Identification of TNFRSF1A as a novel regulator of carfilzomib resistance in multiple myeloma

Multiple myeloma(MM)is a hematological tumor with high mortality and recurrence *** is a new-generation proteasome inhibitor that is used as the first-line therapy for ***,the development of drug resistance is a pervasive obstacle to treating ***,elucidating the drug resistance mechanisms is conducive to the formulation of novel therapeutic *** elucidate the mechanisms of carfilzomib resistance,we retrieved the GSE78069 microarray dataset containing carfilzomib-resistant LP-1 MM cells and parental MM *** gene expression analyses revealed major alterations in the major histocompatibility complex(MHC)and cell adhesion *** upregulation of the tumor necrosis factor(TNF)receptor superfamily member 1A(TNFRSF1A)gene was accompanied by the downregulation of MHC genes and cell adhesion ***,to investigate the roles of these genes,we established a carfilzomib-resistant cell model and observed that carfilzomib resistance induced TNFRSF1A overexpression and TNFRSF1A silencing reversed carfilzomib resistance and reactivated the expression of cell adhesion ***,TNFRSF1A silencing suppressed the tumorigenesis of MM cells in immunocompetent mice,indicating that TNFRSF1A may lead to carfilzomib resistance by dampening antitumor ***,our results indicated that TNFRSF1A overexpression conferred carfilzomib resistance in MM cells and suppressed the expression of MHC genes and cell adhesion *** suppression of MHC genes and cell adhesion molecules may impair the interaction between immune cells and cancer cells to impair antitumor *** studies are warranted to further investigate the signaling pathway underlying the regulatory role of TNFRSF1A in MM cells.

Variation of microbiological and small molecule metabolite profiles of Nuodeng ham during ripening by high-throughput sequencing and GC-TOF-MS

The internal microbial diversity and small molecular metabolites of Nuodeng ham in different processing years(the first,second and third year sample)were analyzed by high-throughput sequencing technology and gas chromatography-time of flight mass spectrography(GC-TOF-MS)to study the effects of microorganisms and small molecular metabolites on the quality of ham in different processing *** results showed that the dominant bacteria phyla of Nuodeng ham in different processing years were Proteobacteria and Firmicutes,the dominant fungi phyla were Ascomycota and Basidiomycota,while Staphylococcus and Aspergillus were the dominant bacteria and fungi of Nuodeng ham,***,252 kinds of small molecular metabolites were identified from Nuodeng ham in different processing years,and 12 different metabolites were screened through multivariate statistical *** metabolic pathway analysis showed that 23 metabolic pathways were related to ham fermentation,of which 8 metabolic pathways had significant effects on ham fermentation(Impact>0.01,P<0.05).The content of L-proline,phenyllactic acid,L-lysine,carnosine,taurine,D-proline,betaine and creatine were significantly positively correlated with the relative abundance of Staphylococcus and Serratia,but negatively correlated with the relative abundance of Halomonas,Aspergillus and Yamadazyma.

Introduction:Laiza and nearby areas(LNA)in Myanmar are identified as the primary malaria hotspots in the bordering regions of yunnan province,***:Six sentinel surveillance sites were established at the China-Myanmar border in LNA to monitor *** from 2019 was used as a baseline to analyze malaria incidence and trends in LNA and Myanmar,as well as the importation of malaria cases into China from 2019 to ***:Plasmodium vivax was the predominant species,representing 99.95%(14,060/14,066)of confirmed malaria cases in LNA.A total of 8,356 malaria cases were identified in 2023,with an annual parasite incidence(API)of 19.78 per 100 *** to 2019,the incidence rate ratio was 21.47(95%confidence interval:18.84,24.48),indicating that the API in 2023 was 21.47 times higher than that in *** yunnan,out of 1,016 reported cases,545 imported cases(53.64%)originated from LNA and spread to 18(13.95%)out of 129 *** provinces in China,including yunnan,reported imported malaria cases from LNA in ***:The increase in population,particularly among internally displaced persons,along with inadequate healthcare services,has led to a notable resurgence of malaria in *** resurgence poses a risk to preventing the re-emergence of malaria transmission in *** is an urgent need for novel collaborative policies,as well as financial and technical assistance,to enhance malaria control efforts in LNA,Myanmar.

The source of Double ITCZ induced by the SST bias over the tropical western Pacific as reflected in CAS-ESM2 Model

“Double ITCZ”is a common precipitation bias over the tropical Pacific in current climate models and Earth system models,but the reasons for its formation are still worth exploring and *** this study,we adopted the second version of Chinese Academy of Sciences Earth System Model(CAS-ESM2),by comparing a set of sea surface temperature(SST)bias correction experiments over the tropical Pacific,to explore the possible mechanism of SST bias on inducing the“Double ITCZ”from the perspective of the climatic annual mean bias in the coupled *** revealed that the simulated climatic annual mean SST bias over the tropical Pacific can affect the bias of latent heat flux through the saturation specific humidity,and the bias of latent heat flux can further affect that of vertical velocity of humid air by the condensation release mechanism,and finally modulate the simulated bias in ***,through the inter-comparison between different experiments,it is found that the source of Double ITCZ might mainly come from the annual mean SST bias over the tropical western Pacific through the proposed air-sea coupled process of“SST-saturated specific humidity-latent heat flux-vertical velocity-precipitation”,indicating a possible way on reducing the couple biases in models over the tropical Pacific to improve the accuracy of CAS-ESM2 for climate simulation.
