Gastric cancer is highly prevalent among digestive tract *** to the intri-cate nature of the gastric cancer immune microenvironment,there is currently no effective treatment available for advanced gastric ***,there is promising potential for immunotherapy targeting the prostaglandin E2 receptor subtype 4(EP4)in gastric *** our previous study,we identified a novel small molecule EP4 receptor antagonist called *** with YY001 alone demonstrated a significant reduction in gastric cancer growth and inhibited tumor metastasis to the lungs in a mouse ***,adminis-tration of YYo01 stimulated a robust immune response within the tumor microenvironment,characterized by increased infiltration of antigen-presenting cells,T cells,and M1 ***,our research revealed that YYo01 exhibited remarkable synergistic ef-fects when combined with the PD-1 antibody and the clinically targeted drug apatinib,rather than *** findings suggest that YYo01 holds great promise as a potential therapeutic strategy for gastric cancer,whether used as a standalone treatment or in combination with other drugs.

As ethnic medicine,the whole grass of plants in Cirsium was used as *** review focuses on the antimicrobial activity of plants in Cirsium,including antimicrobial components,against different types of microbes and bacteriostatic *** results showed that the main antimicrobial activity components in Cirsium plants were flavonoids,triterpenoids and phenolic acids,and the antimicrobial ability varied according to the species and the content of *** them,phenolic acids showed a strong antibacterial ability against Acinetobacter baumannii,Pseudomonas aeruginosa,and Enterococcus *** antibacterial mechanisms include:(1)damaging the cell membrane,cell walls,mitochondria and nucleus of bacteria;(2)inhibiting the synthesis of proteins and nucleic acids;(3)suppressing the synthesis of enzymes for tricarboxylic acid cycle pathways and glycolysis,and then killing the bacteria via inhibition of energy ***,most research results on antimicrobial activity of Cirsium plants are reported based on in vitro *** evidence from clinical data and comprehensive evaluation are needed.

The utilization of enteroviruses engineered with reporter genes serves as a valuable tool for advancing our understanding of enterovirus biology and its applications,enabling the development of effective therapeutic and preventive *** this study,our initial attempts to introduce a NanoLuc luciferase(NLuc)reporter gene into recombinant enteroviruses were unsuccessful in rescuing viable *** hypothesized that the size of the inserted tag might be a determining factor in the rescue of the ***,we inserted the 11-amino-acid HiBiT tag into the genomes of enterovirus A71(EV-A71),coxsackievirus A10(CVA10),coxsackievirus A7(CVA7),coxsackievirus A16(CVA16),namely EV-A71-HiBiT,CVA16-HiBiT,CVA10-HiBiT,CVA7-HiBiT,and observed that the HiBiT-tagged viruses exhibited remarkably high rescue ***,the HiBiT-tagged enteroviruses displayed comparable characteristics to the wild-type viruses.A direct comparison between CVA16-NLuc and CVA16-HiBiT recombinant viruses revealed that the tiny HiBiT insertion had minimal impact on virus infectivity and replication ***,these HiBiT-tagged enteroviruses demonstrated high genetic stability in different cell lines over multiple *** addition,the HiBiT-tagged viruses were successfully tested in antiviral drug assays,and the sensitivity of the viruses to drugs was not affected by the HiBiT ***,our findings provide definitive evidence that the integration of HiBiT into enteroviruses presents a universal,convenient,and invaluable method for advancing research in the realm of enterovirus ***,HiBiT-tagged enteroviruses exhibit great potential for diverse applications,including the development of antivirals and the elucidation of viral infection mechanisms.

Characteristics and quality of clinical practice guidelines for diabetic kidney disease:a systematic review

OBJECTIVE:To assess the quality of clinical practice guidelines(CPGs)in the context of diabetic kidney disease(DKD)and determine whether any factors affect the ***:We searched eight databases along with five international and national organizations to develop or archive guidelines from their inception to July 2023,with an additional search of *** the authoritative organizations related to *** and consensus statements created using direct differential diagnosis or therapy for DKD were included without language *** quality was evaluated by four reviewers using the Appraisal of Guidelines for research and EvaluationⅡ(AGREEⅡ)*** with the item and domain scores,the guideline was also allocated an overall quality score,which ranged from 1(lowest possible quality)to 7(highest possible quality).Moreover,an overall recommendation for use was also assigned(“recommended”,“recommended with modifications”or“not recommended”).RESULTS:A total of 16 CPGs were included,of which 14 were from Asia and the remaining two from *** two CPGs were updated in the third *** CPGs were recommended for use because their primary domains scored in the medium or high ***,five CPGs were recommended with modifications as the stakeholder involvement,applicability,and editorial independence domains were evaluated as low *** all domains,the lowest average score was for rigour of development(33%),followed by application(36%),and stakeholder involvement(51%).The highest average score was for scope and purpose(79%),followed by clarity of presentation(75%).None of the CPGs considered the patient's viewpoint,and six of 16 CPGs did not use any grading system to translate the evidence into ***,only three of 16 CPGs shared search strategy,and eight of 16 CPGs did not declare a funding ***:According to the AGREE II evaluation,more than one in four CPGs for DKD had poo

