Quantification of theophylline or paracetamol in milk matrices by high-performance liquid chromatography

A simple, accurate and sensitive high-performance liquid chromatography （HPLC） method was developed, validated and applied to the determination of either theophylline or paracetamol in milk-based samples. The method allowed drug quantification in fresh and powdered milk with a relatively short run time of analysis and it was also successfully applied to the quantification of the drugs in solid dosage forms intended for pediatric use. Moreover, the main significant advantages over other published works are the simplicity of the sample preparation, reduced assay time and sample loss. The method meets the International Conference on Harmonization guideline for analytical methods validation regarding specificity, linearity, accuracy, precision, specificity and robustness as required by health authorities and applied by industry while designing and marketing new drug products. The technique encompasses the separation of the analytes with a reverse phase C18 column under isocrafic conditions and UV detection at 272 nm and 243 nm, respectively, for theophylline and paraeetamol. The lower limit of quantification for both drugs was determined as 0.2 μg/mL and the between-batch accuracy was 99.7%. This HPLC method allows quantification of theophylline and paracetamol in milk matrices and it can be applied in the design, development and production of milk-based pediatric dosage forms.

An adequate vitamin D(vitD)intake(Recommended Daily Allowance,RDA=5μg)is crucial for health maintenance and its deficiency is associated with several health *** increase in hypovitaminosis D cases and the proliferation of food supplements(FS)that are easily accessible by the population,have led to an unrestrained chronic consumption of *** may accumulate in the body and originate toxicity(Tolerable Upper Limit,UL=100μg).The aim of this study was to evaluate if daily vitD doses mentioned in FS labels are in conformity with RDA.210 solid and liquid FS(for pediatrics and adults)sold in Portuguese pharmacies,supermarkets,health shops and on the internet were examined for indicated daily intake of vitD and compared to RDA and UL values.51.43%of FS have values higher than RDA,8.57%higher than *** average vitD daily dose in FS is 24.48μg,with a high variability between samples(0.25-250μg).Majority of FS labels recommend vitD daily doses above RDA and some even above UL,regardless of being for adults or ***,it is crucial that vitD dose in FS is reviewed to ensure the safety of these products.

Genetic characterization of species using phylogeographic approaches represents a basic reference to understand their evolutionary history as well as to identify conservation priorities to protect areas of particular interest regarding evolutionary *** in well-studied regions such information is lacking for the majority of species,including many endemic species with reduced distribution *** investigate the phylogeographic pattern of the Iberian frog Rana iberica,an endemic amphibian restricted to Central and North-Western Iberian *** mitochondrial sequences,we reconstruct the phylogeographic history of the species to test the effect of Quaternary climate changes on the evolutionary diversification of lineages,that is,the differentiation of mitochondrial lineages and the formation of genetic diversity melting pots,and integrate phylogeographic evidence for future conservation *** results indicate the existence of 3 main mitochondrial lineages differentiated during the Upper *** historical demographic analyses and climatic niche modeling show a strong effect of glacial climate changes,suggesting recurrent range contractions and *** such circumstances,differentiation took place most likely by isolation in allopatric interglacial *** lineage admixture in northern Portugal generated a broad mixed zone with highest nucleotide *** its particular evolutionary potential,its reduced distribution and eventual threats under current climate change scenario,conservation priorities should focus on the isolated lineage from Sierra de Guadalupe.

Effects of voluntary imipramine intake via food and water in paradigms of anxiety and depression in naive mice

Objective: We sought to investigate the efficacy of oral dosing in mice with imipramine(7mg/kg/day) via water or in food pellets, and to compare its effects in the paradigms of learned helplessness, locomotion, hedonic state, and anxiety. Methods: Water and food consumption were measured to determine daily imipramine dosage in C57BL/6N mice. Next, baseline scores for O-maze, dark/light box, and sucrose tests were measured. Mice were then subjected to a 4-week treatment of voluntary ingestion of drinking water or food pellets containing imipramine. Lastly, all groups were subjected to novel cage, open field, O-maze, dark/light box, sucrose test, and forced swim test to assess the effects of the treatment. Results: In na?ve mice, imipramine delivered via food, induced a reduction of total floating and increased latency in the forced swim test, i.e., antidepressant-like effects. No other significant effects were found. Dosing with water did not change behavior in the forced swim, sucrose preference test, anxiety, or locomotor paradigms, but increased exploration in the novel cage. Conclusions: Voluntary ingestion is an effective method of chronic dosing with imipramine in na?ve mice. delivery of imipramine with food pellets elicits antidepressant-like effects in the forced swim test, with no effects on anxiety, locomotion, or preference behaviors. In contrast, no such effects were observed with treatment via drinking water, suggesting that a higher dose may be required. Our work argues for a broader use of oral delivery using food-treated pellets, in small rodent models of pre-clinical depression. It may substantially improve animal welfare and overcome potential confounds in translational research, which are frequently associated with adverse chronic invasive pharmacotherapies.

