Effects of dietary supplementation with tropical microfungi Mucor circinelloides and Trichoderma harzianum powder on growth performance,bacterial communities,and the biomass nutritional profile of white shrimp grow-out culture(Litopenaeus vannamei)

Microfungi have been explored further as supplement constituents in shrimp culture due to their rich protein,lipid,essential amino acids,fatty acids,and various bioactive *** researchers have evaluated the effects of two microfungi powders(Mucor circinelloides and Trichoderma harzianum)as feed supplements on growth performance,bacterial communities,and the biomass nutritional profile of white shrimp(Litopenaeus vannamei).The results showed that commercial/artificial feed(Feng-Li)with supplementation microfungi powder had a different effect on white shrimp growth performance than control after 62 days of the grow-out *** with 1.5%*** powder(M-2)could improve the growth performance of white shrimp,especially the survival rate(81.33%),final body weight(5.21 g),and shrimp biomass(211.35 g).In contrast,supplementation with 1.5%*** powder(T-2)did not improve the growth performance of white shrimp compared to control(p>0.05).Furthermore,it also affected the profile of the bacterial community in culture water as well as in shrimp *** with both microfungi could reduce the total number of bacteria in culture water at the end of the experiment,where the lowest number was obtained in treatment T-2(3.9×10^(5)CFU/mL),followed by M-2(9.9×10^(5)CFU/mL)and control(4.4×10^(6)CFU/mL).In the culture water,20 isolates of culturable bacteria were found during the trial,with the dominant bacteria in M-2:Bacillus cereus,Bacillus velezensis,Gordonia polyisoprenivorans,T-2:Acinetobacter sp.,Gordonia polyisoprenivorans,Micrococcus *** on average well colour development(AWCD),the highest bacterial activity was in the M-2(0.297-0.707),followed by T-2(0.226-0.717)and control(0.094-0.694).The highest substrate group utilised in M-2 and T-2 were polymers(0.896 and 1.348)and control:amines(0.100).The highest Shannon diversity index was obtained in the M-2(2.508),followed by control(2.286)and the T-2(2.157).As many as

Multi-year assessment of paralytic shellfish toxins in hard clam species along the coastline of Jiangsu Province, China

Paralytic shellfish toxins(PSTs) are notorious neurotoxins that threaten public health and food safety *** PST monitoring programs have recently been established throughout China, the profiles and variation of PSTs in important commercial clams(e.g., Mactra veneriformis, Ruditapes philippinarum, and Meretrix meretrix) along the Jiangsu Province coastline remain largely unexplored. In this study, a validated hydrophilic interaction liquid chromatography–tandem mass spectrometry(HILIC-MS/MS) method was used to examine PST profiles and levels in 540 clam samples from natural production areas along Jiangsu Province coastline during2014–2016. Although the PST levels(≤6.38 μg saxitotoxin equivalents(eq)/kg) were consistently below European Union regulatory limits(≤800 μg saxitotoxin eq/kg) during this time period, saxitotoxin, decarbamoylsaxitotoxin,and gonyautoxins 1 and 4 were detected, and nearly 40% of the samples were saxitotoxin-positive. The PST levels also varied significantly by seasons, with peak values observed in May during 2014–2016. This is the first systematic report of PSTs in clams from Jiangsu Province, and additional research and protective measures are needed to ensure the safety of clams harvested in this area.

Zircon U-Pb ages and O-Hf isotopes of Quaternary trachytes from the East Sea:Implications for the genesis of low-δ^(18)O magmas

