Sanhua Tang(三化汤) protects against ischemic stroke by preventing blood-brain barrier injury: a network pharmacology and in vivo experiments

OBJECTIVE: To assess the effect and mechanism of Sanhua Tang(三化汤, SHT) in treating ischemic stroke(IS) through the "Kaitong Xuanfu" theory by using network pharmacology and animal experiments. METHODS: The active ingredients and targets of SHT and IS were screened by public databases such as Traditional chinese Medicine systems pharmacology, GeneCards, and online mendelian inheritance in man. Visual network topographies were constructed using R, Cytoscape 3.6.0, AutoDockTools, a user-sponsored molecular visualization system on an open-source foundation, and other software to analyze the correlation between targets and active ingredients. The middle cerebral artery occlusion(MCAO) model was established by operation. Animals were divided into the Sham group, MCAO group(M group), aloe-emodin(AE) group(MCAO rats treated with aloe-emodin), SHT at low dosage(SL group)(MCAO rats treated with SL), SHT at medium dosage(SM group), and SHT at high dosage(SH group). 2,3,5-triphenyl tetrazolium chloride staining was used to detect the volume of cerebral infarction; Nissl staining was used to observe the morphology of neuronal cells; transmission electron microscopy was used to observe the integrity of the blood-brain barrier(BBB); enzymelinked immunosorbent assay was used to detect the content of interleukin-6(IL-6), IL-10, tumor necrosis factor α(TNF-α) in serum. Western blot was used to detect the expression of vascular endothelial growth factor A(VEGFA) protein in the cerebral ischemic penumbra. RESULTS: Using network pharmacology and molecular docking validation, four active ingredients(lignan, naringenin, aloe-rhodopsin, and β-sitosterol), seven target proteins(protein kinase b 1, IL-6, TNF, VEGFA, TP53, jun proto-oncogene, and cysteinyl aspartate specific proteinase 3), and inflammatory signaling pathways were identified. Persistent Variations in the East Asian Trough from March to April and the Possible Mechanism

The East Asian trough(EAT)profoundly influences the East Asian spring *** this study,the relationship of the EATs among the three spring months is *** analysis shows that the variation in March EAT is closely related to that of April *** empirical orthogonal function(EEOF)analysis also confirms the co-variation of the March and April *** positive/negative EEOF1 features the persistent strengthened/weakened EAT from March to *** investigation indicates that the variations in EEOF1 are related to a dipole sea surface temperature(SST)pattern over the North Atlantic and the SST anomaly over the tropical Indian *** dipole SST pattern over the North Atlantic,with one center east of Newfoundland Island and another east of Bermuda,could trigger a Rossby wave train to influence the EAT in March−*** SST anomaly over the tropical Indian Ocean can change the Walker circulation and influence the atmospheric circulation over the tropical western Pacific,subsequently impacting the southern part of the EAT in March−*** the SST factors,the Northeast Asian snow cover could change the regional thermal conditions and lead to persistent EAT anomalies from March to *** three impact factors are generally independent of each other,jointly explaining large variations in the EAT ***,the signals of the three factors could be traced back to February,consequently providing a potential prediction source for the EAT variation in March and April.

Drug-Target Interactions Prediction Based on Signed Heterogeneous Graph Neural Networks

Drug-target interactions(DTIs) prediction plays an important role in the process of drug *** computational methods treat it as a binary prediction problem, determining whether there are connections between drugs and targets while ignoring relational types information. Considering the positive or negative effects of DTIs will facilitate the study on comprehensive mechanisms of multiple drugs on a common target, in this work, we model DTIs on signed heterogeneous networks, through categorizing interaction patterns of DTIs and additionally extracting interactions within drug pairs and target protein pairs. We propose signed heterogeneous graph neural networks(SHGNNs), further put forward an end-to-end framework for signed DTIs prediction, called SHGNN-DTI,which not only adapts to signed bipartite networks, but also could naturally incorporate auxiliary information from drug-drug interactions(DDIs) and protein-protein interactions(PPIs). For the framework, we solve the message passing and aggregation problem on signed DTI networks, and consider different training modes on the whole networks consisting of DTIs, DDIs and PPIs. Experiments are conducted on two datasets extracted from Drug Bank and related databases, under different settings of initial inputs, embedding dimensions and training modes. The prediction results show excellent performance in terms of metric indicators, and the feasibility is further verified by the case study with two drugs on breast cancer.

