CXCR6-based immunotherapy in autoimmune,cancer and inflammatory infliction

T cells, including both CD4^(+) and CD8^(+)T cells, play a pivotal role in mediating various inflammation and immune disorders. A long-standing challenge in T cell-based immunotherapy is to precisely inactivate or delete the pathogenic T cells in inflammation and autoimmune diseases, or to selectively expand the immunocompetent T cell in tumor or other immune compromised situations, without inducing global immunosuppression or zealous immune activation respectively. To achieve this, a specific marker is needed to differentiate the pathogenic or immunocompetent T cell among the rests. Indeed,recent progress of immunology strongly suggests that CXC chemokine receptor 6(CXCR6, CD186) is such a kind of marker. Here, we review the emerging role of CXCR6 as a novel target for immunotherapy and discuss the underlying mechanism. We propose that CXCR6-based immunotherapy will play a significant role in autoimmune, nonalcoholic steatohepatitis(NAsH), tumor, coronavirus disease 2019(COVID-19) and even ageing-related inflammatory infliction.

Hi-TOM: a platform for high-throughput tracking of mutations induced by CRIsPR/Cas systems

The CRIsPR/Cas system has been extensively applied to make precise genetic modifications in various organisms. Despite its importance and widespread use, large-scale mutation screening remains time-consuming, labour-intensive and costly. Here, we developed Hi-TOM(available at http://***/hi-tom/), an online tool to track the mutations with precise percentage for multiple samples and multiple target sites. We also described a corresponding next-generation sequencing(NGs) library construction strategy by fixing the bridge sequences and barcoding primers. Analysis of the samples from rice, hexaploid wheat and human cells reveals that the Hi-TOM tool has high reliability and sensitivity in tracking various mutations, especially complex chimeric mutations frequently induced by genome editing. Hi-TOM does not require special design of barcode primers,cumbersome parameter configuration or additional data analysis. Thus, the streamlined NGs library construction and comprehensive result output make Hi-TOM particularly suitable for high-throughput identification of all types of mutations induced by CRIsPR/Cas systems.

serum Uric Acid is Associated with the Predicted Risk of Prevalent Cardiovascular Disease in a Community-dwelling Population without Diabetes

Objective To examine the association between serum uric acid levels and cardiovascular disease risk among individuals without *** We investigated the association between serum uric acid levels and the risk of prevalent cardiometabolic diseases, 10-year Framingham risk for coronary heart disease, and 10-year risk for atherosclerotic cardiovascular diseases （AsCVD） among 8,252 participants aged 〉 40 years without diabetes from Jiading district, shanghai, *** Body mass index, waist circumference, blood glucose, glycated hemoglobin, blood pressure, and serum lipids increased progressively across the sex-specific quartiles of uric acid （all P trend 〈 0.05）. Compared with individuals in the lowest quartile, those in the higher quartiles had a significantly higher prevalence of obesity, hypertension, and dyslipidemia （all P trend 〈 0.05）. A fully adjusted logistic regression analysis revealed that individuals in the highest quartile had an increased risk of predicted cardiovascular disease compared with those in the lowest quartile of uric acid. The multivariate adjusted odds ratios （ORs） [95% confidence intervals （C/s）] for the highest quartiles for high Framingham risk were 3.00 （2.00-4.50） in men and 2.95 （1.08-8.43） in women. The multivariate adjusted ORs （95% C/s） for the highest quartile for high AsCVD risk were 1.93 [1.17-3.17） in men and 4.53 （2.57-7.98） in *** serum uric acid level is associated with an increased risk of prevalent obesity, hypertension, dystipidemia, 10-year Framingham risk for coronary heart disease, and lO-year risk for AsCVD among Chinese adults without diabetes.

Background： Manganese-enhanced magnetic resonance imaging （MEMR1） for visual pathway imaging via topical administration requires further research. This study investigated the permeability of the corneal epithelium and corneal toxicity after topical administration of Mn2＋ to understand the applicability of MEMR1. Methods： Forty New Zealand rabbits were divided into 0.05 mool/L, 0.10 mol/L, and 0.20 mol/L groups as well as a control group （n ： 10 in each group）. Each group was i-hrther subdivided into epithelium-removed and epithelium-intact subgroups （n = 5 in each subgroup）. Rabbits were given 8 drops of MnCl2 in 5 min intervals. The Mn2＋ concentrations in the aqueous and vitreous humors were analyzed using inductively coupled plasma-mass spectrometry at different time points. MEMRI scanning was carried out to image the visual pathway after 24 h. The corneal toxicity of Mn2＋ was evaluated with corneal imaging and pathology slices. Results： Between the aqueous and vitreous humors, there was a 10 h lag for the peak Mn2＋ concentration times. The intraocular Mn2＋ concentration increased with the concentration gradients ofMn2＋ and was higher in the epithelium-removed subgroup than that in the epithelium-intact subgroup. The enhancement of the visual pathway was achieved ill the 0.10 mol/L and 0.20 mol/L epithelium-removed subgroups. The corresponding peak concentrations of Mn2. were 5087 ~ 666 ng/ml, 22920 ± 1188 ng/ml ill the aqueous humor and 884 ± 78 ng/ml, 2556 ± 492 ng/ml in the vitreous body, respectively. Corneal injury was evident in the epithelium-removed and 0.20 mol/L epithelium-intact subgroups. Conclusions： The corneal epithelium is a barrier to Mn2＋, and the iris and lens septum might be another intraocular banier to the permeation of Mn2＋. An elevated Mn2＋ concentration contributes to the increased permeation of Mn2＋, higher MEMRI signal, and corneal toxicity.
