利用60头9月龄,初始体重接近277kg的肉牛(安格斯肉牛29头,安格斯一西门塔尔杂交肉牛31头)在日粮含有高水平硫和钼的条件下连续饲喂148天,评估甘氨酸螯合铜来源的铜相对于饲料级硫酸铜形式铜的生物利用率。在同一品系内根据体重将阉牛随机分为5个试验组:(1)对照组(不添加铜);(2)5mg Cu/kg DM(硫酸铜);(3)10mg Cu/kg DM(硫酸铜);(4)5mg Cu/kg DM(甘氨酸螯合铜);(5)10mg Cu/kg DM(甘氨酸螯合铜)。试验阉牛个体分别饲喂青贮玉米为基础的日粮(分析值8.2mg Cu/kg DM),试验前期(第一阶段)添加2mg Mo/kg DM和0.15%的硫元素连续使用120天;在试验后期(第二阶段)添加6mg Mo/kg DM和0.15%的硫元素持续测试28天。结果表明:两种来源的铜均可以显著改善阉牛的日增重和重料比(p=0.01)。与对照组相比,各试验组最终的血浆铜蓝蛋白,血浆铜以及肝脏铜的含量明显提高(P<0.05);10mg Cu/kg DM试验组比5mg Cu/kg DM试验组改善显著(P<0.05)。基于最终血浆铜,肝铜含量和血浆铜蓝蛋白中铜的含量与对应的日粮铜元素摄入量进行的多元回归方法,在试验第一阶段(添加2mg Mo/kg DM拮抗剂),添加甘氨酸螯合铜试验组的生物利用率相对于硫酸铜组(100%)分别为140%(P=0.10),131%(P=0.12)和140%(p=0.01);在试验第二阶段(添加6mg Mo/kg DM拮抗剂),甘氨酸螯合铜试验组的最终血浆铜,肝铜含量和血浆铜蓝蛋白中铜的生物利用率相对于硫酸铜组(100%)分别为144%(P=0.01),150%(P=0.01)和157%(P=0.01)。本研究结果表明,在日粮高水平拈抗因子硫和钼存在条件下,甘氨酸螯合形式的铜比硫酸铜具有更好的利用率。

Auxin homeostasis is pivotal for normal plant growth and development. The superroot2 （sur2） mutant was initially isolated in a forward genetic screen for auxin overproducers, and sUR2 was suggested to control auxin conju- gation and thereby regulate auxin homeostasis. However, the phenotype was not uniform and could not be described as a pure high auxin phenotype, indicating that knockout of CYP83B1 has multiple effects. subsequently, sUR2 was identified as CYP83B1, a cytochrome P450 positioned at the metabolic branch point between auxin and indole glucosinolate metabolism. To investigate concomitant global alterations triggered by knockout of CYP83B1 and the countermeasures chosen by the mutant to cope with hormonal and metabolic imbalances, 10-day-old mutant seedlings were characterized with respect to their transcriptome and metabolome profiles. Here, we report a global analysis of the sur2 mutant by the use of a combined transcriptomic and metabolomic approach revealing pronounced effects on several metabolic grids including the intersection between secondary metabolism, cell wall turnover, hormone metabolism, and stress responses. Metabolic and transcriptional cross-talks in sur2 were found to be regulated by complex interactions between both positively and negatively acting transcription factors. The complex phenotype of sur2 may thus not only be assigned to elevated levels of auxin, but also to ethylene and abscisic acid responses as well as drought responses in the absence of a water deficiency. The delicate balance between these signals explains why minute changes in growth conditions may result in the non-uniform phenotype. The large phenotypic variation observed between and within the different surveys may be reconciled by the complex and intricate hormonal balances in sur2 seedlings decoded in this study.

Better parameters of ventilation-CO2 output relationship predict death in CHF patients

Objective: Measures of ventilation-co_2 output relationship have been shown to be more prognostic than peak O_2 uptake in assessing life expectancy in patients with chronic heart failure(CHF). Because both the ratios(V4e/V4co_2) and slopes(V4e-vs-V4co_2) of ventilation-co_2 output of differing durations can be used, we aim to ascertain which measurements best predicted CHF life expectancy. Methods: Two hundred and seventy-one CHF patients with NYHA class II-IV underwent incremental cardiopulmonary exercise testing(CPET) and were followed-up for a median duration of 479 days. Four different linear regression V4e-vsV4co_2 slopes were calculated from warm-up exercise onset to: 180 s, anaerobic threshold(AT), ventilatory compensation point(VCP); and peak exercise. Five V4e/V4co_2 ratios were calculated for the following durations: rest(120 s), warm-up(30 s), AT(60 s), lowest value(90 s), and peak exercise(30 s). Death or heart transplant were considered end-points. Multiple statistical analyses were performed. Results: CHF patients had high lowest V4e/V4co_2(41.0±9.2, 141±30%pred), high V4e/V4co_2 at AT(42.5±10.4, 145±35%pred), and high V4e-vs-V4co_2 slope to VCP(37.6±12.1, 126±41%pred). The best predictor of death was a higher lowest V4e/V4co_2(≥42, ≥141%pred), whereas the V4e-vs-V4co_2 slope to VCP was less variable than other slopes. For death prognosis in 6 months, %pred values were superior: for longer times, absolute values were superior. Conclusion: The increased lowest V4e/V4co_2 ratio easily identifiable and simply measured during exercise, is the best measurement to assess the ventilation-co_2 output relationship in prognosticating death in CHF patients.