目的:观察运动针法治疗缺血性中风的临床疗效,以及对患者运动功能和日常生活能力的改善作用。方法:将106例脑梗死导致的肢体运动功能障碍患者随机分为试验组和对照组,每组53例。两组患者均给予常规康复治疗,对照组加用头穴透刺治疗,试验组加用运动针法治疗。每周治疗3次,持续治疗4周。于治疗前、治疗2周和4周后,以及随访2个月时,采用Fugl-Meyer评估(FMA)量表、Lovett肌力评定量表、改良Ashworth量表(MAS)、日常生活活动(ADL)量表评分对患者运动功能、肌力、肌张力和日常生活能力进行评定。结果:治疗后两组运动功能和日常生活能力均提高;治疗4周后及随访2个月时试验组FMA及ADL评分高于对照组(P<0.05)。治疗后两组MAS评分均下降;治疗2周和4周后以及随访2个月时,试验组MAS评分低于对照组(P<0.05)。治疗后两组Lovett评分均提高;治疗4周后试验组Lovett评分高于对照组(P<0.05)。结论:在常规康复治疗基础上,运动针法与头穴透刺均能有效改善缺血性中风患者的运动功能,提高其日常生活能力;运动针法作用优于头穴透刺治疗。

目的:从中性粒细胞胞外诱捕网(NETs)参与克罗恩病(CD)进展的角度,阐释隔药灸减轻CD大鼠肠道炎症的效应机制。方法:将大鼠随机分为正常组、模型组、隔药灸组和美沙拉嗪组。除正常组外,其余大鼠采用2,4,6-三硝基苯磺酸制备CD模型。正常组和模型组不治疗仅做与其他组相同的固定。隔药灸组予以隔药灸气海和双侧天枢穴。美沙拉嗪组接受美沙拉嗪肠溶片灌胃治疗。比较各组大鼠一般情况,苏木精-伊红染色观察结肠组织病理学变化并评分。采用ABC酶联免疫吸附测定法检测各组大鼠血清中性粒细胞胞外诱捕网DNA(NETs-DNA)、中性粒细胞弹性蛋白酶DNA(NE-DNA)、过氧化物酶DNA(MPO-DNA)浓度,采用免疫荧光和实时定量聚合酶链反应技术观察各组大鼠结肠组织瓜氨酸化组蛋白3(citH3)、MPO和NE蛋白和mRNA的表达。结果:与正常组比较,模型组结肠黏膜溃疡深达肌层,上皮不完整,杯状细胞明显减少,可见大量炎性细胞浸润,结肠黏膜损伤指数(CMDI)评分显著增加(P<0.01),模型组血清NETs-DNA、NE-DNA、MPO-DNA浓度均增加(P<0.05),模型组结肠组织NE、citH3和MPO蛋白和mRNA表达均上调(P<0.01或P<0.05);与模型组相比,隔药灸组和美沙拉嗪组结肠黏膜上皮修复,杯状细胞明显增多,可见少量炎性细胞浸润,CMDI评分明显降低(P<0.01),隔药灸组和美沙拉嗪组血清NETs-DNA、NE-DNA、MPO-DNA浓度均下降(P<0.05),隔药灸组和美沙拉嗪组NE、citH3和MPO蛋白和mRNA表达下调(P<0.01或P<0.05)。结论:隔药灸能降低血清中性粒细胞NETs复合物的浓度,抑制结肠组织中的NETs复合物的蛋白和mRNA表达,可能是隔药灸缓解CD结肠黏膜炎症的机制之一。

