The comprehensive assessment of epigenetics changes during skin development

Epigenetic regulation is critical to multiple physiological and pathological ***,little is known regarding the epigenetic changes during neonatal skin development and skin aging,and in response to ultraviolet(UV)*** transcriptomes of human skin samples from different ages or irradiated with different types and doses of UV light were analyzed using R(version 4.0.3)*** epigenetic landscape of the skin,including histone modifications,genetic imprinting and m^(6)A modification,which are mainly involved in collagen formation,extracellular matrix organization,immune function and keratinization,underwent significant changes during neonatal to adult *** effectors such as IGF2BP2,GATA2,GATA3,CPA4 and CDK1 were significantly correlated with extracellular matrix organization,and VEGFA,CDK1 and PRKCB with skin immune *** m^(6)A readers such as IGF2BP2,IGF2BP3,HNRNPA2B1 and EIF3G showed significant correlation with extracellular matrix organization,metabolism,or antigen processing and *** doses of UV exposure only induced changes in the expression levels of some epigenetic effectors,without any significant effect on the overall epigenetic ***,the minimal erythema dose of UV exposure altered multiple epigenetic effectors regulating extracellular matrix organization,cell-matrix adhesion,innate immune response,mitochondrial function and mRNA *** addition,epigenetic changes following UV exposure were more pronounced in the elderly skin compared to the younger *** conclusion,histone modifications,genetic imprinting and m^(6)A modification play critical roles during skin development,and a large dose of UV exposure can significantly change the expression of multiple epigenetic effectors.

Precise tumor targeting and therapy is a major trend in cancer ***,we designed a tumor acidic microenvironment activatable drug loaded DNA nanostructure,in which,we made a clever use of the sequences of AS1411 and i-motif,which can partially hybridize,and designed a simple while robust DNA D-strand nanostructure,in which,i-motif sequence was designed to regulate the binding ability of the AS1411 aptamer to target *** the normal physiological environment,i-motif inhibits the targeting ability of *** the acidic tumor microenvironment,i-motif forms a quadruplex conformation and dissociates from AS1411,restoring the targeting ability of *** when acidic condition and tumor cell receptor are present,this nanostructure can be internalized by the tumor ***,the structure change of this nanostructure can realize the release of loaded *** drug loaded A-I-Duplex DNA structure showed cancer cell and spheroid targeting and inhibition ability,which is promising in the clinical cancer therapy.

Fast electron-hole recombination issues during titanium dioxide(TiO_(2))photocatalysis limit its application in preventing bacterial infection during bone defect *** this study,TiO_(2)@reduced graphene oxide(rGO)composites were synthesized using a hydrothermal method in which rGO,which possesses very high electrical conductivity,promotes the separation of photoelectron-hole pairs of TiO_(2),thus improving the efficiency of photocatalytic production of reactive oxygen species(ROS).Subsequently,TiO_(2)@rGO composites were introduced into poly-L-lactic acid(PLLA)to prepare bone scaffolds with photocatalytic antibacterial function via selective laser *** results showed that TiO_(2)grew on the surface of rGO and formed a covalent bond connection(Ti-O-C)with rGO.A decreased electrochemical impedance of TiO_(2)@rGO composites was observed,and the transient photocurrent intensity increased from 0.05 to 0.5μA/cm^(2).Analysis of electron spin resonance found that the photocatalytic products of TiO_(2)were·OH and·O^(2-),two kinds of ROS capable of killing bacteria via disrupting the structure of the bacterial membrane in *** experiments showed that the PLLA/TiO_(2)@rGO scaffolds had good antibacterial properties against Escherichia coli and Staphylococcus ***,we report that these scaffolds exhibited both enhanced mechanical properties due to the addition of TiO_(2)@rGO as a reinforcement material and good biocompatibility during cell proliferation.

