosphorus stress in August 2007. In comparison, the ELF-labeled percentages of dinoflagellates were all above 50% during the four time series, which meant dinoflagellates such as Alexandrium and Scrippsiella, sustained perennial phosphorus stress.

Construction of a Full-Length cDNA Library of Solen grandis Dunker and Identification of Defense- and Immune-Related Genes

The basic genetic characteristics, important functional genes, and entire transcriptome of Solen grandis Dunker were investigated by constructing a full-length cDNA library with the ‘switching mechanism at the 5'-end of the RNA transcript'(SMART) technique. Total RNA was isolated from the immune-relevant tissues, gills and hemocytes, using the Trizol reagent, and cDNA fragments were digested with Sfi I before being ligated to the pBluescript II SK* vector. The cDNA library had a titer of 1048 cfu μL-1 and a storage capacity of 1.05×106 cfu. Approximately 98% of the clones in the library were recombinants, and the fragment lengths of insert cDNA ranged from 0.8 kb to 3.0 kb. A total of 2038 expressed sequence tags were successfully sequenced and clustered into 965 unigenes. BLASTN analysis showed that 240 sequences were highly similar to the known genes(E-value 80%), accounting for 25% of the total unigenes. According to the Gene Ontology, these unigenes were related to several biological processes, including cell structure, signal transport, protein synthesis, transcription, energy metabolism, and immunity. Fifteen of the identified sequences were related to defense and immunity. The full-length cDNA sequence of HSC70 was obtained. The cDNA library of S. grandis provided a useful resource for future researches of functional genomics related to stress tolerance, immunity, and other physiological activities.

Global parameter estimation of the Cochlodinium polykrikoides model using bioassay data

Cochlodinium polykrikoides is a notoriously harmful algal species that inflicts severe damage on the aquacultures of the coastal seas of Korea and Japan. Information on their expected movement tracks and boundaries of influence is very useful and important for the effective establishment of a reduction plan. In general, the information is supported by a red-tide（a.k.a algal bloom） model. The performance of the model is highly dependent on the accuracy of parameters, which are the coefficients of functions approximating the biological growth and loss patterns of the C. polykrikoides. These parameters have been estimated using the bioassay data composed of growth-limiting factor and net growth rate value pairs. In the case of the C. polykrikoides, the parameters are different from each other in accordance with the used data because the bioassay data are sufficient compared to the other algal species. The parameters estimated by one specific dataset can be viewed as locally-optimized because they are adjusted only by that dataset. In cases where the other one data set is used, the estimation error might be considerable. In this study, the parameters are estimated by all available data sets without the use of only one specific data set and thus can be considered globally optimized. The cost function for the optimization is defined as the integrated mean squared estimation error, i.e., the difference between the values of the experimental and estimated rates. Based on quantitative error analysis, the root-mean squared errors of the global parameters show smaller values, approximately 25%–50%, than the values of the local parameters. In addition, bias is removed completely in the case of the globally estimated parameters. The parameter sets can be used as the reference default values of a red-tide model because they are optimal and representative. Growth and antioxidant status of oriental river prawn Macrobrachium nipponense fed with diets containing vitamin E

A feeding trial was carried out to investigate the dietary vitamin E requirement of the oriental river prawn Macrobrachium nipponense(weight of 0.3–0.4 g) and its effect role on antioxidant *** were fed with seven levels of vitamin E(0,25,50,75,100,200,and 400 mg/kg diet) for 60 *** results show that dietary vitamin E supplementation could significantly increased the prawn weight( P 0.05).The contents of vitamin E in the hepatopancreas and in the muscle increased with increasing dietary vitamin *** was a linear correlation between the vitamin E level in diet and that in muscle,and between the vitamin E level in diet and that in the *** the above results indicated that dietary vitamin E can be stored in the hepatopancreas and muscle and lower both the activities of SOD and CAT in the hepatopancreas,suggesting that it is a potential antioxidant in *** line analysis conducted on the weight gains of prawns in each diet group showed that the dietary vitamin E requirement for maximum growth is 94.10 mg/kg.

