Simultaneous inhibition of PI3Kα and CDK4/6 synergistically suppresses KRAS-mutated non-small cell lung cancer

Objective: Activating KRAS mutations are the most common drivers in the development of non-small cell lung cancer(NSCLC).However, unsuccess of treatment by direct inhibition of KRAS has been proven. Deregulation of PI3K signaling plays an important role in tumorigenesis and drug resistance in NSCLC. The activity of PI3Kα-selective inhibition against KRAS-mutated NSCLC remains largely ***: Cell proliferation was detected by sulforhodamine B assay. Cell cycle distribution and apoptosis were measured by flow cytometry. Cell signaling was assessed by Western blot and immunohistochemistry. RNA interference was used to down-regulate the expression of cyclin D1. Human NSCLC xenografts were employed to detect therapeutic efficacy in ***: CYH33 possessed variable activity against a panel of KRAS-mutated NSCLC cell lines. Although CYH33 blocked AKT phosphorylation in all tested cells, Rb phosphorylation decreased in CYH33-sensitive, but not in CYH33-resistant cells, which was consistent with G1 phase arrest in sensitive cells. Combined treatment with the CDK4/6 inhibitor, PD0332991, and CYH33 displayed synergistic activity against the proliferation of both CYH33-sensitive and CYH33-resistant cells, which was accompanied by enhanced G1-phase arrest. Moreover, down-regulation of cyclin D1 sensitized NSCLC cells to CYH33. Reciprocally, CYH33 abrogated the PD0332991-induced up-regulation of cyclin D1 and phosphorylation of AKT in A549 cells. Co-treatment with these two drugs demonstrated synergistic activity against A549 and H23 xenografts, with enhanced inhibition of Rb ***: Simultaneous inhibition of PI3Kα and CDK4/6 displayed synergistic activity against KRAS-mutated NSCLC. These data provide a mechanistic rationale for the combination of a PI3Kα inhibitor and a CDK4/6 inhibitor for the treatment of KRASmutated NSCLC.

The classification and assessment of vulnerability of man-land system of oasis city in arid area

Oasis city system is the center of the man-land relationship in arid area and it is the most influential spatial and temporal multiple dynamic system. Oasis city system is not only the largest area where artificial disturbances occur at a regional scale but also the most concentrated area of human activity in arid area. In this study, we developed an applicable and convenient method to assess vulnerability of man-land system of oasis cities with vulnerability indicator system, respectively evaluating the sensitivity, adaptability and vulnerability of the eco- environment system, the economic system and the social system. The results showed that the sensitivity and vulnerability of oasis cities in Xinjiang, China have significant differences while their adaptability does little. In order to find the inherent differences in the vulnerability of oasis cities, triangle methodology has been adopted to divide Xinjiang oasis cities into five types. Some adaptive developing policies specific for individual cities are also proposed based on their vulnerability type and constrain- ing factors.

Study of Borromean halo nuclei by the neutron wall with simulation

The model of three-body Borromean halo nuclei breakup was described by using standard phase space distributions and the Monte Carlo simulation method was established to resolve the detection problem of two neutrons produced from breakup reaction on the neutron wall detector. For 6He case, overall resolution ~rEk for the Oaussiaal part of the detector response and the detection efficiency including solid angle acceptance with regard to the excitation energy Ek are obtained by the simulation of two neutrons from 6He breakup into the neutron wall. The effects of the algorithm on the angular and energy correlations of the fragments are briefly discussed.

New matrix of MALDI-TOF MS for analysis of small molecules

New matrix, metal-phthalocyanine （MPc）, of matrix-assisted laser desorption/ionization time-of-flight mass spectrometry （MALDI-TOF MS） was used for analysis of small molecules （usually 〈500 Da）. By using MPcs as matrices, small molecular samples were moved to high mass-to-charge region where there was no interference caused by the traditional matrices. The mass of the target analvte was obtained by simple calculation.

Fish gut microecosystem: a model for detecting spatial pattern of microorganisms

The spatial distribution pattern of organisms is a basic issue in understanding the mechanisms of community assembly. Although the spatial distributions of animals and plants have been well studied,those of microorganisms are still being debated. In this study, we used a fi sh gut microecosystem to detect the spatial pattern of microbes, because it can provide a relatively unifi ed and stable environment. Results suggest that the turnover of intestinal bacterial assemblages showed a weak but highly signifi cant negative correlation between similarity and distances in the microbial community, in respect of both grass carp intestinal loci distances and the geographical distance between fi sh sampling sites. Our results also suggest that intestinal bacterial assemblages responded to differences within the external environment and within different parts of the fi sh themselves. These results show that some, or possibly all, microbes are restricted in their distribution and that environmental factors are also important infl uences on the structure of intestinal bacterial assemblages. The fi sh gut microecosystem is useful in promoting study of the spatial distribution patterns of microorganisms.