A prediction model for guiding tumor microwave ablation surgery based on simulation

Purpose: The major limitation of tumor microwave ablation (MWA) operation is the lack of predictability of the ablation zone before surgery. Operators rely on their individual experience to select a treatment plan, which is prone to either inadequate or excessive ablation. This paper aims to establish an ablation prediction model that guides MWA tumor surgical planning. Methods: An MWA process was first simulated by incorporating electromagnetic radiation equations, thermal equations, and optimized biological tissue parameters (dynamic dielectric and thermophysical parameters). The temperature distributions (the short/long diameters, and the total volume of the ablation zone) were then generated and verified by 60 cases ex vivo porcine liver experiments. Subsequently, a series of data were obtained from the simulated temperature distributions and to further fit the novel ablation coagulated area prediction model (ACAPM), thus rendering the ablation-dose table for the guiding surgical plan. The MWA clinical patient data and clinical devices suggested data were used to validate the accuracy and practicability of the established predicted model. Results: The 60 cases ex vivo porcine liver experiments demonstrated the accuracy of the simulated temperature distributions. Compared to traditional simulation methods, our approach reduces the long-diameter error of the ablation zone from 1.1cm to 0.29cm, achieving a 74% reduction in error. Further, the clinical data including the patients' operation results and devices provided values were consistent well with our predicated data, indicating the great potential of ACAPM to assist preoperative planning.

Establishment and validation of a nomogram for predicting esophagogastric variceal bleeding in patients with liver cirrhosis

BACKGROUND Patients with decompensated liver cirrhosis suffering from esophagogastric variceal bleeding(EGVB)face high *** To investigate the risk factors for EGVB in patients with liver cirrhosis and establish a diagnostic *** Patients with liver cirrhosis who met the inclusion criteria were randomly divided into training and validation cohorts in a 6:4 ratio in this retrospective *** analysis,least absolute shrinkage and selection operator regression,and multivariate analysis were employed to establish the nomogram *** curve,the area under the receiver operating characteristic curve(AUC),and decision curve analysis(DCA)were applied to assess the discrimination,accuracy,and clinical practicability of the nomogram,*** A total of 1115 patients were enrolled in this *** nomogram was established based on white blood cells(P<0.001),hemoglobin(P<0.001),fibrinogen(P<0.001),total bilirubin(P=0.007),activated partial thromboplastin time(P=0.002),total bile acid(P=0.012),and ascites(P=0.006).The calibration curve indicated that the actual observation results were in good agreement with the prediction results of the *** AUC values of the diagnostic model were 0.861 and 0.859 in the training and validation cohorts,respectively,which were higher than that of the aspartate aminotransferase-to-platelet ratio index,fibrosis index based on 4 factors,and aspartate aminotransferase-to-alanine aminotransferase ***,DCA indicated that the net benefit value of the model was higher than that of the other *** This research constructed and validated a nomogram with perfect performance for predicting EGVB events in patients with liver cirrhosis,which could help clinicians with timely diagnosis,individualized treatment,and follow-up.

The gut microbiota and the associated metabolism play a pivotal role in maintaining human health,yet their assessment is challenging due to their inherent diversity,variety,and ***,we developed a multichannel sensor array based on a single fluorescent probe,allowing for rapid and robust profiling gut microbiota and their key metabolites including cysteine(Cys),glutathione(GSH),and homocysteine(Hcy).The assay leverages the single fluorescent probe that is with multiple binding sites and the cross-reactive sensing principle to specifically recognize different biological *** analyzing the pattern of biological thiols,the assay is capable of rapidly identifying six gut-derived bacteria including probiotics,neutral,and pathogenic strains based on fluorescent fingerprints within 5 min,and also discriminating bacteria and their mixtures with different *** the assay that enables the simultaneous measurement of multiple gut-derived bacteria and their metabolites,the designed array achieved an accuracy of 0.99 when discriminating colorectal cancer(CRC)patient feces samples from healthy ***,the as-prepared sensor array can also be used to identify various stages of *** simplicity,rapidness,and cost-effectiveness of our approach render it a robust platform for the analysis of gut microbiota.

Auroral Kilometric Radiation (AKR) is a common radio emission,which can contribute to the magnetosphere-ionosphereatmosphere co u *** emissions have been observed in all magnetic planet magnetospheres of the solar *** this study,using observations from the FAST satellite from 30 August 1996 to 9 September 2001,the distribution of AKR in altitude=500-4500 km and invariant latitude (|ILAT|)=60°-80°has been analyzed.63045 AKR samples have been identified with~48%(52%) samples on the dayside (nightside).Of considerable interest,there is a distinct MLT asymmetry with the high occurrence rate in MLT=05-08 and 18-22(02-05 and 12-17) in the northern (southern) *** distinct MLT asymmetry is associated with the direction of Bxof the interplaneta ry magnetic *** addition,the occurrence rate on the nightside clearly increases as the AE^(*) index *** study further enriches the information and understanding of AKR in the magnetosphere as well as other similar radio emissions.
