The Survey of Cronobacter spp.(formerly Enterbacter sakazakii) in Infant and Follow-up Powdered Formula in China in 2012

Objective To determine Cronobacter spp. contamination in infant and follow-up powdered formula in China. Methods All of 2282 samples were collected from the retail markets in China from January 2012 to December 2012, and analyzed for Cronobacter spp. by the Chinese National food Safety Standard. Characterization of the isolates was analyzed by pulsed-field gel electrophoresis（PFGE） with XbaI and Spe I restriction enzymes. Results Cronobacter spp. strains were isolated from 25 samples, and the positive rates in infant powdered formulas and follow-up powdered formulas were 0.90%（10/1011） and 1.18%（15/1271）, respectively. Analysis of variable data regarding different purchasing store formats, seasonality, and production locations as well as comparison of infant versus follow-up formulas did not reveal statistically significant factors. During the sampling period, one of six surveillance zones did exhibit a statistically significant trend towards higher positive rate. PFGE characterization of Cronobacter spp. to elucidate genetic diversity revealed only three pairs of Cronobacter spp. out of 25 having the same PFGE patterns. Conclusion The current investigation indicated a lower positive rate of Cronobacter spp. in the powdered formula in China. This evidence suggested contamination originating from multiple different sources during the manufacturing process.

Comparing invasive effects of five foodborne bacterial pathogens in human embryonic intestine 407 cells and human ileocecum HCT-8 cells

Objective: To refine the infectious doses of enteric bacterial pathogens in animal assays and vaccine clinical trials by studying the invasion kinetics of five bacterial pathogens with human intestinal ***: Utilizing in vitro cultured cell invasion assays with gentamicin-killing step,the invasive effects were analyzed in foodborne pathogens including Salmonella,Shigella, Yersinia, Escherichia coli(E. coli) O157 and opportunistic pathogens Citrobacter in human embryonic intestine 407 cells and ileocecum HCT-8 cells at multiplicities of infection(MOIs) of 0.04–4 000.00 E. coli HS served as a noninvasive ***: The study results showed that the bacterial invasive efficiency and the average number of internalized bacteria per host cell changed with different starting MOIs. Higher starting MOIs did not always produce more bacterial internalization. The bacterial invasion effects varied with different bacterial strains and host cell lines. E. coli O157:H7 did invade human ileocecum HCT-8 ***: This study shows that these bacteria possess different invasive patterns at various starting MOIs and also in different cell lines. The results could help to figure out the appropriate infectious doses of the bacteria in animal assays and in vaccine clinical trials. The bacterial invasion kinetics is also valuable in evaluating the safety and efficacy of live attenuated bacterial vaccines.

Glucose-responsive closed-loop insulin delivery systems represent a promising treatment strategy for diabetes,but current systems generally cannot achieve long-term *** this study,we designed an erythrocyte-biomimetic glucose-responsive system(EGRS)by coupling glucose-responsive nanoparticles(GRNs)to red blood cells;these nanoparticles exhibited the dual functions of glucose-responsiveness and persistent presence in *** are generated by encapsulating with insulin through ion crosslinking,followed by coloading with glucose oxidase(GOx)and catalase(CAT),a process that endows the nanoparticles with ***,the GRNs are coupled with red blood cells to camouflage them from the immune system,therefore,these erythrocyte-coupled GRNs can circulate in the blood for a long *** conditions of hyperglycemia,GOx acts on blood glucose to produce gluconic acid,which causes the rupture of GRNs and efficient release of ***,insulin is only released at the basic rate during ***,EGRS can efficiently and continuously respond to hyperglycemia to maintain blood glucose levels within the normal range.