3D Modal Solution for Tidally Induced Lagrangian Residual Velocity with Variations in Eddy Viscosity and Bathymetry in a Narrow Model Bay

In this study, we examine the results obtained by the Finite-volume coastal Ocean Circulation Model(FVCOM) regarding the effects of eddy viscosity and bathymetry on the three-dimensional(3 D) Lagrangian residual velocity(LRV) in a narrow bay. The results are cast in terms of two nondimensional numbers: the ratio of friction to local acceleration(δ) and the ratio of the minimum depth over shoals to the maximum depth in the channel(ε). The ratio δ depends on the eddy viscosity and mean depth. For a given eddy viscosity, when ε > 0.5, the along-estuary LRV tends to be vertically sheared and when ε < 0.5, the exchange is laterally sheared. When ε << 1, the structure of the 3 D, depth-integrated, and breadth-averaged LRV changes only slightly as δ increases. For ε values between 0.33 and 0.5, the structure of the 3 D LRV is mainly laterally sheared. In the same ε range, the 3 D and depth-integrated LRV exhibit reversed structures from high to low δ values. In addition, the breadth-averaged LRV weakens the typical twolayered circulation when δ decreases. When ε is 1, the two-layered vertical structure reverses direction, and a three-layered vertical structure develops in the outer bay as δ decreases.

Regime shifts of hydrometeorological factors in the Jiaozhou Bay and their potential ecological impacts

The wavelet transform was applied to studying the regime shifts of hydrometeorological factors(i.e.,precipitation,air temperature,sea surface temperature and sea surface salinity)during the period of 1961–2000 in the Jiaozhou Bay(JZB).The results indicated clearly that these factors show variability of multiple timescales,with interannual and decadal *** local abrupt changes such as the 1978–1979 and 1988–1989 shifts feature the physical environment variation,which is consistent with the Southern Oscillation and Arctic Oscillation in the northern *** regard to the JZB ecosystem,the benthic diatom cell abundance(BEN)showed a decrease shift in1978–1979,which is closely related to the precipitation abrupt decrease,while the shellfish mortality disaster in the JZB greatly released the predating pressure of diatom growth,possibly resulting in BEN increase shift in 1995.

Offshore high-grade limemud resources of west coast of India:Economic potential and industrial applications

Limestone is one of the essential raw materials in the cement,paint,steel,ceramic,glass,chemical,pharmaceutical,paper,and fertilizer *** India,only 8%of the limestone resources are placed under the reserve category,of which 97%is of cement ***,India depends on imports to bridge the demand‐supply gap of steel,blast furnace,and chemical‐grade *** of Geological Survey of India(GSI)to locate alternate sources for limestone led to the discovery of enormous quantities of carbonate minerals called limemud from the continental shelf margin of the west coast of *** carried out systematic studies to explore the nature of the disposition,quality,quantity,and suitability of the offshore limemud for various industrial applications.A preliminary estimate of resources using high‐resolution subbottom profiling and sediment core sample studies established the occurrence of more than 172 billion tonnes of high‐grade(The content of CaCO3 is greater than 80 wt%)limemud in 0.4–28.0m thick stratified sediment layers spread over an area of 18000 ***,physical,mineralogical,beneficiation,and agglomeration studies found the offshore limemud as a potential replacement for limestone in the cement,filler,blast furnace,steel melting shop,lime production,paint,and Grade‐I steel *** assessment of mining and transportation costs indicates that the offshore limemud(USD 5–6/ton)is more cost‐effective than that imported from other countries(USD16‐18/ton).With several advantageous factors like low impurity,mode of occurrence in overburden‐free stratified form,fine‐grained slurry nature,and shallow water depth,sustainable mining of offshore limemud could be a future reality with controllable technological,economic,and environmental challenges.

The impact of Kuroshio water on the source water of the southeastern Taiwan Strait:numerical results

Model output from a Pacific basin-wide three-dimensional physical-biogeochemical model during the period of 1991 to 2008 was used to investigate the impact of Kuroshio water on the source water of the southeastern Taiwan Strait. Based on the characteristic salinities of both Kuroshio water and the South China Sea water, a Kuroshio impact index （KII） was designed to measure the degree of impact. The KII correlates significantly with the northeast-southwest component of wind stress, but the former lags the latter by approximately two months. The correlation coefficient between them increases from 0.267 4 to 0.852 9, with a lag time increasing from 0 to 63 days. The impact of Kuroshio Water is greater in winter and spring than in summer and autumn. At the interannual time scale, E1 Nifio and La Nifia events play an important role in impacting the KII. During E1 Nifio events, more Kuroshio water contributes to the source water of the southeastern Taiwan Strait. Conversely, during La Nifia events, less Kuroshio water contributes to the source water.