Quaternary intraplate magmatism formed several volcanic islands and seamounts,including Dokdo(DD),Ulleungdo(UD),Simheungtack(ST),Anyongbok,and Isabu in the southwest of the East Sea back-arc *** this study,we present whole-rock geochemical,zircon U–Pb age,and in situ O–Hf isotope data for the submerged volcanic rocks from DD,UD,and ST to provide new insights into the eruption timing of these volcanoes and constrain the magma evolution *** samples used in this study were trachytes and exhibited ferroan,alkalic,and metaluminous to weakly peraluminous *** showed light rare earth element(REE)-enriched patterns with(La/Yb)N ratios of 25.3–31.7 and mostly negative Eu anomalies in a chondrite-normalized REE *** addition,they were enriched in large-ion lithophile elements and high field strength elements;they exhibited positive Pb anomalies and strongly negative Ba,Sr,P,and Ti *** zircons yielded a weighted-mean 206Pb/238U age of 2.61,0.348–0.704,and 2.76–2.94 Ma for the DD,UD,and ST trachytes,*** zircons exhibited lowerδ^(18)O values than normal depleted mantle values,regardless of the crystallization age and spatial distribution of ***δ^(18)O values showed no correlation with U contents or Th/U ratios,indicating that the lowδ^(18)O signatures were of primary magmatic *** Hf isotopic compositions of the zircons were relatively heterogeneous but predominately characterized by positive eHf *** O–Hf mixing modeling revealed that low-δ^(18)O rocks with positive eHf values from the UD and ST volcanoes were derived from a hybrid source of recycled juvenile crustal materials with low-δ^(18)O and positive eHf signatures and an enriched mantle source with normalδ^(18)O and negative eHf *** juvenile oceanic crust in the source was likely metasomatized by seawater at high temperatures prior to *** contrast,the felsic magma that formed the DD volcanoes may have assimilated with regional

Moringa oleifera is well known as a highly nutritious plant and a water purifier in fish *** fish has many impressive qualities making them very suitable for ***,this study aimed to evaluate the effect of supplementation of Moringa oleifera aqueous extract(MOAE)at varying concentrations(0,100,200,and 400 mg/kg diet for 90 days)on growth performance,hematological,and some biochemical parameters of Nile ***,histological changes and immunohistochemical expression of caspase 3 in gills and livers were *** enhancements(P<0.05)in the final weight,weight gain,specific growth rate,red blood cells,hemoglobin,packed cell volume,white blood cells,and total serum protein were observed in fish fed 200 mg MOAE/kg diet compared with other ***,a significant reduction(P<0.05)of liver and kidney function tests and non-significant changes in lipid profile were *** the other hand,fish fed 400 mg MOAE/kg diet revealed disturbance in the corresponding *** histological degenerative changes were detected in the gills and liver of fish fed 400 mg MOAE/kg *** comparison,fish fed 200 mg MOAE/kg diet showed the best histo-architectures of these organs compared with other *** immune expressions of caspase 3 were determined where gills and liver of fish fed 400 mg MOAE/kg diet revealed extensive number of positive caspase 3 cells indicating severe apoptotic ***,a dietary MOAE at 200 mg/kg diet is recommended to improve the growth performance,hematological parameters,liver and kidney functions in Nile ***,caution must be considered when using MOAE at a concentration over 200 mg/kg diet.

Phylogenetic diversity of culturable bacteria in surface seawater from the Drake Passage,Antarctica

QTL Detection for Albinism-Related Loci in Chinese Tongue Sole(Cynoglossus semilaevis)

The Chinese tongue sole(Cynoglossus semilaevis) is widely cultured in the coastal region of East Asia and has excellent economic value. However, the high albino rate of the breeding population has caused a significant loss to the aquaculture industry. To study the molecular mechanism of albinism, the present study used an albino Chinese tongue sole family to construct three simple sequence repeat(SSR) linkage groups, and draft a preliminary linkage map related to albinism. After albinism-related loci mapping, 18 albinism-related loci were detected under two models(containing 2407 genes) compared to the Chinese tongue sole genome. One of these loci, the tyrosinase related protein(tyrp2), which has been reported previously as an important gene regulating both eumelanin and phaeomelanin levels, was indicated to be the possible cause of albinism. Thirty-five Gene Ontology(GO) terms and 14 Kyoto Encyclopedia of Genes and Genome pathways were annotated via bioinformatic analyses. One GO term with protein tyrosine kinase activity, which contained 10 genes, was previously suggested to affect fish albinism. These results establish a foundation for further in-depth study of albinism in Chinese tongue sole.

