α-synuclein, a 140 amino acid protein richly expressed in the brain, has been strongly implicated in the pathogenesis of neurodegenerative diseases, especially Parkinson's disease. Although the protein was initially nominated as synuelein for its localization in both presynaptic terminals and portions of nuclear envelope in the electric organ of the Pacific electric ray Torpedo californica, the nuclear location has not been consistently confirmed in mammal brain neurons in subsequent studies. In the present study, by using recombinant humanα-synuclein to immune Balb/c mice, we successfully obtained a hybridoma clone that produced monoclonal antibody specifically againstα-synuclein. 研究背景和目的:CMTM是本室于2003年在国际上首次报道的新基因家族。在人类,该家族包括9个基因,CMTM7是其成员之一,定位于抑癌基因富集区3p22.3。本室的前期研究发现CMTM7为抑癌基因,其在多种肿瘤中表达下调,杂合缺失和甲基化修饰参与其表达调控。在肺腺癌细胞中,CMTM7通过增强Rab5活性,促进EGFR转运和降解发挥抑癌作用。表达谱分析发现,CMTM7在免疫系统高表达,Immusort Database分析提示其在正常B细胞高表达,在慢性B淋巴细胞白血病中表达明显下调,提示CMTM7可能参与B细胞相关癌症的发生。本研究的目的为进一步明确CMTM7在成熟B细胞相关疾病中的功能,探索其发病机制,为癌症的防治提供理论依据。方法:我们利用293T细胞包装慢病毒,成功筛选并获得了敲减效率最高的两条shRNA,构建稳定敲减CMTM7的Ramos细胞株。通过western blot、细胞计数、细胞周期、AV-PI细胞凋亡实验研究敲减CMTM7对B细胞增殖、凋亡、周期的影响,并分析相关信号通路;通过western blot、qPCR、流式细胞术等实验研究敲减CMTM7对BCR及细胞总IgM表达水平的影响,探索其表达水平与肿瘤发生的关系。结果:通过筛选获得敲减效率最高的两条shRNA,其在mRNA和蛋白质水平均能有效抑制CMTM7的表达;敲减CMTM7可促进Ramos细胞增殖,抑制凋亡和Caspase-3的活性,但对细胞周期无明显作用;信号通路分析发现敲减CMTM7可明显抑制ERK的磷酸化,但对AKT、p38的磷酸化无明显作用;同时我们发现敲减CMTM7可以下调细胞膜IgM及总IgM表达水平。结论:CMTM7对成熟B细胞来源的肿瘤细胞生长有抑制作用,并参与BCR的表达调控。

A clue to potential therapeutic targets: application of integrative medicine and bioinformatics

Plants of Litsea have been widely used as traditional medicine for thousands of *** china and other Asian country,for example,japan,India,Korera,and Malaysia,Litsea has been used in traditional medicine for influenza,stomach aches,diarrhea,and other *** monopetala（Roxb）*** a medium-sized species,about 7-15 m high with stalked,alternate and oblong to ovate shaped *** leaves are used as a topical medicine for the treatment of arthritis[1,2].However,no systematic study has been reported regarding its phytochemistry and *** 95%Et OH extract of the whole plant of Litsea monopetala was partitioned between Et2O,n-Bu OH and water,successively,and the Et2O-soluble fraction and n-Bu OH fraction was chromatographed over silica gel and Sephadex LH-20 to yield compounds（1–6）.The structures of compounds 1-6 were elucidated as trans-phytol（1）,sitosterol（2）,sitosterol?-D-glucopyranoside（3）,catechin（4）,quercetin 3-O-β-D-glucopyranoside（5）,and quercetin-3-O-?-D-rhamnopyranoside（6）,respectively,on the basis of their MS and 1H NMR and 13C NMR *** displayed that Litsea monopetala was rich in *** is a member of the fragrance structural group Alcohols Branched Chain *** common characteristic structural elements are one hydroxyl group per molecule,a C4–C16 carbon chain with one or several methyl or ethyl side chains and up to four non-conjugated double ***’s a fragrance ingredient used in cosmetics,fine fragrances,shampoos,toilet soaps and other toiletries as well as in non-cosmetic products such as household cleaners and *** worldwide use is 0.1 to 1.0 metric tons per year[3].All these provide important information with regard to a source of the isolation of natural phytol and the industrial applications of Litsea monopetala.

More and more evidence show that the targets of traditional Chinese medicine are a resource pool that contributes to the modern medicine. A key aspect of medicine research is to address the issue how to look for a clue of potential therapeutic molecular targets by interdisciplinary approaches. With the development of high throughput technology, a considerable amount of data has been emerging. The application of these big data is not so efficient as the generation of them. This paper attempts to show that interdisciplinary analysis contributes to the discovery of targets at the initial stage of drug research, indicating that it is necessary to explore the drug research and development with interdisciplinary approaches between integrative medicine and bioinformatics.

