The final analysis of the phase 3 Targeted Investigational Treatment Analysis of Novel Anti-androgen(TITAN)trial showed improvement in overall survival(OS)and other efficacy endpoints with apalutamide plus androgen deprivation therapy(ADT)versus ADT alone in patients with metastatic castration-sensitive prostate cancer(mCSPC).As ethnicity and regional differences may affect treatment outcomes in advanced prostate cancer,a post hoc final analysis was conducted to assess the efficacy and safety of apalutamide in the Asian ***-driven endpoints were OS,and time from randomization to initiation of castration resistance,prostate-specific antigen(PSA)progression,and second progression-free survival(PFS2)on first subsequent therapy or *** endpoints were assessed using the Kaplan–Meier method and Cox proportional-hazards models without formal statistical testing and adjustment for *** Asian patients received once-daily apalutamide 240 mg(n=111)or placebo(n=110)plus *** a median follow-up of 42.5 months and despite crossover of 47 placebo recipients to open-label apalutamide,apalutamide reduced the risk of death by 32%(hazard ratio[HR]:0.68;95%confidence interval[CI]:0.42–1.13),risk of castration resistance by 69%(HR:0.31;95%CI:0.21–0.46),PSA progression by 79%(HR:0.21;95%CI:0.13–0.35)and PFS2 by 24%(HR:0.76;95%CI:0.44–1.29)relative to *** outcomes were comparable between subgroups with low-and high-volume disease at *** new safety issues were *** provides valuable clinical benefits to Asian patients with mCSPC,with an efficacy and safety profile consistent with that in the overall patient population.

We previously reported that expression of an environmentally induced gene,mineral dust-induced gene(mdig),predicts overall survival in breast cancer *** the present report,we further demonstrate the differential roles of mdig between earlier-and later-stage breast *** noncancerous breast,mdig is a proliferation factor for cell growth and cell *** breast cancer,however,higher levels of mdig negatively regulate the migration and invasion of cancer *** of global DNA methylation,chromatin accessibility and H3K9me3 heterochromatin signature suggests that silencing mdig enhances DNA and histone *** immunostaining and data mining,we found that mdig is significantly upregulated in noninvasive and/or earlier-stage breast *** contrast,in triple-negative and other invasive breast cancers,diminished mdig expression was noted,indicating that the loss of mdig expression could be an important feature of aggressive breast *** together,our data suggest that mdig is a new biomarker that likely promotes tumor growth in the early stages of breast cancer while acting as a tumor suppressor to inhibit invasion and metastasis in later-stage tumors.

Cardiovascular disease is the number one cause of death in ***,cardiotoxicity is one of the most important adverse effects assessed by arrhythmia recognition in drug ***,cell-based techniques developed for arhythmia recognition primarily employ linear methods such as time-domain analysis that detect and compare individual waveforms and thus fall short in some applications that require automated and efficient arrhythmia recognition from large *** carried out the frst report to develop a biosensing system that integrated impedance measurement and multiparameter nonlinear dynamic algorithm(MNDA)analysis for druginduced arrhythmia recognition and *** biosensing system cultured cardiomyocytes as physiologically relevant models,used interdigitated electrodes to detect the mechanical beating of the cardiomyocytes,and employed MNDA analysis to recognize drug-induced arrhythmia from the cardiomyocyte beating *** best performing MNDA parameter,approximate entropy,enabled the system to recognize the appearance of sertindoleand norepinephrine-induced arrhythmia in the *** MNDA reconstruction in phase space enabled the system to classify the different arrhythmias and quantify the severity of *** new biosensing system utilizing MNDA provides a promising and alternative method for drug-induced arrhythmia recognition and classification in cardiological and pharmaceutical applications.

CRISPR-Cas12a genome engineering systems have been widely used in plant research and crop breeding. To date, the performance and use of anti-CRISPR-Cas12a systems have not been fully established in plants. Here, we conduct in silico analysis to identify putative anti-CRISPR systems for Cas12a. These putative anti-CRISPR proteins, along with known anti-CRISPR proteins, are assessed for their ability to inhibit Cas12a cleavage activity in vivo and in planta. Among all anti-CRISPR proteins tested, AcrVA1 shows robust inhibition of Mb2Cas12a and LbCas12a in E. coli. Further tests show that Acr VA1 inhibits LbCas12a mediated genome editing in rice protoplasts and stable transgenic lines. Impressively, co-expression of Acr VA1 mitigates off-target effects by CRISPR-LbCas12a, as revealed by whole genome sequencing. In addition, transgenic plants expressing AcrVA1 exhibit different levels of inhibition to LbCas12a mediated genome editing, representing a novel way of fine-tuning genome editing efficiency. By controlling temporal and spatial expression of AcrVA1, we show that inducible and tissue specific genome editing can be achieved in plants. Furthermore, we demonstrate that AcrVA1 also inhibits Lb Cas12a-based CRISPR activation(CRISPRa) and based on this principle we build logic gates to turn on and off target genes in plant cells. Together, we have established an efficient anti-CRISPR-Cas12a system in plants and demonstrate its versatile applications in mitigating off-target effects, finetuning genome editing efficiency, achieving spatial-temporal control of genome editing, and generating synthetic logic gates for controlling target gene expression in plant cells.

AWI-BS: An adaptive weight incentive for blockchain sharding

Secure separable reversible data hiding of halftone image in blockchain

To realize the distributed storage and management of a secret halftone image in blockchain,a secure separable reversible data hiding(RDH)of halftone image in blockchain(SSRDHB)was proposed.A secret halftone image can be used as the original image to generate multiple share images which can be distributed storage in each point of blockchain,and additional data can be hidden to achieve management of each share ***,the secret halftone image was encrypted through Zu Chongzhi(ZUC)algorithm by using the encryption key(EK).Secondly,the method of using odd or even of share data was proposed to hide data,and a share dataset can be generated by using polynomial ***,multiple share images can be obtained through selecting share data,and different additional data can be hidden through controlling odd or even of share data,and additional data can be protected by using data-hiding key(DK).After sharing process,if the receiver has both keys,the halftone image can be recovered and additional data can be revealed,and two processes are *** results show that multiple share images hidden additional data can be obtained through SSRDHB,and the halftone image can be recovered with 100%by picking any part of share images,and one additional data can be revealed with 100%by picking any one share image.