目的:从组蛋白赖氨酸去甲基化酶4D(KDM4D)的角度探究隔药饼灸抑制结肠炎相关性结肠癌(CAC)肿瘤生长的效应机制。方法:将近交系雄性Sprague-Dawley大鼠随机分为正常组、CAC组、隔药饼灸组和抑制剂组。除正常组外,其余3组大鼠采用偶氮甲烷(AOM)联合葡聚糖硫酸钠(DSS)制备CAC大鼠模型。正常组和CAC组不干预;隔药饼灸组大鼠取气海和双侧天枢,每次每穴灸2壮,每日1次,每周休息1 d,共30 d;抑制剂组予以KDM4D抑制剂5-氯-8-羟基喹啉(5-c-8HQ)腹腔注射,每日1次,共30 d。干预后观察各组大鼠的一般情况、结肠长度、瘤体数量和体积及结肠组织病理学变化。采用实时荧光定量聚合酶链反应法检测腺瘤样息肉病基因(APC)、体轴抑制因子(Axin)、细胞周期蛋白D1(cyclin D1)、基质金属蛋白酶(MMP)-7及MMP-9 mRNA的表达。免疫组化法检测结肠组织增殖细胞核抗原(PCNA)、cleaved caspase3、KDM4D、APC及Axin蛋白的表达。结果:与正常组相比,CAC组大鼠一般情况较差,结肠长度显著缩短(P<0.01),结肠组织瘤体数目增多、体积增大(P<0.01),可见腺管结构明显紊乱,出现“背靠背”及共壁现象,高级别腺癌形成;CAC组结肠组织PCNA及KDM4D蛋白表达水平显著升高(P<0.01),cleaved caspase3、APC及Axin蛋白表达水平显著降低(P<0.01),cyclin D1、MMP-7及MMP-9 mRNA表达水平显著升高(P<0.01),APC及Axin mRNA表达水平显著降低(P<0.01)。与CAC组相比,隔药饼灸组与抑制剂组大鼠的一般情况改善,结肠长度显著增加(P<0.01),结肠瘤体数目减少、体积缩小(P<0.05),大鼠的结肠组织可见结肠上皮细胞增生,核增大且深染,异型增生,炎性细胞浸润。隔药饼灸组与抑制剂组大鼠结肠组织PCNA及KDM4D蛋白表达水平显著降低(P<0.01),cleaved caspase3、APC及Axin蛋白表达水平显著升高(P<0.01),cyclin D1、MMP-7及MMP-9 mRNA表达降低(P<0.05),APC及Axin mRNA表达升高(P<0.05)。结论:隔药饼灸能调控CAC大鼠的KDM4D异常表达,激活Wnt/β-catenin通路上游APC和Axin分子,抑制Wnt/β-catenin通路下游分子的异常激活,这可能是隔药饼灸抑制CAC肿瘤生长的关键机制之一。

Compound Shenhua Tablet,a medicine comprising seven herbs,is employed in treating IgA *** study aimed to meticulously analyze its chemical *** on a list of candidate compounds,identified through extensive literature review pertinent to the tablet’s herbal components,the composition analysis entailed the systematic identification,characterization,and quantification of the *** analyte-capacity of LC/ESI-MS-based and GC/EI-MS-based assays was *** identified and characterized constituents were quantified to determine their content levels and were ranked based on the constituents’daily doses.A total of 283 constituents,classified into 12 distinct categories,were identified and characterized in the Compound Shenhua *** constituents exhibited content levels of 1−10982μg·g^(−1),with daily doses of 0.01−395μmol·d^(−1).The predominant constituents,with daily doses of≥10μmol·d^(−1),include nine organic acids(citric acid,quinic acid,chlorogenic acid,cryptochlorogenic acid,gallic acid,neochlorogenic acid,isochlorogenic acid C,isochlorogenic acid B,and linoleic acid),five iridoids(specnuezhenide,nuezhenoside G13,nuezhenidic acid,secoxyloganin,and secologanoside),two monoterpene glycosides(paeoniflorin and albiflorin),a sesquiterpenoid(curzerenone),a triterpenoid(oleanolic acid),and a phenylethanoid(salidroside).Additionally,there were 83,126,and 55 constituents detected in the medicine with daily doses of 1–10,0.1–1,and 0.01–0.1μmol·d^(−1),*** combination of the LC/ESI-MS-based and GC/EI-MS-based assays demonstrated a complementary relationship in their analyte-capacity for detecting the constituents present in the *** comprehensive composition analysis establishes a solid foundation for further pharmacological research on Compound Shenhua Tablet and facilitates the quality evaluation of this complex herbal medicine.