Aim:To determine the inhibitory effect of the synthetic STAT3 siRNA on theexpression of STAT3 gene in human laryngeal cancer cell lines Hep2 and to inves-tigate the effect of STAT3 siRNA on growth and apoptosis in Hep2 ***:A pair of DNA templates coding siRNA against STAT3-mRNA was synthesized toreconstruct plasmid of pSilencer1.0-U6 ***2 cells were trans-fected with RPMI-1640 media (untreated),plasmid (empty),and STAT3 siRNA,*** blot and Western blot analysis of STAT3 and pTyr-STAT3expression in Hep2 cells and Western blot analysis of Bcl-2 expression in the Hep2cell was performed 72 h after ***,flow cytometry,and AO/EBassay were used for determination of cells proliferation and apoptosis in ***:pTyr-STAT3 was markedly expressed in untreated Hep2 cells andthe vector-treated Hep2 cells,whereas pTyr-STAT3 expression was significantlyreduced in STAT3 siRNA-transfected Hep2 cells,indicating that STAT3 siRNAinhibited the activity of *** of Hep2 cells with STAT3 siRNAsignificantly inhibited STAT3 expression at both mRNA and protein level in Hep2cells and the inhibition was characterized by time-dependent ***-ment of Hep2 cells with STAT3 siRNA resulted in dose-dependent growth inhibi-tion of Hep2,this significantly increased apoptotic cell rate,and decreased Bcl-2expression level in Hep2 ***3 siRNA had an effect on induction of eitherearly or late stage ***:This study demonstrates that STAT3siRNA effectively inhibits STAT3 gene expression in Hep2 cells leading to growthsuppression and induction of apoptosis in Hep2 *** use of siRNA tech-nique may provide a novel therapeutic approach to treat laryngeal cancer andother malignant tumors expressing constitutively activated STAT3.

Ultrasound plays an important role not only in preoperative diagnosis but also in intraoperative guidance for liver *** ultrasound(IOUS)has become an indispensable tool for modern liver surgeons,especially for minimally invasive surgeries,partially substituting for the surgeon’s *** fundamental mode,Doppler mode,contrast enhancement,elastography,and real-time virtual sonography,IOUS can provide additional real-time information regarding the intrahepatic anatomy,tumor site and characteristics,macrovascular invasion,resection margin,transection plane,perfusion and outflow of the remnant liver,and local ablation efficacy for both open and minimally invasive liver *** and localization of intrahepatic lesions and surrounding structures are crucial for performing liver resection,preserving the adjacent vital vascular and bile ducts,and sparing the functional liver *** ultrasound can provide critical information for intraoperative decision-making and ***,all liver surgeons must master IOUS techniques,and IOUS should be included in the training of modern liver *** investigation of the potential benefits and advances in these techniques will increase the use of IOUS in modern liver surgeries *** study comprehensively reviews the current use of IOUS in modern liver surgeries.

Expression of osteoclast-related cytokines in mandibular invasion by gingival squamous cell carcinoma

Purpose: Gingival squamous cell carcinoma (GSCC) displays strong potential for local infiltration。Detailed evaluation of bone invasion to the mandible is an important problem clinically。The purpose of this study was to investigate expression of osteoclast-related cytokines, which play an important role in mandibular invasion。Methods: Twenty-three cases of lower GSCC were classified histopathologically as non-invasive group (n=10) or invasive group (n=13)。Expressions of various cytokines in clinical samples were studied by immunohistochemical staining and quantitative real-time reverse transcription-polymerase chain reaction (QRT-PCR)。Cytokines interleukin (IL)-1a, IL-1b, IL-6, IL-11, tumor necrosis factor (TNF)-a, parathyroid hormone-related protein (PTHrP), receptor activator of nuclear factor kB (RANK), RANK ligand (RANKL) and osteoprotegerin (OPG) were selected。Results: Immunohistochemical staining revealed significantly higher levels of IL-1a, IL-1b, IL-6, IL-11, PTHrP and RANK in the invasive group than in the non-invasive group。TNF-a and RANKL also tended to be higher in the invasive group, but not significantly。The mRNA expression of IL-1a, IL-1b, TNFa and RANKL tended to be higher in the invasive group, and OPG tended to be higher in the non-invasive group。Conclusion: These results suggest that IL-1a, IL-1b, IL-6, TNF-a, PTHrP and RANKL play important roles in the mechanisms underlying mandibular invasion。Furthermore, expression analysis of OPG/RANK/RANKL system may contribute to treatment and prevention of GSCC in future。
