experimental investigation on the factors affecting VWF damage based on an in vitro blood-shearing platform

With the increasing number of people suffering from heart failure,ventricular assist devices have gradually become an effective way to treat end-stage heart ***,the blood damage caused by ventricular assist devices has not been effectively solved,which is an obstacle to its clinical *** research focused on erythrocyte damage under shear stress,while few researches were conducted on the interaction between blood under shear stress and the induction of von Willebrand factor(VWF)*** research used a vortex oscillator blood-shearing platform to conduct in vitro experiments and used immunoblotting to quantify VWF damage in sheared samples to study the laws of shear-induced VWF damage under different shear stress,different exposure times,different blood components,and hemolysis *** was found that VWF damage increased with exposure time and shear *** the same time,under lower shear stress,other blood components had little effect on VWF damage,while in a higher shear stress,other blood components would accelerate VWF *** will also affect VWF damage,and the higher the degree of hemolysis,the higher the rate of VWF degradation in the *** results of this research provide a reference for VWF damage evaluation standards and follow-up research and also guide for improving the design of ventricular assist devices to reduce VWF damage.

In sports events,the rapid recovery after high-intensity training or sport competition performance is very important for athletes'performance and *** aim of this study is to evaluate the effect of laser acupuncture and electrical stimulation on the recovery from exercise fatigue,using mice with swimming fatigue as experimental model and the electromyography(EMG)and the Raman spectroscopy of blood as evaluation *** mean square(RMS)and mean power frequency(MPF)of EMG were analyzed after laser acupuncture and electrical *** amplitude frequency combined analysis(JASA)showed that the proportion of muscles in the fatigue recovery area of the control group,the laser acupuncture group,the multi-channel laser acupuncture group and the laser combined with electrical stimulation group were 34.78%,39.13%,39.13%and 43.48%,*** spectroscopy of the mice blood during fatigue recovery showed there is a significant difference between the multi-channel laser acupuncture group and the laser combined with electric stimulation group compared with the recovery period and fatigue period(P<0:05)at the peak of 997 cm^(-1) and the laser combined electrical stimulation group had a statistical difference in the recovery period compared with the fatigue period(P<0.05)at the peak of 1561 cm^(-1).The results showed that laser acupuncture combined with electrical stimulation was beneficial to fatigue recovery in mice,and had the potential value in sports fatigue recovery.

Background: The primary motor cortex (M1) stimulation (MCS) is a useful tool for attenuation of the peripheral neuropathic pain in patients with pharmacologically refractory pain. Furthermore, that neurological procedure may also cause antinociception in rodents with neuropathic pain. Cold allodynia is a frequent clinical finding in patients with neuropathic pain, then, we evaluated if an adapted model of neuropathy induced by chronic constriction injury (CCI) of the ischiadicus nervus (sciatic nerve) produces cold allodynia in an animal model of chronic pain. In addition, we also investigated the effect of the electrical stimulation of the M1 on chronic neuropathic pain condition in laboratory animals. Methods: Male Wistar rats were used. An adapted model of peripheral mononeuropathy induced by CCI was carried out by placing a single loose ligature around the right sciatic nerve. The acetone test was used to evaluate the cold allodynia in CCI or Sham (without ligature) rats. The MCS (M1) was performed at low-frequency (20 μA, 100 Hz) during 15 s by deep brain stimulation (DBS-Thomas Recording device) 21 days after CCI or Sham procedures. The cold allodynia was measured before and immediately after the neurostimulation of M1 in the following time-window: 0, 15 and 30 min after MCS. Results: Cold allodynia threshold increased in animals with chronic neuropathic pain submitted to the acetone test 21 days after the CCI surgery. The M1-stimulation by DBS procedure decreased the cold allodynia immediately and until 30 min after M1-stimulation in rats with chronic neuropathic pain. Conclusion: The current proposal for a CCI model by a single loose ligature of the sciatic nerve can be employed as an experimental model of chronic neuropathic pain in rats submitted to peripheral nervous system injury. The M1-stimulation produced antinociception in rats with chronic neuropathic pain. Thus, we reinforced that the MCS decreases cold allodynia in laboratory animals submitted to

Coastal medicine is a promising field for research and drug development,because several natural products isolated from marine organisms have been found to be therapeutically useful in a clinical *** comparison to terrestrial products,the merit of marine compounds arises from the diversity in their chemical structure and unique bioactivity.A recently discovered marine compound,xyloketal B,displays neuroprotective and antioxidative effects in in-vitro and in-vivo experimental *** elicits these beneficial effects through mitochondrial protection,free radical scavenging,reducing reactive oxygen species production and suppressing apoptotic *** addition,unpublished data from our lab revealed that transient receptor potential melastatin 7 channel activity,implicated in a myriad of neurodegenerative diseases,was inhibited following xyloketal B *** this review,the therapeutic effect of xyloketal B will be evaluated based on existing evidence and its potential for drug development will be also discussed.

