Association of serum vascular endothelial growth factor-C and lymphatic vessel density with lymph node metastasis and prognosis of patients with gastric cancer

AIM： To investigate whether serum vascular endothelial growth factor-C （SVEGF-C）, VEGF-C, and lymphatic vessel density （LVD） in tumor tissues are related to lymph node metastasis （LNM） and prognosis in gastric cancer. METHODS： SVEGF-C levels of 80 gastric cancer patients and 20 healthy donors were examined using ELISA. VEGF-C expression and LVD were examined using immunohistochemical staining. Kaplan-Meier survival analysis was performed to determine their influence on the prognosis of the patients. RESULTS： The SVEGF-C level in gastric cancer patients （595.9 ± 201.0 ng/L） was significantly higher （P = 0.000） than controls （360.0 ± 97.4 ng/L）. Both SVEGF-C and LVD were significantly higher in poorly differentiated adenocarcinomas, T3 and T4, LNM, distant metastasis, and pTNM groups Ⅲ and Ⅳ （P = 0.000）. The sensitivity and specificity of SVEGF-C for predicting LNM were 82.8% and 81.8%, respectively （cut-off = 542.5 ng/L）. The positive expression rate of VEGF-C was significantly higher in cancerous than in normal tissues （65% vs 20%; P = 0.001）. VEGF-C expression up-regulation was significantly related to differentiation, depth of invasion, LNM, distant metastasis, and pTNM stage （P = 0.000）. LVD was 10.7 ± 3.1/200 HP in the experimental group vs 4.9 ± 1.3/200 HP in controls （P = 0.000）; LVD in cancerous tissues with and without LNM was 12.0 ± 2.7/200 HP vs 7.6 ± 0.5/200 HP, respectively （P = 0.000）. SVEGF-C and LVD were significantly higher in VEGF-C positive than in negative patients （P = 0.000）; SVEGF-C level was related to LVD （P = 0.000）. Kaplan-Meier survival analysis factors predicating poor prognosis were： SVEGF-C level （P = 0.001）, VEGF-C expression and LVD （both P = 0.000）. CONCLUSION： SVEGF-C level, VEGF-C and LVD are related to LNI and poor prognosis of patients with gastric cancer, SVEGF-C may be a biomarker for LNI in gastric cancer,

Effect of the hyaluronic acid-poloxamer hydrogel on skin-wound healing: in vitro and in vivo studies

Background: Recent research into skin injury and wound healing has focused mainly on post‐trauma hemostasis, infection prevention, dermal regeneration and angiogenesis. However, less attention has been paid to air permeability and moisture loss prevention which also play important roles in injury healing. Methods: In the present work, we prepared a hyaluronic acid‐poloxamer (HA‐POL) hydrogel and tested the therapeutic effect of the hydrogel on skin‐wound healing. Results: The HA‐POL hydrogel transformed from sol to gel at 30°C, close to body temperature, and had stable moisturizing properties. HA‐POL hydrogel promoted skin‐wound healing and increased protein accumulation in the wound area. HA‐POL hydrogel allowed greater air permeability than Band‐aid, a typical wound covering. Results from transwell assays showed that the HA‐POL hydrogel effectively isolated skin‐wounds from bacterial invasion. Conclusion: This work demonstrates the advantages of using HA‐POL gel materials in the treatment of cutaneous wounds.

Efficacy of long-term rifaximin treatment for hepatic encephalopathy in the Japanese

Proliferative activity of bile from congenital choledochal cyst patients

AIM: To explore the potential carcinogenicity of bile from congenital choledochal cyst (CCC) patients and the mechanism of the carcinogenesis in congenital choledochal cyst ***: 20 bile samples from congenital choledochalcyst patients and 10 normal control bile samples were usedfor this study. The proliferative effect of bile was measuredby using Methabenzthiazuron (MTT) assay; Cell cycle andapoptosis were analyzed by using flow cytometry (FCM),and the PGE2 levels in the supernatant of culturedcholangiocarcinoma cells were quantitated by enzyme-linkedimmunoabsordent assay (ELISA).RESULTS: CCC bile could significantly promote theproliferation of human cholangiocarcinoma QBC939 cellscompared with normal bile (P＝0.001) and negative controlgroup (P＝0.002), and the proliferative effect of CCC bilecould be abolished by addition of cyclooxygenase-2 specificinhibitor celecoxib (20 μM). The QBC939 cells proliferativeindex was increased significantly after treated with 1% bilefrom CCC patient (P＝0.008) for 24 h, the percentage of Sphase (29.48±3.27)% was increased remarkably (P＜0.001)compared with normal bile (11.72±2.70) %, and thepercentage of G0/G1 phase (54.19±9.46) % was decreasedremarkably (P＝0.042) compared with normal bile (69.16±10.88) %, however, bile from CCC patient had no significantinfluence on apoptosis of QBC939 cells (P＝0.719).CONCLUSION: Bile from congenital choledochal cyst patientscan promote the proliferation of human cholangiocarcinomaQBC939 cells via COX-2 and PGE2 pathway.