Effect of moxibustion on expression profile of miRNAs in Tripterygium glycoside-induced decreased ovarian reserve

OBJECTIVE: To explore the possible regulatory mechanism of microRNA(miRNA) in moxibustion treatment for decreased ovarian reserve(DOR). METHODS: The DOR model was constructed by intragastrical Tripterygium glycoside suspension administration, and moxibustion therapy was simultaneously given. The morphological ovarian changes were observed by hematoxylin and eosin staining. The miRNA expression profile was detected by RNA sequencing, and bioinformatics analysis was performed. Cytoscape software 3.6.1 was used to establish a regulatory network and differentially expressed miRNAs were verified by reverse transcription quantitative polymerase chain reaction(RT-qPCR). RESULTS: Decreased number of mature follicles, increased atresia follicles, and abnormal granulosa cell morphology were observed in the model group compared with the control group. The moxibustion group demonstrated increased mature follicles, decreased atretic follicles, and significantly decreased abnormal morphology of granulosa cells compared with the model group. Additionally, RNA sequencing results manifested significantly up-regulated miRNA expressions(miR-92b-3p, miR-26-5p＿R + 1＿1ss10TC, miR-206-3p, miR-9993b-3p＿1ss6GA, miR-7857-3p＿R-1, miR-219a-2-3p＿1ss10GC, miR-3968-p5＿1ss10AT, and PC-5p-6478＿1795) and down-regulated miR-664-2-5p＿R + 1 in the model group, compared with the control group, and the moxibustion group reversed abnormal disorder levels of these miRNAs. Moreover, these differentially expressed miRNAs were mainly involved in the phosphatidylinositol-3-kinase/protein kinase B signaling pathway and nuclear factor erythropoietin-2-related factor 2/heme oxygenase 1 signaling pathway. Finally, network and RT-qPCR verification revealed miR-9993b-3p＿1ss6GA as the most critical miRNA. CONCLUSION: This experiment proved the effectiveness of moxibustion in improving the ovarian reserve of rats by regulating miRNA expression, especially miR-9993b-3p＿1ss6GA.

Artemisia argyi extract subfraction exerts an antifungal effect against dermatophytes by disrupting mitochondrial morphology and function

Consensus on glycemic management for patients with coronary heart disease and type 2 diabetes

The prevalence of patients with coronary heart disease(CHD)and diabetes mellitus is notably high,posing sig-nificant residual cardiovascular risks even after routine interventions such as antihypertensive,lipid-lowering,and antithrombot-ic *** studies have demonstrated that certain glucose-lowering medications confer cardiovascular benefits for pa-tients with type 2 ***,a survey indicates that cardiologists may not be fully acquainted with the optimal screen-ing timing,indicators,and diagnostic criteria for type 2 diabetes,and there is insufficient awareness and a low rate of prescrip-tion of novel glucose-lowering medications with proven cardiovascular efficacy,such as glucagon-like peptide-1 receptor agon-ists(GLP-1 RAs)and sodium-glucose co-transporter-2 inhibitors(SGLT-2i).In this context,based on domestic and international guidelines or consensus and the latest evidence-based evidence,this consensus aims to standardize the glycemic management for patients with acute coronary syndrome,chronic coronary syndrome,and perioperative management for percutaneous coronary *** highlights the key points of screening and diagnosis of type 2 diabetes,and the comprehensive management of cardiovascular risk in patients with *** consensus elaborates on the principles and algorithms of glycemic management for CHD patients,without involving acute complications of diabetes,clarifies the clinical practice of glucose-lowering medications with cardiovascular benefits,and promotes the standardized use of these medications in cardiovascular and other related spe-cialty ***,it addresses the glucose-lowering treatment to comprehensively reduce cardiovascular risks.
