Photodynamic therapy(PDT)has shown great application potential in cancer treatment and the important manifestation of PDT in the inhibition of tumors is the activation of immunogenic cell death(ICD)***,the strategy is limited in the innate hypoxic tumor *** are two key elements for the realization of enhanced PDT:specific cellular uptake and release of the photosensitizer in the tumor,and a sufficient amount of oxygen to ensure photodynamic ***,self-oxygenated biomimetic nanoparticles(CS@M NPs)co-assembled by photosensitizer prodrug(Ce6-S-S-LA)and squalene(SQ)were *** the treatment of triple negative breast cancer(TNBC),the oxygen carried by SQ can be converted to reactive oxygen species(ROS).Meanwhile,glutathione(GSH)consumption during transformation from Ce6-S-S-LA to chlorin e6(Ce6)avoided the depletion of *** co-assembled(CS NPs)were encapsulated by homologous tumor cell membrane to improve the tumor *** results showed that the ICD effect of CS@M NPs was confirmed by the significant release of calreticulin(CRT)and high mobility group protein B1(HMGB1),and it significantly activated the immune system by inhibiting the hypoxia inducible factor-1alpha(HIF-1α)-CD39-CD73-adenosine a2a receptor(A2AR)pathway,which not only promoted the maturation of dendritic cells(DC)and the presentation of tumor specific antigens,but also induced effective immune infiltration of ***,the integrated nanoplatform implements the concept of multiple advantages of tumor targeting,reactive drug release,and synergistic photodynamic therapy-immunotherapy,which can achieve nearly 90%tumor suppression rate in orthotopic TNBC models.

In public health emergencies, the collection of archival information in the area of public health emergencies is very important, which can provide a reference for the early warning and processing mechanism. At the present stage, the unlimited demand of archival users for resources and the convenience of obtaining resources have become the two driving forces to promote the “transformation” and “growth” of archives. Public health emergencies, the transformation of archives collection and service mode, social media has become an indispensable platform for user information exchange, sharing and transmission, which requires archives to change the mode of archives acquisition and storage. Archival users require more interactive, targeted and cutting-edge forms of access and archival services are also developing toward diversified functions and connotations. Archival information resource sharing is also an important link in this development trend. This paper attempts to analyze the collection methods of archives departments in public health emergencies, and then puts forward the corresponding measures for archives departments to play their functions, such as flexibly solving the needs of archives access, strengthening the development of information resources, doing a good job in the collection of relevant archives, and publicizing archives work in combination with hot spots. This paper discusses the completeness of archival data collection, the means of archival management, the scientific classification of archival data, the ways of archival data collection and so on.

The pattern of plasma BCAA concentration and liver Bckdha gene expression in GK rats during T2D progression

Background: This study was conducted to measure the concentration of branched chain amino acid(BCAA) in different species and detect the expression pattern of the liver Bckdha gene in Goto-Kakizaki(GK) rats during type 2 diabetes(T2D)***: We measured the concentration of BCAA in GK rats, induced T2D cynomolgus monkeys and T2D humans by liquid chromatography tandem mass spectrometry, and used real-time quantitative PCR to analyze the gene expression of Bckdha and Bckdk, which encode the rate-limiting enzymes in catabolism of,respectively, branched chain amino acids and branched chain α-keto acid dehydrogenase ***: In this study, we showed that GK rat BCAA concentrations were significantly reduced at 4 and 8 weeks(P < 0.05 and P < 0.01, respectively), while the expression of Bckdha in GK rat liver was increased at 4 and 8 weeks(1.62-fold and1.93-fold, respectively). The BCAA concentrations were significantly reduced in diet-induced T2D cynomolgus monkeys(P < 0.01), but significantly increased in T2D humans(P < 0.001).Conclusions: Our results showed that BCAA concentrations changed at different times and by different amounts in different species and during different periods of T2D progress, and the significant changes of BCAA concentration in the three species indicated that BCAA might participate in the progress of T2D. The results suggested that the increased expression of Bckdha in GK rat liver might partially explain the reduced plasma BCAA concentration at 4 and 8 weeks. Further studies are required to investigate the exact mechanism of BCAA changes in non-obese T2D.

双氢青蒿素(DHA)作为一种新型的抗癌药物引起了全世界的广泛关注.然而,DHA的固有缺陷降低了其在血液系统中的运输效率,而肿瘤细胞中铁含量不足进一步导致疗效不佳.因此,构建一个可同时负载DHA和Fe^(3+)并在肿瘤微环境中特异性释放出DHA/Fe的纳米药物具有重大意义.本文报道了一种智能纳米药物CaCO_(3)@DHA@Fe^(3+)-TA@PEG(CDFP NPs)用于肿瘤协同治疗.Fe^(3+)-TA壳层的包覆赋予CDFP NPs良好的光热性能.此外,CDFP NPs在酸性的肿瘤微环境中可以快速降解并释放出TA,Fe^(3+),DHA和Ca^(2+).Fe^(3+)与TA反应生成Fe^(2+),Fe^(2+)与DHA相互作用生成高毒性的自由基.此外,在氧化应激状态下,Ca^(2+)过载导致线粒体功能紊乱,最终诱导肿瘤细胞死亡.在光热治疗,活性氧,Ca^(2+)过载的协同作用下,CDFP NPs在体内外均表现出良好的治疗效果.

The sequence upstream of the antibody variable region(antibody upstream sequence[AUS])consists of a 5′untranslated region(5′UTR)and a preceding leader *** sequence variations in AUS affect antibody engineering and PCR based antibody quantification and may also be implicated in mRNA transcription and ***,the diversity of AUSs remains *** 5′rapid amplification of cDNA ends and high-throughput antibody repertoire sequencing technique,we acquired full-length AUSs for human,rhesus macaque,cynomolgus macaque,mouse,and *** designed a bioinformatics pipeline and identified 3307 unique AUSs,corresponding to 3026 and 1457 unique sequences for 5′UTR and leader region,*** analysis indicated that 928(63.69%)leader sequences are novel relative to those recorded in the international ImMunoGeneTics information ***,leader sequences are more conserved than 5′UTR and seem to coevolve with their downstream V ***,single-nucleotide polymorphisms are position dependent for leader regions and may contribute to the functional reversal of the downstream V ***,the AUGs in AUSs were found to have little impact on gene *** together,our findings can facilitate primer design for capturing antibodies efficiently and provide a valuable resource for antibody engineering and molecule-level antibody studies.

The purpose of this study was to analyze the components of inpatient costs for coronary artery bypass graft(CABG)according to preoperative risk stratification and to provide evidence for improvement of diagnosis-related groups(DRGs)*** patients(n=458)receiving an isolated CABG between January 2014 and December 2016 in a tertiary referral center,in southwest china,were *** mortality was predicted by the EuroSCORE Ⅱ for each patient. The patients were subdivided into two groups according to the observed mortality(1.97%,9/458):a high-risk group(group H,predicted mortality≥1.97%)and a low-risk group(group L,predicted mortalityresource use,in-hospital direct costs,and reimbursement expenses were compared between the two *** differences existed between group L and group H in postoperative mortality(0.4% vs.3.4%;P=0.02),postoperative complications(10.6% vs.45.7%;Phospital stay(17.5±4.9 days vs.18.8±6.5 days,P=0.01),in-hospital costs($20 256±3096vs.$23 334±6332;Pgeneral,a higher EuroSCORE Ⅱ was significantly associated with a worse clinical outcome and increased *** CABG cost data provide evidence for improvement of DRGs payment.