Liquid nitrogen post-treatment for improved aggregation and electrical properties in organic semiconductors

Organic semiconductors are promising candidates as active layers in flexible and biocompatible electronics owing to their solution processability and molecular design flexibility. However, it remains necessary to establish a green processing approach to acquire desirable electrical properties for scalable industrial applications. Here, a highly efficient and environmentally friendly post-treatment method using liquid nitrogen as a cooling bath is developed to optimize the aggregation structure and electrical performance of organic semiconductors. The carrier mobility has increased by nearly 60% with this treatment, achieving a performance boost comparable to that of traditional annealing methods. This performance improvement is attributable to the denser aggregation structure and enhanced molecular ordering compared with those of as-cast semiconducting polymer films. Impressively, the entire process can be completed within a few minutes without additional vacuum or high-temperature conditions, offering an economical and efficient alternative to traditional methods. Furthermore, the enhancement effect and long-term stability of this treatment are validated across a wide range of organic semiconductors, positioning this green and versatile approach as a promising substitute for conventional post-treatment, thereby facilitating the development of next-generation sustainable electronics.

Clinical significance of preoperative nutritional status in elderly gastric cancer patients undergoing radical gastrectomy:A singlecenter retrospective study

Differentiation of hepatocytes from mouse embryonic stem cells and its significance

BACKGROUND:There have been lots of important discoveries in the research on embryonic stem (ES) cells; but few studies have been involving in the differentiation and differentiation ratio of hepatic stem cells. To provide a new source of hepatocytes in hepatocytes transplantation and liver tissue engineering, we investigated the differentiation and differentiation ratio of hepatocytes in the ES cells differentiating system in a certain length of time.METHODS:BALB/c ES cells from mice were cultured for differentiation in medium without leukemia inhibitory factor (LIF). During the period of differentiation of ES cells, dual immunofluorescence was performed to analyze the expression of hepatic proteins in cytoplasm such as alpha-fetoprotein (AFP) and albumin (ALB). The count of relative hepatocyte-like cells was analyzed by flow cytometry (FCM) and the hepatic differentiation ratio was obtained.RESULTS:The expression of AFP was first determined at day 7 and that of ALB at day 13. The expressions of AFP and ALB became stronger with the extention of the differentiation time. The hepatic differentiation ratio at day 13 and 21 was 5.5% and 10.4%, respectively.CONCLUSION:ES cells could differentiate into hepatocytes, which should be a new source of hepatocytes in transplantation of hepatocytes and liver engineering.

Enhanced therapeutic effects of combined chemotherapeutic drugs and midkine antisense oligonucleotides for hepatocellular carcinoma

AIM: To evaluate the effect of combined antisense oligonucleotides targeting midkine (MK-AS) and chemotherapeutic drugs [cisplatin(DDP), 5-fluorouracil (5-FU) and adriamycin (ADM)] on inhibition of HepG2 cell proliferation, and to analyze the efficacy of MK-AS used in combined ADM in in situ human hepatocellular carcinoma (HCC) model. METHODS: HepG2 cells were treated with MK-AS and/or chemotherapeutic drugs mediated by Lipofectin, and cell growth activity was determined by MTS assay. An in situ HCC model was used in this experiment. MK- AS, ADM and MK-AS + ADM were given intravenously for 20 d, respectively. The animal body weight and their tumor weight were measured to assess the effect of the combined therapy in vivo. RESULTS: Combined treatment with MK-AS reduced the IC50 of DDP, 5-FU and ADM in HepG2 cells. MK-AS significantly increased the inhibition rate of DDP, 5-FU and ADM. Additionally, synergism (Q 1.15) occurred at a lower concentration of ADM, 5-FU and DDP with combined MK-AS. Combined treatment with MK-AS and ADM resulted in the more growth inhibition on in situ human HCC model compared with treatment with chemotherapeutic drugs alone. CONCLUSION: MK-AS increases the chemosensitivity in HepG2 cells and in situ human HCC model, and thecombination of MK-AS and ADM has a much better in vitro and in vivo synergism.

AIM:To analyze differences in ultra-widefield fluorescein angiography(UWFA)findings between dynamic and static images of eyes with diabetic retinopathy(DR).METHODS:This cross-sectional study included 28 eyes of 28 patients with DR undergoing UWFA.A series of UWFA images acquired from each patient were converted into a time-lapse video and used as a dynamic image.A single,clear,arteriovenous phase image was chosen as a static ***-perfusion index(NPI)and its correlation with vascular abnormalities in different zones were compared between dynamic and static UWFA ***:NPI appeared to increase from the center to the far-periphery in both *** NPI was lower in the total retinal area(0.26 vs 0.29,P=0.009)and farperiphery(0.33 vs 0.36,adjusted P=0.042),which was contrary to the static ***-peripheral NPI was associated with intraretinal microvascular abnormality in the posterior area in both ***:Time-lapse dynamic UWFA imaging is a useful modality to differentially diagnose hypofluorescence in the most peripheral *** modality could provide a reliable method for NPI measurement.