Net ecosystem CO_2 exchange and controlling factors in a steppe——Kobresia meadow on the Tibetan Plateau

Knowledge of seasonal variation of net ecosystem CO2 exchange (NEE) and its biotic and abiotic controllers will further our understanding of carbon cycling process, mechanism and large-scale modelling. Eddy covariance technique was used to measure NEE, biotic and abiotic factors for nearly 3 years in the hinterland alpine steppe--Korbresia meadow grassland on the Tibetan Plateau, the present highest fluxnet station in the world. The main objectives are to investigate dynamics of NEE and its components and to determine the major controlling factors. Maximum carbon assimilation took place in August and maximum carbon loss occurred in November. In June, rainfall amount due to monsoon climate played a great role in grass greening and consequently influenced interannual variation of ecosystem carbon gain. From July through September, monthly NEE presented net carbon assimilation. In other months, ecosystem exhibited carbon loss. In growing season, daytime NEE was mainly controlled by photosynthetically active radiation (PAR). In addition, leaf area index (LAI) interacted with PAR and together modulated NEE rates. Ecosystem respiration was controlled mainly by soil temperature and simultaneously by soil moisture. Q10 was negatively correlated with soil temperature but positively correlated with soil moisture. Large daily range of air temperature is not necessary to enhance carbon gain. Standard respiration rate at referenced 10℃(R10) was positively correlated with soil moisture, soil temperature, LAI and aboveground biomass. Rainfall patterns in growing season markedly influenced soil moisture and therefore soil moisture controlled seasonal change of ecosystem respiration. Pulse rainfall in the beginning and at the end of growing season induced great ecosystem respiration and consequently a great amount of carbon was lost. Pruning-aware Sparse Regularization for Network Pruning

Spatial-temporal Patterns and Driving Forces of Water Retention Service in China

Overwhelming water-deficiency conditions and an unbalanced water supply and demand have been major concerns of both the chinese government and the general public during recent decades. Studying the spatial-temporal patterns and impact factors that influence water retention in China is important to enhance the management of water resources in China and other similar countries. We employed a revised Integrated Valuation of Ecosystem Services and Tradeoffs(InVEST) model and regression analyses to investigate the water retention service in China. The results showed that the southeastern China generally performed much better than Northwest China in terms of the spatial distribution of water retention. In general, the efficacy of the water retention service in China increased from 2000 to 2014; although some areas still had a downward trend. Water retention service increased significantly(P < 0.05) in aggregate in the Qinghai-Tibet Plateau, and the Da Hinggan Mountains and Xiao Hinggan Mountains. However, the service in southwestern China showed a decreasing trend(P < 0.05), which would have significant negative impact on the downstream population. This study also showed that in China the changes in water retention service were primarily due to climate change(which could explain 83.49% of the total variance), with anthropogenic impact as a secondary influence(likewise the ecological programs and socioeconomic development could explain 9.47% and 1.06%, respectively). Moreover, the identification of water retention importance indicated that important areas conservation and selection based on downstream beneficiaries is vital for optimization protection of ecosystem services, and has practical significance for natural resources and ecosystem management.

Study on the Crystal of Daomanite

It was not possible to carry out a complete analyses of crystal, as the experiment by Ding and Shi et al.. It＇s analysis precision R=0.25 or more big than this, which value are not satisfied for single crystal study, but we through many test and found the best： [R（int）=14.5%]. The final fullmatix least-squares refinement on F2 converged to R1=0.0791 and wR2=0.1864 for 704 observed reflections [I 3 2s（I）]. Daomanite is orthorhombic system, space group Cmc21, a=3.7520（8））A, b=15.844（4） A, c=5.8516（12） A, α=β=γ=90°. V=347.86（14）A3, Z=4. Daomanite chemical formula is Cu Pt AsS 2. Idealized composition Me＋M2＋M2＋S2=CuS ·PtA s S. There is no other similar mineral in the world.