PURPOsE. To study deactivation of the rod photoresponse in infants using a paired-flash procedure. Rhodopsin content increases and scales the parameters of the activation of rod phototrans duction as rods develop. However, little is known about the kinetics of deactivation in the rods of young infants. METHODs. ERG responses to pairs of flashes were used to study the recovery of the rod response in 4- and 10- week-old infants and mature control subjects. The amplitudes of rod isolated a-wave responses to a probe flash (+ 3.3 log scot tds) presented 2 to 120 seconds after an equal-intensity test flash were measured. The interstimulus interval (IsI) at which the amplitude was half that of the response to the probe flash alone (t50) was determined by linear interpolation. REsULTs. Recovery time (t50) was significantly longer in infants than in adults (F=18.9, df 2, 32; Pshape of the recovery function did not vary with age. The t50 values were inversely proportional to the parameters of activation of rod phototransduction. CONCLUsIONs. These results are evidence that the kinetics of deactivation in infants are slower and may be set by the proportion of rhodopsin isomerized.

Dasatinib is a highly effective second-generation tyrosine kinase inhibitor used to treat chronic myeloid leukemia (CML).In 2007,a pivotal phase-2 study of dasatinib as second-line treatment was initiated in 140 Chinese CML *** report from the 4-year follow-up revealed that 73% of 59 patients in chronic phase (CML-CP) and 32% of 25 patients in accelerated phase (CML-AP) remained under *** initial dosage of dasatinib for CML-CP and CML-AP patients were 100 mg once daily and 70 mg twice daily (total=140 mg/ day),*** cumulative major cytogenetic response (MCyR) rate among patients with CML-CP was 66.1%(versus 50.8% at 18 months),and the median time to MCyR was 12.7 *** CML-CP patients who achieved MCyR after a 4-year follow-up also achieved a complete cytogenetic *** cumulative complete hematological response (chr) rate among patients with CML-AP was 64%(16/25),with three CML-AP patients achieving chr between 18 months and 4 years of follow-up;the median time to chr was 16.4 *** adverse event (AE) profile of dasatinib at 4 years was similar to that at 6 and 18 *** most frequently reported AEs (any grade) included pleural effusion,headache,and *** long-term follow-up data continue to support dasatinib as a second-line treatment for Chinese patients with CML.

Background Pulmonary angiography is widely performed in pulmonary hypertension patients, but its immediate effects on right heart hemodynamics and safety are not well known. The objective of this study was to investigate the right heart hemodynamic effects and safety of pulmonary angiography in Chinese patients with pulmonary hypertension. Methods Between January 2008 and June 2009, pulmonary hypertension patients undergoing pulmonary angiography were consecutively enrolled. Pulmonary angiography was performed during breath-holding after deep breathing. The baseline clinical data, hemodynamic measurements before and after pulmonary angiography and complications occurring within 48 hours after angiography were recorded. Results Ninety-five patients were included. All received non-ionic contrast medium with a volume of （75.7±29.8） ml. Angiography reduced heart rate in patients with baseline mean pulmonary arterial pressure 〉 60 mmHg （change of heart rate：（-3.1±7.0） beats/min, P=0.005）, increased mean right atrial pressure, diastolic and end-diastolic right ventricular pressure in patients with baseline mean pulmonary arterial pressure 〈60 mmHg （all P 〈0.05）. Patients with decreased mean pulmonary arterial pressure （change of mean pulmonary arterial pressure 〈 -10 mmHg） had the highest total pulmonary resistance （P=0.009 vs. no change in mean pulmonary arterial pressure （change of mean pulmonary arterial pressure, -10 mmHg to 10 mmHg）; P=0.03 vs. increased mean pulmonary arterial pressure （change of mean pulmonary arterial pressure 〉 10 mmHg）） and the lowest cardiac output （P=-0.018 vs. no change in mean pulmonary arterial pressure; P=0.013 vs. increased mean pulmonary arterial pressure）. There were 7 complications （7%）, with 6 related to catheter and only 1 directly related to angiography. All complications were mild and no death occurred. Conclusion Pulmonary angiography has minimal effect on right heart hemodynamics and is safe in pulmonary hypertension patients.

Cyst aspiration with gonadotropin suppression in ovarian remnant syndrome with ureteral obstruction: A case report

Background: Ovarian remnant syndrome (ORs) is characterized by functional ovarian tissue following bilateral salpingo-oophorectomy. ORs presents with pelvic pain or mass, but may also present with urinary tract symptoms resulting from enlarging residual tissue. Case: A 37 year old woman presented with pelvic pain radiating to her flank due to ureteral obstruction from an enlarging ovarian cyst in ORs. Transvaginal ultrasound guided, ovarian cyst aspiration accompanied by leuprolide acetate gonadotropin suppression resulted in acute and chronic relief of ureteral obstruction. Conclusion: This case demonstrated successful treatment of ureteral obstruction caused by an ovarian cyst in a patient with ORs via cyst aspiration and leuprolide acetate gonadotropin suppression. This treatment is a viable alternative for management of ORs, but sacrifices pathologic diagnosis when compared to traditional surgical resection.