Proteolysis of seed storage proteins (SSP) during germination provides a steady supply of amino acids to the embryo development into seedling. This process is coordinated by different peptidases that act sequentially and overlaid mode. These enzymes are an ancient group evolved separately in a wide structural and functional diversity and have many applications in medicine, pharmacy and industry. However, the knowledge about seed peptidases during germination was obtained from studies almost restricted to the cultivated species. This restriction implies caution about generalizations made from these studies, as well limits the biological knowledge about plant kingdom and technological use from plant peptidases. In this work, a scan of the proteolytic activity was held in germinating seeds of a leguminous subtropical woody tree. Eleven proteolytic activities were detected in protein extracts from embryonic axis and cotyledons. The presence and intensity of these activities varied over time and between these tissues. There was indication that aspartyl-endopeptidases (phytepsins) and cysteine-carboxypeptidases (plant cathepsins) were involved in A. colubrina SSP hydrolysis. These peptidases differ to that commonly involved in germination of the cultivated leguminous. In addition, one of detected phytepsins showed stability on pH scale, which is important for industrial uses. There was also detected a metallo-carboxypeptidase activity, which has been not described in plants. These peptidases must be isolated to confirm or not these indications. However, these data indicate the biological and technological importance of extending the studies about plant peptidases on a diverse genetic basis.

Quantification of Flavonoids in Brazilian Orange Peels and Industrial Orange Juice Processing Wastes

The flavonoid content in orange peels of different Brazilian citrus varieties such as bahia, lima, lima-of-persian, morcote, pera, ponkan, seleta, cravo, kinkan and pomelo was assessed. Industry processing juice wastes such as bagasse, bagasse residues, animal feeding bagasse, pulp WEUE and CORE-wash were also analyzed. The HPLC analysis indicates that the most abundant flavonoids found in these Brazilian citrus peels are hesperidin and naringin. The solvents used are selective for flavonoid extraction, and depending on their polarity, glycoside or aglycone flavonoids are extracted. The use of multivariate analysis shows that DMSO is the best solvent to extract glycosides flavanones while hexane displays high selectivity in the extraction of polymethoxylated flavones. The flavonoids present in the orange wastes, obtained at different stages of the industrial processing, are qualitative and quantitatively different. The identification and quantification of the flavonoid composition in each Brazilian citrus variety were evaluated and allowed the selection of the best solvent for the extraction of each specific class of flavonoids. These compounds were found to be more abundant in the fruit peels than in their juices, revealing their great industrial potential. The residual portion of the processing juices is also rich in flavonoids, depending on the processing step.

AIM: To study the therapeutic effect of three tubeguides with electrical conductivity associated to mesenchymal stem cells(MSCs) on neuro-muscular regeneration after ***: Rats with 10-mm gap nerve injury were tested using polyvinyl alcohol(PVA), PVA-carbon nanotubes(CNTs) and MSCs, and PVA-polypyrrole(PPy). The regenerated nerves and tibialis anterior muscles were processed for stereological studies after 20 wk. The functional recovery was assessed serially for gait biomechanical analysis, by extensor postural thrust, sciatic functional index and static sciatic functionalindex(SSI), and by withdrawal reflex latency(WRL). In vitro studies included cytocompatibility, flow cytometry, reverse transcriptase polymerase chain reaction and karyotype analysis of the MSCs. Histopathology of lung, liver, kidneys, and regional lymph nodes ensured the biomaterials biocompatibility. RESULTS: SSI remained negative throughout and independently from treatment. Differences between treted groups in the severity of changes in WRL existed, showing a faster regeneration for PVA-CNTs-MSCs(P des induced better nerve fiber regeneration. Functional and kinematics analysis revealed positive synergistic effects brought by MSCs and PVA-CNTs.