Discordant patterns of genetic variation between mitochondrial and microsatellite markers in Acanthogobius ommaturus across the coastal areas of China

Acanthogobius ommaturus, which belongs to the family Gobiidae, is a euryhaline and demersal fish that is widely distributed in the coastal areas, harbors, and estuaries of China, D. P. R. Korea and Japan. In this study, the genetic diversity and genetic structure of five geographical populations of A. ommaturus was assessed using the mitochondrial hypervariable region gene and microsatellite markers. The results of the two genetic markers indicated that the A. ommaturus populations had a high level of genetic diversity. The mitochondrial marker detected weak genetic differentiation among populations, and the Neighbor-Joining tree showed that there was no obvious pedigree branches and geographic structure as well. However, population of Zhoushan showed significant genetic differentiation with other populations by microsatellite markers. The population of *** has not experienced bottleneck effect recently. We speculated that the Pleistocene climate change and juvenile fish dispersal played an important role in the population differentiation of A. ommaturus.

DNA barcoding of fishes from Zhoushan coastal waters using mitochondrial COI and 12S rRNA genes

Accurate species identification is a key component of biodiversity *** barcoding is an effective molecular method used for fish species *** aimed to study the DNA barcoding of fish in Zhoushan coastal waters,explore the differences and applicability of two gene fragments(12S rRNA and COI)of DNA barcoding in fish species identification,and established a comprehensive fish barcoding reference *** hundred and eighty-seven captured fish samples from Zhoushan coastal waters were identified using morphological characteristics and DNA barcoding.A total of 26412S rRNA sequences(belonging to eight orders,31 families,55 genera,and 66 species)and 188 COI sequences(belonging to seven orders,30 families,48 genera,and 58 species)were *** lengths of the 12S rRNA sequences ranged from 165 to 178 bp,and the guanine-cytosine(GC)content was 45.37%.The average 12S rRNA interspecific and intraspecific genetic distances(K2P)were 0.10%and 26.66%,*** length of the COI sequence ranged 574–655 bp,and the content of GC was 45.97%.The average 12S rRNA interspecific and intraspecific genetic distances(K2P)were 0.16%and 27.45%,*** minimum interspecific genetic distances of 12S rRNA and COI(1.23%and 1.86%)were both greater than their maximum intraspecific genetic distances(2.42%and 8.66%).Three molecular analyses(NJ tree,ABGD,and GMYC)were performed to accurately identify and delineate *** errors occurred when the 12S rRNA sequences were delimited using the NJ tree method,and the delimitation results of ABGD and GMYC are consistent with the final species identification *** results demonstrate that DNA barcoding based on 12S rRNA and COI can be used as an effective tool for fish species identification,and 12S rRNA has good application prospects in the environmental DNA(eDNA)metabarcoding of marine fish.

Effects of light intensity and photoperiod on the growth performance of juvenile Murray cods(Maccullochella peelii)in recirculating aquaculture system(RAS)

Murray cod(Maccullochella peelii)is a freshwater percichthyid fish that has a high market value and potential for culture in recirculating aquaculture systems(RASs).Illumination is an important environmental factor that affects the growth and physiological condition of fishes.A comprehensive understanding of the relationship between the light factors and the growth,nutrient composition and stress response of juvenile Murray cod in RAS is important to achieve satisfactory theoretical and practical aquaculture *** Murray cod were randomly assigned to nine RAS tanks with a volume of 1.5 m^(3),with each tank containing 120 fishes(3.5±0.5g).The fish were cultured for 120 days under different light intensities(1200,2400,and 3600 lx)and photoperiods(12L:12D,18L:6D and 24L:0D).The results showed that the final weight and feed conversion ratio of Murray cod under a light intensity of 1200 lx were significantly better than those under 3600 lx(P<0.05).The serum total protein and globulin were higher at the light intensity of 1200lx,compared to other light *** blood urea nitrogen level improved with increasing light intensity at LD12:12 and LD18:6 of photoperiod,as well as the superoxide dismutase level improved significantly from LD18:6 to LD24:0 of *** findings indicated that the optimal light condition to enhance growth at juvenile stage is 1200 lx of the light intensity and LD18:6 of the photoperiod.