Dynamic DNA nanodevices have gained tremendous attention due to their extraordinary inherent functionality and advantages,however,dynamic DNA nanodevices-based biosensors are still challenging due to their high reliance on proteases and limited amplification ***,exploiting bispecific aptamer as initiators for the first time,we developed a three-dimensional(3D)DNA nanomotor biosensor powered by DNAzyme and entropy-driven circuit for sensitive and specific detection of lysozyme,in which walking and rolling strategies are efficiently integrated to achieve excellent signal amplification *** from the high selectivity of bispecific aptamer,the 3D DNA nanomotor biosensor can respond to lysozyme with high specificity and operate at high speed to release *** whole process is independent of protease,avoiding the influence of adverse environment on the operation *** optimal conditions,it can achieve a limit of detection as low as 0.01 pg/mL with an excellent linear range of 0.05 pg/mL–500 ng/mL for ***,the proposed strategy revealed high accuracy in the analysis of real samples,indicating a great potential for the application of nanomotor biosensors to the detection of non-nucleic acid targets.

Synthesis and cytotoxic activities of E-resveratrol derivatives

The present study was designed to synthesize derivatives of E-resveratrol and evaluate their cytotoxic activity in *** functional groups were conjugated with the phenolic hydroxyl group of E-resveratrol,and the double bond of E-resveratrol was *** in vitro cytotoxicity of the synthetic derivatives was evaluated against three tumor cell lines(A549,LAC,and He La) using the MTT ***-six E-resveratrol derivatives were synthesized and their structures were confirmed by 1H NMR,MS,IR,and elemental *** 1-6,12,15-21,and 23-26 were reported for the first *** them,Compounds 1,2,4,5,and 9-11,showed significant cytotoxicity against tumor cells;especially,Compound 1 showed an IC50 value of 4.38 μmol·L-1 in the A549 cells which was 15-fold more active than E-resveratrol;Compound 9 showed an IC50 value of 1.41 μmol·L-1 in the He La cell line which was 90-fold more active than E-resveratrol,and close to *** structure–activity relationships were also *** 1,2 and 9-11 may serve as potential lead compounds for the discovery of new anticancer drugs.

Hantaviruses,such as Hantaan virus(HTNV)and Seoul virus,are the causative agents of Hantavirus cardiopulmonary syndrome(HCPS)and hemorrhagic fever with renal syndrome(HFRS),and are important zoonotic *** has the highest incidence of HFRS,which is mainly caused by HTNV and Seoul *** approved antiviral drugs are available for these hantaviral ***,a chemiluminescence-based highthroughput-screening(HTS)assay was developed and used to screen HTNV pseudovirus(HTNVpv)inhibitors in a library of 1813 approved drugs and 556 small-molecule compounds from traditional Chinese medicine *** identified six compounds with in vitro anti-HTNVpvactivities in the low-micromolar range(EC50values of0.1–2.2μmol/L;selectivity index of 40–900).Among the six selected compounds,cepharanthine not only showed good anti-HTNVpvactivity in vitro but also inhibited HTNVpv-fluc infection in Balb/c mice 5 h after infection by94%(180 mg/kg/d,Pdrugs screening,and shown that cepharanthine is a candidate for HCPS and HFRS *** findings may offer a starting point for the treatment of patients infected with hantaviruses.

Development and Influencing Factors of Compliance Behaviors of Investigators in Clinical Trials

The development and influencing factors of compliance behavior of investigators in clinical trials were explored. According to literature review, a hypothetical model of development of compliance behavior of investigators in clinical trials was established, and the influencing factors of compliance behavior of investigators and their interrelationships were studied based on questionnaire survey of five hundred investigators sampled randomly from one hundred clinical trial institutions in China. Cron- bach＇s alpha coefficient and structural equation modeling were adopted to empirically analyze the re- suits. Six variables in the hypothetical model were included： compliance behavior of investigators, credibility of clinical trial, capability of government regulation, quality control of sponsor, quality con- trol of clinical institution and compliance intention of investigators. Empirical analysis showed that the compliance behavior of investigators in clinical trial was directly affected by compliance intention of investigators, quality control of sponsor and quality control of clinical institution. In addition, credibility of clinical trial and capability of government regulation indirectly affected the compliance behavior of investigators in clinical trial through influencing the compliance intention of investigators, quality con- trol of sponsor and quality control of clinical institution. Quality control of sponsor was affected by credibility of clinical trial and capability of government regulation while quality control of clinical in- stitutinn wan only influenced by capability of government regulation.