The incidence of breast cancer is tending younger globally,and tumor development,clinical treatment,and prognosis are largely influenced by histopathological *** diagnosed patients,the distinction between the cancer nests and normal tissue is the basis of breast cancer *** hyperspectral imaging technology has shown its potential in auxiliary pathological examinations due to the superior imaging modality and data *** paper presents a method for cancer nest segmentation from hyperspectral images of breast cancer tissue microarray *** scheme combines the strengths of the U-Net neural network and unsupervised principal component analysis,which reduces the amount of calculation and improves the recognition *** experimental accuracy of cancer nest segmentation reaches 87.14%.Furthermore,a set of quantitative pathological characteristic parameters reflects the degree of breast cancer lesions from multiple angles,providing a relatively comprehensive reference for the pathologist’s ***-depth exploration of the combined development of deep learning and microscopic hyperspectral imaging technology is worthy to promote efficient diagnosis of breast tumors and concern for human health.

Purpose: Regulated in development and DNA damage response-1(REDD1) is a stress related protein and is involved in the progression of cancer. The role and regulatory mechanism of REDD1 in bladder urothelial carcinoma(BUC), however, is yet unidentified. experimental Design: The expression of REDD1 in BUC was detected by western blot and immunohistochemistry. The correlation between REDD1 expression and clinical features in BUC patients were assessed. The effects of REDD1 on cellular proliferation, apoptosis, autophagy, and paclitaxel sensitivity were determined both in vitro and in vivo. Then the targeted-regulating mechanism of REDD1 by microRNAs was explored. Results: Here the significant increase of REDD1 expression is detected in BUC tissue, and REDD1 is firstly reported as an independent prognostic factor in BUC patients. Silencing REDD1 expression in T24 and EJ cells decreased cell proliferation, increased apoptosis, and decreased autophagy, whereas the ectopic expression of REDD1 in RT4 and BIU87 cells had the opposite effect. Additionally, the REDD1-mediated proliferation, apoptosis, and autophagy are found to be negatively regulated by miR-22 in vitro, which intensify the paclitaxel sensitivity via inhibition of the well-acknowledged REDD1-EEF2 K-autophagy axis. AKT/mTOR signaling initially activated or inhibited in response to silencing or enhancing REDD1 expression and then recovered rapidly. Lastly, the inhibited REDD1 expression by either RNAi or miR-22 sensitizes BUC tumor cells to paclitaxel in a subcutaneous transplant sarcoma model in vivo. Conclusion: REDD1 is confirmed as an oncogene in BUC, and antagonizing REDD1 could be a potential therapeutic strategy to sensitize BUC cells to paclitaxel.

Because China is becoming an aging society,the incidence of diabetes and diabetic foot have been *** foot has become one of the main health-related killers due to its high disability and mortality *** pressure wound therapy(NPWT)is one of the most effective techniques for the treatment of diabetic foot wounds and great progress,both in terms of research and its clinical application,has been made in the last 20 years of its ***,due to the complex pathogenesis and management of diabetic foot,irregular application of NPWT often leads to complications,such as infection,bleeding and necrosis,that seriously affect its treatment *** 2020,under the leadership of Burns,Trauma and Tissue Repair Committee of the Cross-Straits Medicine Exchange Association,the writing group for‘Consensus on the application of negative pressure wound therapy of diabetic foot wounds’was established with the participation of scholars from the specialized areas of burns,endocrinology,vascular surgery,orthopedics and wound *** on evidence-based practice suggested by the latest clinical research,this consensus proposes the best clinical practice guidelines for the application and prognostic evaluation of NPWT for diabetic *** consensus aims to support the formation of standardized treatment schemes that clinicians can refer to when treating cases of diabetic foot.
